        Antibacterial Efficacies of Disinfectants against Salmonella typhimurium Depending on Pre-warming Conditions

        Salmonellosis is a widespread bacterial zoonosis that commonly causes enterocolitis and foodborne poisoning leading to an extensive economic loss in domestic animal industry. Considerably, the emergence of multidrug resistant strains of Salmonella spp. induces further severe problems affecting public health. The present report was designated to investigate the antibacterial efficacies of three common disinfectants including an oxidizing compound disinfectant (OXC), a triple salt (TS) and a quaternary ammonium compound (QAC) against Salmonella typhimurium subjected to the preliminary changes of drug temperature. All solutions of three disinfectants were pre-incubated at different temperature (22, 37 and 63°C) for 1 h prior to exposure to bacteria. The disinfectants and bacteria were diluted with distilled water (DW), hard water (HW) or organic matter suspension (OMS) according to treatment condition. Under the DW condition, the disinfectant efficacy of the QAC at 63°C was higher than that of 22°C. Furthermore, under HW diluent the disinfectant efficacy of the TS pre-warmed at both of 37 and 63°C were increased compared to that of 22°C. Considerably, the efficacy of pre-warmed QAC at both of 37 and 63°C under the OMS diluent were higher than that of 22°C. Conclusively, prewarming at higher temperatures have positive effects on the stability of the antibacterial efficacies of TS and QAC.

        Antibacterial Efficacy of Dental Sealant Containing Phytoncide

        Background: Dental caries prevention is a key research focus in dentistry, requiring advancements in the formulation of dental sealants. This study investigated the physical and antibacterial attributes of dental sealant enriched with phytoncide. Methods: Phytoncide was mixed with a commercially available dental sealant (Clinpro) at concentrations of 0 (control), 1.5%, 3%, and 4.5% by weight (wt%). The flexural strength, curing depth, and wettability of the dental sealant were measured. Antibacterial properties against Streptococcus mutans were evaluated through the enumeration of colony-forming units. Statistical analyses employed one-way variance analysis followed by Tukey’s test (p<0.05). Results: The dental sealant containing 3% phytoncide showed no significant difference in flexural strength and curing depth compared with that in the control group (p>0.05). The flexural strength and curing depth decreased with increasing phytoncide content and significantly differed in sealant containing 4.5 wt% phytoncide (p<0.05). Wettability did not differ between the experimental and control groups (p>0.05). The antibacterial properties of the sealant containing 1.5% phytoncide were the same as those of the control group (p>0.05). The bacterial viability was significantly reduced in groups containing 3% and 4.5% phytoncide compared with that in the control group (p<0.05). Conclusion: Dental sealants incorporating phytoncide have a promising potential as antibacterial dental materials.

        Antibacterial activity of a novel florfenicol-loaded chitosan-sodium tripolyphosphate composite nanogels in vitro and in mice against Staphylococcus aureus small colony variants

        Background: Florfenicol might be ineffective for treating Staphylococcus aureus small colony variants (SCVs) mastitis. Objectives: In this study, florfenicol-loaded chitosan (CS)-sodium tripolyphosphate (TPP) composite nanogels were prepared to allow targeted delivery to SCV infected sites. Methods: The formulation screening, the characteristics, in vitro release, antibacterial activity, therapeutic efficacy, and biosafety of the florfenicol composite nanogels were studied. Results: The optimized formulation was obtained when the CS and TPP were 10 and 5 mg/mL, respectively. The encapsulation efficiency, loading capacity, size, polydispersity index, and zeta potential of the optimized florfenicol composite nanogels were 87.3% ± 2.7%, 5.8% ± 1.4%, 280.3 ± 1.5 nm, 0.15 ± 0.03, and 36.3 ± 1.4 mv, respectively. Optical and scanning electron microscopy showed that spherical particles with a relatively uniform distribution and drugs might be incorporated in cross-linked polymeric networks. The in vitro release study showed that the florfenicol composite nanogels exhibited a biphasic pattern with the sustained release of 72.2% ± 1.8% at 48 h in pH 5.5 phosphate-buffered saline. The minimal inhibitory concentrations of commercial florfenicol solution and florfenicol composite nanogels against SCVs were 1 and 0.25 μg/mL, respectively. The time-killing curves and live–dead bacterial staining showed that the florfenicol composite nanogels were concentration-dependent. Furthermore, the florfenicol composite nanogels displayed good therapeutic efficacy against SCVs mastitis. Biological safety studies showed that the florfenicol composite nanogels might be a biocompatible preparation because of their non-toxic effects on the renal tissue and liver. Conclusions: Florfenicol composite nanogels might improve the treatment of SCV infections.

        양식 어류에 있어서 황산동(copper sulfate pentahydrate)의 안전성과 살균효과에 대한 연구

        Antibacterial efficacy and safety of copper sulfate pentahydrate (5% solution) was evaluated in laboratory and field studies for disinfection of some fish bacterial pathogens in cultured fish, flounder(Paralichthys olivaceus), mirror carp(Cyprinus carpio), gold fish(Carrassius auratus) and eel(Anguilla japonica). Dipping of fishes for acute toxicity was performed for a period of 48h, and the TLm value(median tolerance limit) were 4.0ppm in mirror carp and flounder, 4.4ppm in rainbow trout, and 10.0ppm in gold fish. After exposure below than TLm value for 1 month, no side effects and histopathological changes were observed. The test drug were shown high antibacterial activities against most bacterial pathogens by exposure in the laboratory and field trials. However, Aeromonas salmonicida showed a resistance to the dosage. We can prevent outbreaks of bacterial diseases of fish by the dosage of 1ppm for 6hr and the test drug is thought to be used effectively and widely as a disinfectant against most bacterial fish pathogens.

        Evaluation of the Antibacterial Activity and Treatment of Acne with SiO2-ionized Loess

        SiO2-ionized loess was prepared from the reaction of loess and sodium hydroxide at 1,400°C for 2 h. The antibacterial activity of SiO2-ionized loess against Staphylococcus aureus, Bacillus subtilis, Escherichia coli,Pseudomonas aerusinosa, and Propionobacterium acnes causing acne was examined by comparing the results against those of untreated loess, and treatment efficacy was examined for treating acne using soap containing SiO2-ionized loess. The minimum inhibitory concentration value of SiO2-ionized loess against S. aureus, P. aerusinosa, B. subtilis, E. coli, and P. acne was 10.0, 10.0, 2.5, 5.0, and 2.5 mg/mL, respectively. However, medium containing untreated loess had no antimicrobial activity. A treatment efficacy test revealed that acne symptoms decreased as the duration of using soap containing SiO2-ionized loess increased.

      • Green synthesis of multifunctional silver and gold nanoparticles from the oriental herbal adaptogen: Siberian ginseng

        <P>Pharmacologically active stem of the oriental herbal adaptogen, Siberian ginseng, was employed for the ecofriendly synthesis of Siberian ginseng silver nanoparticles (Sg-AgNPs) and Siberian ginseng gold nanoparticles (Sg-AuNPs). First, for metabolic characterization of the sample, liquid chromatography-tandem mass spectrometry analysis (indicated the presence of eleutherosides A and E), total phenol content, and total reducing sugar were analyzed. Second, the water extract of the sample mediated the biological synthesis of both Sg-AgNPs and Sg-AuNPs that were crystalline face-centered cubical structures with a Z-average hydrodynamic diameter of 126 and 189 nm, respectively. Moreover, Fourier transform infrared analysis indicated that proteins and aromatic hydrocarbons play a key role in the formation and stabilization of Sg-AgNPs, whereas phenolic compounds accounted for the synthesis and stability of Sg-AuNPs. 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H tetrazolium bromide (MTT) assay determined that Sg-AgNPs conferred strong cytotoxicity against MCF7 (human breast cancer cell line) and was only slightly toxic to HaCaT (human keratinocyte cell line) at 10 µg⋅mL<SUP>−1</SUP>. However, Sg-AuNPs did not display cytotoxic effects against both of the cell lines. The disc diffusion assay indicated a dose-dependent increase in the zone of inhibition of <I>Staphylococcus aureus</I> (ATCC 6538), <I>Bacillus anthracis</I> (NCTC 10340), <I>Vibrio parahaemolyticus</I> (ATCC 33844), and <I>Escherichia coli</I> (BL21) treated with Sg-AgNPs, whereas Sg-AuNPs did not show inhibitory activity. In addition, the 2,2-diphenyl-1-picrylhydrazyl assay demonstrated that both Sg-AgNPs and Sg-AuNPs possess strong antioxidant activity. To the best of our knowledge, this is the first report unraveling the potential of <I>Eleutherococcus senticosus</I> for silver and gold nanoparticle synthesis along with its biological applications, which in turn would promote widespread usage of the endemic Siberian ginseng.</P>
