        The role of FGF-2 in smoke-induced emphysema and the therapeutic potential of recombinant FGF-2 in patients with COPD

        <▼1><P>Although the positive effects of recombinant fibroblast growth factor-2 (rFGF-2) in chronic obstructive pulmonary disease (COPD) have been implicated in previous studies, knowledge of its role in COPD remains limited. The mechanism of FGF2 in a COPD mouse model and the therapeutic potential of rFGF-2 were investigated in COPD. The mechanism and protective effects of rFGF-2 were evaluated in cigarette smoke-exposed or elastase-induced COPD animal models. Inflammation was assessed in alveolar cells and lung tissues from mice. FGF-2 was decreased in the lungs of cigarette smoke-exposed mice. Intranasal use of rFGF-2 significantly reduced macrophage-dominant inflammation and alveolar destruction in the lungs. In the elastase-induced emphysema model, rFGF-2 improved regeneration of the lungs. In humans, plasma FGF-2 was decreased significantly in COPD compared with normal subjects (10 subjects, <I>P</I> <I>=</I> 0.037). The safety and efficacy of inhaled rFGF-2 use was examined in COPD patients, along with changes in respiratory symptoms and pulmonary function. A 2-week treatment with inhaled rFGF-2 in COPD (<I>n</I> = 6) resulted in significantly improved respiratory symptoms compared with baseline levels (<I>P</I> <I><</I> 0.05); however, the results were not significant compared with the placebo. The pulmonary function test results of COPD improved numerically compared with those in the placebo, but the difference was not statistically significant. No serious adverse events occurred during treatment with inhaled rFGF-2. The loss of FGF-2 production is an important mechanism in the development of COPD. Inhaling rFGF-2 may be a new therapeutic option for patients with COPD because rFGF-2 decreases inflammation in lungs exposed to cigarette smoke.</P></▼1><▼2><P><B>Lung disease: Inhaling a protein might help</B></P><P>Studies on the role of the protein ‘fibroblast growth factor-2’ (FGF-2) in chronic obstructive pulmonary disease (COPD) suggest that inhaled FGF-2 could help treat the emphysema linked to smoking. Researchers in South Korea led by Young-Koo Jee at Dankook University, Cheonan, and Yeon-Mok Oh at the University of Ulsan, Seoul, studied the role of the reduced FGF-2 levels found in mice with lung inflammation caused by exposure to cigarette smoke. They also uncovered details of a protective effect of inhaled FGF-2, identifying specific cellular and lung structure changes attributed to the administered FGF-2. Reduced FGF-2 levels were also found in patients with COPD. Initial trials revealed some improvement in patients treated with FGF-2, but not at a statistically significant level. Nevertheless, the authors suggest their results justify further investigation of the protein’s therapeutic potential.</P></▼2>

      • 국내 5개 종합병원에서 분리된 주요 원내감염 세균에 대한 Cefepime의 시험관내 항균력

        목적 : Enterobacter, Citrobacter, Serratia, A. baumannii 및 P. aeruginosa 등의 원내 감염균은 AmpC β-lactamase 과량 생산으로 인하여 여러 가지 β-lactamase 항균제에 내성인 균주가 많다. Cefepime은 제 4세대 cephalosporin으로 AmpC β-lactamase 과량 생산균에 대한 항균력이 우수하고, oxacillin 감수성 포도구균과 penicillin 내성 S. pneumoniae에도 항균력 있는 것으로 알려져 있다. 그러나 국내분리균주에서 cefepime의 항균력은 시험된 바 적다. 본 연구에서는 우리나라 병원에서 분리되는 원내감염 세균에 대한 cefepime의 항균력을 다른 광범위 β-lactamase 항균제의 항균력과 비교하고자 하였다. 방법 : 1999년 전국 5개 병원에서 분리한 원내 감염세균에 대한 항균제 감수성을 각각의 병원에서 Etest strip (AB Biodisk, Solna, Sweden)으로 시험하였다. 시험 균주는 균종별로 약 10주씩이었고, 대상균종은 E. coli, Klebsiella spp., C. freundii, Enterobacter spp., Serratia spp., Indole-positive Proteeae, Acinetobacter spp., P. aeruginosa, oxacillin-suspectible S. aureus, and oxacillin-suspectible coagulase-negative Staphylococcus이었다. 시험 항균제는 cefepime, cefpirome, ceftazidime, ceftriaxone, imipenem, oxacillin 그리고 piperacillin-tazobactam이었다. 결과 : 총 407주 그람음성 간균에서 cefepime의 MIC_(90)는 E. coli에 대하여 0.75㎍/mL, Klebsiella spp. 12㎍/mL, C. freundii 2㎍/mL 및 E. cloacae 48㎍/mL로서 imipenem을 제외한 다른 시험항균제의 MIC_(90)보다 낮았다. 전체 균종의 감수성율은 imipenem 89%, cefepime 78%, cefpirome 74%, ceftazidime 66%, piperacillin-tazobactam 65% 및 ceftriaxone 51%이었다. Oxacilin 감수성 포도구균의 감수성율은 cefepime, cefpirome 및 imipenem에 대해 100%이었다. 결론 : Cefepime의 원내감염 세균에 대한 시험관내 항균력은 imipenem을 제외한 다른 광범위 항균제보다 우수하여, 이 항균제는 원내감염증 환자의 치료에 유용할 것으로 판단되었다. Background : Common nosocomial pathogens such as Pseudomonas aeruginosa, Acinetobacter baumannii, and species of Enterobacter, Citrobacter, and Serratia, are often resistant to various β-lactam antibiotics by Amp C β-lactamase hyperproduction. Cefepime, a fourth generation cephalosporin, has been reported to have higher activity against AmpC β-lactamase-producing strains. It is also reported to be active against oxacillin-susceptible S. aureus, and penicillin-resistant S. pneumoniae. However, few studies were reported with Korean isolates. The purpose of this study was to compare the in vitro activities of cefepime and other broad-spectrum β-lactam antibiotics against nosocomial pathogens isolated in Korean Hospitals. Methods: In 1999, each of five participating hospitals in Korea tested the antimicrobial susceptibility for nosocomial pathogens using Etest strip. The number of isolates tested were about ten per species, which were E. coli, Klebsiella spp., C. freundii, Enterobacter spp., Serratia spp., Indole-positive Proteeae, Acinetobacter spp., P. aeruginosa, oxacillin-susceptible S. aureus, and oxacillin-susceptible coagulase-negative Staphylococcus. Antimicrobials used were cefepime, cefpirome, ceftazidime, ceftriaxone, imipenem, oxacillin and piperacillin-tazobactam. Results: A total of 407 isolates were tested. The MIC_(90s) of cefepime were 0.75 ㎍/mL for E. coli, 12 ㎍/mL for Klebsiella spp., 2 ㎍/mL for C. freundii, and 48 ㎍/mL for Enterobacter spp., which were generally lower than those of other antibiotics except imipenem. The following were the susceptibility rates of gram-negative bacilli to various antimicrobials, respectively: imipenem, 89%; cefepime, 78%; cefpirome, 74%; ceftazidime, 66%; piperacillin-tazobactam, 65% and ceftriaxone, 51%. To cefepime, cefpirome and imipenem, 100% of oxacillin-susceptible S. aureus were susceptible. Conclusion: The higher in vitro activities of cefepime than other broad-spectrum β-lactam antibiotics except imipenem suggests cefepime is useful for the treatment of patients with nosocomial infections.

        Workspace Visibility Graph Analysis (VGA) for Concentration Privacy and Group Relations in the Open-Plan Office Environment

        The present study explored the applicability of Visibility Graph Analysis (VGA) techniques to workplace design research. Six types of VGA measures in Depthmap encompassing visual connectivity, three types of visual integration, mean depth, and visual entropy were employed for the analysis of individual privacy for task concentration and group relationship behavior in the open-plan office environment. Data comprised 136 workers in 6 open-plan offices filled with low-paneled (1.2-1.5m) cubicle workspaces. For the statistical analysis, Spearman’s rho correlations and t-tests were applied for the spatial and behavioral measures. The results showed that workspace VGA measures have a potential to be useful information to account for workers’ concentration privacy and, limitedly, also informal relationships with team members. Visual entropy values especially offer reliable information to predict various aspects of office workers’ privacy behavior while visual integration can be used to account for the workers’ sense of trust in group relations. The study also discussed the limitation of VGA applications to the workplace context.

        외상후 스트레스장애에 대한 하타 스타일 요가 치료의 해외 임상연구 동향

        Objectives: The purpose of this study was to review the research trends in the treatment of hatha-style yoga on Post Traumatic Stress Disorder (PTSD). Methods: We searched articles in Pubmed and the China National Knowledge Infrastructure (CNKI) January 2010-December 2019, for studies to treat PTSD using hatha-style yoga. Selected studies were evaluated by the CLEAR-NPT (A Checklist to Evaluate a Report of a Non-pharmacological Trial). Results: Seven randomized controlled clinical trials were selected. PSS-I (PTSD Symptom Scale-Interview) was the most frequently used as diagnostic criteria. The PCL (PTSD Checklist) was also the most commonly used outcome measurement. Of the seven articles, most studies reported that hatha-style yoga was effective to reduce symptoms of PTSD. Conclusions: Hatha-style yoga practice intervention can be used to relieve symptoms of PTSD. More studies should be conducted to make hatha-style yoga as protocol (complementary therapy) for PTSD patients.

        Pheochromocytoma is usually associated with a combination of various clinical manifestations caused by the overproduction of catecholamines. It is frequently accompanied by impaired glucose tolerance operating through 2 - and - adrenergic mechanisms. A 41-year-old-woman was admitted to the hospital because of poorly-controlled diabetes mellitus and hypertension. She had suffered intermittent paroxysmal attacks of headache and chest discomfort and had been treated intermittently over a 2 year period for diabetes mellitus and hypertension. At admission, the levels of serum epinephrine, norepinephrine urinary excretion of total metanephrine, and VMA were all abnormally elevated. Adrenal CT showed a well-defined, homogenous mass in the right adrenal region and the tumor was diagnosed as pheochromocytoma. After tumor resection, the increased blood level of catecholamines, the urinary excretion of total metanephrine, and VMA were normalized, as was the hyperglycemia state. Diabetes mellitus of the patient was considered permanently resolved after tumor removal by the result of glucose tolerance in 75 g oral glucose tolerance test

      • 벌꿀제품에 있어서 잔류항생물질에 관한 조사연구

        꿀벌의 무저병을 치료하기 위하여 테트라싸이클린 계열의 항생물질(TCs)이 널리 사용되고 있으며,이러한 사용은 벌꿀중 Cs의 잔류문제를 일으킬 수 있다. 따라서 벌꿀중 잔류하는 TCs의 분석법이 팔요하며, 본 연구에서는 벌꿀클의 옥시테트라싸이클린(OTC), 테트라싸이클린fC), 클로르테트라싸이 클린(CTC), 독시싸이클린(DC)끌 분석하기 위해 두가지 HPLC방법을 사용하였다. 아카시아꿀, 잡화꿀,토종꿀, 방꿀등 4가지 종류의 댈꿀에 대한 회수율 및 검출한계를 측정하였다. 형광검출기를 이웅한 분석법에서는 0.Sppm의 수준에온 회수율은 OTC는 91.4~98.2%, TC는 84.B~94.4%, CTC는 뚬.I~105.9%, DC는 뽀.5~져.5%이었으며, 검출한계는 OTC와 TC는 0.01ppf CTC차 OC는 0.05ppm이었다. 자외선검출기를 이용한 분석법에퍼는 0.Sfpm의 수준에서 회수율은 OTC는 78.3~97.0%, TC는 76.8~86.8%, CTC는 89.6~99.5%, Df는 91.I~93.8%이었으며, 검출한계는 OTC와 TC는 0,01ppm, CTC와 DC는 0.02ppm이었다. 두 ITPLC법으로 국내산 벌꿀 140건과 외국산 벌꿀 29건을 조사한 결과 OTC,TC, CTC, BC 모두 검출되지 않았다. ln order to prevent foul brood of honey-bees, tetracycline antibiotics(TCs) are widely used in honey-bee culture. Such usage may resu)t in residues of TCs in honey.Therefore, the establishment of a method for residue analysis of TCs in honey is required.Two HPLC methods for oxytetracycline(OTC), tetracycline(TC), chlortetracycline(CTC) anddoxycycline(DC) in honey have been established. The recoverles of OTC, TC, CTC and BCfrom 4 kinds of honelr(acacia honey, mixed flower honey, native honey, chestnut honey) spikedat a level of 0.Sppm by HPLC with fluorescence detection are 91.4 ~98.2%, 84.B~94.4%, 85.1 ~105.9%, 86.5 ~94.5% refpectively. The limits of detection in honey b.# HPLC with fluorescencedetection are 0.01ppm for OTC and TC, and 0.05ppm for CTC and DC respectively. And therecoveries of OTC, TC, CTC and DC from 4 kinds of honey spiked at a level of 0,Sppm byHPLC with UV detection are 78.3 ~97.0%, 76,8 ~86.8%, 89.6 ~99.5%, 91,1 ~93,8% respectively.The Ihuts of detection in honey by HPLC with UV detection are 0.01ppm for OTC and TC,and 0.02ppm for CTC and DC respectively The residual TCs(OTC, TC, CTC, DC) were notde7ected in the 169 samples of honey(domestic honey 140, imported honey 29) by two HPLCmethods.
