        Determination of Heparin Using Norfloxacin-cerium Complex as a Fluorescence Probe by Spectrofluorimetry

        A simple, rapid, practical and sensitive spectofluorimetric method was developed for the determination of trace amount of heparin (Hep). Under the Optimum conditions, we studied the interaction between NFLX-Ce$^{3+}$-Hep complex by using absorption and fluorescence spectra. It was observed that Hep remarkably enhance the fluorescence intensity of the NFLX-Ce$^{3+}$ complex at ${\lambda}$= 356 nm in the buffer solution of pH = 7.60 and the enhancement effect is shown to relate with the concentration of Hep. The linear range and detection limit for the determination of Hep was obtained. By the Rosenthal graphic method, the association constant (K) and binding numbers (N) of Hep with probe were investigated. This method is relatively free of interference from coexisting substances and successfully applied for the determination of heparin in heparin sodium injection samples. A suitable mechanism of fluorescence enhancement between NFLX-Ce$^{3+}$ and the NFLX-Ce$^{3+}$-Hep systems were proposed and discussed.

        Structural studies of Mn doped ZnO nanoparticles

        Mn substituted ZnO nanoparticles with compositional formula Zn_(1-x)Mn_xO where x = 0.00, 0.04 and 0.08were synthesized by sol-gel route. All the samples sintered at 650 ℃ for 12 h in a furnace followed by furnace cooling up to room temperature. X-ray diffraction (XRD) studies shows the presence of hexagonal crystal structure as same as parent compound (ZnO) in all samples. The lattice parameters ‘a’ and ‘c’were determined from XRD data and found that they increase linearly with the Mn content, which suggests that doped Mn ions go to Zn sites. Grain size, X-ray density and Atomic packing fraction (APF)were evaluated using XRD data and found that grain size increases while X-ray density and Atomic packing fraction (APF) decreases with dopant concentration increases. The functional groups and chemical interactions of Mn substituted Zinc oxide samples were also determined at various peaks using FTIR data and observed the presence of function groups in the samples. Results of such an investigation presented in this paper.

        Investigation of Single Nucleotide Polymorphisms in Porcine Chromosome 2 Quantitative Trait Loci for Meat Quality Traits

        Several studies have reported quantitative trait loci (QTL) for meat quality on porcine chromosome 2 (http://www.animalgenome.org/QTLdb/pig.html). For application of the molecular genetic information to the pig industry through marker-assisted selection, single nucleotide polymorphism (SNP) markers were analyzed by comparative re-sequencing of polymerase chain reaction (PCR) products of 13 candidate genes with DNA from commercial pig breeds such as Berkshire, Yorkshire, Landrace, Duroc and Korean Native pig. A total of 34 SNPs were identified in 15 PCR products producing an average of one SNP in every 253 bp. PCR restriction fragment length polymorphism (RFLP) assays were developed for 11 SNPs and used to investigate allele frequencies in five commercial pig breeds in Korea. Eight of the SNPs appear to be fixed in at least one of the five pig breeds, which indicates that different selection among pig breeds might be applied to these SNPs. Polymorphisms detected in the PTH, CSF2 and FOLR genes were chosen to genotype a Berkshire-Yorkshire pig breed reference family for linkage and association analyses. Using linkage analysis, PTH and CSF2 loci were mapped to pig chromosome 2, while FOLR was mapped to pig chromosome 9. Association analyses between SNPs in the PTH, CSF2 and FOLR suggested that the CSF2 MboII polymorphism was significantly associated with several pork quality traits in the Berkshire and Yorkshire crossed F2 pigs. Our current findings provide useful SNP marker information to fine map QTL regions on pig chromosome 2 and to clarify the relevance of SNP and quantitative traits in commercial pig populations.

        Hydroalcoholic Extract of Ashwagandha Improves Sleep by Modulating GABA/Histamine Receptors and EEG Slow-Wave Pattern in In Vitro - In Vivo Experimental Models

        Withania somnifera (ashwagandha) has been used traditionally as a remedy for insomnia and to enhance cognitive function. The effects of ashwagandha extract (AE, 35% withanolide glycosides, Shoden<SUP>Ⓡ</SUP>) on the expression levels of γ-aminobutyric acid (GABA)Aρ1 and histamine H3 receptors in Rattus norvegicus glioblastoma (C6) cell lines were studied using semiquantitative reverse transcriptase-polymerase chain reactions. The effects of AE on sleep onset and duration were studied in Swiss albino mice using the pentobarbital-induced sleep model. Furthermore, the effects on nonrapid eye movement (NREM) and rapid eye movement sleep patterns were studied in Wistar rats with electroencephalogram (EEG) to support the improvement in sleep quality. There was an increase in gene expression levels of GABAAρ1 receptor (1.38 and 1.94 folds) and histamine H3 (1.14 and 1.29 folds) receptors induced by AE at doses of 15 and 30 μg/mL compared to control. AE at doses of 10, 25, and 50 mg/kg body weight showed a significant decrease in time to sleep onset and increased total sleep duration in the pentobarbital-induced sleep model. At 50 mg/kg body weight dosage level, a 34% decrease (P<0.0001) in sleep onset time and 47% increase (P<0.0001) in sleep duration was observed. The EEG study showed significant improvement in alpha, beta, theta, delta, and gamma bands at doses of 10, 25, and 50 mg/kg body weight with delta waves showing increases of 30%, 46% (P<0.05), and 34%, respectively. The induction of sleep, GABA-mimetic action, NREM sleep, and the effects on slow-wave cycles support the calming property of AE in improving the quality of sleep.

        In vivo and invitro experimentation for scientific validation of the traditional use of Chromolaena odorata (L.) against envenomation of honey bee sting

        Honey bees are well known for their economic importance but despite of their domicile nature they often become a threat when swarm of bees attacks a victim by their sting. Mellitin and Phospholipase A2(PLA2) in the bee venom causes pain, inflammation, mild hemorrhage and anaphylactic reaction. Common line of treatment includes hydrocortisone cream and no anti-venom developed yet. Herbal bioactive therapy and complementary medicine remains the option for symptomatic relief of the pain and inflammation associated with insect bites and sting. Chromolaena odorata (L.) (C. odorata) is investigated in the present study to scientifically validate its folkloric use in insect bites. Gel of C. odorata was applied for 7 days in the bare skin of each albino mice after allowing 6 live honey bees to sting on day 1. In egg yolk plate assay, percent inhibition of PLA2 by C. odorata was done. In L929 assay, cell viability was observed after challenge with PLA2 and protection by C. odorata was observed. In the in-vivo mice experiment, significant reduction in erythema and oedema was observed visually and histopathologically. In the In vitro PLA2 inhibition assay, the concentration of 25, 50 and 100 µg of C. odorata extract showed 28.5%, 54.9% and 76.7% inhibition respectively. In the L929 assay, C. odorata neutralised the PLA2 when incubated together and minimize cell death by inhibition of lactate dehydrogenase enzyme. The above results prove its candidature as an alternative medicine for healing from honey bee sting envenomation.
