Molecular characterization of chemosensory protein genes in Bactrocera minax (Diptera: Tephritidae)

Chemoreception is of great importance for survival of insects. Chemosensory proteins (CSPs) are believed to be involved in the perireceptor events of chemosensory system in many insect species, but this has not been clarified in the citrus fruit fly, Bactrocera minax, a serious dipteran pest of citrus crops. Previous studies indicated that four CSP genes were identified in many fly speices in Diptera. In the present study, we also identified four CSP genes in B. minax,namely BminCSP1–4, from a transcriptome database. All CSP proteins encoded by these genes bear the typical hallmarks of the CSP family: an N-terminal signal peptide and the four highly conserved cysteine residues. Phylogenetic analysis comparing with other dipteran CSPs indicated that dipteran CSPs may evolved from three ancestral CSP genes, and revealed the sequence diversities of BminCSPs and showed that BminCSP1–4 are clustered in separate groups, indicating the possibility of their contrasting function in B. minax. Furthermore, the tissue distribution of the four CSP genes in adult B. minax was analyzed by real-time quantitiative PCR. The results demonstrated that BminCSP3 are significantly transcriptionally enriched in antennae; BminCSP4 expressed primarily in heads; BminCSP1 and BminCSP2 showed high expressions in different tissues, such as antennae, abdomens and wings. Based on these findings, the different implications for the functions of BminCSPs are discussed. This study will offer a significant indication for further functional studies of the CSPs in B. minax
Surface Modification with Pluronic P123 Enhances Transfection Efficiency of PAMAM Dendrimer

To improve gene delivery efficiency and decrease cytotoxicity of polyamidoamine (PAMAM) polymers,P123-g-PAMAM was synthesized by modifying PAMAM with pluronic P123. The structure of the synthesized polymers was analyzed using proton nuclear magnetic resonance. The polymers were able to self-assemble with DNA,forming nanometer-scale complexes. Particle size measurement confirmed that the mean diameter of the polyplexes was 100-250 nm. The cytotoxicity and transfer efficiency were measured and compared with those of PEI and PAMAM. All of the polyplexes showed significantly low cytotoxicity in the MCF-7, HepG2, and 293T cell lines. In addition, the low level of P123 grafting to PAMAM showed significantly higher transfection efficiency than unmodified PAMAM at the optimal N/P ratio. These results suggest that P123-ylated PAMAM may prove as a useful carrier for gene delivery.
Investigation on MRR and Machining Gap of Micro Reciprocated Wire-EDM for SKD11

In this paper, the investigational work is carried out on the micro reciprocated wire electrical discharge machining (wire-EDM) of SKD11 using circularly travelled wire with Φ30 μm as tool electrode, aiming to achieve the fabrication of micro parts with high cost-effect, efficiency and precision. The experiments are systematically performed using central composite design approach of response surface methodology to study the effect of open voltage, discharge capacitance, pulse duration and feed rate on material removal rate (MRR) and machining gap. The mathematical models of MRR and machining gap are developed by analysis of variance, and whilst verified by confirmation experiments with ≤ 8.38% and 6.70% prediction errors for MRR and machining gap, respectively. Besides, multi-objective optimization of parameter combinations is attempted by non-dominated sorting genetic algorithm-II. Finally, a typical experimental sample, micro gear is successfully machined with 1194.38 μm tip diameter under the condition of No. 14 optimal solution.

A new and efficient technique is described for the production of diacylglycerols (DAGs) by the esterification of oleic acid with glycerol catalyzed by diatomite-loaded SO4 2/TiO2. DAGs show some potential health benefits compared to triacylglycerols, and also can be used to produce the novel industrial plasticizer epoxy fatty acid methyl ester in material science. Diatomite-loaded SO4 2/TiO2 catalyst was prepared and characterized, and the selected conditions for the synthesis of DAGs were determined to be: reaction time = 6.0 h, temperature = 210 C, catalyst loading = 0.1% of the oleic acid weight, and mass ratio of oleic acid to glycerol = 2:1. Under these conditions, DAGs yield reached 59.6% with a purity of 69.6% after a one-stage molecular distillation. Diatomite-loaded SO4 2/TiO2 as a solid catalyst could be recycled and reused with high catalytic efficiency. Under the same conditions, diatomite-loaded SO4 2/TiO2 showed a better catalytic performance than the commercial solid acid SO4 2/ZrO2-Al2O3. Based on this, a two-step reaction method for the production of DAGs was performed and provided a yield similar to the one-step method (58.3% vs. 59.6%), but with a shorter reaction time (4 h vs. 6 h). It is concluded that a two-step reaction method could be a better alternative to the one-step production of DAGs in the presence of diatomite-loaded SO4 2/TiO2.

A series of dimethine cyanine dyes were synthesized in a fast, efficient and high yield by the condensation of quaternary salts with 1H-indole-3-carbaldehyde in the presence of piperidine under solvent-free microwave irradiation. The products were identified by 1H NMR, IR, UV-Vis spectra and elemental analysis. The absorption and fluorescence properties of these dyes were investigated both experimentally and theoretically. Calculations performed at a combination of time-dependent density functional theory (TD-DFT) and the polarizable continuum model (PCM) reproduced the p-p* type absorption bands of the dyes. Regression analysis was used for studying theoretical results of the absorption maxima in different solvents. Compared with experimental counterparts, estimated overall uncertainties in the absorption maxima were about ?2%.
Construction and Expression of an Eukaryotic Expression Vector Containing the IER3 Gene

Background: More and more research indicate that the immediately early response gene 3 (IER3) is involved inmany biological provesses, such as apoptosis and immunoreaction, as well as viral infection, tumorigenesis and tumour progression. Methods: Here we describe the construction of an eukaryotic expression vector containing IER3 gene and its expression in A549 cells as assessed through fluorescence microscopyand Western-blotting. Results: Fluorescence detection displayed that GFP in cytoplasm was high during 48 and 72 hours post-transfection. In addition, Western blotting showed significant increase in IER3 gene expression in the transfected cells compared with controls. Conclusion: The recombinate plasmid expression vector was constructed successfully, which may provide a basis for further exploration of function of IER3 in lung cancer.

The present study aimed to investigate the function of translationally controlled tumor protein (TCTP) in the regulation of Oct4 in mouse embryonic carcinoma P19 cells and mouse J1 embryonic stem (ES) cells. The mRNA level of endogenous TCTP in somatic cells was 2-4 folds higher than that in pluripotent P19 and J1 ES cells. Overexpression of TCTP in mouse pluripotent cells not only reduced the level of Oct4 transcription, but also decreased the pluripotency of stem cells. The N-terminal end of TCTP (amino acids 1-60) played an important role in suppressing the Oct4 promoter. Moreover, overexpression of TCTP in P19 cells suppressed the Oct4 promoter activity in a dose- and a time-dependent manner. In addition, knockdown of TCTP by small interfering RNA increased the expression of Oct4. Our study indicates that TCTP downregulates the Oct4 expression by binding the Sf1 site of Oct4 promoter in mouse pluripotent cells. [BMB reports 2012; 45(1): 20-25]
