      • Planococcus屬 菌株가 生成하는 Alkaline Lipase

        A microorganism producing alkaline lipase strongly was isolated. The optimal condition to produce alkaline lipase by the selected strain was as follows; the medium was composed of 2% sucrose, 3% soybean meal, 0.6% K_2HPO_4 0.1%(NH_4)_2SO_4 0.1% urea, and 0.1% MgSO_4, 7H_2O., and the initial pH was 8.0 to 8.5 and culture time was 48hr. The lipase of the culture supernatant was purified by fraction-with ammonium sulfate at 30-50% saturation, followed by DEAE-cellulose chromatography. The specific activity of the purified enzyme was about 7.8 times over that of the starting centrifugate, and the yield was about 8.05% of the original activity. The enzymatic characteristics is included as follows; optimal pH of the enzyme was 9.0, and the stable pH range was between 6 to 10, and optimal temperature was 55C. And more than 85% of the enzyme activity remained after exposure at 60C for 30 min. Lipase activities were stimulated in the presence of Ca^++ and Mg^++, and inhibited in the presence of Hg^++ and Cu^++ The enzyme was apt to hydrolyze oils containing natural triglycerides plentifully. On the basis of the taxonomical studies, the selected strain was identified as Planococcus sp.

      • SIIM 퍼지 PID 제어기를 이용한 3축 푸마 로봇의 경로 제어에 관한 연구

        This paper describes the design of the Fuzzy PID controller using simplified indirect inference method. First, the fuzzy digital PID controller are derived from the conventional continuous time linear ones. Then the fuzzification, control-rule base, and defuzzification using SIIM in the design of the fuzzy digital controller are discussed in detail. The resulting controller is a discrete time fuzzy version of the conventional digital PID controller, which has the same linear structure, but are nonlinear functions of the input signals. The proposed controller enhances the self-tuning control capability, particularly when the process to be controlled is nonlinear. When the SIIM is applied, the fuzzy inference results can be calculated with splitting fuzzy variables into each action component and are determined as the functional form of corresponding variables. So the proposed method has the capability of the high speed inference and adapting with increasing the number of the fuzzy input variables easily. Computer simulation results have demonstrated the superior to the control performance of the conventional digital PID controller. Lastly, it is applied to the trajectory control of the PUMA robot with three-axis.

        들깨 유전자원의 단백질함량과 지방 및 지방산 특성

        공시된 들깨 지방재래종 90개 품종의 단백질함량, 지방 및 지방산을 분석한 결과 다음과 같다. 1. 숙기별로는 중ㆍ만생종의 평균단백질 함량은 22.4%로 비교적 낮았고, 품종분포가 대부분 평균치에 가까웠으며, 조생종은 평균단백질 함량이 24.1%로 비교적 높았다. 2. 평균단백질함량은 소립종(천립중3.0~4.0g이하)이 23.0%로, 중립종(천립중 4.0~5.0g)이나 대립종(천립중 5.0~6.0g)의 22.1%보다 높은 경향이었다. 3. 기름함량은 조생종이 42.6%로 가장 적었고, 중생종이 44.5%, 만생종이 45.5%로 기름함량이 많았으며, 립중별로는 소립종이 44.2%, 중립종이 43.1%, 대립종이 41.5% 대립종일수록 낮은 경향이었다. 4. 지방산조성은 숙기가 늦어짐에 따라 Oleic acid와 Linolenic acid는 다소 많았고, Linoleic acid는 적어지는 경향이었다. Linoleic acid는 극대립종에서 15.5%로 가장 높았고 립중이 가벼울수록 높아지는 경향이었다. 5. 기름함량과 지방산 비율간의 상관 정도를 보면 기름함량과 Palmitic acid, Linoleic acid는 負의 상관이나, Stearic acid, Linolenic acid는 正의 상관을 보였다. Stearic acid는 Oleic acid와 유의적인 정의 상관을 , Linoleic acid와는 부의 상관을 보였다. Oleic acid는 Linoleic acid 및 Linolenic acid와 고도로 유의한 부의 상관을, Linoleic acid는 Linolenic acid와, 포화지방산은 불포화지방산과 유의한 부의 상관을 보였다. Korean local collected strains or varieties of perilla analyzed on their protein and oil content and fatty acids composition in the seed. The strains analyzed were 90 of which grown in the experimental field of Chungbuk Agricultural Research and Extension Services at Cheongwon and Umseong from 2001 to 2002. The seed protein contents of 90 perilla varieties ranged from 19.1% to 29.0% and the average content was 22.8%. There differed by the seed coat color showing 1.9% higher content in light gray than that of dark brown, which may suggest that the seed coat color could be used as a marker gene for breeding high protein varieties. Oil content of medium and late varieties in maturity showed higher as compared to early varieties, and appeared differences by seed coat color, and small seed varieties showed higher seed oil content than in large seed varieties by seed size. Linoleic and linolenic acid content of which major fatty acids in perilla seed were 74.4% in average of tested varieties. Fatty acid composition of perilla varieties were not significantly different by maturity, seed coat color and 1,000 seed weight. Oil content showed high positive correlation, with stearic and linolenic acids and negative correlation with linoleic acid.

      • 세계산 Rumiceae Dum.족 (마디풀과)의 진화계통학적 연구

        세계산 Rumiceas Dum.족을 구성하는 4속(Rumex속의 4절, Rheum속의 3절, Oxyria속, Emex속) 분류군간의 진화적 유연관계에 대한 가설을 제공하고, 형질진화의 경향을 밝히기 위한 분자분식(cladistic analysis)을 수행하였다. 최소절약의 원리를 이용하여 24개의 두단계 혹은 다단계 형질을 분석하였다. 분석 결과 계통수의 길이는 36, 일관성지수는 0.714, 보존지수는 0.843인 4개의 등가질약 계동수가 산출되었다. Rumex속의 4개 절과 Emex속은 하나의 Clade를 구성하며, 이는 여러 개의 공유파생형질(익상수과, 오용 존서, 만곡의 배, 등)에 의해 지지된다. 결과적으로 Emex속은 Rumex속의 하나의 절(section)로 처리함이 타당하다. 또한 Oxyria속은 3개의 독립파생형질(화사수:2개, 렌즈형의 수과, 4개의 화피핀)에 의해 다른 속과 잘 구별되었다. Cladistic analysis of four genera (4 sections of the genus Rumex, 3 sections of the genus Rheum, Oxyria and Emex) of the tribe Rumiceae Dum. was undertaken to provide hypotheses about evolutionary relationships and to elucidate patterns of character evolution. A set of 24 binary and multistate morphological characters were analyzed using maximum parsimony algorithms. With the given data set. four equally parsimonious trees 36 tree length with a consistency index 0.714 and a retention index of 0.843 were produced. Four sections of Rumex and Emex came together as one clade and was strongly supported by several synapomorphies (e.g.. winged achene. trichome present. curved embryo. etc.). Thus: it is considered that Emex is better treated as one of section in Rumes. Oxyria is also well distinguished by its automorphic characters (by having 2 styles. lenticular achene. 4 perianth segment number).

      • 생쥐 안구에서 산화적 손상에 의한 superoxide dismutases와 metallothionein의 나이에 따른 유도의 차이

        The basal levels and inducibility of superoxide dismutases (SODs; Mn-SOD and Cu/Zn-SOD) and metallothionein (MT) was investigated in the eyes of mice with age. Oxidative stress was induced by paraquat, an intracellular superoxide generator, in 1, 4, 8, and 12 months of age and then mRl\As of SODs and MT in the eyes were determined by RT-PCR assay. The basal level of Mn-SOD mRNA increased from 1 month to 8 months but decreased thereafter. Mn-SOD mRNA was induced by paraquat until 4 month but not after 8 months. On the other hand, the basal level of Cu/Zn-SOD mRNA decreased with age. Cu/Zn-SOD mRNA was not induced by paraquat until 4 months. The basal level of MT mRNA declined with age whereas its inducibility by paraquat was intact at all ages. Taken together, the results indicate that SODs and MT are differentially expressed and induced according to the age. It could be also implicated that failure in induction of Mn-SOD with age may be one of causative factors in the aging process whereas induction of Cu/Zn-SOD and MT may be one of important factors in defense against oxidative stress in the eyes. 서론: 최근 노화는 반응성 산소종에 의한 산화적 손상의 축적결과라는 설이 각광을 받고 있다. 노화과정에서 노화와 관련된 질병을 비교적 잘 나타내는 안구를 선정하고, 정상생쥐의 안구에서의 superoxide dismutases (SODs) 와 metallothionein (MT) 의 함량 뿐 아니라 산화성 스트레스를 유발하는 paraquat에 의한 SODs와 MT의 유도능과 나이와의 상호관계를 조사하여 이들의 노화과정에서의 역할을 규명하고자 하였다. 재료 및 방법 : 본 연구에서 1, 4, 8, 12개월된 정상생쥐의 안구에서 반응성 산소종 생성의 초기에 중요한 방어기전인 SODs mRNA와 hydroxy radical (OH·)에 의한 산화적 손상의 마지막 방어기전인 MT mRNA의 함량과 산화성 스트레스를 유발하는 paraquat에 의한 유도능을 역전사 중합효소 연쇄반응 (RT-PCR) 으로 조사하였다. 결과: 정상생쥐의 안구의 노화과정에서 Mn-SOD와 Cu/Zn-SOD, 그리고 MT mRNA의 항정상태 함량을 조사한 결과, Mn-SOD mRNA는 8개월까지는 증가하다가 12개월에서 감소하였으나, Cu/Zn-SOD mRNA는 나이와 함께 감소하였다. 그러나 MT mRNA 는 8개월까지는 감소하다가 12개월에서 약간 증가하였다. 산화성 스트레스를 유발하는 parraquat에 의하여 Mn-SOD mRNA는 1개월과 4개월에서만 유도되었고, Cu/Zn-SOD mRNA는 8개월과 12개월에서만 유도되었으며, MT mRNA는 관찰한 전 기간에서 유도되었다. 결론: 이상의 결과를 종합하면 SODs와 MT는 안구에서 나이에 따라 서로 다르게 발현되고 유도되는 것으로 생각되며, 노화과정에서 Mn-SOD유도의 이상이 노화의 원인이 되는 인자로 작용하고, 반면에 Cu/Zn-SOD와 MT의 정상적인 유도능이 산화적 스트레스에 대하여 방어역할을 하는 것으로 생각된다. 이와 같이 연령이 증가함에 따라 Mn-SOD, Cu-Zn-SOD, 및 MT의 발현과 유도능의 차이가 있게 되고, 그 결과 반응성 산소종에 의한 산화적 손상의 정도의 차이로 노화의 속도가 조절되지 않나 생각된다.

      • 초등학생의 패스트푸드 이용횟수에 따른 식행동 및 영양지식 비교

        The present study surveyed elementary school students' fast food consumption patterns, dietary behavior, nutrition knowledge, and examined if these variables were different according to the frequency of fast food consumption. The subjects of this study were a total of 589 fifth and sixth grade students from two elementary schools in Seoul, and according to the frequency of fast food consumption, they were classified into frequent-users (1-2 times or more a week, 14.9%), moderate-users (2-3 times a month, 48.2%) and non-users (fewer than once a month, 36.8%). Frequent-users consumed fast food when they felt hungry, and moderate-users and non-users consumed it on special days. As to whether they consider nutrition and health when choosing fast food, 10.2% of the frequent-users replied 'Not consider at all' and the percentage was higher than that of the moderate-users (7.4%) and the non-users (7.4%). Father's education level was higher in non-users than in frequent-users and moderate-users. The amount of monthly pocket money was larger in frequent-users and moderate-users than in non-users. The mean score of dietary habit (out of 75 points) was 55.40 in frequent-users, 58.11 in moderate-users and 62.10 in non-users, and the differences were statistically significant (p<0.001). Frequent-users showed more severe unbalanced diet and more frequent dining out. The mean score of nutrition knowledge (out of 20 points) was 15.90 in frequent-users, 16.24 in moderate-users and 16.86 in non-users, and the differences were statistically significant (p<0.05). According to the results, when the frequency of fast food consumption is higher the amount of monthly pocket money is larger, and when the frequency of fast food consumption is lower father's education level is higher and the students have more desirable dietary habits and correct nutrition knowledge. Furthermore, students with correct nutrition knowledge showed more desirable dietary habits. Accordingly, there should be systematic and continuous nutrition education at home and in school so that elementary school students lead a healthy dietary life by refraining themselves from indiscreet eating of fast food and developing desirable dietary habits.

However, on treatment of the unsubstituted or phenyl substituted allylic cyclic carbonates 1e and 1f with Palladium(0) catalyst, deprotonation-elimination of π-allyl Palladium complex followed by condensation with the other π-allyl Palladium complex provided the coupled products 3a and 3b, respectively.
