        멸치젓의 呈味成分에 관한 기초자료를 얻고자 생멸치와 이를 原料로 하여 제조된 멸치젓 중의 核酸關聯物質, 유리아미노산, betaine, TMAO, TMA 및 總creatinine을 分析하였다. 核酸關聯物質 중 생멸치에서는 IMP가 乾物量基準으로 2.8 μmole/g로 가장 많았으나 멸치젓에서는 hypoxanthine이 4.2 μmole/g으로 가장 많았고 , ATP는 흔적량에 불과 하였다. 멸치젓 중의 유리아미노산 중 함량이 많은 것은 leucine, isoleucine, phenylalanine, lysine, tyrosine, alanine, histidine, valine, methionine의 順이었고 glycine, proline, glutamic acid, arginine, threonine, serine 등은 함량이 적었다. 그리고 全유리아미노산은 젓갈 숙성중생원료에 비하여 약 2배로 增加 하였다. Betaine 은 생멸치 및 멸치젓 중에 各各 10.2㎎/100g, 30.0㎎/100/g였으며 이들이 엑스分室素에 대한 比率은 各各 0.6% 및 1.2%였다. TMAO 含量은 생멸치가 13.9㎎/100g, 멸치젓이 9.0㎎/100g였으며 TMA 含量은 各各 22.7㎎/100g 및 30.9㎎/100g였다. 總 creatinine 含量은 생멸치 및 멸치젓에서 各各 433.1㎎/100g, 575.8㎎/100g로서 엑스分室素에 대해 27.2% 및 22.8%였다. Omission test 結果 멸치젓의 呈味成分으로서는 유리아미노산 및 核酸關聯物質이 중요한 구실을 한다는 것을 알았다. This study was conducted to evaluate the taste compounds of fermented anchovy. The contents of such compounds as nucleotides and their related compounds, free amino acids, betaine, TMAO and total creatinine raw and fermented anchovy were analysed. The contents of IMP in raw anchovy appeared higher than other nucleotides and tended to decrease rapidly during fermentation. Therefore, in the case of fermented anchovy, hypoxanthine was abundant and IMP was very low in content. In the free amino acid composition of fresh samples abundant amino acids were leucine, isoleucine, phenylalanine, lysine, tyrosine, alanine, histidine, valine, methionine in order. Such amino acids as glycine, proline, glutamic acid, arginine, threonine and serine were poor in content. The contents of total free amino acids reached approximately about two times as compared with that of raw sample. The amount of betaine and TMA increased, while TMAO decreased during fermentation. The content of total creatinine in raw and fermented anchovy was occupied 22.8% and 27.2% of total of extractive nitrogen, respectively. As the results of omission test, the taste-active compounds of fermented anchovy are assumed to be free amino acids and nucleotides.

        Background: The purpose of this project is to elucidate the effect of dexamethasone on hypoxia-induced EMT in colon cancer cells. Methods: Human colon cancer HCT116 and HT29 cells were exposed to normoxic (21%) or hypoxic (1%) conditions. In order to measure the expression levels of EMT markers (Snail, Slug, E-cadherin, integrin aVß6) treated with dexamethasone, quantitative real-time polymerase chain reaction and Western blot analysis were performed. Furthermore, effect of dexamethasone on the invasiveness and migration of colon cancer cells was elucidated using matrigel invasion and wound healing migration assays. Results: We found that dexamethasone inhibits HIF-1a protein level and its downstream gene, vascular endothelial growth factor (VEGF) mRNA level, in hypoxic condition. In addition, dexamethasone down-regulates not only hypoxia-induced Snail and Slug mRNA levels, which play a part in transcriptional level of EMT, but also hypoxia- induced integrin aVß6 protein level, a well-known EMT marker for colon cancer cells. Furthermore, reduced E-cadherin in hypoxic condition is found to be recoverable by treating with dexamethasone in colon cancer cell lines; dexamethasone blocks the migration and invasion of cancer cells in hypoxia. Conclusions: Our study suggests that dexamethasone has inhibitory effects on cell migration and invasion by suppressing EMT of colon cancer cell lines in hypoxic condition and may serve a new chemotherapeutic agent for colon cancer patients.

      • 석병산지구 태백산분지 조선누층군 원평층의 퇴적학적 연구

        석병산지구에 분포하는 원평층은 주로 충식상의 석회암과 암갈색의 세립질 사암, 암회색의 셰일, 회색 슬레이트로 구성된다. 이러한 원평층은 1962년 태백산지구 지하자원조사단의 연구에서 처음으로 층서명이 제시된 바 있다. 원평층은 태백층군에 속하나 전형적인 태백층군의 층서명을 따르지 않았는데, 본 연구는 이에 대한 궁금점에서 기인하여 시작되었으며 전형적인 태백층군의 층서들과 원평층의 암층서의 특징을 비교하고자 한다. 이 연구에서는 석병산지구에 위치한 등갈산(794 m)의 능선을 따라 노출된 노두를 조사하고, 암상을 구분하여 퇴적환경을 해석하고자 하였다. 원평층의 최하부 층원은 세립질 괴상의 녹색 사암과 유백색의 괴상 입자암이 호층을 이루며 나타나고, 하부 층원은 괴상의 암회색 석회암으로 구성된다. 중부 층원은 적색 내지 녹색의 중립질 층리 사암으로 관찰되며, 최상부 층원은 유백색의 엽층리 석회암 및 유백색의 석회암과 암갈색의 셰일이 호층을 이루고 있다. 최하부 층원은 세립질 괴상 사암과 입자암의 호층 및 엽층리 석회암이 특징인 중부 내지 내부 완사면의 환경으로, 하부 층원은 엽층리 석회암 및 유백색의 석회암과 암갈색의 셰일이 호층, 괴상의 석회암이 특징적인 얕은 조하대 환경으로 해석하였다. 중부 층원에서 나타나는 중립질 층리 사암의 적색 내지 녹색의 색상은 근해 환경을 지시한다. 최상부 층원은 유백색 엽층리 및 괴상의 석회암 특징이 나타나 조하대 환경으로 해석하였다. 이러한 해석을 종합하여 전형적인 태백층군과 비교해본 결과, 원평층의 각 층원은 세송층부터 두무골층에 나타나는 퇴적학적 특징과 유사하다고 판단하였다. 따라서 석병산지구의 원평층이 전형적인 태백층군보다 얕은 환경에서 퇴적되었다고 해석하였다.

        Clinical Features of Acute Subdural Hematomas Caused by Ruptured Intracranial Aneurysms

        Objective : Spontaneous acute subdural hematomas (aSDH) secondary to ruptured intracranial aneurysms are rarely reported. This report reviews the clinical features, diagnostic modalities, treatments, and outcomes of this unusual and often fatal condition. Methods : We performed a database search for all cases of intracranial aneurysms treated at our hospital between 2005 and 2010. Patients with ruptured intracranial aneurysms who presented with aSDH on initial computed tomography (CT) were selected for inclusion. The clinical conditions, radiologic findings, treatments, and outcomes were assessed. Results : A total of 551 patients were treated for ruptured intracranial aneurysms during the review period. We selected 23 patients (4.2%) who presented with spontaneous aSDH on initial CT. Ruptured aneurysms were detected on initial 3D-CT angiography in all cases. All ruptured aneurysms were located in the anterior portion of the circle of Willis. The World Federation of Neurosurgical Societies grade on admission was V in 17 cases (73.9%). Immediate decompressive craniotomy was performed 22 cases (95.7%). Obliteration of the ruptured aneurysm was achieved in all cases. The Glasgow outcome scales for the cases were good recovery in 5 cases (21.7%), moderate disability to vegetative in 7 cases (30.4%), and death in 11 cases (47.8%). Conclusion : Spontaneous aSDH caused by a ruptured intracranial aneurysm is rare pattern of aneurysmal subarachnoid hemorrhage. For early detection of aneurysm, 3D-CT angiography is useful. Early decompression with obliteration of the aneurysm is recommended. Outcomes were correlated with the clinical grade and CT findings on admission.

      • Large Deformation Formulation of a Hypoelasto-plastic Constitutive Model for Soils

        A constitutive equation was implemented in order to model the behavior in overall ranges from small to large strains, which is based on anisotropic hardening rule and total stress concept. The constitutive model was implemented in ABAQUS code in which large deformation analysis can be performed accurately and efficiently. The formulation includes (1) finite strain plasticity on the basis of Jaumann stress rate, (2) implicit stress integration and (3) consistent tangent moduli. A large deformation analysis was performed with the constitutive model using ABAQUS program. In the analysis of an actual embankment, it was found that the proposed model was formulated accurately and efficiently. 미소변형에서 대변형에 이르는 전변형도 영역의 거동을 모델하기 위하여 비등방 경화규칙과 전응력개념에 의거한 구성관계를 이용하였다. 이러한 구성관계는 ABAQUS 코드에 구현되었으며 대변형해석시 정확도와 수렴속도를 확보하도록 하였다. 이 정식화는 Jaumann 응력속도에 의거한 유한 변형도 소성론, 내재적 응력적분, 일관된 접선계수를 포함한다. ABAQUS를 이용한 대변형해석을 통하여 알고리즘의 정확도 및 수렴도 해석을 할 수 있었다.

      • Heterogeneous surface saponification of suspension-polymerized monodisperse poly(vinyl acetate) microspheres using various ions

        <P>Poly(vinyl alcohol) (PVA)/poly(vinyl acetate) (PVAc) microspheres with a skin/core structure were prepared through the heterogeneous surface saponification of PVAc microspheres suspension-polymerized. The PVA skin formed through the heterogeneous saponification was hydrogel swellable in water. In addition, to obtain monodisperse PVA/PVAc microspheres having various skin/core ratios and morphologies, the ion-specificities to the heterogeneous saponification were investigated using SO<EM><SUB>4</SUB><SUP>2−</SUP></EM>, Cl<SUP>−</SUP>, NO<EM><SUB>3</SUB><SUP>−</SUP></EM>, Br<SUP>−</SUP>, and I<SUP>−</SUP> for anions and Li<SUP>+</SUP>, Na<SUP>+</SUP>, and K<SUP>+</SUP> for cations, respectively. The ions were not specific significantly to the rate of the heterogeneous saponification, while were related to the degree of saponification (DS). DSs had different values between by weight loss (DS<SUB>w</SUB>) and by proton nuclear magnetic resonance spectroscopy (DS<SUB>NMR</SUB>) measurements. The order of DS<SUB>w</SUB>s was SO<EM><SUB>4</SUB><SUP>2−</SUP></EM> < Cl<SUP>−</SUP> < NO<EM><SUB>3</SUB><SUP>−</SUP></EM> < Br<SUP>−</SUP> < I<SUP>−</SUP> for anions and K<SUP>+</SUP> < Na<SUP>+</SUP> < Li<SUP>+</SUP> for cations, and that of DS<SUB>NMR</SUB>s, I<SUP>−</SUP> < Br<SUP>−</SUP> < NO<EM><SUB>3</SUB><SUP>−</SUP></EM> < Cl<SUP>−</SUP> < SO<EM><SUB>4</SUB><SUP>2−</SUP></EM> for anions and Li<SUP>+</SUP> < Na<SUP>+</SUP> < K<SUP>+</SUP> for cations. The differences in values between DS<SUB>w</SUB>s and DS<SUB>NMR</SUB>s were caused by the dissolution of PVA skin and were significantly decreased for SO<EM><SUB>4</SUB><SUP>2−</SUP></EM>.
