        주정 중독 흰쥐에서 총담관 결찰이 간의 β-D-Glucuronidase활성에 미치는 영향

        The activities of the liver and serum β-D-glucuronidase were studied when cholestasis was induced by common bile duct ligation in rats and chronic ethanol intoxication developed, or cholestasis following acute ethanol intoxication for manifestation of the biochemical background of alcohol intoxication in hepatobiliary disease. When common bile duct was ligated in the rats, lysosomal and microsomal β-D-glucuronidase activities in the cholestatic liver and activity of β-D-glucuronidase in the serum showed a considerable increase. When common bile duct was ligated after chronic ethanol intoxication, the activities of liver lysosomal β-D-glucuronidase decreased more significantly than in the group with common bile duct ligation, while the activities of liver microsomal β-D-glucuronidase and serum β-D-glucuronidase increased more significantly than in the group with common bile duct ligation performed. The activities of liver lysosomal and microsomal β-D-glucuronidase and serum β-D-glucuronidase in the group with 14th day after common bile duct ligation increased markedly, and the same was seen in tile group with acute ethanol intoxication after common bile duct ligation. At the 24th hour following the acute intoxication with ethanol done after 140days of the common bile duct ligation, the rats showed more remarkable increase in the serum β-D-glucuronidase activity than the group only with the 14th day after the common bile duct ligation. In summary, the liver lysosomal β-D-glucuronidase seem to be an enzyme which decreases activities in chronic ethanol intoxication with cholestasis more than in cholestasis. And the liver microsomal β-D-glucuronidase seem to be an enzyme which increases activities in chronic ethanol intoxication with cholestasis more than in cholestasis. The causes of the increase or the decrease seems to be the development or the retardmrnt of biosynthesis. Especially, when the acute and chronic ethanol intoxication with cholestasis occurred, the sera β-D-glucuronidase are higher than in cholestasis because of increased liver cell damage, which causes the enzyme to leak into the blood in great quantity. Accordingly, these results will be the data supporting alcoholic drink is enzymologically harmful in hepatobiliary disease.

      • 골관절염과 류마치스성 관절염 환자를 대상으로 한 β-glucuronidase 활성도의 분석

        목적 : 골관절염과 류마치스성 관절염에서 질병에 따른 체액내에서 β-glucuronidase의 활성도에 차이가 있다는 것을 규명하여 질병의 진단 및 연구에 이용하고자 함이 이 논문의 목적이다. 방법 : 골관절염, 류마치스성 관절염 환자에서 채취한 소변, 혈청, 관절액에서 β-glucuronidase 활성도를 표준화된 방법에 의해 측정하여 표준곡선으로 환산한 후 다음의 결과를 얻었다. 결과 : 1) 정상대조군 성인에서의 소변 및 혈청에서 β-glucuronidase 활성도는 0.09u/l. 0,075u/l였다. 2)골관절염 환자의 소변, 혈청, 관절액의 β-glucuronidase 활성도는 0.09u/l, 0.14u/l, 0.06u/l로서 혈청에서의 활성도가 가장 높았다. 3) 류마치스성 관절염 환자의 소변, 혈청, 관절액의 β-glucuronidase 활성도는 0.19u/l, 0.17u/l, 0.07u/l로서 소변에서의 활성도가 가장 높았다. 4) 두 질병에 있어서 소변, 혈청, 관절액의 β-glucuronidase 활성도는 남자에 있어서 여자보다 높았고, 류마치스성 관절염에서 골관절염에 비하여 효소 활성도가 높았다. 5) 두 질병에 있어서의 소변, 혈청, 관절액의 β-glucuronidase 활성도는 연령에 따라서의 변화는 의미가 없었고, 소변에서의 활성도 변화는 남자 P=0.0041. 여자 P=0.0001로 현저히 의미있는 차이가 있었다. 결론 : 위의 결과에 따르면 골관절염과 류마치스성 관절염이 각각 β-glucuronidase의 활성도 변화에 영향을 미침을 알 수 있으며 그 외 다른 정형외과적 질병과 검체에 따라서도 β-glucuronidase 활성도에 차이가 있을 수 있음을 결론지을 수 있다. Objectives : In ostroarthritis and rheumatoid arthritis, β-glucuronidase activity was measured in body fluid to diagnose this special disease entities. Methods : The β-glucuronidase activity was studied in the urine, serum and joint fluid. The level of this specimens were estimated by statistical sections. Results : 1) The β-glucuronidase activity in urine, serum and joint fluid of osteoarthritis patients were 0.09U/l urine, 0.14U/l serum, 0.06U/l joint fluid. The highest level of activity was in the serum. 2) The β-glucuronidase activity on rheumatoid arthritis was 0.19U/l urine, 0.17U/l serum, 0.07U/l joint fluid. The highest increased level of activity was in the urine. 3) In the joint fluid and serum, there is the no mean of stistical analysis. But in the urine, there is marked meaning of statistical analysis(Male : p=0.0041, Female : p=0.0001). Conclusions : β-glucuronidase activity was influenced by osteoarthritis and rheumatoid arthritis. Especially in the urine, β-glucuronidase activity was outstanding statistical meaning. According to disease entities, The level of activity was different from each specimen.

        담즙울체 쥐에서 간 라이소솜의 α-D-Glucosidase와 β-D-Glucuronidase 활성도에 미치는 Taurocholate의 정맥 내 투여의 영향

        이 연구는 담즙울체간에서 라이소솜의 α-D-glucosidase와 β-D-glucuronidase의 활성도 증가 기전의 일부를 알아내기 위하여 쥐에게 총담관 결찰로 담관을 폐쇄시킨 직후 TCA 또는 TUDCA를 정맥 내에 주입시킨 다음 경시적으로 간 라이소솜의 α-D-glucosidase 및 β-D-glucuronidase와 혈청의 라이소솜 α-D-glucosidase, 혈청의 총 β-D-glucuronidase의 활성도를 측정하여 이 효소들의 활성도 변동에 대한 이들 담즙산의 효과를 알아 보았다. 쥐에게 총담관 결찰을 시킨 직후 TCA를 주입시켜 1일 및 2일 경과시켰을 때 간 라이소솜의 α-D-glucosidase 및 β-D-glucuronidase 활성도와 혈청의 라이소솜 α-D-glucosidase 및 혈청의 총 β-D-glucuronidase 활성도는 대조군인 총담관 결찰만 시킨 군보다 통계학적으로 유의한 증가를 나타내었다. 이상의 결과로 보아 담즙울체간에서 활성도가 증가되는 라이소솜의 α-D-glucosidase 및 β-D-glucuronidase 활성도 증가는 담즙산 중 TCA에 의해 이들 효소의 합성이 증가되어 나타난 결과로 생각되며 아울러 담즙울체시 라이소솜 α-D-glucosidase 및 총 β-D-glucuronidase의 혈중 활성도 증가는 TCA에 의한 간의 괴사로 간 세포막의 투과성이 항진되어 나타난 결과로 생각된다. The effects of intravenously administered of high concentration of taurocholic acid (TCA) on α-D-glucosidase and β-D-glucuronidase activities in rat liver lysosomes were studied. These liver lysosomal enzymes, and serum lysosomal α-D-glucosidase and total β-D-glucuronidase activities were also determined in experimental rats with common bile duct ligation (CBDL). The activities of liver lysosomal α-D-glucosidase and β-D-glucuronidase as well as the activities of serum α-D-glucosidase and total β-D-glucuronidase were found to be significantly increased in the CBDL plus TCA injected group than in the control group such as CBDL alone group. On the other hand, these serum and hepatic enzymes activities did not change in the CBDL plus tauroursodeoxycholic acid injected group. The above results suggest that TCA induces biosynthesis of the lysosomal α-D-glucosidase and β-D-glucuronidase in the liver and that the elevated lysosomal α-D -glucosidase and total β-D-glucuronidase activities in the serum are most likely due to increased hepatocyte membrane permeability caused by TCA mediated liver cell necrosis.

        김치에서 분리된 젖산균의 β-glucuronidase 활성 탐색

        전통발효식품인 김치에서 분리된 젖산균으로부터 β-glucuronidase 효소활성이 높은 균주를 선발하였다. 김치에서 분리된 156점의 젖산균 중 52점의 균주가 glucuronic acid를 탄소원으로 대사하였으며, 대부분의 젖산균은 세포내 β-glucuronidase 활성이 세포외 활성보다 유의적으로 높았다. 순무김치에서 분리된 Leu. mesenteroides KFRI 73007 균주가 0.77 ± 0.01 U/mg protein로서 가장 높은 세포내 β-glucuronidase 효소활성을 나타내었다. 최적 반응조건은 pH 7, 37℃이었으며 1.14 ± 0.01 U/mg protein의 효소활성을 나타냈다. 양이온 금속이온은 β-glucuronidase 효소활성을 약 70% 이상 저해하였으며, 균주 배양에 사용한 탄소원 중 β-glucuronidase 생산을 위한 최적의 탄소원은 glucuronic acid이었다. Lactic acid bacteria (LAB) isolated from kimchi were studied for their β-glucuronidase activity. Among the 156 strains tested, 52 strains utilized glucuronic acid as a carbon source and their intracellular β-glucuronidase activities were significantly higher than their extracellular activities. Leuconostoc mesenteroides KFRI 73007 isolated from turnip kimchi exhibited the highest intracellular β-glucuronidase activity of 0.77 ± 0.01 U/mg protein, which was further increased to 1.14 ± 0.01 U/mg protein under optimized reaction conditions (pH 7, 37℃). The activity of β-glucuronidase was notably decreased by the addition of divalent cations, and glucuronic acid was the best carbon source to produce β-glucuronidase in Leu. mesenteroides KFRI 73007.

      • β-Glucuronidase-Inhibitory Activity and Hepatoprotective Effect of Ganoderma lucidum

        To prove the relationship between the fluctuation in serum β-glucuronidase level and hepatoprotective activity was investigated. The ether fraction of G.lucidum, which had potent β-glucuronidase-inhibitory activity, protected against CCI₄-induced liver injury. From this ether fraction, ganoderenic acid A, was isolated as the potent inhibitor of β-glucuronidase. It had a potent hepatoprotective effect against CCI₄-induced liver injury. These results suggest that the glucuronidase seems to be closely related to liver injury, which could be prevented by glucuronidase inhibitors.

        비병원성 야생효모 Candida oleophila WP5-19-1로부터 장내유용세균 유해 효소 β-glucuronidase 저해제의 생산 및 특성

        We aimed to produce a potent β-glucuronidase inhibitor from wild yeast that could inactivate toxic substances in the colon. Culture supernatants and cell-free extracts of non-pathogenic wild yeasts were prepared and their β-glucuronidase inhibitory activities were measured. Cellfree extract from Candida oleophila WP5-19-1 showed the highest β-glucuronidase inhibitory activity (49.0%). The β-glucuronidase inhibitor was maximally produced (IC50 value; 8.4 mg) when C. oleophila WP5-19-1 was cultured in potato dextrose medium containing 5% dextrose (initial pH; 6.0) at 30  C for 24 hours. β-glucuronidase inhibitor of C. oleophila WP5-19-1 was partially purified by trypsin hydrolysis, ultrafiltration (3 kDa), and Sephadex G-50 filtration. The partially purified β-glucuronidase inhibitor was stable from 30℃ to 60℃ and at pH 6.0 9.0, and showed residual inhibitory activity of about 80%.

      • β-glucuronidase 효소치 활성의 변화에 있어서 흡충류 감염자 소변을 대상으로 한 분석

        The author studied the β-glucuronidase activity in the urine of 46 males and 19 females with trematodes such as Clonorchis sinensis, Stellantchasmus falcatus, metagonimus yokogawai, paragonimus westermani, Heterophyes heterophyes. Specific activity of β-glucuronidase was analyzed statistically by sex, age, intensity of infection and mixed infection. The results obtained in this studies were summarized as follows. In the 41-50 years of age groups of males, β-glucuronidase activity in the urine was elevated the highest with 1.56U/ml, 1.21U/ml of females and 1.49U/ml of male in the 30-40 years of age groups, but activity of β-glucuronidase in the urine according to age and sex was not statistical significance. It was gradually increased β-glucuronidase activity in urine of males and females according to number of mixed infection of trematodes species(p<0.01). β-glucuronidase activity in urine of males and females according to intensity of trematodes infection appeared 0.92U/ml in the simple infection, 1.14U/ml in the medium infection and 1.54U/ml in the heavy infection, respectively(p<0.01). On the basis of results, it was suggested that β-glucuronidase activity in the human urine was significantly increased in the intensity of infection and mixed infection groups than that of normal groups.

        정형외과 특정질환에 따른 β-glucuronidase의 활성에 관한 연구

        Objectives: The purpose of this study was to get help in order to diagnose orthopaedic disease, measure its activity and determine treatment plan by measuring the β-glucuronidase activity in urine, serum and joint fluid. Methods: The β-glucuronidase activity was determined in the serum, urine and joint fluid of the patients with degenerative arthritis, rheumatoid arthritis, osteomyelitis and osteogenic sarcoma, and some other disease to study the change of the enzyme activity. These values of each specimen were calculated by standard curve and treated by statistical analysis. Results: The results obtained were summarized as follows. 1. The β-glucuronidase activity in the serum, urine and joint fluid was increased in patients with degenerative arthritis, rheumatoid arthritis, osteomyelitis and osteogenic sarcoma etc. 2. The increased β-glucuronidase activity in the serum and joint fluid of each disease does not show a specific finding about respective disease, but the increased β-glucuronidase activity was statistically significant in the urine of all disease groups (male: p=0.0041, female: p=0.0001). Conclusions: On the basis of these results, it was suggested that β-glucuronidase activity was affected by the orthopaedic disease and differed according to each specimen.

      • 생약의 β-Glucuronidase 저해와 간장보호효과

        Inhibitory effect of water and ethyl acetate extract of 60 kinds of herbal medicines was investigated on β-glucuronidase. Among water extract of them, Galla Rhois had the most potent β-glucuronidase-inhibitory activity. Termaliae Fructus, Amomi Tsa-ko Fructus and Arecae Semen were also potent inhibitors. Among ethyl acetate extract of them, Galla Bhois had the most potent β-glu-curonidase-inhibitory activity. Nelumbinis Semen. Ephedrae Radix and Termaliae Fructus were also potent inhibitors. The extract of Galla Rhois had potent hepa-toprotective effect on CCl₄-induced hepatotoxicity of rats. These results suggest that the β-glucuronidase seems to be closely related to the liver injury, which could be prevented by the inhibitor of β-glucuronidase.
