        Phytotoxicity and Volatile Monoterpenes of Leaves from Artemisia capillaris and Artemisia iwayomogi Used as Korean Herbal Injin

        Artemisia capillaris and Artemisia iwayomogi are weeds used as medicinal plants in Korea under the name ”Injin“. I collected leaves of A. capillaris and A. iwayomogi, examined them for phytotoxic effects from volatile substances and determined the composition of monoterpenes in the leaves. The effects of volatile substances from each species on seed germination and radicle elongation in each of the two Artemisia species were assessed. The volatile substances of A. capillaris did not negatively affect the seed germination of A. capillaris, but they did inhibit radicle elongation. Rates of seed germination of A. iwayomogi decreased when the seeds were exposed to high concentration of A. capillaris volatile substances. The inhibition of seed germination and radicle elongation by volatile substances from both Artemisia species was stronger for A. iwayomogi than for A. capillaris. I identified the monoterpenoids from the leaves with a gas chromatograph-mass spectrometer (GC-MS). The main constituents of A. capillaris were acenaphthylene (37.91%), β-pinene (12.08%), 4-carene (10.61%) and γ-curcumene (9.92%), while those of A. iwayomogi were germacrene-δ (32.15%), borneol (21.24%), camphor (20.45%) and trans-caryophyllene (7.75%).         인진(茵蔯)으로 쓰이는 사철쑥과 더위지기 추출물의 항미생물활성 및 total polyphenol함량

        We investigated the antimicrobial activity of ether and ethylacetate fractions from Artemisia capillaris and Artemisia iwayomogi used as Injin. The antimicrobial activity of the ethylacetate fractions from Artemisia capillaris and Artemisia iwayomogi for tested microbes were stronger than those of ether fractions of the two Artemisia plants. The antimicrobial activity of fractions of Artemisia iwayomogi was higher than that of fractions of Artemisia capillaris for the tested microbes. The extracts of young shoots and leaves showed stronger antimicrobial activity than those of young leaves. The range of minimum inhibitory concentrations(MICs) of ethylacetate fractions from the part of Artemisia capillaris and Artemisia iwayomogi were 0.25~2.0 mg/ml. The MICs of ether fractions were showed higher concentration than those of ethylacetate fractions. The fractions of Artemisia iwayomogi showed lower MICs than fractions of Artemisia capillaris. The highest total polyphenol content was found in young shoots and leaves of A. capillaris. The young shoots and leaves of the two kinds of Artemisia plants showed higher content of total polyphenol. 본 실험은 인진으로 쓰이는 사철쑥과 더위지기의 항미생물활성을 검정한 것으로 추출용매에 따른 항미생물활성은 gram양성균 3종과 gram음성균 2종 모두에서 ethylacetate추출물들이 ether추출물들보다 전반적으로 높았다. 실험미생물 중 B. subtilis와 B. cereus에 대한 추출물들의 활성이 다른 실험미생물에 비하여 강하게 나타났으며 더위지기 추출물들이 사철쑥 추출물들에 비하여 높은 활성을 보였다. 추출용매에 따른 최소저해농도는 ethylacetate추출물이 ether추출물보다 낮은 농도에서 대부분의 미생물들의 생육을 억제하였고, 더위지기와 사철쑥 부위별로는 잎과 줄기에서 잎에 비하여 낮은 농도에서 미생물들의 생육을 억제하였으며 더위지기 추출물들의 최소저해농도가 사철쑥 추출물들보다 낮았다. 총 폴리페놀함량은 사철쑥 잎과 줄기 추출물에서 27.57 mg/g으로 가장 높게 나타났으며 대체로 잎과 줄기 추출물에서 잎 추출물에서보다 높은 함량을 나타내었다.

        에탄올 투여 흰쥐의 지질대사 및 간 독성 저하에 미치는 인진쑥 및 눈꽃동충하초의 영향

        본 연구는 에탄올을 투여한 흰쥐에서 인진쑥과 눈꽃동충하초의 단독 또는 병행투여가 혈청 및 간의 지질대사와 간기능 및 항산화효소 활성에 미치는 효과를 살펴보고자 생후 4주령된 흰쥐에게 표준식이를 급여한 정상식이군, 에탄올 투여 대조군, 6%인진쑥과 6% 눈꽃동충하초를 각각 단독투여한 군, 4% 인진쑥과 2% 눈꽃동충하초 병행투여군, 3%인 진쑥과 3% 눈꽃동충하초 병행투여군, 2% 인진쑥과 4% 눈꽃동충하초 병행투여군 등 7군으로 나누고 각 실험식이로 5주간 사육하였다. 혈청 총 콜레스테롤, 중성지방 농도 및 동맥경화지수(AI)는 정상식이군에 비해 에탄올 투여 대조군에서 유의하게 증가되었으나 4% 인진쑥과 2% 눈꽃동충하초 병행투여군과 3% 인진쑥과 3% 눈꽃동충하초 병행투여군에서 대조군에 비해 유의하게 감소되는 것으로 나타났다. 에탄올 투여한 흰쥐의 간장 내 총 지질과 총 콜레스테롤 및 중성지방의 함량은 정상식이군보다 증가하였으나 인진쑥과 눈꽃동충하초의 투여로 감소하였으며 특히 4% 인진쑥과 2% 눈꽃동충하초를 병행투여한 군에서 유의하게 감소하였다. 인진쑥과 눈꽃동충하초의 투여로 ALT의 활성이 대조군에 비해 저하되었으며 6% 인진쑥 투여군과 4% 인진쑥과 2% 눈꽃동충하초 병행투여군에서는 유의하게 감소되었고, 인진쑥과 눈꽃동충하초의 단독투여와 비율을 달리한 병행투여는 catalase, SOD등 항산화효소의 활성을 증가시키는 것으로 나타났다. 과산화지질은 에탄올의 투여 시 정상식이 군에 비해 유의하게 증가되었으며 이는 6% 인진쑥 투여군과 4% 인진쑥과 2% 눈꽃동충하초 병행투여군에서 유의하게 감소되었다. 이상의 결과를 보아 인진쑥과 눈꽃동충하초의 투여는 흰쥐의 혈청과 간의 지질 대사를 개선하고, 간의 항산화효소 활성을 증가시킴과 동시에 과산화지질을 감소시키는 것으로 나타나 혈중지질 대사를 개선하고, 에탄올에 의한 지방간 및 간 경변과 같은 간 손상을 완화시키는데 효과적일 것으로 사료된다. 또한 본 연구의 결과 인진쑥과 눈꽃동충하초를 단독으로 투여하는 것보다 병행투여 하는 것이 효과적이며 인진쑥과 눈꽃동충하초의 동량투여와 4% 인 진쑥과 2% 눈꽃동충하초를 병행투여 하는 것이 효과적인 것으로 사료된다. The purpose of this study was to identify the inhibitory effect of hepatic toxicity and liver lipid metabolism after administration of Artemisia capillaris and Paecilomyces japonica. Along with the control, SD rats were divided into ethanol treated group with subgroups of 6% Artemisia capillaries (6A), 4% Artemisia capillaris+2% Paecilomyces japonica (4A2P), 3% Artemisia capillaris+3% Paecilomyces japonica (3A3P), 2% Artemisia capillaris+4% Paecilomyces japonica (2A4P) and 6% Paecilomyces japonica (6P). In this study we also intended to verify the optimum ratio of Artemisia capillaris and Paecilomyces japonica which can reduce hepatotoxicity. Artemisia capillaris and Paecilomyces japonica reduced cholesterol and triglyceride levels which were increased by ethanol. Total-cholesterol level was decreased the most in the groups of 4A2P and 3A3P. On the other hand, activity of superoxide dismutase (SOD) was enhanced significantly (p<0.05). Malondialdehyde (MDA) activity was decreased significantly (p<0.05) in the subgroup of 6A and 4A2P. When the ratio of Artemisia capillaris and Paecilomyces japonica was 2:1, the improvement of the rat serum and liver lipid metabolism and the alleviation of hepatic damage by ethanol were the most effective in this study. Therefore, it can be considered that the symptoms of severe chemically induced hepatotoxicity could be reduced by Artemisia capillaris and Paecilomyces japonica administration.

      • 인진쑥의 scopoletin함량 분석 및 뇌혈류량에 미치는 영향

        In this study, Artemisia capillaris, which has been used as oriental medicine area and folks remedy, was investigated for effective substance (scopoletin) analysis. As an attempt to develop new functional beverage by using medicinal herb, Artemisia capillaris. Therefore we investigate the effect of scopoletin in Artemisia capillaris on vessel and regional cerebral blood flow (rCBF) of rats. Artemisia capillaris extract was significantly decreased vessel constraction and significantly increased rCBF. Artemisia capillaris has often been cited in medical literature for its medicinal effects. Although the Korean Food and Drug Administration indicated Artemisia capillaris as the In-Jin-Sook which was allowed as a food stuff. From the view point of the subjects who eat In-Jin-Sook products, most of them ingested it for a certain pharmacological efficacy rather than as a beverage or a food. According to this facts, we need develop functional foods using In-Jin-Sook. Also scopoletin, get known to smooth muscle relaxation content was 326.13 ug/g. These results demonstrate that Artemisia capillaris has pharmaceutical effects and balanced nutritional contents. In according with Artemisia capillaris has a sufficiency of the terms use of food stuff in functional foods.

        사염화탄소를 투여한 흰쥐에서 인진쑥 및 동충하초의 지질대사촉진 및 간독성저하효과

        본 연구는 CCl₄를 투여한 흰쥐에서 인진쑥과 동충하초를 서로 다른 혼합비율로 투여했을 때 간 손상에 대한 생리활성 기능을 조사하기 위해 혈청 및 지질대사에 미치는 영향을 측정하였다. 사염화탄소의 투여는 간독성을 나타내 간의 무게 및 ALT, AST 수준을 증가시켰으며 이는 인진쑥과 동충하초의 투여에 의해 개선되며 인진쑥과 동충하초를 병행 투여 시 더욱 효과적인 것으로 나타났다. 사염화탄소 투여로 인해 증가된 혈청 콜레스테롤과 중성지방은 인진쑥, 동충하초의 투여로 저하되었으며 혈청 중성지방은 인진쑥과 동충하초를 동량으로 투여한 군에서 유의적으로 낮게 나타났고, 혈청 콜레스테롤은 전체 인진쑥, 동충하초 투여군에서 모두 유의적으로 감소되었으며 인진쑥과 동충하초를 병행 투여하는 경우 더 효과적인 것으로 나타났다. 또한 동맥경화지수는 사염화탄소의 투여 시 정상식이군보다 유의적으로 증가되었으나 인진쑥, 동충하초의 투여로 감소되었으며 4% 인진쑥과 2% 동충하초 병행 투여가 동맥경화지수를 낮추는데 가장 효율적인 것으로 사료된다. 사염화탄소를 투여한 흰쥐의 간장 내 총 지질과 총 콜레스테롤 및 중성지방의 함량은 정상군보다 증가하였다. 인진쑥, 동충하초의 투여는 사염화탄소 투여로 높아진 중성지방과 콜레스테롤을 감소하는 효과를 보였으며 4% 인진쑥과 2% 동충하초를 병행 투여 시 가장 효과적인 것으로 나타났다. 본 연구 결과에 의하면 인진쑥과 동충하초의 투여는 사염화탄소로 유발된 간 독성을 저하시키며 혈청과 간지질 대사를 개선하는 효과가 있는 것으로 사료되며 간 기능 개선과 지질대사 개선효과를 가지고 있는 인진쑥과 동충하초를 단독으로 투여하는 것보다 병행 투여하는 것이 더 효과적이며 그 혼합비율이 4% 인진쑥과 2% 동충하초에서 가장 효율적으로 나타났으나 혼합비율에 대해서는 앞으로 지속적인 연구가 필요하다고 사료된다. When the ratio of Artemisia capillaris and Paecilomyces japonica was 2:1, the improvement of rat serum and liver lipid metabolism and the alleviation of hepatic damage induced by CCl₄ were shown to be the most effective in this study. It is considered that the symptoms of severe chemically induced hepatotoxicity could be lessened by Artemisia capillaris and Paecilomyces japonica administration.

      • SPME법을 이용한 인진쑥(Artemisia capillaris)의 향기성분 분석

        This study was performed to analyze volatile flavor compounds in Artemisia capillaris. Artemisia capillaris is a perennial plant of the Compositae family and is widely distributed in the world. This plant is characterized by fresh minty and herbaceous flavor. The volatile flavor compounds, isolated from dried Artemisia capillaris using solid phase microextraction(SPME) with three types of fibers, were analyzed by GC/MSD with INNOWAX column. Identification of volatile flavor compounds was based on the linear retention indices(RIs) and the comparison of their mass spectra with those of on-computer library. Forty six compounds, comprising 22 hydrocarbons(84.94ppm), 12 alcohols(68.20ppm), 3 esters(12.53ppm), 6 ketones(104.88ppm), 1 aldehyde(1.71ppm), and 2 miscellaneous ones(143.34ppm), were tentatively identified by the PDMS fiber. For the CAR/PDMS fiber, sixty-four compounds, comprising 25 hydrocarbons(77.02ppm), 15 alcohols(50.23ppm), 3 esters(8.70ppm), 10 ketones(66.97ppm), 4 aldehydes(4.88ppm), and 7 miscellaneous ones(86.86ppm), were identified. Fifty compounds, comprising 21 hydrocarbons (88.28ppm), 14 alcohols(83.26ppm), 2 esters(12.25ppm), 8 ketones(105.29ppm), 2 aldehydes(1.96ppm), and 3 miscellaneous ones(96.95ppm), were tentatively identified by the CW/DVB fiber.

        The solid-state fermentation of <i>Artemisia capillaris</i> leaves with <i>Ganoderma lucidum</i> enhances the anti-inflammatory effects in a model of atopic dermatitis

        <P><I>Artemisia capillaris</I>, which belongs to the Asteraceae family and the genus <I>Artemisia</I>, has been reported to exert inhibitory effects on diabetes, cancer and inflammation. In this study, in order to enhance the bioactivity potential of the leaves of <I>Artemisia</I> by <I>Ganoderma lucidum</I> mycelium, we prepared aqueous samples of <I>Artemisia capillaris</I> (<I>Ac</I>) leaves, <I>Ganoderma lucidum (Gl</I>) and aqueous fractions produced by the solid fermentation of <I>Ganoderma lucidum</I> on <I>Artemisia capillaris</I> leaves (<I>afAc/Gl</I>). Thereafter, we evaluated whether these samples have potential to attenuate inflammation-related symptoms in an amimal model of 2,4-dinitrofluorobenzene (DNFB)-induced atopic dermatitis. We found that <I>afAc/Gl</I> exhibited enhanced anti-inflamamatory activity following the solid fermentation process when compared with <I>Ac</I> or <I>Gl</I> on ear thickness, ear epidermal thickness and eosinophil infiltration in the skin tissues. The expression of nitric oxide (NO) synthases (NOSs) was measured by immunohistochemical staining. The results revealed that <I>afAc/Gl</I> decreased endothelial NOS and inducible NOS expression compared with the DNFB group, while neuronal NOS expression was not altered. By comparing NO production, we found that as opposed to <I>Ac, afAc/Gl</I> has potential to inhibit atopic dermatitis-related symptoms during the inflammatory event. As regards matrix metalloproteinase (MMP) expression patterns, <I>afAc/Gl</I> exerted potent inhibitory activity on the mRNA expression of MMP-2, -7, -9, -12, -14 and -19. Taken together, these results suggest that the solid state fermentation of <I>Ac</I> by <I>Gl</I> is an effective strategy to obtaining useful ingredients which are converted into valuable compounds during an atopic inflammatory insult.</P>

        사철쑥과 그늘쑥의 다당류 연구

        Two polysaccharides, ACP-UMF and ACP-ULF, were purified from the aerial part of Artemisia capillaris by anion-exchange chromatography, ultrafiltration, and gel filtration chromatorgraphy. The polysaccharides appeared to be homogenious from the results of HPLC. The molecular weights of ACP-UMF and ACP-ULF were estimated to be 16305.92 D and 3292.23 D, respectively, by MALDI-TOF MS. The sugar compositions were determined by GC to be arabinose 10.05%, xylose 1.67%, mannose 5.45%, galactose 39.06%, glucose 15.43% for ACP-UMF and arabinose 11.60%, xylose 11.15%, mannose 6.37%, galactose 32.47%, glucose 18.35% for ACP-ULF. A polysaccharide, SP-M, was also purified from Artemisia sylvatica in the same manner as in A. capillaris. The molecular weight of SP-M was determined to be 2462.52 D by MALDI-TOF MS. SP-M consisted mainly of rhamnose 36.49%, arabinose 29.00%, and glucose 19.38%. Incubation of CCl4-intoxicated hepatocytes with ACP-UMF reduced the levels of glutamic pyruvic transaminase(GPT) and cellular malondialdehyde(MDA) to 62.8% and 23.8%. ACP-ULF also reduced the levels of GPT and MDA to 46.1% and 38.1% and SP-M to 38.8% and 26.3%, respectively.

        Discrimination between Artemisia princeps and Artemisia capillaris Based on Near Infrared Spectroscopy Combined Multivariate Analysis

        The Artemisia princeps (Compositae) has been used in traditional Korean medicine for the treatment of microbial infections and inflammatory diseases. Since A. princeps is generally difficult to be discriminated from A. capillaris, A. caplillaris has been misused in place of A. princeps. To solve this problem, a rapid and nondestructive method for discrimination of A. princeps and A. capillaris samples was developed using near infrared spectroscopy (NIRS) in the present study. A principal component analysis (PCA) and a partial least squares discrimination analysis (PLS-DA) were performed to discriminate two species. As a result, with the use of PLS-DA, A. princeps and A. capillaris were clustered according to their genus. These outcomes indicated that the NIRS could be useful for the discrimination between Artemisia princeps and Artemisia capillaris.
