        Efficient Interleukin-21 Production by Optimization of Codon and Signal Peptide in Chinese Hamster Ovarian Cells

        Interleukin-21 is a common γ-chain cytokine that controls the immune responses of B cells, T cells, and natural killer cells. Targeting IL-21 to strengthen the immune system is promising for the development of vaccines as well as anti-infection and anti-tumor therapies. However, the practical application of IL-21 is limited by the high production cost. In this study, we improved IL-21 production by codon optimization and selection of appropriate signal peptide in CHO-K1 cells. Codon-optimized or non-optimized human IL-21 was stably transfected into CHO-K1 cells. IL-21 expression was 10-fold higher for codon-optimized than non-optimized IL-21. We fused five different signal peptides to codon-optimized mature IL-21 and evaluated their effect on IL-21 production. The best result (a 3-fold increase) was obtained using a signal peptide derived from human azurocidin. Furthermore, codon-optimized IL-21 containing the azurocidin signal peptide promoted IFN-γ secretion and STAT3 phosphorylation in NK-92 cells similar to codon-optimized IL-21 containing original signal peptide. Collectively, these results indicate that codon optimization and azurocidin signal peptides provide an efficient approach for the high-level production of IL-21 as a biopharmaceutical.

        배추의 형질전환용 선발항생제로서 Paromomycin의 이용

        정상' 배추와 '서울' 배추의 배축절편을 선발마커로서 hygromycin 저항성유전자를 갖고 있는 pCAMBIA1301와 paromomycin저항성유전자를 갖고 있는 pPTN290으로 각각 형질 전환된 LBA4404 또는 EHA101균주와 공동배양한 후 선발배지에서 배양하면서 형질전환체를 선발하였다. 형질전환빈도는 사용된 항생제와 품종에 따라서 현저하게 차이가 있었으며, 특히 paromomycin은 hygromycin보다 효과적이었고 정상 배추는 서울배추보다 양호하였다. 가장 높은 형질전환빈도는 (0.70%) 100mg/L paromomycin이 첨가된 선발배지에서 정상배추의배축을 배양할 경우 얻어졌다. GUS양성반응으로 확인 한 결과 정상배추에서 9개체와 서울배추에서 3개체를 각각 얻었으며, 온실에서 생장한 후 $T_1$종자를 수확하였다. $T_1$ 종자를 다시 발아시켜 유식물체를 얻은 후 GUS양성반응을 확인함으로서 외래유전자가 안정적으로 발현하고 있음을 확인하였다. Hypocotyl explants of Chinese cabbage (us. 'Jeong Sang' and 'Seoul') produced adventitious shoots on Murashige and Skoog (MS) basal medium supplemented with 4mg/L $AgNO_3$, 5 mg/L acetosyringone, 4 mg/L 6-benzyladenine and 3mg/L alpha-naphthaleneacetic acid (SI) after cocoultivation with strains of Agrobacterium tumefaciens (LBA4404) harboring the pCAMBIA1301 and the $_PPTN290$ containing hygromycin-resistance gene and paromomycin-resistance gene as a selectable marker genes, respectively. There was a significant difference in the frequency of transgenic plants depending on antibiotics and cultivars used. Paromomycin was better than hygromycin, and cultivar 'Jeong-sang' was higher than 'c.v. Seoul' in the frequency of transgenic plants. In particular, the highest frequency (0.70%) of transgenic plants was obtained from selection medium (SI) containing 100mg/L paromomycin in c.v., 'Jeong-sang' GUS positive response were obtained 9 plants and 3 plants from the cultivars, 'Jeong-sang' and 'Seoul', respectively. They were grown to maturity in a greenhouse and normally produced $T_1$ seeds. GUS histochemical assay for progeny $(T_1)$ revealed that the transgenes were expressed in the plant genome.

      • 동종 반월상 연골 이식술의 임상적 결과 - 내측과 외측 및 동반 손상의 유무에 따른 비교 -

        목적: 동종 반월상 연골 이식술의 결과를 내측과 외측 및 동반손상 유무에 따라 비교하고자 하였다. 대상 및 방법: 1999년 12월부터 2007년 6월까지 동종 반월상 연골 이식술을 시행하고 1년이상 추시가 가능했던 52례를 대상으로 하였다. 내측 이식군은 19례, 외측 이식군은 33례였고, 단독 손상군은 18례 동반 손상군은 34례였다. 평균나이는 34.2세(18세~51세) 평균 추시기간은 42.2개월(12개월~90개월)이었다. 임상적 결과의 판정을 위하여 슬관절 운동범위, VAS 점수, IKDC 주관적 평가 점수, Lysholm 점수, Tegner 점수, 환자 만족도 및 이차 관절경 또는 추시 MRI를 시행하였다. 결과: 수술 후 평균 관절운동 범위는 130.3도 였다. VAS 점수는 수술 전 5.96점에서 3.05점으로, IKDC 주관적 평가 점수 46.5점에서 64.52점으로(p<0.001), Lysholm 점수는 61.94점에서 79.58점으로(p=0.0019) Tegner점수는 수술 전 2.97에서 3.62점으로 향상되었으며 수술의 만족도는 '매우만족', '대체로 만족'이라고 대답한경우가 61.5% 였다. 수술 후임상적 점수의 향상이 있었지만 내측과 외측 이식술 군간 그리고 단독 손상군과 동반 손상군 간의 통계적 유의한 차이는 없었다. 이차 관절경을 시행한 18례중 10례에서 이식된 동종 반월상 연골의 변연부의 치유 소견이 관찰되었고 6례에서 부분파열이 2례에서는 복합 파열이 관찰되었다. 추시 MRI 촬영이 가능하였던 16례에서 아탈구는 12례에서 관찰되었다. 결론: 반월상 연골 이식술 후 내측과 외측 이식군 간의 임상적 결과는 차이가 없었으며 동반된 연골 손상이나 불안정성에 대한 치료를 병행한 경우 동반손상의 유무는 결과에 영향을 미치지 않았다. Purpose: To compare the clinical outcomes after meniscus allograft transplantation between lateral and medial or isolated and combined procedure groups. Materials and Methods: Of the patients who had undergone arthroscopic meniscal allograft transplantation between Dec. 1997 and Jun. 2007, 52 patients were available for retrospective evaluation. Patients were grouped into lateral(33 cases) and medial(19 cases) transplant groups as well as those with isolated(18 cases) and combined(34 cases) procedure. The average age was 34.2 years and the mean follow-up period was 42.2 months. Postoperative range of motion (ROM), visual analog scale (VAS), International Knee Documentation Committee (IKDC) subjective score, Lysholm score, Tegner score, patient's subjective satisfaction, $2^{nd}$ look arthroscopy and MRI were evaluated retrospectively. Results: Mean postoperative ROM was $130.3^{\circ}$. The VAS showed an improvement from 5.96 to 3.05 at the last follow up. IKDC subjective score and Lysholm score also showed an improvement from 46.5 to 64.5 and from 61.9s to 79.58 respectively. Tegner score was improved from 2.9 to 3.6. Overall, 61.5% of patients reported they were completely or mostly satisfied with procedure. There were no significant differences noted between lateral and medial groups as well as isolated and combined groups. In 2nd look arthroscopy, 10 of 18 cases showed good pheripheal healing and there were 6 cases of partial and 2 of complex tear. We observed graft subluxation or extrusion in 12 of 16 cases who were evaluated with follow-up MRI. Conclusion: Meniscus allograft transplantation alone or in combination with other procedure showed an improvement in knee pain and clinical score. But there were no significant difference between lateral and medial groups or isolated and combined procedure groups.

        Variation in IR and Raman Spectra of CD3CN upon Solvation of InCl3 in CD3CN: Distinctive Blue Shifts, Coordination Number, Donor-Acceptor Interaction, and Solvated Species

        Notable blue shifts of the ν2 C≡N stretching, ν4 C-C stretching and ν8 CCN deformation bands of CD3CN are observed upon solvation of InCl3, resulting from the donor-acceptor interaction. The Raman spectrum in the ν2 region shows further details; at least two new bands emerge on the blue side of the ν2 band of free CD3CN, whose relative intensities vary with concentration, suggesting that there exist at least two different cationic species in the solution. The strong hydrogen bonds formed between the methyl group and InCl4 − result in a large band appearing on the red side of the ν1 CD3 symmetric stretching band. The solvation number of InCl3, determined from the Raman intensities of the C≡N stretching bands for free and coordinated CD3CN, increases from ~1.5 to ~1.8 with decreasing concentration.

        Establishment of a Resource Population of SLA Haplotype-Defined Korean Native Pigs

        The highly polymorphic porcine major histocompatibility complex (MHC), or the swine leukocyte antigens (SLA), has been repeatedly associated with variations in swine immune response to pathogens and vaccines as well as with production traits. The SLA antigens are also important targets for immunological recognition of foreign tissue grafts. We recently established a resource population of Korean native pigs as models for human transplantation and xenotransplantation research. In this study, 115 animals derived from three generations of the Korean native pigs were genotyped for three SLA class I (SLA-2, SLA-3 and SLA-1) and three SLA class II loci (DRB1, DQB1, DQA) using PCR with sequence-specific primers (PCR-SSP) at the allele group resolution. A total of seven SLA haplotypes (Lr-5.34, Lr-7.23, Lr-31.13, Lr-56.23, Lr-56.30, Lr-59.1, Lr-65.34), comprising six unique class I and five unique class II haplotypes, were characterized in the founding animals. Class I haplotype Lr-65.0 and class II haplotype Lr-0.34 were novel; and together with Lr-56.0 these haplotypes appeared to be breed-specific. In the progeny population, Lr-7.23 and Lr-56.30 appeared to be the most prevalent haplotypes with frequencies of 34.7% and 31.6%, respectively; the overall homozygosity was 27.4%. This resource population of SLA-defined Korean native pigs will be useful as large animal models for various transplantation and xenotransplantation experiments, as well as for dissecting the roles of SLA proteins in swine disease resistance and production traits.

        A Study on the Synthesis of Organophilic [TEACOOH]-Montmorillonite Intercalations Complex and its Swelling Properties

        In this research an organic cation, [TEACOOH] Br, with a long alkyl chain was synthesized which will replace the metal ion between the layers of Na-Montmorillonite and an organophilic [TEACOOH]-Montmorillonite intercalations complex was formed by the cation exchange reaction between the Na-Montmorillonite and the synthesized [TEACOOH] Br. After drying of this intercalations complex in high vacuum we have tried to experiment on the probability whether it will form complexes with various swelling solutions such as dist. water, methano, ethanol, toluene, acetonitrile and propionitrile and the corresponding basal sp acings measured were $17.41{\AA}$, $31.90{AA}$, $34.42{AA}$, $30.52{AA}$ and $32.36{AA}$, respectively.

        Effect of Dietary Selenium on the Colon Carcinogenesis in Male ICR Mice

        본 연구에서는 azoxymethane (AOM)과 dextran sodium sulfate (DSS)로 유도된 대장 발암과정에 대한 셀레늄의 방어 효과를 조사하였다. 셀레늄 결핍(0.02 ppm Se), 정상(0.1 ppm Se), 과다(0.5 ppm Se)사료를 12주간 식이로 급여하여 혈액검사와 대장암 발생의 초기단계인 aberrant crypt foci (ACF)수를 측정했으며, 암 발생율을 조사하였다. ICP-AES를 사용하여 간의 셀레늄 농도를 측정하였으며, 또한 셀레늄포함 항산화효소인 glutathione peroxidase (GPx) 활성을 알아보았다. 또한 TUNEL assay와 PCNA, $\beta$-catenin에 대한 면역조직 염색을 수행하였다. ACF 수 및 종양 발생률에 있어서, 셀레늄과다사료를 급여한 군이 정상셀레늄사료를 급여한 군보다 낮았으며, 셀레늄결핍사료를 급여한 군은 오히려 ACF 수 및 종양 발생률이 높았다. GPx 활성은 셀레늄의 섭취가 과다한 군에서 높게 나타났으며, 이 때, TUNEL에서 apoptotic positive cell이 증가하는 것을 확인했다. 또한 셀레늄의 섭취가 과다한 군에서 PCNA와 $\beta$-catenin의 발현이 감소됨을 볼 수 있었다. 본 마우스 모델실험에서 셀레늄은 여러 기전에 의해 대장암 발생을 억제할 수 있을 것으로 사료된다. Selenium is an essential micronutrient for normal body function and functions as an essential constituent of selenoproteins. This study was carried out to investigate effect of selenium on the formation of colonic aberrant crypt foci (ACF) and tumor formation in a mouse model. Five-week old ICR mice were acclimated for one week and fed different selenium diet (0.02, 0.1, and 0.5 ppm) for 12 weeks. Animals received three intraperitoneal injections of azoxymethane (10 mg/kg B.W. in saline for 3 weeks), followed by 2% dextran sodium sulfate in the drinking water for a week. There were four experimental groups, including a normal control group and three different selenium levels groups. After sacrifice, the total numbers of aberrant crypt (AC) and ACF were measured in the colonic mucosa after methylene blue staining. The number of tumors was noted for tumor incidence. Liver selenium concentration was measured using ICP-AES method. Gutathione peroxidase (GPx) activity was determined using a GPx assay kit in the liver and colon. TUNEL assay and proliferating cell nuclear antigen (PCNA) staining were performed to examine the cell apoptosis and cell proliferation, respectively. Immunohistochemistry of $\beta$-catenin was also performed on the mucous membrane tissue of colon. The activity of GPx in the liver and colon was decreased in the selenium-deficient diet group while it was increased in the selenium-overloaded diet group. Apoptotic positive cells were increased in the selenium-overloaded diet group but decreased in the selenium-deficient diet group. PCNA staining area was decreased in the selenium-overloaded diet group. In addition, the $\beta$-catenin protein level in the selenium-deficient diet group was increased but decreased in the selenium-overloaded diet group. These results indicate that dietary selenium might exert a modulating effect on colon cancer by inhibiting the development of ACF and colon tumor formation in this mouse model.

        Prediction of Cohesive Sediment Transport and Flow Resistance Around Artificial Structures of the Beolgyo Stream Estuary

        To predict changes in the marine environment of the Beolgyo Stream Estuary in Jeonnam Province, South Korea, where cohesive tidal flats cover a broad area and a large bridge is under construction, this study conducted numerical simulations involving tidal flow and cohesive sediment transport. A wetting and drying (WAD) technique for tidal flats from the Princeton Ocean Model (POM) was applied to a large-scale-grid hydrodynamic module capable of evaluating the flow resistance of structures. Derivation of the eddy viscosity coefficient for wakes created by structures was accomplished through the explicit use of shear velocity and Chezy's average velocity. Furthermore, various field observations, including of tide, tidal flow, suspended sediment concentrations, bottom sediments, and water depth, were performed to verify the model and obtain input data for it. In particular, geologic parameters related to the evaluation of settling velocity and critical shear stresses for erosion and deposition were observed, and numerical tests for the representation of suspended sediment concentrations were performed to determine proper values for the empirical coefficients in the sediment transport module. According to the simulation results, the velocity variation was particularly prominent around the piers in the tidal channel. Erosion occurred mainly along the tidal channels near the piers, where bridge structures reduced the flow cross section, creating strong flow. In contrast, in the rear area of the structure, where the flow was relatively weak due to the formation of eddies, deposition and moderated erosion were predicted. In estuaries and coastal waters, changes in the flow environment caused by artificial structures can produce changes in the sedimentary environment, which in turn can affect the local marine ecosystem. The numerical model proposed in this study will enable systematic prediction of changes to flow and sedimentary environments caused by the construction of artificial structures.
