<P>A Bacillus species that produces 2,3-butanediol (2,3-BD), termed BRC1, was newly isolated, and a 2,3-BD dehydrogenase (Bdh) from this species was identified and characterized at the molecular and biochemical level. Sequence analysis revealed that Bdh is homologous to D-2,3-BD dehydrogenases. An analysis of the enzymatic properties of Bdh overexpressed in Escherichia coli confirmed the molecular results, showing preferred activity toward D-2,3-BD. Optimum pH, temperature, and kinetics determined for reductive and oxidative reactions support the preferential production of 2,3-BD during cell growth. Overexpression of bdh under the control of a xylose-inducible promoter resulted in increased enzyme activity and enhanced 2,3-BD production in Bacillus sp. BRC1. Additionally, a hydrolysate of cellulosic material, (empty palm fruit bunches), was successfully used for the enhanced production of 2,3-BD in the recombinant Bacillus strain.</P>

강원도 동해시 시멘트공장과 북평산업단지 인근에서 측정한 PM<SUB>2.5</SUB>의 특성

PM<SUB>2.5</SUB> has negative effects on human health and the environment. The major components of PM<SUB>2.5</SUB> are composed of ionic components, carbonaceous components, and trace metallic elements, and ionic and carbonaceous components account for approximately 50% and 30%, respectively. The purpose of this study was to collect PM<SUB>2.5</SUB> and determine its major chemical components in two different functional areas. PM<SUB>2.5</SUB> was collected at Samhwa Elementary School (SES), located near a cement factory in Donghaesi, Gangwon-do and Bukpyeong-dong Administrative Welfare Center (BAWC), located near the industrial complex. NO₃- showed the highest concentration at both locations. Since [NO₃-/SO₄<SUP>2-</SUP>] showed relatively higher values in BAWC than in SES, it was anticipated that PM<SUB>2.5</SUB> in BAWC was more affected by mobile sources than in SES. It was observed that SO₄<SUP>2-</SUP> and NO₃- mainly existed in the form of (NH₄)₂SO₄ and NH₄NO₃ at both sites. In addition, high concentrations of PM<SUB>2.5</SUB> appeared when NH₄+ was deficient, indicating that NH₃ was likely to play an important role in forming secondary NH₄NO₃. At both sites, good correlation between organic carbon (OC) and elemental carbon (EC) was found. Relatively high OC/EC ratios observed at both sites indicate that a significant portion of OC was formed secondarily in atmosphere.

Host plants of the Henosepilachna vigintioctomaculata are known as Solanaceae plants such as potato, tomato, eggplant and Lycium chinense. Henosepilachna vigintioctomaculata (Motschulsky) cause continuous damage from early June to mid-August in leaves of L. chinense under environmental-friendly management in Cheongyang, Chungnam. The developmental period of egg and larva of H. vigintioctomaculata were investigated. H. vigintioctomaculata were reared on L. chinense at several temperatures, for example, 15, 20, 25 and 30°C in the laboratory. In addition, the control effect of 5 eco-friendly agricultural materials were examined. As results, hatching rate is the highest to 91% at 25°C. Egg periods were 10.8, 7.7, 5.1 and 3.7 days at each tested temperature, respectively. And the total developmental periods from egg to adult were 42.7, 26.3, 18.4 and 19.4 days at each tested temperature, respectively. Otherwise, their eclosion rates from pupa to adult were showed 58%, 77%, 85%, 65% at each tested temperature, respectively. As a result of the control effect, both adults and larvae of H. vigintioctomaculata were shown a high mortality to mixture with extract of Sophora flavescens, Chenopodium ambrosioides and Melia azedarach. Additional, larvae is shown high mortality to extract of Azadirachta indica and Cymbopogon nardus. Thus H. vigintioctomaculata might be controled by using plant extract of eco-friendly agricultural materials in due season.
