        Genome-wide analyses reveal the extent of opportunistic STAT5 binding that does not yield transcriptional activation of neighboring genes

        <P>Signal Transducers and Activators of Transcription (STAT) 5A/B regulate cytokine-inducible genes upon binding to GAS motifs. It is not known what percentage of genes with GAS motifs bind to and are regulated by STAT5. Moreover, it is not clear whether genome-wide STAT5 binding is modulated by its concentration. To clarify these issues we established genome-wide STAT5 binding upon growth hormone (GH) stimulation of wild-type (WT) mouse embryonic fibroblasts (MEFs) and MEFs overexpressing STAT5A more than 20-fold. Upon GH stimulation, 23 827 and 111 939 STAT5A binding sites were detected in WT and STAT5A overexpressing MEFs, respectively. 13 278 and 71 561 peaks contained at least one GAS motif. 1586 and 8613 binding sites were located within 2.5 kb of promoter sequences, respectively. Stringent filtering revealed 78 genes in which the promoter/upstream region (−10 kb to +0.5 kb) was recognized by STAT5 both in WT and STAT5 overexpressing MEFs and 347 genes that bound STAT5 only in overexpressing cells. Genome-wide expression analyses identified that the majority of STAT5-bound genes was not under GH control. Up to 40% of STAT5-bound genes were not expressed. For the first time we demonstrate the magnitude of opportunistic genomic STAT5 binding that does not translate into transcriptional activation of neighboring genes.</P>

        Successful Treatment of Psychiatric Symptoms after Traumatic Brain Injury Using Deep Brain Stimulation to the Anterior Limb of Internal Capsule-Nucleus Accumbens

        Psychiatric symptoms are common after traumatic brain injury (TBI), and some patients have poor drug therapeutic efficacy. We report a successfully treated case of psychiatric symptoms after TBI using deep brain stimulation (DBS) to the anterior limb of internal capsule (ALIC)-nucleus accumbens (NAc) in a 76-year-old woman. The patient suffered from auditory hallucination, mood changes, and insomnia caused by TBI. Psychological test assessment showed the scores of Hamilton Anxiety Scale, Hamilton Depression Scale and Positive and Negative Syndrome Scale were 30, 35, and 96 respectively. Head magnetic resonance imaging scan showed right temporal lobe encephalomalacia. Head magnetic resonance spectroscopy (MRS) showed bilateral basal ganglia choline increased relatively. After DBS to the ALIC-NAc, the target parameters were adjusted. The psychiatric symptoms were completely improved and the result of head MRS was normal in the end. The current report declares that DBS is reversible, adjustable and safe in the treatment of psychiatric symptoms caused by TBI. DBS to the ALIC-NAc should be considered as a possible treatment choice once a patient showed psychiatric symptoms after TBI.

        Influence of K2TiF6 in electrolyte on characteristics of the microarc oxidation coating on aluminum alloy

        Black and gray microarc oxidation (MAO) coatings were prepared in a phosphate electrolyte with and without K2TiF6 on 2A70 aluminum alloy, respectively. Voltageetime curves were recorded during the MAO process. The effects of K2TiF6 on the morphology, composition, abrasive resistance and corrosion resistance of MAO coatings were investigated. The results showed that the MAO coating produced in the electrolyte with K2TiF6 was thicker, and more uniform than that produced in the electrolyte without K2TiF6. Ti was detected in the surface of the MAO coating formed in the electrolyte with K2TiF6. The results of abrasive resistance and corrosion resistance showed that the MAO coating formed in the electrolyte with K2TiF6 exhibited better abrasive resistance and corrosion resistance.

        DMRUT-MCDS: Discovery Relationships in the Cyber-Physical Integrated Network

        In recent years, we have seen a proliferation of mobilenetwork- enabled smart objects, such as smart-phones and smartwatches, that form a cyber-physical integrated network to connect the cyber and physical worlds through the capabilities of sensing, communicating, and computing. Discovery of the relationship between smart objects is a critical and nontrivial task in cyber-physical integrated network applications. Aiming to find the most stable relationship in the heterogeneous and dynamic cyber-physical network, we propose a distributed and efficient relationship-discovery algorithm, called dynamically maximizing remaining unchanged time with minimum connected dominant set (DMRUT-MCDS) for constructing a backbone with the smallest scale infrastructure. In our proposed algorithm, the impact of the duration of the relationship is considered in order to balance the size and sustain time of the infrastructure. The performance of our algorithm is studied through extensive simulations and the results show that DMRUT-MCDS performs well in different distribution networks.

      • The transcription factors signal transducer and activator of transcription 5A (STAT5A) and STAT5B negatively regulate cell proliferation through the activation of cyclin‐dependent kinase inhibitor 2b (<i>Cdkn2b</i>) and <i>Cdkn1a</i> expression

        <P><B>Abstract</B></P><P>Although the cytokine‐inducible transcription factor signal transducer and activator of transcription 5 (STAT5) promotes proliferation of a wide range of cell types, there are cell‐specific and context‐specific cases in which loss of STAT5 results in enhanced cell proliferation. Here, we report that loss of STAT5 from mouse embryonic fibroblasts (MEFs) leads to enhanced proliferation, which was linked to reduced levels of the cell cycle inhibitors p15<SUP>INK4B</SUP> and p21<SUP>CIP1</SUP>. We further demonstrate that growth hormone, through the transcription factor STAT5, enhances expression of the <I>Cdkn2b</I> (cyclin‐dependent kinase inhibitor 2B) gene and that STAT5A binds to interferon‐gamma–activated sequence sites within the promoter. We recently demonstrated that ablation of STAT5 from liver results in hepatocellular carcinoma upon CCl<SUB>4</SUB> treatment. We now establish that STAT5, like in MEFs, activates expression of the <I>Cdkn2b</I> gene in liver tissue. Loss of STAT5 led to diminished p15<SUP>INK4B</SUP> and increased hepatocyte proliferation. <I>Conclusion:</I> This study for the first time demonstrates that cytokines, through STAT5, induce the expression of a key cell cycle inhibitor.         Synthesis and photoelectrochemical behavior of CdS quantum dots-sensitized indium–tin–oxide mesoporous film

        In this paper, we reported an investigation on a new photoelectrode of quantum dots-sensitized solar cell (QDSC) combining indiumetineoxide (ITO) mesoporous film and CdS quantum dots (QDs). The ITO mesoporous film was prepared by doctor-blade technique and CdS QDs attached on ITO mesoporous film were synthesized by successive ionic layer adsorption and reaction method. X-ray diffraction, field emission scanning electron microscopy, transmission electron microscope, X-ray spectroscopy and UV evis spectroscopy were used to characterize the samples. The results indicated that the ITO mesoporous film was uniform, crack-free and highly porous. And absorbance in visible region was enhanced after the deposition of CdS QDs on ITO mesoporous film. The photoelectrochemical property of the CdS QDssensitized ITO mesoporous film photoelectrode was investigated by forming a photoelectrochemical cell. Photocurrentevoltage measurement showed that the photoelectrode was efficient in the cell as working electrode.

        A Relay Selection and Power Allocation Scheme for Cooperative Wireless Sensor Networks

        This paper investigates optimal relay selection and power allocation under an aggregate power constraint for cooperative wireless sensor networks assisted by amplify-and-forward relay nodes. By considering both transmission power and circuit power consumptions, the received signal-to-noise ratio (SNR) at the destination node is calculated, based on which, a relay selection and power allocation scheme is developed. The core idea is to adaptively adjust the selected relays and their transmission power to maximize the received SNR according to the channel state information. The proposed scheme is derived by recasting the optimization problem into a three-layered problem ― determining the number of relays to be activated, selecting the active relays, and performing power allocation among the selected relays. Monte Carlo simulation results demonstrate that the proposed scheme provides a higher received SNR and a lower bit error rate as compared to the average power allocation scheme.

        Fast fluorescent blood sugar sensing using phenylboronic acid functionalized N, S-doped carbon dots

        A phenylboric acid functionalized carbon dot (2-FPBA-CD) for rapid fluorescent sensing of glucose in blood was synthesized by simply mixing N, S-doped carbon dots (CDs) with phenylboric acid at room temperature. At pH 7.4, the response of 2-FPBA-CD to glucose could reach equilibrium in a very short time (10 min), with a wide responsive linear range of 19.70 µM to 2.54 mM, which can be applied to the detection of glucose in serum. The mechanism studies showed that the layered carbon film of 2-FPBA-CD aggregated after adding glucose, thereby leading to the fluorescence quenching of 2-FPBA-CD.

        fects of Dietary Soy Intake on Maternal Thyroid Functions and Serum Anti-Thyroperoxidase Antibody Level During Early Pregnancy

        Soy and its isoflavones have been suggested to suppress thyroperoxidase (TPO), induce goiter, inhibit deiodinase, and modulate immune functions. This study initially investigated the effects of dietary soy consumption on maternal thyroid functions and anti-TPO antibody (TPOAb) production during early pregnancy. Data were collected through questionnaire from 505 women enrolled during early pregnancy by random sampling in Shenyang, China. Based on soy intake frequency, the subjects were divided into three groups (frequent [three or more times per week], conventional [more than twice per month but less than three times per week], and occasional [two or fewer times per month]). Serum thyrotropin (TSH), free thyroxine (FT_4), and TPOAb were measured by chemiluminescence immunoassay. Additionally, the concentrations of two primary isoflavones (daidzein and genistein) and creatinine were assessed in the spot urine samples from representative subjects (about 20%) randomly selected from the three groups. The percentages of frequent, conventional, and occasional consumers were 18.6%, 62.6%, and 18.8%, respectively. No difference was found in age, medical records, family history of thyroid diseases, serum FT_4, TSH, and TPOAb levels, TPOAb-positive percentages, or prevalence of thyroid dysfunctions among the groups. Both urinary daidzein and genistein levels were significantly higher in the frequent consumers compared with the other two groups. No correlations were found between urinary isoflavone levels and serum FT_4 or TSH. Urinary isoflavone levels were not significantly different between TPOAb-positive and -negative women among the randomly selected representative subjects. On the whole, our findings suggest dietary soy consumption during early pregnancy is not associated with the development of thyroid dysfunction or autoimmunity.
