        New polymorphic microsatellite markers in the Korean mi-iuy croaker, $Miichthys$ $miiuy$, and their application to the genetic characterization of wild and farmed populations

        Eighteen new polymorphic microsatellite markers were developed for the Korean mi-iuy croaker ($Miichthys$ $miiuy$, Perciformes, Sciaenidae), and allelic variability was compared between a wild population in Mokpo, Korea, and a hatchery population in Tongyeong, Korea. All loci were amplified readily and demonstrated allelic variability, with the number of alleles ranging from 5 to 37 in the wild population, and from 4 to 12 in the farmed population. The average observed and expected heterozygosities were estimated, respectively, to be 0.74 and 0.78 in the hatchery population samples, and 0.79 and 0.86 in the wild samples. These results indicate lower genetic variability in the hatchery population compared with the wild population, and significant genetic differentiation between the wild population and the hatchery samples ($F_{ST}$=0.058, P<0.001). These microsatellite loci may be valuable for future population genetic studies, monitoring changes in the genetic variation within stocks in a commercial breeding program, conservation genetics, and molecular assisted selective breeding of the mi-iuy croaker in the future.

        천연 생약재 열수 및 알코올 추출물의 어병 세균에 대한 항균력

        감초 외 48종의 천연 생약제로부터 열수 및 알콜 추출액을 제작하여 19 균주 어병 세균을 대상으로 항균력을 조사한 결과, 알콜 추출액은 22종, 열수 추출액은 16종이 어병 세균에 대하여 항균력을 나타내며 이 중 13종의 생약재는 알콜 및 열수 추출액 모두 8㎜ 이상의 저지대를 나타내어 항균력이 있는 것으로 나타났다. 열수 추출물에서 항균력을 나타내는 약재 중 그람 음성균에 감수성이 양호한 약재는 저지대 8 ㎜ 이상을 나타내는 애엽, 개옻나무, 연교, 지유, 파엽, 대황 및 황금이었으며, 그람 양성균에 항균력이 양호한 약재는 삼지구엽초, 육계 및 보골지 등 이었다. 그리고 오매, 황련, 계혈등, 상백피, 오배자 및 오미자는 저지대가 8㎜ 이상으로 그람 음성,양성균 모두에 항균력이 있었다. 알콜 추출물중에서 그람 음성균에 항균력을 나타내는 것은 백작약, 오매, 선모 및 황금 등으로 저지대가 8㎜ 이상으로 측정되었으며, 그람 양성균에 항균력을 나타내는 것은 감초, 계혈등, 단삼, 상백피, 육계 및 보골지 등으로 저지대가 8㎜ 이상이었다. 또한 그람 음성, 양성균에 모두 8㎜ 이상의 저지대를 나타내는 것은 애엽, 개옻나무, 황련, 지유, 오배자 및 오미자 등이었다. 그러나 이 중에서 오배자 열수 추출물의 어병 세균에 대한 발육 저지대가 32㎜로 그람 음성, 양성균 모두에 가장 뛰어난 항균력을 나타내어 다른 생약재와 다른 광범위 생약재인 것으로 나타났다. 애엽, 황금, 지유, 오매, 황련 및 오배자 열수 ·알콜 추출물은 tetracycline에 내성을 나타내는 균주에 감수성을 나타내었다. Hundreds of medicinal herbs have been using for the purpose of diseases treatment and immune enhancement for human being and other animals including fishes. Among them, 49 species of medicinal herbs were selected and tested for antibacterial activities against 19 strains of fish pathogenic bacteria in different 4 species. The 49 medicinal herbs were extracted by water and ethyl alcohol. The extracts were freeze dried and some paper discs from the extracts were prepared for the evaluation of antibacterial activity. The tested pathogenic bacteria were 5 strains of Edwardsiellu tarda, 5 strains of Vibrio sp., 4 strains of Lactococcus gawieae, 1 strain of Lactococcus raffinose. 1 strain of Streptococcus parauberius, and 3 strains of Streptococcus iniue. The Galla rhois (Obaeja), Gaeonnamu and Hwangleyon showed antibacterial activities on both gram negative and gram positive fish pathogenic bacteria. The Youkgae, Sangbaekpi, Bogolji and Gamcho showed very effective antibacterial activities on gram positive pathogens while Jiyu, Aeyoeb and Yeonkyo showed very effective on gram negative pathogens.

Tourism event is important procuctions in order to succeed. Tourism events is to productions, tourists is satisfied and will visit destinations again. Resultly, tourism event has a advantage to enhance the tourism destiantion' image, to attract tourists. This purpose of study is to suggest that is effective production strategies of tourism event. In order to production strategies of tourism event, the major strategies are as follow: to develop a production program formation and production process. Therefore I will research very hard to the dephts of a complex tourist's needs, will try to develop region' traditional and different traits.

        In those days, Cultural Tourism is emerging touist'need and as alternative tourism Specially, Traditional Cultural Tourism is interesting and is attracting many tourists. The utilizational of Traditional Cultural Tourism resources such as tradtional dress, wedding ceremony, traditional dancing and music, regional festivals and arts contest, traditional life styles have become very popular everyone. Therefore it is important to develop and use cultural assets as experience tourism products. Tourism behavior is a phenomenon as tourists try to find and experience attractions of tourism products. This purpose of study is to suggest that traditional cultural streets as, tourism resources is to develop experience tourist's products. In order to develop experiences tourist's product of traditional cultures are as follows: First, to manage fields of experiences of traditional culture products. and to play in the stage everyday Second, to train experts : guides and interpreters, producers, maketers. Third, to operate puplicity and promotion strateges. In the future study, it is important to develop several experience tourism products of traditional cultural streets. Therefore, I will research very hard to the depths of a complex tourist's needs, will try to develop traditional cultures and will develop several experience tourist's products.

        The purpose of this study is to research the New Recognition and Merchandising of Traditional Foods in Korea. First, this study focused on the relationship between traditional aspects of Korea foods and its meaning in the setting. Second, desire of sight-seeing in increasing sue to extension of sight-seeing experience, so interest about merchandising of food culture necessity of Korean traditional food as sight-seeing goods are being increased. The results of the study as follows : First, Korean foods can be divided in groups by usage, and is distinguished by strong local flavor. Korea regard "traditional foods" as being passed from their ancestors. Second, development of related character in necessary, summarized merchandising way and wrote comment about future studies and these bounds.

      • 대학생의 수면양상과 수면장애요인에 관한 연구

        The results of this Study are as follows:33.6% of all participants have insomnia; 22.5% of those who have insomnia are DIS(difficulty in initiating sleep), 17.3% are DMS(difficulty returning to sleep once awakened) and 7.0% awakened too early. 3 4.8% experience sleepiness during daytime. Type 1, experiencing insomnia and sleepi ness during daytime together, is 12.0%, Type 2, with insomnia only, is 21.6%, Type 3, with sleepiness during daytime only, is 22.8% and 43.5% experience no sleeping disturbances. After studying only those with 3 types of sleeping disturbances, it is found that the most common cause of such disturbance is stress 88.4%, anxiety 56.0%, no apparent reason 33.8%, anxiety/fear/terror 29.3%, hurry 23.6%, alcohol/caffeine 16.9%, bedroom tem perature 11.1%, urination during nighttime and persons living together 10.7%, noise from inside 8.9%, illumination 8.0%, and pain/itch 5.8%. The one group revealed significant differences in residential environment(p=0.003). Sex, age, education level, medicine, monthly earning revealed no meaningful differences. Of sleeping behavior, mean duration of sleep latency(p=0.000), whether or not feeling freshness(p=0.000), whether taking enough sleep(p=0.029), whether taking regular sleep(p=0.005) showed significant differences depending on whether or not having insomnia, and mean duration of sleep time, time to sleep, time of rising, whether taking naps did not reveal significant differences. Of sleep behavior, time to sleep(p=0.000), whether taking naps(p=0.000), indicated significant differences. Of sleeping behavior, mean duration of sleep latency(p=0.000), whether or not feeling freshness(p=0.000), and whether taking enough sleep(p=0.000), time of going to bed (p=0.002), whether or not taking nap(p=0.000), whether or not taking regular sleep(p=0.010) indicated significant differences among the sleeping disturbance types.

        Trichostatin A-mediated upregulation of p21WAF1 contributes to osteoclast apoptosis

        <P>Histone deacetylase inhibitors (HDIs), a new class of anti-cancer agents, have been reported to suppress formation of osteoclast precursors and their fusion into multinucleated cells. However, little is known about the effect of HDIs on mature osteoclasts, which may have significance for their therapeutic use. Here, we demonstrate a novel action of HDIs on osteoclast apoptosis. Primary multinucleated mature osteoclasts were prepared from mouse bone marrow cells. Treatment of osteoclasts with the HDI trichostatin A (TSA) caused apoptosis, as confirmed by annexin V staining and caspase activation. TSA caused the upregulation of p21WAF1 in osteoclasts. To understand the role of p21(WAF1) upregulation in TSA-treated osteoclasts, shRNA against p21(WAF1)-containing lentivirus was introduced into osteoclasts. The suppression of p21(WAF1) decreased TSA-directed osteoclast apoptosis. Collectively, our results provide evidence that TSA causes osteoclast apoptosis, which involves, in part, TSA-induced upregulation of p21(WAF1), and strongly supports HDIs as potential therapeutic agents for excessive bone resorption.</P>

        대추 추출물이 장내 미생물의 생육에 미치는 영향 및 항산화 활성

        본 연구에서는 대추 추출물과 장내 유익균의 생육을 조절하여 장내 부패를 방지하기 위한 probiotics로서 유산균을 조합하기 위하여 대추 추출물이 장내 유익균 중에서도 유산균의 생육에 미치는 영향을 in vitro 상에서 알아보고자 하였다. 또한 증식인자로서 대추 추출물에 존재하는 당이 유산균의 생육에 어떠한 영향을 미치는가를 알아보기 위하여 추출방법을 달리하여 당 함량이 다른 대추 1차 및 2차 열수, 에탄올 추출물을 각각 제조하였다. 이와 더불어 지역 특산물인 보은대추를 기능성식품 소재로 활용하기 위하여 페놀성 화합물과 항산화성을 검색하여 기능성식품으로 상품화하기 위한 기본 자료를 확보하고자 하였다. TLC와 HPLC 분석을 통해 장내 유익균의 증식인자로 작용할 수 있는 대추 추출물의 당 조성은 fructose, glucose, sucrose인 것을 확인하였고, 1차 추출물에 비하여 2차 열수 추출물은 fructose, glucose, sucrose가 각각 69, 74, 89% 감소하였으며 2차 에탄올 추출물은 63, 62, 83% 감소한 것을 확인할 수 있었다. 인체 장내세균에 대하여 대추 추출물을 50 mg/mL 농도로 하여 생육증진 효과를 분석한 결과, 대추 1차 열수 추출물에서 B. bifidum과 B. adolescentis가 다른 시험균주들에 비하여 대조군과 대수기에 도달하는 시간은 비슷하였지만, 대조군보다 높은 흡광도 값을 나타내어 생육을 증진시키는 결과를 보였다. 이는 대추 추출물에 존재하는 당이 prebiotics로 작용하여 Bifidobacterium 속의 생육을 증진시킨 것으로 사료되며 인체 장내 균총 조성을 바람직한 방향으로 개선시켜 장내 기능을 향상시킬 것으로 기대된다. 또한 당이 아닌 다른 기능성 성분에 의해 생육이 증진되는지를 알아보기 위해 기능성 물질인 페놀류의 함량과 항산화성을 분석하였다. 총 폴리페놀과 총 플라보노이드 함량을 측정한 결과, 대추 1차 열수 추출물에서 각각 11.63 mg/g, 247.45 μg/g으로 다른 추출물에 비해 높은 함량을 보여 주었다. DPPH에 의한 라디칼 소거능을 살펴본 결과, 대추 추출물의 농도가 높을수록 우수한 전자공여능을 나타내었으며 대추 1차 열수 추출물의 경우 다른 추출물에 비해 가장 높은 85.60%의 전자공여능을 보여 천연 항산화제인 ascorbic acid에 견줄 수 있는 기능성이 있음을 확인하였다. ABTS free 라디칼 소거활성을 통해서도 대추 1차 열수 추출물은 679.91 mg AA eq/g의 항산화력을 지니고 있어 다른 추출물에 비해 가장 좋은 항산화 활성을 나타내었다. 따라서 대추 1차 열수 추출물에는 당뿐만 아니라 항산화능, 폴리페놀, 플라보노이드 함량도 높아 B. bifidum과 B. adolescentis의 생육이 증진된 것으로 사료되며, 기능성 식품소재로 생리활성 성분을 함유하고 있는 대추추출물에 Bifidobacterium 속을 이용하여 유산발효를 진행함으로써 기능성 발효 음료 개발 및 이유식과 같은 다양한 식품 개발에 응용될 수 있을 것으로 판단된다. This study was carried out to investigate the effects of jujube extracts on intestinal microflora, along with their antioxidant activities, according to extraction method. The antimicrobial activities of the extracts were measured using the agar diffusion method with a jujube extract concentration of 50 mg/mL. Neither the first nor second jujube extracts were inhibitory against the tested intestinal bacteria. However, water extracts of jujube significantly enhanced the growth of lactic acid bacteria, especially Bifidobacterium bifidum and Bifidobacterium adolescentis. Total phenol compounds and flavonoid compounds were higher in the 1st than in the 2nd water extracts. The EDA values of both water and ethanol extracts increased in proportion to the extract concentration. The 1st water extract showed the highest value among all the others, which was 85.60% at the concentration of 0.05 mg/mL. Furthermore, the 1st water extract showed stronger antioxidant activity than the other samples with an activity of 679.91 mg AA eq/g. These results support the potential use of jujube water extracts as a functional food component and a valuable resource for the development of nutraceutical foods, to increase the growth of Bifidobacterium spp. in the human intestine.
