YPL-001 is a drug substance of Pseudolysimachion rotundum var. subintegrum and has been reported to be a potent COPD inhibitor. For the first time, this study demonstrated a correlation among the iridoid constituents, antioxidants, and MUC5AC inhibition activities in P. rotundum during different growth stages (5 to 11 weeks). Single-factor extraction was used to optimize the plant extraction conditions to maximize the major iridoid constituents (70% ethanol, 40 °C, 1 h); isolated metabolites 1–6 were identified using nuclear magnetic resonance spectroscopy (NMR) and mass spectrometry (MS). The contents of each metabolite and antioxidant/MUC5AC inhibition effects were markedly changed according to the growth stages, especially for catalposide (2, 5.97 → 10.99 mg/g, 1.8-fold) and isovanillyl catapol (5, 4.42 → 20.00 mg/g, 4.5-fold), which were the predominant substances in August. Our results indicated that YPL-001 could potentially contribute to enhancing the P. rotundum value in accumulated iridoids at the growth stage and the biological effect aspects to develop industrial medicinal crops.

Literature Review: A Development of Long-Term Evolution Scenarios in Biosphere Assessment

The post-closure safety assessment of a repository is typically conducted over an extensive timescale from ten thousand to a million years. Considering that biosphere ecosystems may undergo significant changes over such lengthy periods, it is essential to incorporate the long-term evolution of the biosphere into the safety assessment. Climate change and landscape development are identified as critical drivers with the potential to impact the hydrogeological and hydrogeochemical characteristics of the biosphere. These changes can subsequently alter the migration patterns of radionuclides through the biosphere and influence human exposure doses. Therefore, this study formulates scenarios within the context of long-term biosphere evolution. We examine biosphere assessment processes employed in other countries and conduct a comparative study on scenario conditions. For example, biosphere assessment in Finland has identified sea-level changes and land-use alterations as significant factors in the long-term evolution of the biosphere. These factors are linked to Features, Events, and Processes (FEPs) associated with climate change and human activities. Sea-level changes are related to FEPs regarding climate change, land uplift, and shoreline displacement, while land-use changes are based on human activity-related FEPs (e.g., crop type, livestock and forest management, well construction, and demographics). Based on the literature review, this study has configured long-term evolution scenarios for the safety assessment of a deep geological repository for spent fuels.

<P>A mild and practical cobalt-catalyzed defluoroborylation of fluoroarenes is presented for the first time. The method permits straightforward functionalization of fluoroarenes, with high selectivity for borylation of C-F over C-H bonds, and a tolerance for aerobic conditions. Furthermore, two-step <SUP>18</SUP>F-fluorination was achieved for expanding the scope of <SUP>18</SUP>F-positron emission tomography probes.</P> [FIG OMISSION]</BR>

Ulmus parvifolia 잎의 약리학적 지표 성분을 제시하기 위해 간단하고 신뢰할 수 있는 HPLC 분석방법을 개발하였다. 지표 성분들은 NMR 및 UPLC-QTof-MS 분석에 의해 neochlorogenic acid (trans-5-O-caffeoylquinic acid, 5-CQA)과 chlorogenic acid (trans-3-O-caffeoylquinic acid, 3-CQA)로 구조 동정 되었다. HPLC를 이용하여 두개 화합물을 포함한 잎 추출물의 정성/정량 분석 방법을 평가하였다. 잎 30% 에탄올 추출물과 지표성분의 안정성 테스트는 6개월 동안 평가되었다. 그러나 안정성 조사 기간 동안 추출물과 지표 성분들 각각의 함량에 대한 유의한 변화는 관찰되지 않았다. A simple and reliable HPLC method was developed to determine pharmacologically standard marker compounds of Ulmus parvifolia leaves. Standard markers were characterized with neochlorogenic acid (trans-5-O-caffeoylquinic acid, 5-CQA) and chlorogenic acid (trans-3-O-caffeoylquinic acid, 3-CQA) using NMR and UPLC-QTof-MS analysis. A method for qualitative/ quantitative analysis of the leaves extracts were evaluated including two compounds by using HPLC. The stability test of 30% ethanolic extracts of the leaves sample and standard markers have been evaluated for six months. However, no significant changes in the content of the marker compounds of each extract was observed during the time of investigation.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder associated with impaired social behavior and communication, repetitive behaviors, and restricted interests. In addition to genetic factors, environmental factors such as prenatal drug exposure contribute to the development of ASD. However, how those prenatal factors induce behavioral deficits in the adult stage is not clear. To elucidate ASD pathogenesis at the molecular level, we performed a high-resolution mass spectrometry-based quantitative proteomic analysis on the prefrontal cortex (PFC) of mice exposed to valproic acid (VPA) in utero, a widely used animal model of ASD. Differentially expressed proteins (DEPs) in VPA-exposed mice showed significant overlap with ASD risk genes, including differentially expressed genes from the postmortem cortex of ASD patients. Functional annotations of the DEPs revealed significant enrichment in the Wnt/β-catenin signaling pathway, which is dysregulated by the upregulation of Rnf146 in VPA-exposed mice. Consistently, overexpressing Rnf146 in the PFC impaired social behaviors and altered the Wnt signaling pathway in adult mice. Furthermore, Rnf146-overexpressing PFC neurons showed increased excitatory synaptic transmission, which may underlie impaired social behavior. These results demonstrate that Rnf146 is critical for social behavior and that dysregulation of Rnf146 underlies social deficits in VPA-exposed mice.

<P>We present two algorithms to predict the activity of AsCpf1 guide RNAs. Indel frequencies for 15,000 target sequences were used in a deep-learning framework based on a convolutional neural network to train Seq-deepCpf1. We then incorporated chromatin accessibility information to create the better-performing DeepCpf1 algorithm for cell lines for which such information is available and show that both algorithms outperform previous machine learning algorithms on our own and published data sets.</P>
