The antioxidant properties of amla extracts and their effects on the oxidative stress in streptozotocin-induceddiabetes were examined in rats. Amla in the form of either the commercial enzymatic extract SunAmla (Taiyo Kagaku Co.Ltd., Yokkaichi, Japan) (20 or 40 mg/kg of body weight/day) or a polyphenol-rich fraction of ethyl acetate extract (10 or 20mg/kg of body weight/day) was given orally for 20 days to the streptozotocin-induced diabetic rats. Amla extracts showedstrong free radical scavenging activity. Amla also showed strong inhibition of the production of advanced glycosylated endproducts. The oral administration of amla extracts to the diabetic rats slightly improved body weight gain and also signifi-cantly alleviated various oxidative stress indices of the serum of the diabetic rats. The elevated serum levels of 5-hydrox-ymethylfurfural, which is a glycosylated protein that is an indicator of oxidative stress, were significantly reduced dose-de-pendently in the diabetic rats fed amla. Similarly, the serum level of creatinine, yet another oxidative stress parameter, wasalso reduced. Furthermore, thiobarbituric acid-reactive substances levels were significantly reduced with amla, indicating areduction in lipid peroxidation. In addition, the decreased albumin levels in the diabetic rats were significantly improved withamla. Amla also significantly improved the serum adiponectin levels. These results form the scientific basis supporting theefficacy of amla for relieving the oxidative stress and improving glucose metabolism in diabetes.

To ascertain the principal active peroxynitrite (ONOO−) scavenging components of heat-processed Panax ginseng C.A. Meyer (sun ginseng [SG]), the ONOO− scavenging activities of fractions and components of SG were compared. The results demonstrated that the ONOO− scavenging ability of SG was due to its ether fraction containing phenolic compounds. High-performance liquid chromatography analysis and ONOO− scavenging activity tests of the phenolic acids contained in SG identified vanillic acid, ferulic acid, p-coumaric acid, syringic acid, and maltol as the main active ONOO− scavenging components of SG. The ONOO− scavenging activities of phenolic acids and maltol were dependent on the degrees of their proton donating ability.

Modulation of age-related NF-@?B activation by dietary zingerone via MAPK pathway

Zingerone, a major component found in ginger root, has been known as anti-mutagenic and anti-carcinogenic activities that are often associated with its anti-oxidative and anti-inflammatory activities. In recent studies, we examined molecular mechanism of zingerone treatment on pro-inflammatory NF-κB activation via the redox-related NIK/IKK and MAPK pathways. Action mechanism of zingerone on NF-κB signaling was investigated in aged rat kidney and endothelial cells. The results showed that zingerone had not only the antioxidant effect by constitutive suppression of ROS, but also anti-inflammatory effects by suppression of nuclear factor (NF)-κB activation in aged rat. In addition, zingerone treatment suppressed gene activation of pro-inflammatory enzymes, COX-2 and iNOS, which were upregulated with aging through NF-κB activation and IKK/MAPK signaling pathway. These experiments strongly indicate that zingerone treatment exerts a beneficial efficacy by suppressing both oxidative stress and age-related inflammation through the modulation of several key pro-inflammatory genes and transcription factors. Thus, the significance of our findings is that the zingerone treatment may provide some preventive measure against chronic inflammatory conditions that underlie many age-related inflammatory diseases, such as metabolic syndrome, cardiovascular disease, dementia, arthritis, diabetes, osteoprosis, and cancers.
