A pfl ldhA double mutant Escherichia coli strain NZN 111 was used to produce succinic acid by overexpressing the E. coli malic enzyme gene (sfcA). This strain, however, produced a large amount of malic acid as well as succinic acid. After the analyses of the metabolic pathways, the fumB gene encoding the anaerobic fumarase of E. coli was co-amplified to solve the problem of malic acid accumulation. A plasmid, pTrcMLFu, was constructed, which contains an artificial operon (sfcA-fumB) under the control of the inducible trc promoter. From the batch culture of recombinant E. coli NZN 111 harboring pTrcMLFu, 7 g/L of succinic acid was produced from 20 g/L of glucose, with no accumulation of malic acid. From the metabolic flux analysis the strain was found under reducing power limiting conditions by severe reorientation of metabolic fluxes.

The self-diffusion experiment of THO was performed across a series of copolymer hydrogel membranes at different temperatures. Copolymer hydrogel membranes were prepared by copolymerizing 2-hydroxyethyl methacrylate (HEMA) and 2-aminoethyl methacrylate (AEMA) in the presence of the solvent and the crosslinker, ethylene glycol dimethacrylate (EGDMA). By changing the crosslinker content and the ratio of HEMA and AEMA monomer, two series of copolymer hydrogel membranes were synthesized. The tagging material was THO and efflux of THO was counted on a Liquid Sc-intillation Counter. The experimental data show that the permeability decreases as the amount of EGDMA and the mole fraction of HEMA increase, and the permeability is proportional to the temperature. The partition coefficient shows a parallel trend with permeability. Using the relationship between viscosity and diffusivity, the viscosity of water within the membrane was obtained. According to the result, the viscosity of watler within the membrane has the same value with those of supercooling water. And we obtained the activation energy of THO for transport in the membrane by using Arrhenius plotting.

Dimeric sesquiterpenoid shizukaol B (SKB) was isolated from Chloranthus japonicus Sieb. Except that SKB inhibited adhesion molecule expression in monocytes and endothelial cells, no more biological and pharmacological activity of SKB had been reported until now. In this study, we examined immunosuppressive activity of SKB. SKB strongly inhibited lipopolysaccharide (LPS) induced B cell proliferation with IC50 of 137 ng/ml, but slightly or not concanavalin A-induced T cell proliferation, LPS-induced macrophage NO production, and LPS-induced dendritic cell maturation. As a mechanism, SKB strongly induced apoptotic death of B cells, but not other cell types. These results suggested that SKB induced toxicity-mediated immunosuppression against B cells.

Objective: To evaluate the association of MRI features with the major genomic profiles and prognosis of World Health Organization grade III (G3) gliomas compared with those of glioblastomas (GBMs). Materials and Methods: We enrolled 76 G3 glioma and 155 GBM patients with pathologically confirmed disease who had pretreatment brain MRI and major genetic information of tumors. Qualitative and quantitative imaging features, including volumetrics and histogram parameters, such as normalized cerebral blood volume (nCBV), cerebral blood flow (nCBF), and apparent diffusion coefficient (nADC) were evaluated. The G3 gliomas were divided into three groups for the analysis: with this isocitrate dehydrogenase (IDH)-mutation, IDH mutation and a chromosome arm 1p/19q-codeleted (IDHmut1p/19qdel), IDH mutation, 1p/19q-nondeleted (IDHmut1p/19qnondel), and IDH wildtype (IDHwt). A prediction model for the genetic profiles of G3 gliomas was developed and validated on a separate cohort. Both the quantitative and qualitative imaging parameters and progression-free survival (PFS) of G3 gliomas were compared and survival analysis was performed. Moreover, the imaging parameters and PFS between IDHwt G3 gliomas and GBMs were compared. Results: IDHmut G3 gliomas showed a larger volume (p = 0.017), lower nCBF (p = 0.048), and higher nADC (p = 0.007) than IDHwt. Between the IDHmut tumors, IDHmut1p/19qdel G3 gliomas had higher nCBV (p = 0.024) and lower nADC (p = 0.002) than IDHmut1p/19qnondel G3 gliomas. Moreover, IDHmut1p/19qdel tumors had the best prognosis and IDHwt tumors had the worst prognosis among G3 gliomas (p < 0.001). PFS was significantly associated with the 95th percentile values of nCBV and nCBF in G3 gliomas. There was no significant difference in neither PFS nor imaging features between IDHwt G3 gliomas and IDHwt GBMs. Conclusion: We found significant differences in MRI features, including volumetrics, CBV, and ADC, in G3 gliomas, according to IDH mutation and 1p/19q codeletion status, which can be utilized for the prediction of genomic profiles and the prognosis of G3 glioma patients. The MRI signatures and prognosis of IDHwt G3 gliomas tend to follow those of IDHwt GBMs.

This paper describes design and characteristic analysis of long primary type linear synchronous motor (LSM) for high speed train system. LSM is designed using loading distribution method and magnetic equivalent circuit. For characteristic analysis of LSM, analytical and numerical methods are applied. Analytical method for solving the magnetic field distribution of the analytic model is based on the Maxwell’s equations. Using the characteristic equation and magnetic equivalent circuit, we analyze the effect of variation of parameters, and then we validate the result by comparing with numerical method by finite element method (FEM). We compare the analytical method with numerical method for analyzing the effect by variable parameters. This result will be useful of design and forecast of performance without FEM.

Background/Aim: The prognosis of hepatocellular carcinoma (HCC) is affected by stage as well as liver function. We analyzed the outcome of early stage HCC in Child-Pugh class C patients and advanced HCC in Child-Pugh class A patients. Methods: Among 453 HCC patients, group I included consecutive 33 patients with early stage (I,II) in Child-Pugh class C and Group II included 45 consecutive patients with advanced stage (III, IV) in Child-Pugh class A. We compared clinical characteristics, complications of cirrhosis, overall survival between two groups. Results: Younger age, male, presence of HBV, α-fetoprotein (AFP) (>400ng/ml), and MELD score (<15) were significantly high in advanced stage HCC in Child-Pugh class A patients (Group II) (p<0.05). Complications of cirrhosis such as variceal hemorrhage, ascites, and hepatic encephalopathy were increased in the Group I (p<0.05). Median overall survival was 24.5 months (95% CI 10.6-38.4) for Group I and 13.5 months (95% CI 4.6-22.3) for Group II (p=0.281). AFP was an independent prognostic factor in early stage in Child-Pugh class C patients (Group I). MELD score and tumor stage were independent prognostic factors in advanced stage in Child-Pugh class A patients (Group II). Conclusion: Overall survival was not different between two groups. AFP was a prognostic factor in early stage in Child-Pugh class C patients (Group I), whereas MELD score and tumor stage were prognostic factors in advanced stage in Child-Pugh class A patients (Group II).
