In this research 26 Shigella isolates were examined by PCR and direct nucleotide sequencing for genetic alterations in the quinolone-resistance determining regions (QRDRs). We tested for the presence of qnr genes by PCR in 91 strains, but no qnr genes were found. The results did show, however, some novel mutations at codon 83 of gyrA ($Ser{\rightarrow}Ile$) and codon 64 of parC ($Ala64{\rightarrow}Cys,\;Ala64{\rightarrow}Asp$), which were related to fluroquinolone resistance.

Isoindolin-1-ones from the stems of Nicotiana tabacum and their antiviral activities

In previous studies, several isoindolin-1-oneanalogs that exhibited signifi cant anti-tobacco mosaic virus(anti-TMV) activities were isolated from Nicotiana tabacum . Since gene-editing mutants provide a new sample for thediscovery of active metabolites, we focused on the stems ofYN-18–23 (a mutant N. tabacum for gene editing with thealkaloid metabolic pathway cultivated by Yunnan TobaccoCompany), which led to the isolation of four new ( 1–4 )and four known ( 5–8 ) isoindolin-1-ones. To the best of ourknowledge, nicindole C ( 3 ) is the fi rst subclass of isoindolin-1-one bearing a pentacyclic ketone, while nicindole D ( 4 )is the fi rst example of isoindolin-1-one bearing a methylpyridin-2-(1 H )-one moiety. Compounds 1–4 were testedfor their anti-TMV activities, and the results revealed thatcompounds 1 , 3 , and 4 exhibited high anti-TMV activities atconcentrations of 20 μM with inhibition rates of 48.6, 42.8,and 71.5%, respectively. These rates are higher than the inhibitionrate of the positive control (33.2%). The mechanisticstudy of compound 4 , which had the highest anti-TMV activityrevealed that increased potentiation of defense-related enzyme activities and downregulation of expression of theNtHsp70 protein may induce resistance in tobacco againstthe viral pathogen TMV. Molecular docking studies alsorevealed that the isoindolin-1-one substructure is fundamentalfor anti-TMV activity. The methyl-pyridin-2-(1 H )-onemoiety in compound 4 and the 2-oxopropyl groups in compounds1 and 3 at the N -2 position may increase inhibitoryactivities. This study of the structure–activity relationshipis helpful for fi nding new anti-TMV activity inhibitors. Tostudy whether the isoindolin-1-ones have broader antiviralactivities, compounds 1–4 were also tested for their antirotavirusactivities. Compound 4 exhibited high anti-rotavirusactivity with a therapeutic index (TI) value of 20.7. This TI value is close to that of the positive control (20.2).
Two New Diphenylethylenes from Arundina graminifolia and Their Cytotoxicity

Two new diphenylethylenes, gramniphenols H and I (1 and 2), together with six known diphenylethylenes (3-8), were isolated from Arundina graminifolia. The structures of 1-8 were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1 and 2 were evaluated for their cytotoxicity against five human tumor cell lines. Compound 1 showed cytotoxicity against PC3 cells with $IC_{50}$ value of 3.5 ${\mu}M$. Compound 2 showed cytotoxicity against NB4 and PC3 cells with $IC_{50}$ values of 3.6 and 3.8 ${\mu}M$, respectively.
THE PERIODIC JACOBI MATRIX PROCRUSTES PROBLEM

The following "Periodic Jacobi Procrustes" problem is studied: find the Periodic Jacobi matrix X which minimizes the Frobenius (or Euclidean) norm of AX - B, with A and B as given rectangular matrices. The class of Procrustes problems has many application in the biological, physical and social sciences just as in the investigation of elastic structures. The different problems are obtained varying the structure of the matrices belonging to the feasible set. Higham has solved the orthogonal, the symmetric and the positive definite cases. Andersson and Elfving have studied the symmetric positive semidefinite case and the (symmetric) elementwise nonnegative case. In this contribution, we extend and develop these research, however, in a relatively simple way. Numerical difficulties are discussed and illustrated by examples.
Three homologous genes encoding functional D8-sphingolipid desaturase in Populus tomentosa

Δ8-sphingolipid desaturase is characterized by itsability to catalyze desaturation at the C8 position of the longchainbase of sphingolipids in plants. No previous studieshave been conducted on genes encoding Δ8-sphingolipiddesaturases in the woody plant Populus tomentosa. In thisstudy, three genes that encode Δ8-sphingolipid desaturasewere isolated fromP. tomentosa. Among these genes, PtD8Aand PtD8B showed high sequence similarity; whereas PtD8Cexhibited large sequence divergence.RT-PCRresults showedthat PtD8A and PtD8B were expressed in all tissues detected,whereas PtD8C was not expressed in roots. Heterologousexpression in yeast revealed that PtD8A/B/C were functionalΔ8-sphingolipid desaturases, and can catalyze the C18-phytosphingeninedesaturation to produce 8(Z)- and 8(E)-C18-phytosphingenine.However, the conversion rate and ratios ofthe two products differed. Compared with control cells,transgenic yeasts expressing PtD8A/B/C exhibited enhancedaluminum tolerance. Our findings further elucidated thebiochemical functions and evolutionary history ofΔ8-sphingolipid desaturases in plants. Candidate genes forbreeding new poplar germplasm resources with enhancedtolerance ability to aluminium were also provided.

Histomorphological study on embryogenesis of the honeybee Apis cerana

As important pollination species, honeybees play substantial impacts on the balance of global ecosystem, including two best-known honeybees Apis mellifera and Apis cerana. Embryogenesis is a fundamental stage of honeybee development and plays important roles in supporting the whole-life developmental process. However, few studies were reported on honeybee embryonic morphology using egg section, possibly due to the fragility of honeybee eggs and the difficulty of making embryonic sections. In this study, we reported a simply equipped method of frozen sectioning and PI (propidium iodide) staining to show the inner structure and cell distribution of A. cerena embryos at the different embryonic developmental stages. We found that the stages of A. cerena embryogenesis could also be typically classified into ten developmental stages, which are similar with the sister honeybee species, A. mellifera. To be noted, besides the cell distribution in the whole egg, we clearly observed the migration route of embryonic cells during the early embryonic development in A. cerena. This study provides a new insight into the whole process of honeybee embryogenesis from the perspective of egg sectioning, a histological basis for genetic manipulation using A. cerena eggs, and a reference method for egg sectioning for other insect species.
