CIDE domains form functionally important higher-order assemblies for DNA fragmentation

<P>Cell death-inducing DFF45-like effector (CIDE) domains, initially identified in apoptotic nucleases, form a family with diverse functions ranging from cell death to lipid homeostasis. Here we show that the CIDE domains of Drosophila and human apoptotic nucleases Drep2, Drep4, and DFF40 all form head-to-tail helical filaments. Opposing positively and negatively charged interfaces mediate the helical structures, and mutations on these surfaces abolish nuclease activation for apoptotic DNA fragmentation. Conserved filamentous structures are observed in CIDE family members involved in lipid homeostasis, and mutations on the charged interfaces compromise lipid droplet fusion, suggesting that CIDE domains represent a scaffold for higher-order assembly in DNA fragmentation and other biological processes such as lipid homeostasis.</P>

Background: Outcomes of postoperative radiotherapy(PORT) for squamous cell carcinoma of the tonsil were analyzed focusing on management and control of the neck.Methods: Medical records of 380 pathologically-confirmed tonsil cancer patients, treated with surgery and PORT from February 1993 to January 2011 at 15 institutions, were reviewed. Neck metastasis was absent(pN0) in 53(13.9%) and present(pN+) in 327(86.1%), of which 279(85.3%), 2(0.6%), and 46(14.1%) were ipsilateral, contralateral, and bilateral, respectively. Results: Neck irradiation(NI) was done in 346(91.1%) patients, while data was unavailable in 34(8.9%). After a median follow-up of 53 months, neck recurrence occurred in 28(7.4%) patients. Among the 53 pN0 patients, NI was ipsilateral-only, bilateral, and unknown in 25(47.2%), 21(39.6%), and 7(13.2%). One patient suffered from contralateral neck failure after an ipsilateral-only NI. For the 279 ipsilateral pN+ patients, NI was ipsilateral-only, bilateral, and unknown in 67(24.0%), 193(69.2%), and 19(6.8%). Neck recurrence occurred in 18(6.5%): 11(3.9%), 6(2.2%) and 1(0.4%) failed in the ipsilateral-only, contralateral-only, and bilateral neck. The 2 contralateral pN+ patients received bilateral NI and remained free of regional recurrence. NI data was available for 38(82.6%) of the 46 bilateral pN+ patients and all received bilateral NI. Three(6.5%), 2(4.3%) and 2(4.3%) patients recurred in the ipsilateral, contralateral, and bilateral neck. Conclusions: Because contralateral neck failure is rare in pN0 patients, elective NI may safely be confi ned to the ipsilateral neck-only. For pN+ patients, however, the risk of contralateral neck failure highly varies (3 to 10%). Thus clinicopathologic factors should be considered to justify the omission of contralateral NI for such heterogenous groups.
Design and Implementation of the RF Systems for Bi-directional Wireless Capsule Endoscopes

This paper explains that the RF systems for hi-directional wireless capsule endoscopes were designed and implemented. The designed RF systems for a capsule endoscope can transmit the images of intestines from the inside to the outside of a body and the behavior of the capsules can be controlled by an external controller simultaneously. The hi-directional wireless capsule endoscope consists of a CMOS image sensor, FPGA, LED, battery, DC to DC Converter, transmitter, receiver, and antennas. The transmitter and receiver which were used in the hi-directional capsule endoscope, were designed and fabricated with $10mm(diameter){\times}3.2mm(thickness)$ dimensions taking into the MPE, power consumption, system size, signal to noise ratio and modulation method. The RF systems designed and implemented for the hi-directional wireless capsule endoscopes system were verified by in-vivo experiments. As a result, the RF systems for the hi-directional wireless capsule endoscopes satisfied the design specifications.
A band-rejection type RF switch based on a dual-mode microstrip ring resonator

<P>A band-rejection type RF switch, which has very high off-state isolation, low on-state insertion loss, and adjustable bandwidth, is proposed using a dual-mode ring resonator and two PIN diodes. To enable sharp band stop and dual-mode transmission, the ring resonator is directly fed using nonorthogonal feeding. Two PIN diodes are mounted in shunt after a quarter-wavelength transmission line from each feeding point, to turn the switch on and off and to reduce the size of the ring resonator. To estimate the practicable transmission characteristics of the proposed switch in relation to the angle of feeding and the wire-bonded PIN diodes, a lossy transmission-line equivalent circuit model is proposed, and even–odd mode analysis is carried out. The calculated frequency response shows good agreement with the measurement. The fabricated switch exhibits off-state isolation of 43.08 dB, and on-state insertion loss of 0.74 dB, within the range of 2.298–2.401 GHz. © 2010 Wiley Periodicals, Inc. Radiotherapy would be the choice of treatment for human cancers, because of high cost-effectiveness. However, a certain population of patients shows a resistance to radiotherapy and recurrence. In an effort to increase the efficacy of radiotherapy, many efforts were driven to find the genes causing the unresponsiveness to ionizing radiation. In this paper, we compared the gene expression profiles of two lung cancer cell lines, H460 and H1299, which showed differential responses to ionizing radiations. Each cell were irradiated at 2 Gy, and harvested after 0, 2, 4, 8, 12 and 24 hours to examine the expressions. Two-way ANOVA analysis on time-series experiments of two cells could select 2863 genes differentially expressed upon ionizing radiation among 32,321 genes in microarray (p<0.05). We classified these genes into 21 clusters by SOM clustering according to the interaction between cell types and time. Two SOM clusters were enriched with apoptosis-related genes in pathway analysis. One cluster contained higher levels of phosphatidyl inositol 3-phosphate kinase (PI3K) subunits in H1299, radio-resistant cells than H460, radiosensitive cells. TRAIL receptors were expressed in H460 cells while the decoy receptor for TRAIL was expressed in H1299 cells. From these results, we could characterize the differential responsiveness to ionizing radiation according to their differential expressions of apoptosis-related genes, which might be the candidates to increase the power of radiotherapy.
