      • Histone deacetylase 3 is selectively involved in L3MBTL2-mediated transcriptional repression

        <P><B>Abstract</B></P><P>This is the first report that L(3)mbt-like 2 (L3MBTL2) specifically interacts with the histone deacetylase domain of histone deacetylase 3 (HDAC3) via its MBT domain. Here, we show that L3MBTL2 selectively interacts with HDAC3, but not other class I HDACs. An in vitro peptide-binding assay demonstrated the specific association of HDAC3 with methylated histone-K20 tail and L3MBTL2. Furthermore, depletion of HDAC3 resulted in a decrease of methylated K20-H4, as well as an increase in acetylated histone H3. Consequently, HDAC3 knock-down selectively suppressed L3MBTL2-mediated transcriptional repression.         Southward shift of the Intertropical Convergence Zone due to Northern Hemisphere cooling at the Oligocene-Miocene boundary

        <P>The Mi-1 glaciation (ca. 23 Ma), which marks the Oligocene-Miocene boundary, was an aberrant cooling event that led to a build-up of the Antarctic ice sheet, which reached the near-modern volume (or greater) from its ephemeral or partial existence. An increase of ∼1‰ in the δ<SUP>18</SUP>O of benthic foraminifera during this interval has been attributed to the development of Antarctic ice sheets and deep-water cooling. Without definitive evidence, Northern Hemisphere (NH) glaciation has not been a material consideration for the δ<SUP>18</SUP>O increase. Here we investigate the interhemispheric temperature contrast during Mi-1, with the movement of the Intertropical Convergence Zone (ITCZ) at a site (10°31′N) in the East Pacific (Integrated Ocean Drilling Program Site U1333), to understand NH cooling and the possibility of NH glaciation. The measured <SUP>143</SUP>Nd/<SUP>144</SUP>Nd, <SUP>87</SUP>Sr/<SUP>86</SUP>Sr, and clay mineral compositions of eolian dust fractions indicate unequivocally the deposition of Asian dust during Mi-1, and of Central American and South American dust before and after Mi-1. This is attributed to the southward displacement of the ITCZ over Site U1333 during Mi-1. The ITCZ shifts toward the warmer hemisphere. Thus our results suggest that the cooling during Mi-1 was more significant in the NH than in the Southern Hemisphere, which underwent a sudden expansion of continental ice sheets. Our data call for a forcing mechanism to drive significant NH cooling during this episode. Based on the available data, we propose that the widespread growth of NH ice sheets and/or changes in the production of North Atlantic–origin deep water could be possible causes of the NH cooling at that time.</P>

        <P><B>Abstract</B></P><P>Calmodulin-regulated protein phosphorylation plays a pivotal role in amplifying and diversifying the action of calcium ion. In this study, we identified a calmodulin-binding receptor-like protein kinase (CBRLK1) that was classified into an S-locus RLK family. The plasma membrane localization was determined by the localization of CBRLK1 tagged with a green fluorescence protein. Calmodulin bound specifically to a Ca<SUP>2+</SUP>-dependent calmodulin binding domain in the C-terminus of CBRLK1. The bacterially expressed CBRLK1 kinase domain could autophosphorylate and phosphorylates general kinase substrates, such as myelin basic proteins.         조혈모세포이식 환자에서 발생한 Cytomegalovirus 질환의 특징 : 일개 대학변원에서 최근 10년간의 경험

        Background : Studies on cytomegalovirus (CMV) diseases in Korean hematopoietic stem cell transplant (HSCT) recipients are lacking and do not reflect the recent trends of advances and changes. Therefore, we tried to analyze the clinical features of CMV diseases in HSCT recipients over the past 10 years at a tertiary university hospital in Korea. Methods : Retrospective review of medical records was done for all adult HSCT patients who received transplant at the Catholic HSCT Center from January 1998 to January 2008. Results : Forty-four cases (2.2%) of CMV diseases were identified. CMV pneumonia was diagnosed in 17 patients, retinitis in 16 patients, enterocolitis in 7 patients, esophagitis 1 patient, gastritis in 1 patient, duodenitis in 1 patient, and hepatitis in 1 patient. The median onset of symptom was 90 days after transplantation. Late CMV diseases accounted for 47.7%. CMV related death varied from 0 to 58.8% according to the involved organ. CMV retinitis was diagnosed relatively later in the course of transplantation mostly in patients who had chronic graft versus host disease (GVHD). On the contrary, CMV enterocolitis mainly occurred in patients who suffered from acute GVHD. The overall concurrent CMV reactivation was documented to be 63.6%: the concurrent CMV reactivation was observed only in 37.5% of patients with retinitis. Conclusions : We observed some differences in the pattern of CMV disease manifestation according to the involved organ and reconfirmed the fact that CMV pneumonia is the most common and fatal disease in HSCT recipients. Additionally, CMV retinitis was not uncommon in HSCT recipients. Since specific marker does not exist in predicting retinitis, regular ocular examination should be done thoroughly, especially in patients with chronic GVHD.

        조혈모세포이식 환자에서 침습성 진균 감염에 대한 Micafungin의 예방 효과 및 안전성

        Background: Micafungin, a potent inhibitor of 1,3-β-D-glucan synthase, is a novel antifungal agent of the echinocandin class. In vitro study showed that micafungin was effective against Aspergillus species as well as Candida species, but clinical data on the prophylactic efficacy against invasive fungal infections (IFIs) other than candidiasis are still lacking. Materials and Methods: We identified 60 consecutive adult hematopoietic stem cell transplantation (HSCT) recipients who received at least 3 doses of micafungin during neutropenic period. Micafungin was started as an alternative in patients who were intolerant or had adverse events (AEs) to primary prophylactic antifungal agents. We retrospectively reviewed the medical records and analyzed the efficacy and safety of micafungin for prophylaxis against IFIs. Results: The patients either had autologous (n=9) or allogeneic (n=51: 1 syngeneic, 24 sibling, 26 unrelated donor) HSCT. Itraconazole oral solution (n=58) was the most frequently used first line antifungal agent for prophylaxis and was administered for median 11 days. The most frequent cause of switch to micafungin was vomiting (n=42). The duration of neutropenia and micafungin administration was median 13 and 12 days, respectively. A successful outcome was achieved in 45 (75%) patients. Empirical antifungal therapy was initiated in 13 (22%) patients. There were 2 cases (3.3%) of breakthrough fungal infections which comprised a probable invasive pulmonary aspergillosis and a possible invasive fungal sinusitis. There was no case of invasive candidiasis. A total of 53 (88%) patients experienced at least one AE regardless of causality during micafungin administration. The most frequent AEs were hypokalemia, vomiting, diarrhea, and elevated serum aspartate aminotransferase or alanine aminotransferase. Among the aforementioned AEs, only 1 case of diarrhea could be classified as a probable relation with micafungin when causality was assessed. There was no AEs that caused discontinuation of micafungin. Conclusions: Micafungin seems to be a safe and effective agent for prophylaxis of IFIs including aspergillosis as well as candidiasis in HSCT recipients. However, further large, prospective, and randomized comparative studies are warranted for aspergillosis.

      • Purge & Trap-GC를 이용한 의약품 필름코팅 정제 중 잔류용제에 관한 연구

        의약품은 약물을 생체에 적풋하기 위하여 유효성분의 효과가 언제나 일정하게 확보되고 사응에 편리하도록 만들어지는 것이므로 유효썽분 이외에 약효에 영향을 주지 않는 성분이 첨가되는 경운가 많다. 이 때 사용되는 용매들은 제피의 광택 및 건쪼시간의 단축 등을 위하여 휘발점이 낮을 용매들이 주로 사용되어진다. 본 연구는 의약품 필름코팅정제 중 잔류용매 4종(chlorofonr benzen, trichloro ethylen, 1,4-dioxane)에 대한 변형된 pirge & trap-GC 장치를 이용한 동시분석방법을 개발하였으며, 각 표준품의 RSD 값은 chloroform 3.03%, benzen 3.17%, trichloroethylen 3.69% and 1,4-dioxane 3.41%였다. 또한 시중 유통중인 의약품 50종에 대하여 잔류웅매 양을 측정하였으며, 검출되는 잔류용매는 한 건도 없었다. This study nras carried out to develope the analytical method for the mixture of chlorefonn, benzen, trichloroethylen and 1,4-dioxane simultaneously and determine the remainingorgauic solvents in coating tablets by Purge & Trap-GC. The results were as follouFs ; 1. Chloroform, benzen, trio:tloroethylen and 1,4-dioxane separated by tenax #5 trap by HP-624GC column by terrlperature programming. The peaks were separated completely at retentiontime of 6.88min for chloroform, 8.21min for benzen, 10.38miu for trichloroethylen and 11.95minfor 1,4-dioxane. 2. Standard RSD were individually chloroform 3.03%, benzen 3.17%, trichloroethylen 3.69%and 1,4-diorane 3.41%. 3. 60 samples were not detrcted chloroform, benzen, trichloroethylen and 1,4-dioxane.

        Plant regeneration from callus of Iris odaesanensis Y. N. Lee native to Korea via organogenesis

        Iris odaesanensis Y. N. Lee. is an important endangered and native plant belonging to the family Iridaceae in Korea. This study describes a method for rapid micropropagation of this species via from leaf, rhizome and root explants derived calli. Leaf, rhizome and root explants were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (2,4-D) for callus induction. Rhizome explants yielded calli at a frequency of 72% when cultured at 1.0 mg/l 2,4-D. Calli were maintained at 1.0 mg/l 2,4-D. These calli were transferred to MS medium supplemented with 0, 0.5, 1.0, and 2.0 mg/l 2,4-D in combination with 0, 0.5, 1.0, and 3.0 mg/l BA for adventitious shoot induction. The highest number of adventitious shoot (228.9 per petri-dish) were formed at 1.0 mg/l 2,4-D and 1.0 mg/l BA. WPM medium was the best to convert calli into plantlets, where up to 98.2% of calli were regenerated into plantlets. This in vitro propagation protocol should be useful for conservation of this endangered plant.

      • 부산 지역 미용실의 실내공기 오염

        In order to grasp how the indoor air levels affect on job satisfaction of beauty salon workers, we analyzed air environment levels of 30 beauty shops in Pusan and surveyed questionnaires of 130 persons who have worked there. The indoor temperature, relative humidity, discomfort index, and affective temperature were in comfort zone. Most measured values in the afternoon were higher than in the morning. Carbon dioxide concentration(1,115.6±766.7ppm even for A.M.) and monoxide(13.0±58.78ppm only for P.M.) were higher than safety standards(1,000 and 10 ppm, respectively). The indoor dry bulb temperature was positively correlated with discomfort index, globe and effective temperature(r=0.961, 0.665, and 0.957, respectively), while air velocity negatively with carbon di- mono-oxide and suspended dust quantity(r=0.416, -0.276, and -0.372, respectively). Backward stepwise regression equation showed that job satisfaction was affected by indoor air levels and environment recognition: Job satisfaction = 106.101 - 1.451(Environment recognition) + 4.137(Job responsibility) + 3.051(Job recommendation) - 4.008(Job renunciation) + 12.329(Relative humidity in the afternoon) + 12.378(Noise in the afternoon). From these results we conclude that the indoor physical environment plays a significant role in job satisfaction of beauty salon workers.

        Background: High-sensitivity C-reactive protein (hsCRP), the classic acute-phase reactant, is an extremely sensitive systemic marker of inflammation. A variety of factors seems to influence hsCRP levels. The aim of this study was to elucidate the gender differences in the association between hsCRP and features of the metabolic syndrome (MS) among Korean. Methods: The study population included 1,283 men and 3,265 women aged 40 years and over who were enrolled in Korean Health Examinee Cohort. They were examined in 10 General hospitals from November 2004 to September 2005. The height, weight, waist circumference, and systolic and diastolic blood pressure of the subjects were examined and on concentrations of fasting blood glucose, total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride, and hsCRP in peripheral venous blood were measured. Results: In both sexes, there were significant positive correlations between age, smoking, body mass index, waist circumference, systolic and diastolic blood pressure, triglyceride, and fasting glucose and hsCRP, whereas significant negative correlations were present between drinking, and HDL cholesterol and hsCRP. Mean concentrations of hsCRP in men with 0, 1, 2, 3, and ≥4 components of MS were 0.13, 0.16, 0.18, 0.20, and 0.22 mg/L, respectively. The respective mean concentrations of hsCRP for women were 0.08, 0.09, 0.12, 0.14 and 0.14 mg/L. Results of stratified analyses by the number of components of the metabolic syndrome of 0, 1, 2, 3, and ≥4 revealed that an increase in hsCRP concentrations was greater in men than women with an increased number of components of the MS. Conclusion: There was a difference in concentrations of hsCRP between men and women, and especially an increase in hsCRP concentrations associated with risk factor-clusteringof MS was more prnounced in men.

      • 항결핵 치료과정에서 호전되었다가 악화된 뇌 결핵종 1예 보고

        In spite of nationmide tuberculosis control program, tuberculosis is still prevalent in Korea. Accordingly, tuberculous meningitis also poses one of the main trouble some problems. Recently, We experienced a perplexing case of tuberculous menin-gitis and tubercdoma in a 24-year old female patient. She was admitted to our hospital owing to meningeal irritation sign, aphasia, VI nerve palsy, and ,fever. CSF analysis showed a finding consistent with that of presumptive tuberculous meningitis.. Initial MRI of the brain revealed a large mass(tuhercu1oma) on left parieto-temporal area. Antitubeculous regimen(1soniazid (INH), rifampin(RMP), ethambutol(EMB), pyrazinamide(PZA)} of meningeal dose was given immediately, and her symptoms and signs were dramatically disappeared with a few days after treatment. She was discharged with improved condition and under maintenance treatment of tuberculosis. Follow-up one year after treatment. revealed that the previous lesion was nearly diminished. She showed no symptom at all until she recognized a total blindness of sudden onset on her left eye a t the 18th month of treatment. Brain MRI checked at that time showed multiple masses of tuberculoma scattered over the brain base and one of them invaded left optic nerve. In our opinion, the cause of relapse in this patient might he explained by the selection and ultimate replication of a minor population of resistant strain. Although such relapse is an extremely rare event, we think that we should always pay attention to this possible resistance problem every time we meet patients with tuberculous meningitis.
