Molecular cloning and analysis of the <i>Thermus caldophilus</i> ADP-glucose pyrophosphorylase

<P><B>Abstract</B></P><P>Previously, we have purified a highly thermostable ADP-glucose pyrophosphorylase (EC 2.7.7.27; AGPase) from <I>Thermus caldophilus</I> GK-24. In the present paper, we further report the molecular cloning and characterization of the thermostable bacterial AGPase. Using a 0.6-kb DNA probe obtained by polymerase chain reaction (PCR) with a primer set deduced from the <I>N</I>-terminal and internal amino acid sequence, the AGPase gene was cloned. The cloned AGPase gene had a 1245-bp open reading frame that encodes a protein of 414 amino acids. Then, the full-length AGPase gene was further cloned into the pHCE19T(II) vector and was expressed in <I>Escherichia coli</I> DH5α. Western analysis of the recombinant enzyme showed the same immunity as the wild-type enzyme with a molecular mass of ca. 46kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The kinetic parameters of the recombinant AGPase were basically similar to the wild type. The <I>N</I>-terminal region of <I>T. caldophilus</I> AGPase showed a partial similarity to that of <I>E. coli,</I> and R336 and P295 were identical to those of <I>E. coli</I> AGPase. In addition, sequence comparison revealed that R177 and P235 of <I>T. caldophilus</I> AGPase were aligned with K195 and K247 of <I>E. coli,</I> and K376 with R386, while K195 residue of <I>E. coli</I> was reported to be located in the glucose-1-phosphate (Glc-1-P) substrate binding region.</P>

The Ti-Al-V-N films have been deposited on various substrates by d.c and r.f reactive magnetron sputtering from a Ti-6Al-4V alloy target in mixed Ar-N₂discharges. The films were investigated by means of XRD, AES, SEM/EDX, microhardness, TG and scratch test. The XRD and SEM results indicated that the films were of single B1 NaCl phase having dense columnar structure with the (111) preferred orientation. The composition of Ti-Al-V-N film was the Ti-7.1Al-4.3V-N(wt%) films. Adhesion and microhardness of Ti-Al-V-N films deposited by r.f magnetron sputtering method were better than those deposited by d.c magnetron sputtering method. The anti-oxidation properties of Ti-Al-V-N films were also superior to that of Ti-N film deposited by the same deposition conditions.

<P>A necrotizing retinitis in children is a rare but vision-threatening ocular complication of chickenpox. We report a 10-year-old girl who developed chickenpox 1 month before presenting with panuveitis and necrotizing retinitis. After prompt antiviral treatment, her inflammatory signs were resolved. Early detection and treatment of varicella zoster-associated necrotizing retinitis after chickenpox can achieve good visual outcome.</P>

<P>A crude oil and the coastal sediments that were affected by the Hebei Spirit Oil Spill (HSOS) of Taean, Korea were investigated for thyroid hormone disruption potentials. Water-accommodated fractions (WAFs) of Iranian Heavy crude oil, the major oil type of HSOS, and the porewater or leachate of sediment samples collected along the coast line of Taean were tested for thyroid disruption using developing zebrafish and/or rat pituitary GH3 cells. Major polycyclic aromatic hydrocarbons (PAHs) and their alkylated forms were also measured from the test samples. In zebrafish larvae, significant decreases in whole-body thyroxine (T-4) and triiodothyronine (T-3) levels, along with transcriptional changes of thyroid regulating genes, were observed following 5 day exposure to WAFs. In GH3 cells, transcriptions of thyroid regulating genes were influenced following the exposure to the sediment samples, but the pattern of the regulatory change was different from those observed from the WAFs. Composition of PAHs and their alkylated homologues in the WAFs could partly explain this difference. Our results clearly demonstrate that WAFs of crude oil can disrupt thyroid function of larval zebrafish. Sediment samples also showed thyroid disrupting potentials in the GH3 cell, even several years after the oil spill. Long-term ecosystem consequences of thyroid hormone disruption due to oil spill deserve further investigation.</P>
