The present study was aimed to scrutinise the preventative role of broccoli sprouts powder (BSP) rich in glucoraphanin, mustard seed powder (MSP) with high activity myrosinase, and mustard and broccoli mixture powder (MBP) on bisphenol-A-induced obese in C57BL/6J mice. Study included six different groups of male C57BL/6J mice: control, bisphenol-A (BPA), garcinia cambogia+bisphenol-A (GAR+BPA), mustard seed powder+bisphenol-A (MSP+BPA), broccoli sprout powder+bisphenol-A (BSP+BPA) and mustard and broccoli mixture powder + bisphenol-A (MBP+BPA). Results demonstrated that MBP-treated groups of mice showed reduced body weights and adipose tissue weight through the downregulation of peroxisome proliferator-activated receptor γ (PPARγ), CCAAT/enhancer-binding protein (C/EBPα), and fatty acid-binding protein (aP2) and fatty acid synthase (FAS). Moreover MBP activated phosphorylation of AMPK and ACC and upregulated the protein expression of carnitine palmitoyltransferase-1 (CPT-1) in bisphenol-A-induced obese mice. These results suggested that MBP may be a potentially effective therapeutic agent for bisphenol-A-induced lipid metabolism disorders.

Environmental endocrine disruptors, including bisphenol-A, have been recently involved in obesity by dysregulating adipose tissue function. This study investigated the effects of BPA on adipocyte differentiation and the effects of the broccoli sprouts powder extract rich in glucoraphanin, mustard seed powder extract with high activity myrosinase, and mustard and broccoli mixture powder (MBP) as a functional food ingredient on obesogen-induced lipid production and adipogenesis in 3T3-L1 cells. The results presented BPA increased lipid accumulation and transcription of the peroxisome proliferator-activated receptor γ (PPARγ), CCAAT-enhancer-binding protein-alpha (C/EBPα), fatty acid-binding protein (aP2), fatty acid synthase (FAS). Which also inhibits the expression of carnitine palmitoyltransferase-1 (CPT-1), and uncoupling protein-1 (UCP-1) lipolysis-related genes. However, MBP suppressed the BPA-induced effects on adipogenesis and inhibits the protein expression of fatty acid synthase gene protein through AMP-activated protein kinase. These results suggested that MBP may be a potentially effective therapeutic agent for obesity and its associated complications.

Glucoraphanin can be hydrolyzed into sulforaphane with biological activity under the action of myrosinase. Sulforaphane has strong anti-cancer activity, however, the effect and the mechanism of anti-obesity activity are not yet clear. In this study, we evaluated the anti-obesity effects of broccoli sprouts powder rich in glucoraphanin, mustard seed powder with high activity myrosinase, and mustard and broccoli mixture powder (MBP) on 3T3-L1 adipocytes, respectively. The results showed that MBP can significantly inhibited the expression of adipogenic genes and increased the expression of genes related to lipolysis and lipid metabolism. MBP inhibits the protein expression of adipogenic transcription factor peroxisome proliferator-activated receptor γ (PPARγ), CCAAT/enhancer-binding protein (C/EBPα), and fatty acid-binding protein (aP2) in 3T3-L1 adipocytes, and inhibits the protein expression of fatty acid synthase gene protein through AMP-activated protein kinase. Which also increases the expression of carnitine palmitoyltransferase-1 (CPT-1) lipolysis-related genes. These results suggest that MBP may have anti-obesity effects on adipogenesis and lipid metabolism.
Properties of a Bacteriocin Produced by Bacillus subtilis EMD4 Isolated from Ganjang (Soy Sauce)

A Bacillus species, EMD4, with strong antibacterial activity was isolated from ganjang (soy sauce) and identified as B. subtilis. B. subtilis EMD4 strongly inhibited the growth of B. cereus ATCC14579 and B. thuringiensis ATCC33679. The antibacterial activity was stable at pH 3-9 but inactive at pH 10 and above. The activity was fully retained after 15 min at 80°C but reduced by 50% after 15 min at 90°C. The activity was completely destroyed by proteinase K and protease treatment, indicating its proteinaceous nature. The bacteriocin (BacEMD4) was partially purified from culture supernatant by ammonium sulfate precipitation, and QSepharose and Sephadex G-50 column chromatographies. The specific activity was increased from 769.2 AU/mg protein to 8,347.8 AU/mg protein and the final yield was 12.6%. The size of BacEMD4 was determined to be 3.5 kDa by Tricine SDS-PAGE. The N-terminal amino acid sequence was similar with that of Subtilosin A. Nucleotide sequencing of the cloned gene confirmed that BacEMD4 was Subtilosin A. BacEMD4 showed bactericidal activity against B. cereus ATCC14579.
Compound Heterozygous Mutations in the DUOX2/DUOXA2 Genes Cause Congenital Hypothyroidism

The mutations in the dual oxidase 2 (DUOX2) and dual oxidase maturation factor 2 (DUOXA2) genes can cause congenital hypothyroidism(CH). This study reports the pedigree with goitrous congenital hypothyroidism (GCH) due to the coexistence of heterozygousmutations in the DUOX2 and DUOXA2 genes. The two sisters with GCH were diagnosed with CH at neonatal screeningand were enrolled in this study. The DUOX2, DUOXA2, and thyroid peroxidase (TPO) genes were considered for genetic defects screening. Family members of the patients and normal controls were also enrolled and evaluated. The two girls harbored compound heterozygous mutations, including a new mutation of c.2654G>T (p.R885L) in the maternal DUOX2 allele and c.738C>G (p.Y246X) in the paternal DUOXA2 allele, that has been previously reported. The germline mutations from the families were consistent with an autosomal recessive inheritance pattern. No mutations in the TPO gene and the controls were observed.

In this paper, the autolysis process of Saccharomyces cerevisiae FX-2 (S. cerevisiae FX-2) via, a variety of endogenous enzyme, was investigated systematically by analyzing changes in physicochemical parameters in autolysate, surface morphology and the internal structure of the yeast cells. As an explicit conclusion, the arisen autolysis depended on the pH and the optimal pH was found to be 5.5. Based on the experimental data and the characteristics of mycelia morphology, a hypothesis is put forward that simple proteins in yeast vacuolar are firstly degraded for utilization, and then more membrane- bound proteins are hydrolyzed to release hydrolytic enzymes, which arouse an enzymatic reaction to induce the collapse of the cell wall into the cytoplasm.
