        Tourette 장애 소년에서 자기공명영상을 이용한 기저핵의 용적 분석

        Objectives : The purpose of this study was to investigate the regional differences of the basal ganglia volume between the boy subjects who have Tourette's disorder and the control group who don't have this disorder by using Brain MRI. Methods : Sixteen subjects who were diagnosed as Tourette's disorder using DSM-IV criteria, and sixteen healthy subjects were selected. For analysis the coronal slice images of the basal ganglia were collected and collected MR images were analyzed with NIH Image (version 6.1 ppc). Resuls : The total brain volume in the subjects with Tourette's disorder were smaller than control group by 6.2% (F=6.2, d.f.=1, P=0.019). The right putamen volume in boys with Tourette's disorder were significant smaller than control group statistically (F=3.2, d.f.=3, P=0.040). The asymmetry (1eft>right) in the globus pallidus decreased significantly in the boys with Tourette's disorder (p=0.06) were found from this experiment. Conclusion : These findings suggest that boys with Tourette's disorder may have neuropathological abnormalities in the basal ganglia. Moreover, the present findings point out the important role of basal ganglia, especially putamen, in Patho-physiology of Tourette's disorder. More studies about the structural and functional analysis of the basal ganglia in Tourette's disorder are highly required.

      • 출아효모에서 TAR 클론닝법을 이용한 고등동물의 게놈으로부터 특정 염색체 부위의 분리

        복잡한 게놈 분석에 용이하도록 효모의 인공 염색체(YAC) 클론닝 시스템은 발달되어왔다. YAC은 박테리아의 인공 염색체(BAC)보다 더 큰 단편을 클론닝할 수 있고, 또한 클론된 단편을 쉽게 변형시킬 수 있다. 형질전환과 연계된 재조합법(Transformation-Associated Recombination; TAR)은 게놈 라이브러리를 만들지 않고 직접 게놈 DNA로부터 분리하고자 하는 유전자나 특정 염색체 부분을 클론닝 할 수 있는 방법이다. 이 방법은 spheroplast transformation을 수행하는 동안, 목적으로 하는 유전자의 5' 그리고 3' 염기 배열(hooks)을 지닌 TAR 벡터와 게놈 DNA 사이에서 일어나는 상동성 재조합에 의해 이루어진다. 효모 내의 in vivo 재조합을 이용한 TAR 클론닝법은 복잡한 게놈으로부터 목적의 염색체 부분을 원형 YAC의 형태로서 선택적으로 분리할 수 있다. 그러므로 TAR 클론닝 법은 특정 염색체로부터 YACs을 만드는데 매우 유용하여, 전체 게놈으로부터 특이적 유전자나 유전자의 family를 분리하는데 효과적인 방법으로 사료된다. Yeast artificial chromosome (YAC) cloning systems have advanced the analysis of complex genomes considerably. They permit the cloning of larger fragments than do bacterial artificial chromosome systems, and the cloned material is more easily modified. Transformation-associated recombination (TAR) is a cloning technique that allows specific chromosomal regions or genes to be isolated directly from genomic DNA without prior construction of a genomic library. This technique involves homologous recombination during spheroplast transformation between genomic DNA and a TAR vector that has 5' and 3' gene targeting sequences (hooks). Using in vivo recombination in yeast, TAR cloning selectively isolates, as circular YACs, desired chromosome segments or entire genes from complex genomes. We propose that TAR cloning can provide an efficient means for generating YACs from specific chromosomes and that TAR cloning may be useful for isolating families of genes and specific genes from total genome DNA.

      • 食用油脂類에 對한 免疫生物學的 硏究

        食用油脂는 健康維持에 必須的인 營養素이다. 그러나 食用油脂는 脂肪을 構成하는 脂肪酸의 種類와 攝取하는 量에 따라 健康을 保衛하는 必須的인 食品의 成分이 되기도 하고 有害한 影響을 끼칠 수도 있어서 脂質代謝와 關連된 疾病 뿐만아니라 老化를 促進하고, 乳房癌, 大腸癌等의 癌發生과도 關連된 것으로 밝혀져 있다. 本 硏究는 우리生活環境에서 食用으로 接할 수 있는 10種의 油脂에 對한 免疫生物學的인 活性에서 究明한 結果 對照群인 참기름食餌群에 比해 올리브유食用群은 全般的으로 體液性 및 細胞性免疫을 有意性있게 增加시켰으나, 大食細胞의 活性과 末梢循環白血球數는 有意性없는 減少를 보였고, 動物性 油脂인 牛脂食餌群과 植物性 油脂인 옥수수기름食餌群, 들기름食餌群, 米糠油食餌群, 大豆油食餌群, 菜種油食餌群 및 고추씨기름食餌群에서는 全般的으로 體液性 및 細胞性免疫, 大食細胞의 活性 및 末梢循環白血球數를 有意性있게 減少시켰다. This study was performed to investigate the effects of edible oils and fats on the immunobiological responses in ICR male mice. Ten groups of experimental diets, such as sesame oil diet, beef tallow diet, lard oil diet, olive oil diet, corn oil diet, perilla oil diet, rice polishing oil diet, soybeen oil diet, rape seed oil diet, and red pepper seed oil diet were fed adlibitum to the ICR male mice for 27 days. The results of this study were summarized as followings. 1. Both humoral and cellular immune responses, phagocyte activity, and number of leukocytes in sesame oil group were increased. However, the increasing rate of body weight and the weight retios of spleen and thymus to body were generally decreased. 2. Hemagglutination titer(HA) and rosette forming cell(RFC) of the beef tallow group and the lard group were significantly lower than the sesame oil group(P<0.01), but the lard group significantly increased Arthus reaction and delayed type hypersensitivity(DTH)(P<0.01). 3. Arthus reaction, DTH, and RFC of the olive oil group were significantly higher than the sesame oil group(P<0.01). 4. DTH of the corn oil group was significantly higher than the sesame oil group(P<0.01) whereas phagochyte activity was significantly lower(P<0.01). But no statistically significant differences were found between the two groups in the humoral immune response. 5. HA, PFC, DTH, RFC, phagocyte activity, and number of leukocytes of the perilla oil group and the rice polishing oil group were significantly lower than the sesame oil group(P<0.01). 6. Both humoral and cellular immune responses, phagocyte activity, and number of leukocytes of the soybeen oil group, the rape seed oil group, and the red pepper oil group were significantly lower than the sesame oil group(P<0.01).

      • Behavior of Rural Ozone in East Asia: Case Study of Matsumoto, Japan

        With the ever-increasing severity of the urban air pollution problem there is an urgent need to clearly understand the factors governing the formation and transport of photochemical ozone in the big cities of Korea. Many studies have shown that the interaction between the urban air and the surrounding rural air plays a critical role in the temporal behavior of urban ozone. However, since there is a lack of any rural ozone monitoring in Korea it is very difficult to ascertain the impact of rural ozone influx on the concentrations of ozone in the big city. Therefore, characteristics of rural ozone behavior is studied using a site in Japan and compared with the big city situation in Korea. In addition, the relative effect of photochemistry on rural ozone behavior is analyzed using various parameters and compared to the impact of photochemistry for urban precursor concentrations. From the comparisons it is very clear that in order for such policies as the ozone warning system or the air pollution forecast system to operate properly the present air pollutant monitoring system is quite inadequate and many sites in rural areas surrounding the big cities must be carefully selected to aid in understanding the behavior of ozone in the Greater Seoul Area and other urban centers.

      • Potential Environmental Applications for Multi-Purpose Korean Satellites

        최근에 부각되고 있는 환경 문제들, 특히 대기오염 문제들은 그 공간적 시간적 규모가 너무 커서 지표면에서의 측정만 해서는 충분한 연구가 어려운 실정이다. 그러므로, 지구 또는 지역 규모에서 일어나는 환경 현상들을 자세히 분석하기 위해서는 위성 측정이 불가피하다. 한국의 위성 사업은 이제 초기 단계다. 또, 여러 “환경파괴 사건들”과 누적된 자연자원의 파괴로 인해 환경에 대한 인식이 고조된 상태이다. 따라서, 앞으로 추진될 다목적 위성 사업을 환경 연구와 접목시키는 것은 시기 적절하다고 사료된다. 아울러 현재 선진국에서 진행되고 있는 또는 앞으로 수행될 위성을 사용한 환경사업을 조사하고 동아시아의 대기환경 문제에 위성 자료를 적용한 사례를 제시한다. 그리고, 한국의 다목적 위성에 적합하다고 생각되는 환경 사업을 건의하고 이를 수행하는 데 있어서 극복해야 될 몇 가지 문제점들에 대해 고찰해 본다. The spatial and temporal scales of environmental problems, especially for those related to the atmosphere, make it a very arduous task to adequately study using only surface-based observations. Therefore, the utilization of satellite monitoring for global- or regional-scale analyses of environmental phenomena is rapidly becoming a prerequisite for comprehensive environmental projects. Korea is in the initial stages of her satellite program. Coincidentally, environmental awareness has also recently peaked due to several severe ramifications (i.e., pollution “disasters”) of rapid industrialization and the cumulative neglect of our natural resources. The time is apropos for a look into potential environmental applications of our multi-purpose satellites. This paper reviews the current status of environmental studies being carried out by or those that are being planned for satellite programs in the developed countries. A case study of satellite usage for an environmental application in the East Asian region is introduced. Finally, initial suggestions are made regarding potential environmental projects that seem appropriate for Korea multi-purpose satellites and a discussion is included regarding difficulties and barriers that must be overcome for the endeavor to be a success.

      • 폐기물 적정처리와 양질의 음용수 공급을 위한 환경정책

        Regarding solidwaste disposal, collective complaints by residents who are living nearby solidwaste land - fill sites existing or to he existing are raised very often, Major causes for the collective complaints are both poorly constructed facilities, the poor manangement practices of the facilities and the intensified phenomenon of "not in my backyard". In Pusan city and Kyung-sang-nam-do region, among accross the country, the collective complaints were frequently raised recently and the construction of the landfill -sites, especially those for industrial solid wastes, have been extremely difficult, thereby resulting in misdisposal of solid wastes. To break through this situation Pusan Regional Environment Office has imposed from 1990 a very strict guideline for the construction of all new industrial solid waste land -fill sites, thereby minimizing possible damage in the future to nearby residents and enviromnent. For instance the ground of the land - fill sites should be leveled-off with clay or sand of 10 centimeters or above, and covered over it with a shock-protect polyester of 2mm and High Density Polyethylene (HDPE) sheet of 1.5mm or thicker. In addition soft material such as coal ash of at least 10 centimeters should be covered over HDPE sheet before landfill work started in order t o prevent damages of the HDPE sheets which can result in infiltration of wastewater into ground. There have been also a general recognition that there are plenty of room for the improvement of current solidwaste management system. To meet this recognition the Ministry of Environment has recently noticed are visional draft of the So1idwaste Management Law. Recently general public's apprehension for the quality of drinking water has been heightened. To meet the demand for the provision of safe drinking water the Government has planned to improve purifying faci1ities of intaken water and distribution systems. In addition the Government has established a schedule to construct or launch to construct sewage treatment plants at 84 sites accross the country by 1996, thereby increasing treatment rate up to 40% from 25% as of 1989. Nightsoil treatment plants will be constructed at 78 sitess which will raise nights oil treatment rate up to 100 perceant from 89% as of 1987. Livestock waste water treatment facilities has been in stalled for medium-size live stock farms. The enforcement of wastewater discharge standards through compliance checks of industrial wastewater discharge behavior will be stregthened. All of these efforts are exppected to contribute to provide cleaner drinking water in Korea, where most of the drinking water sources are in-land fresh water.
