        유제화에 의한 경구용 항암제인 테가푸르의 장관 임파수송

        W/O and O/W emulsions of tegafur (50 mg/5 ml/kg) were orally administered to rats to compare with their mesenteric lymphatic delivery effects. And also in order to demonstrate the lymph targeting associated to the oral route, it was deemed necessary to investigate the fate of solution after oral administration as a control. Lymph and plasma samples were periodically taken from each subject of mesenteric lymphatic duct cannulated rats. Then, lymph and plasma levels of tegafur and its active metabolite, 5-FU, were simultaneously observed. Also pharmacokinetic parameters were compared with each others. On the other hand, most previous studies of lymphatic transport have not addressed the question of whether an increase in mesenteric or thoracic lymph transport by the manipulation of a suspected variable was due to a selective delivery to the intestinal lymphatics or an overall increase availability. Therefore, based on a physiologically based pharmacokinetic model which represents the characteristics of lymphatic systems, we are also going to determine the contributions of mesenteric lymph transport versus thoracic lymph transport of tegafur reported in reference(13). In comparison with tegafur solution, AUC and mean residence time of plasma tegafur were significantly increased in W/O emulsion but significantly decreased in O/W emulsion. Lymph flow rates were similar in both solution and W/O emulsion but half in O/W emulsion. AUC of tegafur in mesenteric lymph and in plasma for W/O emulsion were 3.7 times and 2.9 times more than those for O/W emulsion, respectively. And AUC of 5-FU in thoracic lymph for W/O emulsion was 3.7 times more than that for O/W emulsion. These results suggested that lymphatic delivery or tegafur by W/O emulsion was more effective than that by on emulsion due to its differences or formation ability of chylomicrons.

        상황버섯, 발아현미, 발아현미 상황버섯균사체의 IgE 생합성에 대한 면역조절에 관한 비교연구

        Background and objectives: To compare the immunomodulatory effects on the amount of IgE synthesis, the distribution of T-cells and the amount of cytokine secretion in the MLN (Mesenteric Lymph Node) between Phellinus linteus (PL), Germinated Brown Rice (BR) and Phellinus linteus Grown on Germinated Brown Rice (PB) and thereby to examine their relationships with atopic dermatitis. Materials and methods: All the extracts of PL, BR and PB were dissolved in the propylene glycol. Then, male mice aged 4 weeks were divided into four experimental groups, each of which comprised ten mice. These mice were provided with experimental foods in a certain environment and then orally given each extract. Through a retro-orbital bleeding, the peripheral blood was sampled. This blood sample was used for an analysis of IgE. Results: The concentration of serum IgE was significantly decreased in the experimental groups where PL, BR or PB were administered as compared with the control group (Fig, 1-1). Besides, the concentration of serum IgE was also significantly decreased in the experimental groups where PL, BR or PB were administered in the MLN as compared with the control group (Fig 1-1). The number of CD4+ T-cells was significantly greater in the experimental groups where PL and PB were administered as compared with the control group. But the number of CD8+ T-cells was the smallest in the experimental group where PL was administered(Table 1). Besides, of all the extracts, BR had an effectiveness in enhancing the concentration of IFN-γ (Interferon-γ) to the greatest extent in the MLN (Fig, 1-2). Conclusions: It has been reported that Th2 cytokines and serum IgE are relatively higher in more than 80% of patients with atopic dermatitis. Our results showed that PB reduced the amount of the secretion of Th2 cytokine. Thus, by modulating the amount of IgE synthesis, it had an immunosuppressive effect on various types of inflammatory markers. It is also presumed that PB might reduce the amount of the secretion of pro-inflammatory cytokines in MLN. It can therefore be inferred that PB might also have a treatment effect on atopic dermatitis. Further studies are warranted to examine the relationship between PB and atopic dermatitis. 상황버섯, 발아현미, 발아현미 상황버섯균사체가 각각 생쥐의 MLN(Mesenteric Lymph Node) 림프구 내에서 형성되는 IgE의 생성량, T-세포 분포 및 cytokine 분비량 등에 미치는 면역조절 효과를 서로 비교하므로 아토피피부염과의 관련성 여부를 알아보았다. 데이터는 모두 평균±SEM으로 나타내었고 각 실험 그룹의 평균 차는 던칸의 다중범위검정(Duncan's multiple range test)과 함께 실시된 일원 공분산분석(one-way ANOVA)으로 그 값을 구하였다. 이들 분석을 위해 SPSS 10.0(SPSS institute, Chicago, IL)을 사용하였다. 그 결과, 혈청 IgE 농도는 PL, BR, 또는 PB를 투여한 실험군이 대조군에 비해 유의하게 감소한 것으로 나타났다(Fig, 3-1). 또한 모든 추출물 특히 BR은 MLN 림프구 내에서 IFN-γ농도를 크게 높이는 효과가 있었으며(Fig, 3-2), 모든 추출물은 Th1(IFN-γ 제외) cytokine 및 Th2 cytokine의 분비량을 크게 감소시키는 효과가 있었다(Fig, 3-2). 아토피피부염 환자의 80% 이상에서 Th2 우세와 혈청 IgE가 증가하는 것으로 보고되고 있다. 따라서 아토피피부염에서도 그 효과를 볼 수 있을 것으로 사료되며 향후 아토피피부염과의 관련성 여부에 대한 더 많은 연구가 이루어져야 할 것이다.

        Phellinus linteus (PL), one of the immune-regulatory substances, is recognized to play the role in the metabolic process on inflammation and immunity. It has been traditionally used in the oriental medicine to treat inflammatory related disease. The purpose of this study was to evaluate the effects of water extracts of PL on the mesenteric lymph node lymphocytes immune function in the ICR male mice. Control mice received vehicle only. The PL treated mice were administered the respective extract by oral gavages for 4 weeks. IgE concentrations in serum and MLN lymphocytes were significantly lower in PL treated mice than in control mice. PL increased the proportion of $CD4^+\;and\;CD8^+$ T cells in MLN lymphocytes. PL significantly decreased Th2 cytokine concentrations and mRNA expression levels in cytokine secretions. Therefore, water extracts of PL modulate inflammatory parameters through regulation of immunoglobulin production resulting from decreased Th2 cytokine secretion and mRNA expression levels and reduce pro-inflammatory cytokine secretion and mRNA expression in MLN lymphocytes.

        Feasibility and oncologic safety of low ligation of inferior mesenteric artery with D3 dissection in cT3N0M0 sigmoid colon cancer

        Purpose: There is no standard consensus for the ligation level of the inferior mesenteric artery during radical resection of sigmoid colon cancer. Especially, there is little research about low ligation combined with D3 dissection. The study was performed to compare feasibility and oncologic safety between low ligation with D3 dissection to high ligation in intermediately advanced sigmoid colon cancer. Methods: From January 2008 to December 2013, 134 patients who underwent radical surgery for cT3N0M0 sigmoid colon cancer were evaluated retrospectively. Clinicopathologic factors and oncologic outcomes of high ligation (HL, n = 51) and low ligation (LL, n = 83) groups were compared. Results: The mean operative time was significantly shorter in LL, and there was no difference in complications, distal margin or number of retrieved lymph node. The tumor size was significantly larger in HL, but there was no difference in number of metastatic lymph node, pT or pN stage. There was no difference in overall survival, disease-free survival, or local and systemic recurrence. Conclusion: In cT3N0M0 sigmoid colon cancers, we suggest that low ligation with D3 dissection can be performed with feasibility and oncological safety.

        Further Characterization of Activin A-induced IgA Response in Murine B Lymphocytes

        We have recently shown that activin A, a member of TGF-$\beta$ superfamily, stimulates mouse B cells to express IgA isotype but other isotypes. In the present study, we further characterized effects of activin A on B cell growth and IgA expression. We found that activin A did not have effect on LPS-stimulated cell viability. In parallel, CFSE staining analysis revealed that activin A did not alter cell division. An increase of IgA secretion by activin A was completely abrogated by anti-activin A Ab but not by anti-TGF$\beta$1 Ab. In the same conditions, no other isotypes are significantly affected by each antibody treatment. Finally, activin A, as similar to TGF-$\beta$1, increased IgA secretion by mesenteric lymph node cells. These results suggest that activin A can specifically stimulate IgA response, independent of TGF-$\beta$ in the gut.

        Dietary β-Glucan Regulates the Levels of Inflammatory Factors, Inflammatory Cytokines, and Immunoglobulins in Interleukin-10 Knockout Mice

        β-Glucan is known to have anti-inflammatory properties, and several studies have demonstrated the beneficial effects of dietary β-glucan on inflammatory bowel disease (IBD). However, it is unknown how β-glucan mediates its protective effects on IBD. Therefore, we used a well-established mouse model for IBD, interleukin (IL)-10^(–/–) mice, to explore the protective effects of β-glucan on IBD-like symptoms caused by IL-10 deficiency. The mice were divided into two groups: IL-10^(–/–) and IL-10^(–/–) + β-glucan treatment groups. IL-10^(–/–) mice treated with dietary β-glucan exhibited less inflammation within the colon. The levels of immunoglobulins A and E were lower in the serum, spleen, mesenteric lymph nodes, and Peyer's patches in the IL-10^(–/–) mice compared with the IL-10^(–/–) + β-glucan mice. Also, the expression of pro-inflammatory cytokines was lower in the IL-10^(–/–) + β-glucan mice compared with the IL-10^(–/–) mice. Histological analysis also revealed that administration of dietary β-glucan in IL-10^(–/–) mice reduced colonic tissue damage. Finally, the expression of the pro-inflammatory cytokine tissue necrosis factor-α was significantly lower with dietary β-glucan treatment in IL-10^(–/–) mice. In conclusion, dietary β-glucan reduces the inflammation associated with IBD caused by IL-10 deficiency.

        芎梔化痰煎과 淸神化痰煎의 면역조절효과

        This study was performed to evaluate the immunoregulatory effects of Gungchihwadarn-Jeon(芎梔化痰煎, GCHDJ) and Cheongsinhwadarn-Jeon(淸神化痰煎, CSHDJ). GCHDJ or CSHDJ was administered(p.o.) once a day for 14 days to mice. By the treatment of GCHDJ or CSHDJ was increased the cell viability of cultured mice splenocytes, thymocnes and mesenteric lymph node cells. Administration of GCHDJ or CSHDJ was increased the splenic T lymphocyte, especially the Tc cell subpopulation was increased by the GCHDJ, on the while the TH cell was increased by the CSHDJ. The administration of GCHDJ or CSHDJ was significantly increased the apoptosis of transplanted-L1210 leukernia cells to mice peritoneal cavity. These results suggest that GCHDJ and CSHDJ have an immunoregulatory action
