Poly(vinyl pyrrolidone) was used successfully to control the size and distribution of silver nanoparticles generated on inorganic silica nanofibers. The inorganic nanofibers were electro spun using sol-gel chemistry of silicates, and the diameter of the prepared nanofibers was unaffected by adding up to 7% of poly(vinyl pyrrolidone). The silver ions, in the form of silver nitrate, were introduced into the silica nanofibers and reduced to metallic silver by ultraviolet irradiation with a subsequent thermal treatment. The size of the generated silver particles was decreased dramatically by adding poly(vinyl pyrrolidone). The size of the silver nanoparticles was 73 nm when no poly(vinyl pyrrolidone) was added but 23 nm with the addition of only 1% of poly(vinyl pyrrolidone). The extent of reduction could be checked by determining the concentration of silver ions leached into water from the silica nanofibers. After thermal treatment of the silica nanofibers, more than 99% of the silver remained in the nanofibers, indicating almost complete reduction of the silver ions to silver metal.
Preparation of Inorganic Silica Nanofibers Containing Silver Nanoparticles

Silica nanofibers containing silver nanoparticles were successfully prepared using sol-gel chemistry and electro-spinning technique. Solution of tetraethly orthosilicate in ethanol containing silver nitrate was aged to have sufficient viscosity and electrospun to form nanofibers. Upon thermal treatment, the gelation reaction between silanols was completed in the prepared silica nanofibers, and at the same time, silver ions in the nanofiber changed to metallic silver or silver oxides. The reduction of silver ions could be also achieved by UV irradiation, and the generated silver nanoparticles were present preferentially on the surface of the silica nanofibers. On testing release behavior of silver ions, it was found that most of silver remained in the silica nanofiber. The silica nanofibers containing silver nanoparticles exhibited excellent antibacterial and deodorant properties.
고콜레스테롤 식이 투여 마우스에서 복분자 미숙과 추출물과 홍삼농축액 복합 투여의 콜레스테롤 개선 효과

본 연구는 복분자 미숙과 물 추출물과 홍삼농축액으로 C57BL/6J 수컷 마우스를 이용하여 콜레스테롤 억제 효과 및 지질대사에 미치는 영향을 살펴보고, 기능성 식품으로의 가능성을 알아보았다. 실험동물은 C57BL/6J 수컷 마우스를 사용하였으며, 12주 동안 고콜레스테롤 식이를 급여하면서 복분자 미숙과 물 추출물과 홍삼농축액을 경구투여 하였다. 그 결과로 고콜레스테롤 식이군과 비교하였을 때 복분자 미숙과 추출물과 홍삼농축액 식이군에서 체중증가량, 식이섭취량에는 변화가 없었다. 콜레스테롤 합성의 가장 중요한 기작인 LDL receptor와 SREBP2 발현에서는 복분자 미숙과 추출물 투여군과 복분자와 홍삼 복합 투여군 모두 유의적으로 증가하여 세포 안으로의 LDL 흡수를 촉진시킴으로써 체내 콜레스테롤을 낮추는 것으로 판단된다. HDL 관련 유전자 발현에서도 복분자 미숙과 추출물 투여군과 복분자와 홍삼 복합 투여군에서 HDL uptake 유전자인 SR-B1의 발현을 증가시켜 콜레스테롤 개선 효과를 보였다. 또한 간의 형태학적 변화에서는 고콜레스테롤 식이군과 비교하였을 때 복분자 미숙과 물 추출물과 홍삼농축액 복합 투여군에서 지방 형성이 현저히 줄어들었고, leptin과 FAS의 농도를 측정한 결과 유의적으로 감소하였다. 결과적으로 복분자 미숙과 물 추출물과 홍삼농축액 복합 투여가 콜레스테롤과 고지혈증 예방에 긍정적인 효과를 보일 것으로 사료된다. We examined the effects of unripe black raspberry (UBR) and red ginseng (RG) extracts on cholesterol improvement in C57BL/6J mice fed a HCD (high cholesterol diet) for 12 weeks. Hepatic total lipid and total cholesterol contents were significantly induced in hyperlipidemic mice. However, supplementation with UBR, RG and simvastatin effectively reduced these lipid profiles. Further, UBR and co-treatment with UBR and RG increased expression of LDL receptor, SREBP2, and SR-B1 mRNA compared with HCD. The ApoB/ApoA1 ratio was reduced by co-treatment with UBR and RG compared to treatment with UBR. In addition, histopathologic evaluation showed that co-treatment with UBR and RG suppressed lipid accumulation as well as FAS and leptin expression in plasma. These results indicate that co-treatment with UBR and RG may be useful for the prevention of hypercholesterolemia.
복분자와 홍삼 발효 추출물의 복합투여가 고지방 고콜레스테롤 식이를 섭취한 흰쥐의 지질대사 및 비만에 미치는 영향

이 연구는 고지방 고콜레스테롤 식이를 12주 동안 랫트에게 공급하여 복분자와 홍삼, 복분자와 홍삼 발효 추출물의 지질대사 및 비만 개선효과를 조사하였다. 정상식이, 고지방 고콜레스테롤식이, 양성대조군, 유산균발효군, 복분자와 홍삼, 복분자와 홍삼발효 추출물 투여군의 체중증가, 음수, 식이 섭취량은 그룹간의 유의적인 차이를 보이지 않았다. 하지만 혈중 HDL-콜레스테롤은 고지방 고콜레스테롤 식이와 비교 했을 때 복분자와 홍삼 발효추출물에서 유의적인 증가를 보였으며, LDL-콜레스테롤, 중성지방은 유의적인 감소율을 보였다. 또한, 간 조직에서 복분자와 홍삼 발효추출물 처치에 의해 HMG-CoA reductase, LDL receptor 및 SREBP-2 mRNA의 발현 증가와 지방생성 억제를 확인하였다. 그리고 복분자와 홍삼 발효 추출물 투여군은 비만인자인 혈중 leptin과 FAS의 농도를 유의적으로 감소시켰을 뿐만 아니라 대변에서 체내 콜레스테롤 배출을 증가시켰다. 이러한 결과로 미루어보아 복분자와 홍삼 발효 추출물은 혈중 지질 대사 및 비만의 예방에 기여할 것으로 판단된다. This study investigated the effects of fermented black raspberry (BR) and red ginseng (RG) extract co-treatment on lipid metabolism and obesity in rats fed with a high fat/high cholesterol diet (HFHCD) for 12 weeks. Compared to the corresponding values in rats fed with a HFHCD, total cholesterol and low-density lipoprotein (LDL)-cholesterol and triglyceride levels decreased whereas high-density lipoprotein (HDL)-cholesterol levels increased in rats treated with fermented BR and RG extracts. These extracts significantly increased the expression of HMG-CoA reductase, LDL receptor, and sterol regulatory-element-binding protein-2 (SREBP-2) mRNA, but decreased the mRNA expression of SREBP-1. Additionally the serum levels of leptin and fatty acid synthase were decreased. Moreover, supplementation with fermented BR and RG effectively increased fecal cholesterol excretion. These results suggest that fermented BR and RG extracts might be effective at preventing hypercholesterolemia and obesity.

<P>There are multiple lines of evidence supporting that inflammation is causally linked to carcinogenesis. Abnormal upregulation of cyclooxygenase-2 (COX-2), a rate-limiting enzyme in the prostaglandin biosynthesis, has been implicated in carcinogenesis. Trans-3,4,3',5'-tetrahydroxystilbene (piceatannol), a naturally occurring hydroxylated stilbene with potent anti-inflammatory and antioxidative activities, has been shown to inhibit the proliferation of several cancer cells by inducing apoptosis or blocking cell cycle progression. In this study, we examined the effect of piceatannol on activation of the nuclear transcription factor NF-kappa B, one of the major transcription factors that regulate proinflammatory COX-2 gene transcription, in human mammary epithelial (MCF-10A) cells treated with the tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA). When pretreated to MCF-10A cells, piceatannol markedly inhibited TPA-induced NF-kappa B DNA binding to a greater extent than resveratrol and oxyresveratrol, stilbene analogs structurally related to piceatannol. Piceatannol also inhibited TPA-induced phosphorylation and degradation of Ikappa Balpha as well as nuclear translocation of the phosphorylated form of p65, the functionally active subunit of NF-kappa B. Likewise, TPA-induced expression of COX-2 was abrogated by piceatannol pretreatment. The thiol reducing agent dithiothreitol abolished the inhibitory effects of piceatannol on NF-kappa B DNA binding activity, suggesting that piceatannol may directly modify NF-kappa B or its regulator through reaction with the cysteine thiol(s).</P>
