      • Inhibition by Imatinib of Expression of O-glycan-related Glycosyltransferases and Tumor-associated Carbohydrate Antigens in the K562 Human Leukemia Cell Line

        Objective: To study changes of tumor associated carbohydrate antigen (TACAs) expression and mRNA levels for tumor associated glycosyltransferases, and assess subcellular localizations of N-acetyl galactosyltransferases (GalNAc-Ts) in the K562 leukemia cell line after imatinib treatment. Methods: RT-PCR was performed to analyze the expression of glycosyltransferases which synthesize O-glycan in tumor-associated carbohydrate antigens (TCTAs). The expression of Tn antigen, T antigen and sialyl T antigen on K562 cell membranes was measured by flow cytometry after treatment with different concentrations of imatinib. Co-localization of GalNAc-Ts and ER (endoplasmic reticulum) was determined by confocal laser scanning microcopy. Results: Transcript expression levels of several glycosyltransferases related to TCTAs were decreased after imatinib ($0-0.3{\mu}M$) treatment. Expression of Tn antigen and T antigen was increased while that of sialyl T antigen was decreased. Co-localization of GalNAc-Ts and ER was reduced by $0.2{\mu}M$ of imatinib. Conclusion: Imatinib inhibited the expression of O-glycan related TACAs and several related glycosyltransferases, while decreasing the co-localization of GalNAc-Ts and ER and normalizing O-glycosylation in the K562 human leukemia cell.

      • ANXA2 Regulates the Behavior of SGC-7901 Cells

        ANXA2, a member of the annexin family, is overexpressed and plays important roles in tumor development. However, the significance of ANXA2 expression in gastric carcinoma has not been clarified.To elucidate its roles in growth of gastric cancer, ANXA2 expression in SGC-7901 cells was inhibited with a designated siRNA, then cell proliferation, cell cycling, apoptosis and motility were determined by MTT assay, flow cytometry, Hoechst 33342 staining and wound healing assay, respectively. To further assess the behavior of ANXA2 deleted SGC-7901 cells, changes of microstructures were observed under fluorescence microscopy, laser scanning confocal microscopy and electron microscopy. We found that inhibition of ANXA2 expression caused cell proliferation to decrease significantly with G1 arrest, motility to be reduced with changes in pseudopodia/filopodia structure and F-actin and ${\beta}$-tubulin expression, and apoptosis to be enhanced albeit without significance. At the same time, ANXA2 deletion resulted in fewer pseudopodia/filopodia, non-stained areas were increased, contact inhibition among cells reappeared, and expression of F-actin and ${\beta}$-tubulin was decreased, with induction of polymerized disassembled forms. Taken together, these data suggest that ANXA2 overexpression is important to maintain the malignancy of cancer cells, and this member of the annexin family has potential to be considered as a target for the gene therapy of gastric carcinoma.

        Bias load analysis and asymmetric modification of rollers in the main drive bearing of large roadheader

        An asymmetric modification method for the roller profile was proposed to address the bias load effect of rollers in the main drive bearing of a large roadheader. A contact mechanics model of the bearing was solved, and line contact theory was used to explore the effect of combined load and convexity on the bias load effect. The contact stress was found to be asymmetrically distributed along the contact line. This trend tended to be remarkable with the decrease in axial load or the increase in overturning moment. Symmetrically modified rollers can be used to improve the bias load effect by increasing convexity. However, this process led to the contact stress concentrating in the roller center. The influence of the design parameters for asymmetric modification on the roller profile and the contact stress was considered. The ideal design parameters were determined by using a genetic algorithm. The contact condition and fatigue life were remarkably increased.

        Overexpression of miR-155-5p Inhibits the Proliferation and Migration of IL-13-Induced Human Bronchial Smooth Muscle Cells by Suppressing TGF-β-Activated Kinase 1/MAP3K7-Binding Protein 2

        Purpose: Molecular mechanisms leading to asthma is still ill-defined. Though the function of microRNAs (miRNAs) in asthma was previously reported, the involvement of miR-155 in important features of this disease remains unknown. The present study was designed to uncover the probable involvement of miR-155-5p in the proliferation and migration of IL-13-induced human bronchial smooth muscle cells (BSMCs) and the intrinsic regulatory mechanism. Methods: The effects of different concentrations of IL-13 on the proliferation and migration of BSMCs as well as the expression of miR-155-5p and its predicted target transforming growth factor (TGF)-β-activated kinase 1/MAP3K7-binding protein 2 (TAB2) were investigated. The effects of miR-155-5p on the proliferation and migration of interleukin (IL)-13-induced BSMCs was determined in vitro using BSMCs transfected with miR-155 mimic/inhibitor and induced by a high concentration of IL-13. The quantitative real-time polymerase chain reaction (qRT-PCR) was employed for determining the expression of miR-155-5p and TAB2. Western blotting was applied to analyze the expression of TAB2 at the protein level. Cell proliferation and migration were assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and Transwell assays, respectively. Results: The proliferation and migration of BSMCs were dose-dependently increased with IL-13 treatment. Contrariwise, IL-13 dose-dependently inhibited the expression of miR-155-5p in BSMCs. Mechanistic studies showed that inhibition of miR-155-5p further promoted the stimulatory effects of IL-13, whereas overexpression of miR-155 significantly inhibited these effects. In silico studies and luciferase reporter assays indicated that TAB2 was a negatively regulated miR-155-5p target. Conclusions: These results suggested that miR-155-5p-inhibit the IL-13-induced proliferation and migration of BSMCs by targeting TAB2 and that the IL-13/miR-155/TAB2 pathway could serve as a therapeutic target for pulmonary diseases, especially asthma.

        Inhibitors of Bone Resorption from Halenia corniculata

        Eleven xanthones (1-11), three flavonoids (12-14) and three secoiridoids (15-17) were isolated from the aerial parts of Halenia corniculata. Among those compounds, 1-hydroxy-2,3,4,5- tetramethoxyxanthone (1), 1-hydroxy-2,3,4,7-tetramethoxyxanthone (2), 1-hydroxy-2,3,4,5,7-pentamethoxyxanthone (3), 1-hydroxy-2,3,5-trimethoxyxanthone (4), 1,8-dihydroxy-3,5-dimethoxyxanthone (7), and luteolin (12), at the concentration of 1 μg/mL, effectively inhibited the osteoclast differentiation in a co-culture system with mouse osteoblastic calvarial cells and bone marrow cells. Notably, compounds 1, 3, and 4 exhibited, in a dose-dependent manner, significant inhibition of osteoclast differentiation even at a low concentration (0.01 μg/mL). All the inhibitory compounds, except for compound 7, significantly reduced the pit formation on the dentine slice compared with the control group. For the survival of the mature osteoclasts, compounds 1-4 and 12 (1 μg/mL), significantly decreased the survival number through induction of cell apoptosis, and compound 4 exhibited a significant inhibitory effect on osteoclast survival even at the low concentration of 0.1 μg/mL.

        Active Speed Control of Belt Conveyor with Variable Speed Interval Based on Fuzzy Algorithm

        Aiming at the problems of too fast adjustment frequency, unstable material flling rate and fuctuation of material fow at the boundary of speed interval caused by unreasonable setting of speed interval in traditional active speed control strategy based on hierarchical speed regulation. An active speed control strategy based on fuzzy algorithm is proposed for belt conveyor. The material flling rate of the belt is taken as input, and the double-layer fuzzy boundary is set according to the expected value of the material flling rate to adjust the speed of the belt conveyor. The adjustment time of diferent speed intervals is given according to the estimation-calculation-optimization method. The simulation results of the double belt conveyor system show that compared with the active speed control strategy based on hierarchical speed regulation, the active speed control strategy can not only stabilize the material flling rate of the downstream belt conveyor near the expected value and improve the stability of transportation, but also reduce the energy by about 20.69% per hour on average. The subsequent experimental results also verify the efectiveness of the proposed active speed control strategy, which can achieve more sustainable operation of the belt conveyor.

        N-doped porous carbons with increased yield and hierarchical pore structures for supercapacitors derived from an N-containing phenyl-riched copolymer

        N-doped porous carbon-based materials (NPCMs) with hierarchical pore structures have beenconsidered to be a suitable alternative to meet the ever-increasing demands for supercapacitors;however, the universally low yield of the NPCMs has restricted their practical applications. Herein, aseries of NPCMs with hierarchical pore structures are synthesized with significantly increased yieldsthrough the carbonization of the copolymer made from 2,4,6-tris(chloromethyl)mesitylene and pphenylenediamine. The development of the hierarchical pore structures and the N content of the NPCMsshow opposite dependences on the increasing carbonization temperature. The NPCM exhibits the bestcapacitive ability only if the sufficiently developed hierarchical structures and moderate N content areachieved simultaneously. Therefore, NPCM-600 that is carbonized at 600 C with an excellent yield of53.6% (wt.), large specific surface area of 1778 m2 g 1, and N content of 4.13% (wt.) yields an ideal specificcapacitance of 298 F g 1 at the current density of 1 A g 1 and a perfect cycling stability of the capacitanceafter 10,000 cycles at 10 A g 1. The yield of the NPCM-600 is considerably higher than those for manyother recently reported NPCMs. NPCM-600 also shows better capacitance than those of the otherreported NPCMs, such as NOPC-bis-CN-3 (167 F g 1) and CHCPB-K-600 (260 F g 1).

        A Case of Simultaneously Diagnosed Lung Adenocarcinoma and Endobronchial Inflammatory Myofibroblastic Tumor with Two Distinct Types of ALK Translocation

        A 61-year-old male patient was simultaneously diagnosed with lung adenocarcinoma and inflammatory myofibroblastic tumor (IMT). The lung adenocarcinoma and IMT harbored two distinct types of ALK translocation, LOC101927285-ALK, and TPM3-ALK, respectively. The ALK Ventana showed strong positivity on both lesions. The patient was therefore given an endobronchial cryotherapy and ALK inhibitor crizotinib. The tumors showed durable response however the left lung adenocarcinoma relapsed at 17th month post-crizotinib treatment. Tissue re-biopsy on the resistant tumor revealed an ALK exon 23 C1156Y missense mutation in addition to LOC101927285-ALK mutation. Further RNA-based sequence uncovered that the noncoding region rearrangement is the fusion mutation of EML4-ALK. The patient was therefore received alectinib, and the tumor exhibited partly response. Overall, it is very rare that two types of pulmonary tumors exist in one patient driven by two distinct ALK fusions, which emphasizes the necessity of gene sequencing in clinical decision-making and individualized therapy.
