        제조 藥針의 抗癌作用에 關한 硏究

        최근 과학기술의 놀라운 발전에 힙입어 질병이 하나씩 정복되어 가고 있으나 평균수명의 연장 및 생활수준의 향상으로 인한 고지방식 과다섭취, 스트레스 등으로 인하여 질병으로 인하여 종양으로 인한 사망률이 증가하고 있는 실정이므로 종양의 치료방법이 다양한 방법으로 시도되고 있으며 약침을 이용한 방법도 이중 하나이다. 제조는 예로부터 破血, 行瘀, 散結, 通乳하는 효능이 있는 것으로 알려져 있어 이를 산지별로 다양한 방법으로 유효성분을 추출한 후 중완과 족삼리에 약침처리를 하여 담암생쥐의 생존능, 종양의 억제능, 시험관내세포독성능 실험을 수행하여 항암능을 살펴보고자 하였다. Median survival time에서 Normal군에서는 22.5일을, Control군에서는 24일을 보여 약침 자극만으로도 6.67%의 생존율이 연장되었으며 Hd군과 Jd군은 26.5일을 보여 Control군에 비해 10.42%의 증가율을 보였으며 Hw군은 28일에 16.67%의 증가율을, Jw군은 29일에 20.83%의 증가율을 보여주어 약침제제 사용의 경우뿐만 아니라 단순한 경혈 자극만으로도 생존율이 증가하였음을 알수 있었다. 담암생쥐의 종양을 적출하여 무게를 잰 결과 Control군에 비해 Hwt군만을 제외하고는 모두 유의성 있는 억제율을 보여주고 있었으며 Hw군과 Jw군이 각각 1.851±0.027g, 1.827±0.015g으로 가장 높은 효과를 보여주었다. Sarcoma-180 세포를 이용한 시험관내 세포독성능 실험에서 생존율에 의한 IC_50은 약침제조액을 1로 보았을 경우 Ha군은 0.6을 Ja군은 0.375를 보였다. 이를 종합하면 중완과 족삼리에 단순히 자극을 준 것만으로도 생존율이 어느 정도 연장되어 경혈의 효과를 입증한 것으로 보여지며 여러 추출법에 있어서 물추출알콜 침전법이 다른 추출법에 비해 상대적으로 유효한 성분을 많이 얻었던 것으로 보아 추후 유효성분의 규명과 규격화 및 안전도에 관한 후속 연구가 필요할 것으로 사료된다. In order to investigate the antitumor effect by Aqua-acupuncture liquor made by precipitation method in alcohol from Holotrichia after B-16 cells were transplanted in C57BL/6 mice. Experimental studies were performed for measurance of metastasis, cell cytotoxicity in vitro, life extention, weight of cancer. The first station investgated the effect of Aqua-acupuncture liquor made by precipitation method in alcohol from Holotrichia boiled with water and Aqua-acupuncture Holotrichia liquid by distilled method on the survival time of the mice transplanted by sarcoma-180 tumor cell, inhibition of Holotrichia and Saline on B016 Tumor in Mice. The measurement of In vitro cytotoxicity showed a tendency to inhibite cell, the size of edema showed significant value and the survival time. The results were summarized as follows: 1. Mean survival time in Hd(10.42%), Jd(10.42%), Hw(16.67%), Jw(20.83%)group was prolonged, as compared with control group significantly(P< 0.05). 2. On the weight of solid tumor, Holotrichia-treated group was less than control group with significance(P< 0.05). 3. On the MTT assay, cell viability was significantly inhibited by 1/well, 0.5/well, 0.25/well, 0.125/well. 0.625/well of Aqua-acupuncture liquor made by precipitation method in alcohol from Holotrichia(produced in Korea) boiled with water, 1/will, 0.5/well, 0.25/well, 0.125/well of Aqua-acupuncture liquor made by precipitation method in alcohol from Holotrichia(produced in China) boiled with water and in case of korea and china of Aqua-acupuncture Holotrichia liquid by distilled method concentration inhibited cell viability significantly(P< 0.05). IC_50 for cell viability was 2.17 ㎍/well.

      • 제조 抽出物이 腦組織의 acetylcholine esterase 및 monoamine oxidase 活性에 미치는 영향

        Holotrichia have tested to study the effects concerning acetylcholine esterase and monoamine oxidase activities in rats brain. In vitro, the level of lipid peroxide and activity of acetylcholine esterase in tissues of brain were propertionally decreased to concentration of extracts prepared from Holotrichia. In vii o, after the extracts of Holotrichia was administered to the rat, the level of lipid peroxide was decreased according to treatment days and dose. And, enzyme activities and ratio of type conversion of xanthine oxidase were decreased, too. Also, enzyme activities of acetylcholine esterase were lowered. These results suggest that the extract of Holotrichia might have an effect on treatment of dementia according to decreasing the activities of acetylcholine esterase, monoamine oxidase in brain.

        제조 추출물의 혈전 용해 활성에 대한 실험적 연구

        1. Objectives This study was performed to find the activities and characteristics of purified thrombolytic enzymes from Holotrichia extracts. 2. Methods In the first time, a coarse enzyme fluid was made by using the freedried Holotrichia extracts. After manufacturing total soluble proteins and purifing enzymes, it was evauluated the activities and characteristics of this enzyme's dissolving capability to fibrin and thrombus. This study was taken using azocasein assay, fibrin-plate method, native-PAGE and fibrin zymography. 3. Results A soluble proteins were efficiently extracted form freezedried Holotrichia extracts. And, this purified enzyme had a ten times fibrinolytic capability compare with ustulation Holotrichia sample. In native PAGE and fibrin zymography, Holotrichia extracts showed the respectable fibrinolytic activity. Also, It had higher thrombolytic activities compared with general thrombolytic enzyme 'plasmin'. In experiment of various protease inhibitors of the purified enzyme from Holotrichia extracts on the azocaseinolytic activity, the enzyme was strongly inhibited by EDTA ${\cdot}$ EGTA, and weakly by APMSF ${\cdot}$ PMSF ${\cdot}$ TPCK. 4. Conclusion Holotrichia extracts has the thrombolytic activities, and it will operate directly th fibrin-clot and thrombus.

In Adjuvant arthritis of rats, a chronic inflammatory model, extracts of Holotrichia diompharia demonstrated the significant inhibitory effects on hind paw diameter, arthritis score, pain score, duration time on rotarod tredmill.

        제조가 초산납으로 유발한 흰쥐의 뇌독성에 미치는 영향

        Objectives: This study was undertaken to investigate the action mechanism of Holotrichia (HT) at parameter related to dementia. Methods: HT was tested for the effects on acetylchonine esterase and monoamine oxidase activities, lipid peroxidation, antioxidation in brain of lead acetate-treated rats. Lead acetate were treated firstly into samples for 10 days, and then lead acetate and HT were set with them for 20 days. Results: The level of lipid peroxide, acetylcholine esterase and monoamine oxidase activities, enzyme activities and ratio of type conversion of xanthine oxidase increased in lead acetate-treated rats were decreased as highly as normal group by HT. Superoxide dismutase, catalase and glutathione peroxidase activities, the level of acetylcholine decreased in lead acetate-treated rats were increased as lowly as normal group by HT. Conclusions : These results suggest that HT might have an effect on treatment of dementia according to decreasing the activities of acetylcholine esterase, monoamine oxidase and level of lipid peroxide in brain.

      • 제조가 사염화탄소에 의해 유도된 흰쥐의 지질함량에 미치는 영향

        사염화탄소(CCl_4)는 가축의 구충제, 용매, 지용성, 비가연성, 비휘발성 세제 및 소화제로 널리 사용되어온 할로겐 화합물^14)로 사람을 비롯한 모든 종에 대하여 간손상을 일으키는 대표적인 약물로 알려져 있다. 사염화탄소의 간장 독작용에 관한 기전은 명확히 규정되어 있지 않으나 alkylating radical을 지닌 간장 독성물질로 규정하는 세포 파괴설^15)로 사염화탄소를 투여하면 cytochrome P450 monooxygenase에 의하여 전자를 공급받아 trichloromethyl free radocal(CCl_3·)로 대사되어 세포내 단백질, 지질 및 기타 거대분자 등과 결합하여 단백질을 불활성화시키거나 효소활성을 감소시키거나^16~19) trichloromethylperoxy free radical(CCl_3OO·)로 산화되어 세포막의 불포화 지방산을 과산화시켜 막의 구조와 기능을 파괴하여 간에서 단백합성을 저해하고^20,21) 간장 glycogen양을 감소시키며, 혈중으로 AST 및 ALT 등의 유출을 일으키고 또한 조직학적으로는 간장조직의 지방변성과 괴사 등을 일으킨다고 알려져 있다.^22,23) 한편 혈액의 분산과 기관지 해소작용 등이 있어 각종 염증 및 기관지 치료에 사용되는 한방약제인 제조을 사용하여 흰쥐에게 사염화탄소를 투여하여 혈청 지질함량에 미치는 영향을 알아보고자 실험을 하였다. 사염화탄소 투여에 의하여 흰쥐 혈청의 지질함량이 전반적으로 증가하였는데 특히 중성지방의 증가가 현저하였으며 증가한 지질함량은 제조 수침액을 투여함으로써 양호하게 감소하였다. 그러나 동맥경화의 중요한 유발인자로 알려진 HDL-cholesterol^24)은 별다른 변화가 관찰되지 못하였다. 결국 흰쥐에게 사염화탄소를 투여함으로써 혈청에서 지질함량과 동맥경화지수가 증가하였으며 이것은 동맥경화증과 관련된 혈관의 순환기 질환 발병의 위험성이 높아진 것이며 제조 수침액은 혈청 지질함량을 감소시킴으로써 이들 질환의 치료가능성이 있다고 할 수 있을 것이다. The purpose of this study was to investigate the effect of Holotrichia diomphalia Bates on serum lipid contents in carbon tetrachloride-treated The water extract of ones was obtained and administrated to rats. The results that lipid contents in serum were increased by intra peritoneal injection of carbon tetrachloride that of control group, but they were significantly decreased by orally administration of Holotrichia diomphalia Bates water extract. But HDL-cholesterol content was not changed and arteriosclcrotic index was increased almost twice in carbon tetrachloride-treated group than in normal group. Therefore, it syggested that Holotrichia diomphalia Bates could be promising agents in a circulatory disorder including arteriosclerosis.

        신장(腎臟) 조직(組織)에서 제조의 항산화(抗酸化) 효과(效果)의 기전(機轉) 연구(硏究)

        This study was carried out to examine mechanisms by which Holotrichia (HTC) produces protective effect against renal cell injury. HTC extraction (5%) prevented $H_2O_2(50mM)$-induced LDH release and lipid peroxidation in renal cortical slices. When slices were treated with 5% HTC extraction, cellular glutathione content and the superoxide dismutase activity were not altered in control and $H_2O_2$-treated tissues. When slices were treated with 50 mM $H_2O_2$, the catalase activity was significantly inhibited, which was completely restored by addition of 5% HTC. Treatment of slices with 5% HTC extraction increased the glutation peroxidase activity in $H_2O_2$-treated tissues. These results suggest that HTC prevents oxidant-induced cell injury and lipid peroxidation by increasing the activities of catalase and glutathione peroxidase in renal cortical slices.

        "동의수세보원(東醫壽世保元)"에서 이용된 제조의 기원(起源)에 대한 문헌적(文獻的) 고찰(考察)

        1. Objectives and Methods The original insect of $Q\'{\i}C\'{a}o$ is described as the larvae of Holotrichia diomphalia Bates (Scarabaeidae) in oriental medicine literatures. Traditionally $Q\'{\i}C\'{a}o$ inhabit on rotting wood, compost, and organic debris of soil or thatched roofs. They have the very important distinguishing mark that wriggle along on their back through rotting wood, compost etc. and they never do inflict the living crops. but, Holotrichia diomphalia Bates as the $Q\'{\i}C\'{a}o$ is not right because of many questions, such as Holotrichia are the harmful insects eat into farm products, and those don't match well with korean farmers' moods if their cultivated lands are turned over for collecting $Q\'{\i}C\'{a}o$ larvae, and especially they don't go on using their back. 2. Results and Conclusions Accordingly, in the conclusion of this subject it is reported that the original insect of $Q\'{\i}C\'{a}o$ is the larvae of Korean Cetoniidae(Coleoptera; Scarabaeoidea) insects including Genus Protaetia Burmeister(Protaetia orientalis submarmorea Burmeister, P.brevitarsis seulensis Kolbe, etc.) and the larvae of Korean Cetoniinae insects. Also, those were the original larvae of $Q\'{\i}C\'{a}o$ that were applied to not only 'DongEuiBoGam' but also 'Dongyi Suse Bowon prescriptions'. Those feed on rotting wood, compost, and organic debris of soil or thatched roofs.

        인삼을 가해하는 큰검정풍뎅이(Hozotrichia morose Waterhouse)의 산란선호성

        인삼의 주요 해충의 하나인 큰검정풍뎅이의 발생요인을 구명하여 효율적인 방제대책을 수립하기 위해 몇 가지 환경요인에 대한 산란선호성을 조사한 결과 큰검정풍뎅이는 지피식물이 없는 나지에는 전혀 산란하지 않았고 식물의 종류에 따라서도 차이를 보여 쑥. 망초, 바랭이, 쇠비름 등에는 잔디나 달맞이꽃에 비해 산란수가 많았으며, 토끼풀에는 산란하지 않았다. 지피식물이 바랭이일 때 지피도 높은곳, 토양경도가 낮은 곳에서 산란수가 많았다. 지피물이 볏짚인 곳은 산란수가 있어 나지와 유의차를 보이지 않아 볏짚은 산란유인으로 작용하지 않는 것으로 생각되며, 토양내의 부숙볏짚 역시 산란수에 영향을 미치지 않는 것으로 보였다. 또한 과건 또는 과습토양에서는 산란수가 현저히 감소하였다. The ovipositional preferences of Holotrichia morosa were investigated to defermine several environmental factors related to its incidence in ginseng fields. H. moroia paid no eggs in naked soil without ground color plants, and shouted ovipositional preference among weed plants in the order to Aurmisin prinrcps var oripntnlis, Epigrren canadensis Diuitarin sanguinalis, and Porttulaca oleracea. More oviposition was observed in the denser vegetation of D. sanguinalis and in soft soil. The egg-laying females were seldom attracted to fresh rice straw mulched on soil surface or decayred rite straw mixed into soil. The number of eggs laid by a female decreased considerably when the soil moisture content was below 5% or above 35%.

        Effects of Holotrichia diompharia on angiogenesis were investigated. In adjuvant-induced granuloma pouch of mice, extract of Liocolae vermiculus represented the prominant effects on angiogenesis and granuloma pouch formation. The results show the inhibitony effects on angiogenesis of Liocolae vermiculus.
