Lou Ying,Su-Ho Lee,Shou Jian-Min 한국항해항만학회 2016 한국항해항만학회지 Vol.40 No.2
With the advent of Internet era, e-commerce has become the focus of human's life. It leads a new direction of social development . As the representative of traditional industry, shipping industry is confronted with a series of difficulties, which have to break through the traditional and existing model to make their business for survival. With the increasing pricking up of market competition, shipping industry is now facing development bottle neck, but e-commerce provides a new way to solve the problem. This paper firstly describes the existing forms of the e-commerce shipping platform. Secondly analyzes the data for the situation of shipping industry in China, the data for expected functions of an e-commerce shipping platform and the data for how to choose a specific e-commerce shipping platform. Thirdly analyzes the potential risks of establishing e-commerce shipping platform in China. Based on the above researched, the paper provides a suggested model of the shipping e-commerce shipping platform in China.

Objectives: The present study aimed to examine the cytogenetic and genetic mutation features of acute myeloid leukemia (AML) in elderly Chinese patients. Methods: A retrospective analysis of cytogenetics and genetic mutations was performed in 113 cases (age range 50-82 years) with de novo AML. Results: The most frequent cytogenetic abnormality was t (15;17) (q22;q21), detected in 10.0% (n = 9) of successfully analyzed cases, followed by t (8;21) (q22;q22) in 8.89% (n = 8), and complex karyotypes in 5.56% (n = 5). Those with complex karyotypes included 4 cases (4.44%) of monosomal karyotypes. The frequencies of NPM1, FLT3-ITD, c-kit, and CEBPA mutations were 27.4% (31/113), 14.5% (16/110), 5.88% (6/102), and 23.3% (7/30), respectively. The complete remission rates of patients in low, intermediate, and high risk groups were 37.5%, 48.6%, and 33.3%, respectively (${\chi}^2$ = 0.704, P = 0.703) based on risk stratification. Conclusion: Cytogenetics and genetic mutations alone may not be sufficient to evaluate the prognoses of elderly AML patients. The search for a novel model that would enable a more comprehensive evaluation of this population is therefore imperative.

Purpose The prevalence of frailty among patients with heart failure is about 45%. Frailty may result in patients' functional decline, falls, disability, and decreased quality of life. Qualitative studies can explore older patients' perceptions of frailty and help patients cope with it. However, a qualitative approach that explores the experience of frailty in older patients living with heart failure is lacking. This study aimed to explore the lived experience of frailty in older patients with heart failure. Methods This qualitative study applies Giorgi's phenomenological method. Data were collected from October 2019 to August 2020. Thirteen older patients with heart failure aged at least 60 years were recruited using purposive sampling from a medical center in Taiwan. The participants participated in an in-depth interview using a semistructured interview guide. Results Seven themes were identified: “being reborn at the end of the road but having difficulty recovering”, “living with a disease with an ineffable feeling”, “feeling like being drained: physical weakness and a dysfunctional body”, “struggling with impaired physical mobility and facing unexpected events”, “suffering from mental exhaustion”, “receiving care from loved ones”, and “turning over a new leaf”. Conclusions Frailty in older patients with heart failure was obscure and difficult to describe. Frailty could be improved by medical intervention, self-management, and social support but was difficult to reverse. Patients with heart failure should be evaluated for frailty using multidimensional assessment tools at first diagnosis and provided frailty-related information so that patients have proper insight into their disease as early as possible.

In order to examine dispersion properties of magnetorheological (MR) fluids, characteristics of its medium oil such as their rheological property and density are being focused in this study. Carbonyl iron (CI) based MR fluid with an aqueous polymeric solution using poly(ethylene oxide) (PEO) as a medium was prepared. MR characteristics as a function of applied magnetic field strength was investigated using a rotational rheometer with MR devices attached, demonstrating that the characteristics of the CI–PEO based MR fluids were affected by a medium viscosity in a steady shear flow.

The cytokinin response factor 5 (CRF5) belongs to a family of plant-specific APETALA2 (AP2)/ ethylene-responsive element binding proteins (EREBPs). The novel role of Arabidopsis CRF5, previously identified as a mediator of cytokinin signaling, has been suggested to increase pathogen resistance in this study. Endogenous CRF5 transcripts are expressed in all tissues, including the seedlings, leaf, stem, flower, silique and root, and were found to be induced at 1 h after infection with the bacterial pathogen, Pseudomonas syringae pv. tomato DC3000 (Pst DC3000). The results of a yeast one-hybrid assay revealed that an acidic region of CRF5, including the C-terminal 28 amino acids, functions as a transcriptional activator. Overexpression of CRF5 in transgenic Arabidopsis increases pathogen resistance and concomitantly activates the expression of a large number of GCC-box pathogenesis-related genes. These results indicate that CRF5 may be involved in disease resistance as a transcription activator, thus providing a mechanistic link between the plant pathogen response and cytokinin signaling.

In this study, we attempted to isolate novel mast cell-stimulating molecules from Staphylococcus aureus. Water-soluble extract of S. aureus cell lysate strongly induced human interleukin-8 in human mast cell line-1 and mouse interleukin-6 in mouse bone marrow-derived mast cells. The active molecule was purified to homogeneity through a C18 reverse phase HPLC column. By determination of its structure by MALDI-TOF and 1H- and 13C-NMR, adenosine was revealed to be responsible for the observed cytokine induction activities. Further studies using 8-sulfophenyl theophylline, a selective adenosine receptor blocker, verified that purified adenosine can induce interleukin-8 production via adenosine receptors on mast cells. Moreover, adenosine was purified from S. aureus-engulfed RAW264.7 cells, a murine macrophage cell line, used to induce phagocytosis of S. aureus. These results show a novel view of the source of exogenous adenosine in vivo and provide a mechanistic link between inflammatory disease and bacterial infection. [BMB reports 2011; 44(5): 335-340]
A Novel Approach to Cloning and Expression of Human Thymidylate Synthase
Lv, Ying-Tao,Du, Pei-Juan,Wang, Qiao-Yan,Tan, Yuan,Sun, Zong-Bin,Su, Zhong-Liang,Kang, Cong-Min Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.12
Thymidylate synthase (TS) catalyzes the transfer of a methyl group from methylenetetrahydrofolate to dUMP to form dTMP. It is a primary target in the chemotherapy of colorectal cancers and some other neoplasms. In order to obtain pure protein for analysis of structure and biological function, an expression vector TS-pET28b (+) was constructed by inserting wild-type human thymidylate synthase (hTS) cDNA into pET28b (+). Then an expression strain was selected after transformation of the recombined plasmid into Rosetta (DE3). Fusion protein with His-tag was efficiently expressed in the form of inclusion bodies after IPTG induction and the content was approximately 40.0% of total bacteria proteins after optimizing expression conditions. When inclusion bodies were washed, dissolved and purified by Ni-NTA under denatured conditions, the purity was up to 90%. On SDS-PAGE and West-blotting, the protein band was found to match well with the predicted relative molecular mass-36kDa. Bioactivity was 0.1 U/mg. The results indicated that high-level expression of wild-type hTS cDNA can be achieved in prokaryotes with our novel method, facilitating research into related chemotherapy.
