        에탄올 농도에 따른 미나리 추출물의 영양성분 변화

        본 연구는 미나리의 최적 추출 조건을 확립하기 위해 다양한 에탄올추출 농도가 미나리의 영양학적 성분 변화에 미치는 정도를 관찰함으로써 진행되었다. 본 실험에서는 열수추출물, 에탄올추출물(50%, 80%, 95%)로 추출 농도를 다르게하여 미나리를 추출하였다. 수율을 측정한 결과 50% 에탄올 추출물에서 44.67%로 가장 높게 나타났다. 일반성분 분석결과, 50% 에탄올추출물에서 단백질 함량이 6.76%, 탄수화물 함량이 19.60%로 가장 높게 나타났다. 지방 함량은 열수추출물과 비교하여 에탄올추출물에서 유의적으로 증가하였으며(P<0.05), 에탄올추출 농도에 따라 유의차가 있었다(P<0.05). 식이섬유를 측정한 결과, 50% 에탄올추출물을 제외한 에탄올추출물(80%, 95%)에서 열수추출물과 유의차가 나타났으며(P<0.05), 두 실험군 간에도 유의적 차이가 있었다(P<0.05). 총당 함량을 측정한 결과, 열수추출물에 대해 에탄올추출물은 통계적 유의차가 있었고(P<0.05), 에탄올 농도에 따라서도 유의차를 나타냈으며(P<0.05), 95%에탄올추출 시 5.35%로 가장 낮게 측정되었다. 비타민 A함량은 열수추출물에 대해 에탄올추출물이 유의하게 증가하였으며(P<0.05), 에탄올추출 농도에 따라 유의차가 있었다(P<0.05). 무기성분 측정 결과, 나트륨, 칼슘, 철의 경우 열수추출물에 대해 에탄올추출물이 유의하게 감소하였고(P<0.05), 에탄올추출 농도가 증가함에 따라 유의적으로 감소하였다(P<0.05). 따라서 에탄올추출 방법을 사용 목적에 따라 적용함으로써 미나리를 이용한 우수한 가공식품 개발가능성과 이용률 향상에 도움이 될 것으로 판단된다. This study evaluated the nutritional compositions of dropwort (Oenanthe javanica) extracts depending on the ethanol concentrations. Extractions were performed with hot water, 50% ethanol, 80% ethanol, and 95% ethanol for 4 hours. Changes in yield, as well as total carbohydrate, crude protein, crude fat, total dietary fiber, free sugar, and mineral (Na, Fe, and Ca) contents were investigated. The highest extraction yield of ethanol extracts was 44.67% in 50% ethanol extract of dropwort. Crude protein content reached a maximum of 6.70% while carbohydrate content was highest at 19.6%, in 50% ethanol extract of dropwort. Crude fat content irregularly increased according to ethanol concentration as compared with hot water extract. Total dietary fiber content decreased in ethanol extract, but these changes were not concentration-related. Total sugar contents were highest in hot water and 80% ethanol extracts. Vitamin A content of ethanol extract was higher than that of hot water extract. Mineral (Na, Ca, and Fe) contents were significantly reduced in ethanol extract according to concentration of ethanol, whereas mineral contents were higher in ethanol extract than in hot water extract. Based on this study, ethanol extract of dropwort is more efficient for development of desirable processed foods.

        미세먼지로 인한 피부 각질 세포 손상에서 몰약 에탄올 추출물의 항염증 효과

        Objectives: Myrrh have been used as a traditional remedy to treat infectious and inflammatory diseases. However, it is largely unknown whether myrrh ethanol extract could exhibit the inhibitory activities against particulate matter (PM)-induced skin injury on human keratinocytes, HaCaT cells. Therefore, this study was aimed to investigate the inhibitory activity of myrrh ethanol extract on PM-induced skin injury in HaCaT cells. Methods: To investigate the inhibitory effects of myrrh ethanol extract in HaCaT cells, the skin injury model of HaCaT cells was established under PM treatment. HaCaT keratinocyte cells were pre-treated with myrrh ethanol extract for 1 h, and then stimulated with PM. Then, the cells were harvested to measure the cell viability, reactive oxygen species (ROS), pro-inflammatory cytokines including interleukin (IL) 1-beta, IL-6, and tumor necrosis factor (TNF)-α, hyaluronidase, collagen, MMPs. In addition, we examined the mitogen activated protein kinases (MAPKs) and inhibitory kappa B alpha (Iκ-Bα) as inhibitory mechanisms of myrrh ethanol extract. Resuslts: The treatment of myrrh ethanol extract inhibited the PM-induced cell death and ROS production in HaCaT cells. In addition, myrrh ethanol extract treatment inhibited the PM-induced elevation of IL-1beta, IL-6, and TNF-α. Also, myrrh ethanol extract treatment inhibited the increase of hyaluronidase, MMP and decrease of collagen. Furthermore, myrrh ethanol extract treatment inhibited the activation of MAPKs and the degradation of Iκ-Bα. Conclusions: Our result suggest that treatment of myrrh ethanol extract could inhibit the PM-induced skin injury via deactivation of MAPKs and nuclear factor (NF)-κB in HaCaT cells. This study could suggest that myrrh ethanol extract could be a beneficial agent to prevent skin damage or inflammation.

        Eco-frendly Control of Culex pipiens (mosquito) Larvae by Acorus calamus (sweet flag) and Acorus gramineus (Grassy-leaved sweet flag) Extracts

        Mosquitoes are carriers of malaria and encephalitis. This study performed for eco-friendly control of mosquitos by using genus Acorus. Several solvents were used for the extraction of genus Acorus; water, ethanol, and methanol. Grinded leaves and roots were also included. Acorus extracts killed mosquito larvae and the ethanol extract showed the best result. Autoclaved Acorus water needed long time to kill mosquito larvae. $LT_{50}$ of 1 % Acorus calamus decoction was 13.6 hrs and 1 % autoclaved Acorus water was 53.6 hrs. $LT_{50}$ of 0.05% Acorus calamus rhizome powder was 28.5 hrs. $LT_{50}$ of 0.5% Acorus calamus leaf powder was 10.8 hrs. $LT_{50}$ of 0.1 % Acorus calamus decoction was 63.4 hrs and 0.1 % Acorus calamus ethanol extracts was 48.6 hrs and 0.1% Acorus calamus methanol extracts was 53.9 hrs. $LT_{50}$ of 0.4% Acorus gramineus decoction was 45.5 hrs, 0.4% ethanol extracts was 10.9 hrs, 0.4% methanol extracts was 10.2 hrs. $LT_{50}$ of ethanol extracts was shorter than other extracts. Acorus calamus rhizome powder could be used for the eco-friendly control of the mosquito larvae.

        새싹보리 에탄올 농도별 추출물의 산화방지 활성

        Phenolic compounds and antioxidant activities of ethanol extracts from barley sprouts were evaluated in this study. Barley sprouts were extracted using water and ethanol in various concentration (25, 50, and 75%) using reflux extraction methods. Ultra performance liquid chromatography (UPLC) analysis showed that barley sprouts are mainly composed of rutin, gallic acid, ferulic acid, and p-coumaric acid. The 75% ethanol extracts had higher total polyphenol contents (44.01±1.32 mg/g) and total flavonoid contents (102.96±2.49 mg/g). 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (EC50 value: 1.65±0.02 mg/mL) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging activity (EC50 value: 1.67±0.02 mg/mL) of the 75% ethanol extracts of barley sprouts were found to be the most effective. The 75% ethanol extracts of barley sprouts exhibited a strong reducing activity and ferric reducing antioxidant activity. As a result, the 75% ethanol extracts of barley sprouts showed stronger antioxidant activity than other extracts.

        Melanogenesis-Promoting Effects of Rhynchosia nulubilis and Rhynchosia volubilis Ethanol Extracts in Melan-a Cells

        We evaluated the antioxidant activity and melanogenic effects of black soybean ethanol extracts, including Rhynchosia nulubilis bean ethanol extract (RNBEE), R. nulubilis leaf ethanol extract (RNLEE), R. volubilis bean ethanol extract (RVBEE), and R. volubilis leaf ethanol extract (RVLEE). The total polyphenol contents of RNBEE, RNLEE, RVBEE, and RVLEE were 16.0, 57.7, 365.9, and 260.1 mg/g, respectively. The total flavonoid contents of RNBEE, RNLEE, RVBEE, and RVLEE were 40.4, 91.7, 84.7, and 216.5 mg/g, respectively. The electron-donating abilities of RNBEE, RNLEE, RVBEE, and RVLEE at $1,000{\mu}g/mL$ were 32.4%, 12.7%, 83.5%, and 84.5%, respectively. RNBEE, RNLEE, RVBEE, and RVLEE at $50{\mu}g/mL$ significantly increased (p < 0.01) melanin contents by 30.4%, 32.1%, 35.5%, and 37.4%, respectively, compared to that of the control. RNBEE, RNLEE, RVBEE, and RVLEE at $50{\mu}g/mL$ significantly increased (p < 0.01) intracellular tyrosinase activity by 18.4%, 21.8%, 21.5%, and 21.1%, respectively, compared to that of the control. These results demonstrated that black soybean ethanol extracts promote melanogenesis in melan-a cells. Among the black soybean ethanol extracts, R. volubilis was found to be more effective than R. nulubilis, and leaf extract was found to be more effective than bean extract. The potential mechanism underlying the hyperpigmentation effects of black soybeans is the promotion of tyrosinase activity.

        사플라워와 마더워트 등 에탄올복합추출물의 항균활성과 항염증 효과

        we are investigated the anti-inflammatory effects of Safflower and Mother wort Ethanol Complex Extracts(SEC) on lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results demonstrated that pretreatment of SEC(500μg/mL) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects by Safflower and Mother wort Ethanol Complex Extracts were observed in the following. Safflower and Mother wort Ethanol Complex Extracts inhibited the translocation of NF-κB from the cytosol to the nucleus via the suppression of IκB-α phosphorylation and also inhibited LPS-stimulated NF-κB transcriptional activity. These findings suggest that Safflower and Mother wort Ethanol Complex Extracts exert anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of Safflower and Mother wort Ethanol Complex Extracts. Therefore, Safflower and Mother wort Ethanol Complex Extracts could be regarded as a potential source of natural anti-inflammatory agents. 사플라워와 마더워트 등 에탄올복합추출물(SEC)을 lipopolysaccharide(LPS)로 유도된 마우스 대식세포주인 RAW264.7 세포와 마우스 폐포 대식세포주인 MH-S 세포에서 사플라워와 마더워트 등 에탄올복합추출물((SEC)의 항균과 항염증 활성 효과를 조사하였다. 그 결과 SEC(500 μg/mL)의 전처리 LPS 자극 세포에서 iNOS 단백질 및 염증성 사이토카인 mRNA 발현을 크게 억제하였다. 사플라워와 마더워트 등 에탄올복합추출물(SEC)에 의한 항염증활성 효과는 다음과 같이 관찰되었다. 사플라워와 마더워트 등 에탄올복합추출물(SEC)은 IκB-α 인산화의 억제를 통해 시토졸에서 핵으로의 NF-κB의 전좌를 억제하고 또한 LPS로 자극된 NF-κB 전사 활성을 억제 하였다. 이러한 결과는 사플라워와 마더워트 등 에탄올복합추출물(SEC)이 항염증 작용을 발휘 하고 사플라워와 마더워트 등 에탄올복합추출물(SEC)의 잠재적인 치료 가치성 및 기본 메커니즘을 규명 할 수 있었다. 결론적으로 사플라워와 마더워트 등 에탄올복합추출물(GRP)이 다양한 항염증 활성성분 함유 및 천연 항염증제의 잠재적 공급원으로 활용 가능성이 있음을 시사 하였다.

        에탄올 농도에 따른 돌외 잎 차 추출물의 생리활성

        본 연구는 페놀 및 플라보노이드의 총 함량과 생리 활성에 대한 다양한 농도의 에탄올 추출 효과를 비교하기 위해 수행하였다. 추출물의 총 페놀 함량은 35.54 ~ 71.52 mg GAE / g 범위였다. 용매의 에탄올 농도가 증가함에 따라 페놀 함량이 높았고 80%와 99.5%의 에탄올 추출물에서 최고 함량이 확인되었다. 추출물의 총 플라보노이드 함량은 페놀 함량의 경향과 유사하였다. 항산화 실험으로 DPPH, nitric oxide, superoxide, hydroxyl radical 소거능과 TEAC, FRAP, ORAC를 측정하였다. Radical 소거능은 물이나 99.5% 에탄올 추출물 보다 에탄올 수용액 추출물의 활성이 우수하였다. TEAC와 FRAP은 에탄올 농도 의존적인 경향으로 높은 값이 나타났고 ORAC는 40 ~ 80% 에탄올 추출물이 높은 활성을 보였다. 돌외 잎 차의 항산화능을 검토한 결과 측정 방식에 따라 다른 경향이 확인되었다. 그리고 대부분의 실험에서 물 또는 99.5% 에탄올 추출물의 활성이 상대적으로 낮았다. ${\alpha}$-Glucosidase 저해활성은 80%, 99.5% 에탄올 추출물에서 가장 높은 활성이 확인되었고, 미생물 저해활성 또한 동일한 결과를 보였다. 따라서 차의 항산화 활성과 생리활성을 복합적으로 고려할 때 80% 에탄올로 추출하는 것이 적절한 것으로 사료된다. 이러한 결과를 토대로 돌외 차의 식품개발에 있어 기초자료로 활용될 수 있다고 생각된다. The objective of this study was to compare the effects of different concentration of ethanol extraction on the total phenol and flavonoid contents and physiological activities. The total phenol content of the extracts ranged from 35.54 to 71.52 mg GAE/g. An increase in the ethanol concentration of the solvent led to an increase in the phenol content, with the highest content being found in the 80 and 99.5% ethanol extract. The same trend was observed for flavonoid content. DPPH, nitric oxide, superoxide, hydroxyl radical scavenging activity and TEAC, FRAP and ORAC were measured by antioxidant assay. Radical scavenging activity of aqueous ethanol extracts was better than that of water and 99.5% ethanol extracts. TEAC and FRAP were highly dependent on ethanol concentration and ORAC showed high activity in 40 ~ 80% ethanol extract. Antioxidant activity of Dolwoe leaves tea showed different results among the assay systems. In most experiments, the activities of water and 99.5% ethanol extracts was relatively low. ${\alpha}$-Glucosidase inhibitory activity and microorganism inhibitory activity were highest in the 80% and 99.5% ethanol extracts. Therefore, it was considered that extraction with 80% ethanol was appropriate when considering the antioxidative and physiological activities of Dolwoe leaves tea. Based on these results, it can be used as a basic data for the development of food of Dolwoe leaves tea.

        전처리 방법을 달리한 섬애약쑥 에탄올 추출물의 이화학적 특성

        본 연구에서는 전처리 조건(음건, 숙성, 덖음)과 추출 용매 중 에탄올의 혼합비율(30, 50, 70, 90%)에 따른 섬애약쑥 추출물의 이화학적 특성과 항산화 활성을 비교하여, 최적의 전처리 조건 및 추출용매를 확인하고자 하였다. 추출물의 수율은 음건쑥추출물에 비해 숙성쑥과 덖음쑥 추출물이 더 높았고, 에탄올 농도에 따른 경향은 확인되지 않았다. 색도는 L, a, b값 모두 음건쑥 추출물에서 가장 높았고 열처리 시간이 가장 길었던 숙성쑥 추출물에서 유의적으로 낮았다. 가용성 고형분 함량은 덖음쑥, 숙성쑥, 건조쑥 순으로 높고 에탄올의 비율이 증가할수록 그 함량 또한 증가하였다. 총 페놀, jaceosidin, eupatilin 및 총 플라보노이드 함량 또한 덖음쑥, 숙성쑥, 건조쑥 순으로 높았으며, 전반적으로 50%와 70% 에탄올 추출물이 30%와 95% 에탄올 추출물에 비해 유의적으로 높은 함량이었다. DPPH radical 소거 활성과 ABTS 라디칼 소거활성은 덖음쑥 추출물이 음건쑥과 숙성쑥 추출물에 비해 높았는데, DPPH radical 소거 활성은 덖음쑥 70% 에탄올 추출물이 77.71%로 유의하게 가장 높았으며, ABTS radical 소거 활성은 덖음쑥 30%, 50% 및 70% 에탄올 추출물이 96% 이상의 높은 활성을 나타내었다. 이상의 결과로부터 섬애약쑥은 음건한 것을 덖음 처리를 한 후 50%~70% 에탄올로 추출할 때 유효성분의 함량과 라디칼 소거활성이 높음을 확인하였다. This study was performed to determine the ethanol ratios (30%, 50%, 70%, and 90%) of extraction solvent of Seomaeyakssuk (Artemisia Argyi H.) prepared using different methods (drying at room temperature [D], aging at 60℃ for 7 days after drying [AD], and roasting for 30 min at 160℃ after drying [RD]). The extract yield of the D extracts was lower than that of the AD and RD extracts, but the ethanol concentration of extract solvent did not affect. The L, a, and b values of the D extracts were highest, whereas those of the AD extracts were lowest. No clear trend was observed in the ethanol ratios. The soluble solids, total phenol, total flavonoid, jaceosidin, and eupatilin contents of each extract varied significantly, with RD > AD > D. The soluble solids significantly increased by ethanol ratio of extraction solvent, but other phytochemicals contents of 50% and 70% ethanol extracts were higher than others without affecting the processing methods. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was highest (77.71%) in the 70% ethanol extract obtained from RD. 2,2-azinbis-(3-ethylbenzo-thiazoline-6-sulphonate(ABTS) radical scavenging activity was significantly higher in 30-70% ethanol extract than 90% ethanol extract from RD. The results suggest that the contents of active ingredients and radical scavenging activity of ethanol extracts from Seomaeyakssuk were highest in the RD extract using 50-70% ethanol.
