        The Candidate Tumor Suppressor Gene SLC8A2 Inhibits Invasion, Angiogenesis and Growth of Glioblastoma

        Glioblastoma is the most frequent and most ag-gressive brain tumor in adults. Solute carrier family 8 member 2 (SLC8A2) is only expressed in normal brain, but not present in other human normal tissues or in gliomas. Therefore, we hy-pothesized that SLC8A2 might be a glioma tumor suppressor gene and detected the role of SLC8A2 in glioblastoma and explored the underlying molecular mechanism. The glioblas-toma U87MG cells stably transfected with the lentivirus plasmid containg SLC8A2 (U87MG-SLC8A2) and negative control (U87MG-NC) were constructed. In the present study, we found that the tumorigenicity of U87MG in nude mice was totally inhibited by SLC8A2. Overexpression of SLC8A2 had no effect on cell proliferation or cell cycle, but impaired the invasion and migration of U87MG cells, most likely through inactivating the extracellular signal-related kinases (ERK)1/2 signaling pathway, inhibiting the nuclear translocation and DNA binding activity of nuclear factor kappa B (NF-κB), reducing the level of matrix metalloproteinases (MMPs) and urokinase-type plasminogen activator (uPA)-its receptor (uPAR) system (ERK1/2-NF-κB-MMPs/uPA-uPAR), and altering the protein levels of epithelial to mesenchymal transitions (EMT)-associated proteins E-cardherin, vimentin and Snail. In addition, SLC8A2 inhibited the angiogenesis of U87MG cells, probably through combined inhibition of endothelium-dependent and endothelium-nondependent angiogenesis (vascular mimicry pattern). Totally, SLC8A2 serves as a tumor suppressor gene and inhibits invasion, angiogenesis and growth of glioblastoma.

        Immobilized Bioactive Agents onto Polyurethane Surface with Heparin and Phosphorylcholine Group

        Heparin (HEP) and phosphorylcholine groups (PC) were grafted onto the polyurethane (PU) surface in order to improve biocompatibility and anticoagulant activity. After the surface grafting sites of PU were amplified with the primary amine groups of polyethylenimine (PEI), heparin was covalently linked onto the surface by the reaction between the amino group and the carboxyl group. PC groups were covalently immobilized on the PU-PEI surface through the reaction between the amino group and the aldehyde group of phosphorylcholine glyceraldehyde (PCGA). The surface density of primary amine groups was determined by a ninhydrin assay. The amino group density reached a maximum of 0.88 μmol/cm2 upon incorporation of 10 wt% PEI. The amount of heparin covalently immobilized on the PU-PEI surface was determined by the toluidine blue method. The grafting chemistry resulted in the comparatively dense immobilization of HEP (2.6 μg/cm2) and PC to the PU-PEI surfaces. The HEP and PC modified surfaces were characterized by water uptake (PU 0.15 mg/cm2, PU-PEI 3.54 mg/cm2, PU-HEP 2.04 mg/cm2, PU-PC 2.38 mg/cm2), water contact angle (PU 95.3º, PU-PEI 34.0º, PU-HEP 39.5º, PU-PC 37.2º), attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS),atomic force microscopy (AFM), and scanning electron microscope (SEM). The results demonstrated that the PUPEI surface was successfully grafted with HEP and PC. The hydrophilicity and hemocompatibility of these grafted surfaces were significantly improved. These results suggested that the PU-HEP and PU-PC composite films are promising candidates for blood contacting tissue engineering.

        Optimization of dyeing parameters of cotton standardized samples for laundry test of dye transfer inhibition program

        In order to prevent light-colored clothes from being stained by dyes released from dark clothes during the washing process, some new-type washing machines have developed the dye transfer inhibition washing program. However, there is no certified reference materials for the test of dye transfer inhibition function. To this end, cotton fabric and reactive dyes are used as experimental materials to prepare standardized samples to evaluate the dye transfer inhibition function of washing machines. Firstly, the single factor analysis method is used to analyze the significance of the dyeing parameters including dye dosage, dyeing temperature, sodium sulfate dosage and sodium carbonate dosage. Secondly, a 4-factor 5-level experimental design and theoretical prediction of the best dyeing parameters are successively carried out through central composite design and response surface method. Two evaluation indicators, the dye release amount of the standardized sample of dyed fabric and the color difference value of the standardized sample of white fabric after washing, are proposed as the response values for response surface analysis to search the optimal dying parameters in theory. The optimal dyeing parameters obtained through response surface analysis are that the dosage of dye is 5.63% (owf), the dyeing temperature is 60 °C, the dosage of sodium sulfate is 93.60 g/L, and the dosage of sodium carbonate is 15 g/L. Experimental results indicate that the standardized samples prepared with optimal dyeing parameters can effectively distinguish the dye transfer inhibition function of washing machines.

        Influence of K2TiF6 in electrolyte on characteristics of the microarc oxidation coating on aluminum alloy

        Black and gray microarc oxidation (MAO) coatings were prepared in a phosphate electrolyte with and without K2TiF6 on 2A70 aluminum alloy, respectively. Voltageetime curves were recorded during the MAO process. The effects of K2TiF6 on the morphology, composition, abrasive resistance and corrosion resistance of MAO coatings were investigated. The results showed that the MAO coating produced in the electrolyte with K2TiF6 was thicker, and more uniform than that produced in the electrolyte without K2TiF6. Ti was detected in the surface of the MAO coating formed in the electrolyte with K2TiF6. The results of abrasive resistance and corrosion resistance showed that the MAO coating formed in the electrolyte with K2TiF6 exhibited better abrasive resistance and corrosion resistance.

        Achieving fixed-time synchronization of the Kuramoto model via improving control techniques

        This paper further analyzes the fixed-time (FxT) phase agreement (PA) and the frequency synchronization (FS) of the Kuramoto model with identical and non-identical oscillators, respectively. The non-chattering control protocol is carefully picked to overcome the deficiency of the signum function contained in the conventional finite-time (FT)/FxT controllers, which improves the synchronized performance of networked systems. Furthermore, for reducing control cost, the switching control scheme is taken into consideration. Through the utilization of FxT stability theory, several sufficient conditions are derived successively for achieving the FxT PA and FS, in which the upper bound time estimates for arriving at PA and FS are given regardless of the initial states of the Kuramoto model. Finally, some simulation results lead to the conclusion that the proposed theoretical analysis is correct and effective.

        Mechanical Behavior of Nine Tree-Pool Joints Between Large Trees and Buildings

        Nine types of tree-pool joints designed to plant large trees on building structures were proposed in this study. Solid finite elementmodel considering plastic damage constitutive relationship for the tree-pool joints were built using ABAQUS software platform. Based on comparative analysis of six different mesh generation methods of the 3D tree-pool joint model, an optimized meshgeneration method was determined and verified considering both the calculation time and accuracy. The mechanical behavior of thenine tree-pool joints, such as peak tension/compression stress, hysteretic energy dissipation performance, plastic damageperformance and corresponding crack development process, were studied in detail under horizontal and vertical loads. The resultsshow that the tree-pool joints with steel strengthened form and composite strengthened form have superior horizontal and verticalbearing capacity. Similarly, they also have stable hysteretic energy dissipation performance, minimal plastic damage and crackdevelopment relatively in vertical ultimate load. However, the tree-pool joints with other strengthened forms, such as forms of innerbeamand inner-beam combined with ring bracket, show poor mechanical properties. Although there is a certain degree ofperformance improvement for these tree-pool joints relative to non-strengthened joints, narrow hysteretic curves, apparent strength/stiffness degradation characteristics, extensive material damage and crack development can be found. Results of this paper do shinesome lights on how to design reasonable and reliable tree-pool joints in building structures.

        Structural and component characterization of the B4C neutron conversion layer deposited by magnetron sputtering

        Neutron conversion detectors that use 10B-enriched boron carbide are feasible alternatives to 3 He-based detectors. We prepared boron carbide films at micron-scale thickness using direct-current magnetron sputtering. The structural characteristics of natural B4C films, including density, roughness, crystallization, and purity, were analyzed using grazing incidence X-ray reflectivity, X-ray diffraction, X-ray photoelectron spectroscopy, time-of-flight secondary ion mass spectrometry, and scanning electron microscopy. A beam profile test was conducted to verify the practicality of the 10B-enriched B4C neutron conversion layer. A clear profile indicated the high quality of the neutron conversion of the boron carbide layer.

        Circ_0068655 Promotes Cardiomyocyte Apoptosis via miR-498/PAWR Axis

        Background: The cardiomyocyte apoptosis is considered as one of major contributions to cardiac remodeling after myocardial infarction (MI). Numerous studies find that circular RNAs (circRNAs) play pivotal roles in a variety of biological functions. However, the role of circ_0068655 in MI and human induced pluripotent stem-derived cardiomyocytes (HCMs) remains unknown. Methods: The expression of circ_0068655, miR-498, and PRKC apoptosis WT1 regulator (PAWR) in human MI heart tissues and hypoxia subjected HCMs was evaluated with qRT-PCR and Western blot. The effects of circ_0068655 on hypoxia-induced apoptotic death and cell migration in HCMs were evaluated with qRT-PCR, cell viability, cell death ELISA (POD), and Caspase-3 activity assay, and Trans-well assay, respectively. Furthermore, luciferase assay, qRT-PCR, biotin-labeled miRNA pulldown assay, and Western blot were employed in the functional studies. Results: We found that the expression of circ_0068655 and PAWR was enhanced in MI patients and hypoxia subjected HCMs; by contrast, the expression of miR-498 decreased. Inhibited expression of circ_0068655 in HMCs counteracted hypoxia-induced apoptotic death and impaired cell migration, in sharp contrast to circ_0068655 knockdown. We identified that circ_0068655 sponged an endogenous miR-498 to sequester and inhibit its activity, leading to the increased PAWR expression. Conclusions: Our findings reveal that the expression of circ_0068655 can promote cardiomyocyte apoptosis through the modulation of miR-498-PAWR axis in vitro, which highlights the diagnostic and therapeutic value of circ_0068655 in patients with MI.
