<P>One of the most popular approaches to parameter adaptation in hidden Markov model (HMM) based systems is the maximum likelihood linear regression (MLLR) technique. In this letter, we extend MLLR to factored MLLR (FMLLR) in which the MLLR parameters depend on a continuous-valued control vector. Since it is practically impossible to estimate the MLLR parameters for each control vector separately, we propose a compact parametric form of the MLLR parameters. In the proposed approach, each MLLR parameter is represented as an inner product between a regression vector and transformed control vector. We present an algorithm to train the FMLLR parameters based on a general framework of the expectation-maximization (EM) algorithm. The proposed approach is applied to adapt the HMM parameters obtained from a database of reading-style speech to singing-style voices while treating the pitches and durations extracted from the musical notes as the control vectors. This enables to efficiently construct a singing voice synthesizer with only a small amount of singing data.</P>
Forkhead box O-class 1 and Forkhead box G1 as Prognostic Markers for Bladder Cancer

<P>Forkhead box O-class 1 (<I>FOXO1</I>) is a key regulator of glucose homeostasis, cell-cycle progression, and apoptosis. Its functions are modulated by forkhead box G1 (<I>FOXG1</I>), which acts as a transcriptional repressor with oncogenic potential. Real-time PCR and immunohistochemical staining were performed in 174 primary bladder cancer specimens and 21 normal bladder mucosae to evaluate these genes. <I>FOXO1</I> and <I>FOXG1</I> mRNA expression in cancer tissues were higher than in normal mucosae (each <I>P</I><0.001). <I>FOXO1</I> mRNA levels were significantly higher in samples of non-progressed patients (<I>P</I><0.001), but <I>FOXG1</I> were enhanced in those of progressed patients (<I>P</I>=0.019). On univariate analysis, <I>FOXO1</I> mRNA expression was significantly associated with grade, stage, recurrence, progression and survival (each <I>P</I><0.05). On multivariate analysis, increased <I>FOXO1</I> mRNA expression was associated with both reduced disease progression (odds ratio [OR], 0.367; 95% confidence interval [CI], 0.163-0.826, <I>P</I>=0.015) and enhanced disease-free survival (OR, 3.262; 95% CI, 1.361-7.820, <I>P</I>=0.008). At a median follow-up of 33 months (range 2 to 156), the patients with a high <I>FOXO1</I> mRNA expression had a significantly prolonged survival (<I>P</I>=0.001). Immunohistochemical findings of <I>FOXO1</I> were generally concordant with mRNA expression levels. In conclusion, <I>FOXO1</I> may be a promising marker for predicting progression in human bladder cancers.</P>

<B>Graphic Abstract</B> <P>A transparent PS-b-P4VP copolymer/SWNTs nanocomposite film is used as both source and drain electrode in a pentacene OTFT with transmittance and electricresistance values of 85% and 6000 Ω/□, respectively. FoLT-PCR 기술을 인체체질의하적 응용을 위하여 당뇨병연구에 사용하였다. 당뇨병은 제1형 및 제2형으로 나뉘는데 제1형은 인슈린비의존성(NIDDM)으로 당뇨병환자의 약60%이상을 차지하며, 제2형은 인슈린의존성(IDBM)으로 당뇨병환자의 30%미만을 차지한다. 이들은 대부분 후천적으로 환경중에서의 인슈린관련 유전자의 돌연변이에 의해 발병하는 것으로 알려져 있다. 이에, 본 연구에서는 당뇨병의 병인을 유전적변이와 체질의학적 관계에서 고찰하기 위하여 수행되었다. NIDDM환자와 IDDM환자를 대상으로 인슈린유전자를 증폭하여 제한효소절단 양상과 염기배열분석을 하였다. 비암호영역중 4개 위치 +216, +1045, +1367, 및 +1380에서 다형성을 보였으며 새로운 α4, α5, α6 및 β2이 α1과 β1가 이형(heterozygous)에서만 검출되며 α1은 우성이며 신규형들과 β1은 열성이었다. 이러한 당뇨병병인은 유전학적으로 체질의학과 깊은 관계를 가지는 것을 시사하였다. A simple and rapid FoLT(formamide low temperature)-PCR, whereby human genomic DNA from blood can be amplified without DNA preparative stps, is described using human insulin genes. By applicatin of FoLT-PCR in human insulin genes, intragenic polymorphism in non-coding regions of the human insulin gene was shown after amplification and analysis by restriction enzyme digestion. All nucleotide sequences were the same as the reported, and four necleotides, at 4 different positions were polymorphic, and polymorphic alleles α4, α5, α6, and β2 were identified. The new alleles were originated from homologous recombination between the α1 and β1 alleles, and the alleles were founded in heterozygotes only. Althouughallele α1 was dominant, the new alleles and β1 were recessive. From this results, it was suggested that the new method of FoLT-PCR was highly applicable in genetic variation analysis.

<P>Summary</P><P>Bortezomib is a therapeutic proteasome inhibitor with antimyeloma activity and polyphenols are well known compounds that exert antiproliferative effects against tumuors. We attempted to co-treat myeloma cells with bortezomib and polyphenols, anticipating a synergistic effect. However, the anticancer activity of bortezomib was blocked by the polyphenols. The structural features of the polyphenols correlated strikingly with their antagonistic effect; in particular, the presence or absence of a vicinal diol moiety was the key element for effective blockage of the anticancer function of bortezomib. We speculated that the vicinal diols in the polyphenols interact with the boronic acid of bortezomib and convert the active triangular boronic acid of bortezomib to an inactive tetrahedral boronate, thus abolishing the antimyeloma activity of bortezomib. We confirmed this hypothesis by <SUP>11</SUP>B nuclear magnetic resonance spectroscopy and an <I>in vitro</I> assay on multiple myeloma (MM) cell lines and primary myeloma cells from patients. Based on these findings, restriction of the intake of natural polyphenols in foods or vitamin supplements during bortezomib treatment in MM patients should be considered.</P>

<P>We demonstrated enhanced localized surface plasmon resonance (SPR) biosensing based on subwavelength gold nanoarrays built on a thin gold film. Arrays of nanogratings (1D) and nanoholes (2D) with a period of 200 nm were fabricated by electron-beam lithography and used for the detection of avian influenza DNA hybridization. Experimental results showed that both nanoarrays provided significant sensitivity improvement and, especially, 1D nanogratings exhibited higher SPR signal amplification compared with 2D nanohole arrays. The sensitivity enhancement is associated with changes in surface-limited reaction area and strong interactions between bound molecules and localized plasmon fields. Our approach is expected to improve both the sensitivity and sensing resolution and can be applicable to label-free detection of DNA without amplification by polymerase chain reaction. </P>

Meta- and gene set analysis of stomach cancer gene expression data.

<P>We generated gene expression data from the tissues of 50 gastric cancer patients, and applied meta-analysis and gene set analysis to this data and three other stomach cancer gene expression data sets to define the gene expression changes in gastric tumors. By meta-analysis we identified genes consistently changed in gastric carcinomas, while gene set analysis revealed consistently changed biological themes. Genes and gene sets involved in digestion, fatty acid metabolism, and ion transport were consistently down-regulated in gastric carcinomas, while those involved in cellular proliferation, cell cycle, and DNA replication were consistently up-regulated. We also found significant differences between the genes and gene sets expressed in diffuse and intestinal type gastric carcinoma. By gene set analysis of cytogenetic bands, we identified many chromosomal regions with possible gross chromosomal changes (amplifications or deletions). Similar analysis of transcription factor binding sites (TFBSs), revealed transcription factors that may have caused the observed gene expression changes in gastric carcinomas, and we confirmed the overexpression of one of these, E2F1, in many gastric carcinomas by tissue array and immunohistochemistry.
