        질적지표를 활용한 국내 지적장애 대상 비디오 모델링 중재특성 분석

        연구목적: 본 연구는 국내 지적장애 대상 비디오 모델링 중재연구를 종합하여 그 특성을 파악하고, 질적지표에 근거한 분석을 통해 증거기반실제의 기초를 마련함으로써 향후 연구와 실제를 위한 시사점을 얻는 것을 목적으로 한다. 연구 방법: 이를 위해 2000년도 이후 국내 학술지에 발표된 논문 중, 선정기준을 충족시키는 연구 총 21편을 선정하고 질적지표를 활용하여 중재특성을 분석하였다. 연구 결과: 2000년 이후 지적장애인을 대상으로 한 비디오 모델링 중재연구는 2006년과 2015년 사이에 가장 많이 이루어졌으며, 연구의 형태는 단일대상 연구의 비율이 높게 나타났다. 비디오 모델링 중재는 초등학교 급에서 일상생활기술 증진을 위해 단독중재형태로 학교에서 가장 많이 이루어졌다. (준)집단설계연구의 질적지표 분석 결과, 3편 중 1편만이 충분한 질적 수준을 보인 반면, 단일대상연구 18편 중 17편이 평균 2.0점 이상으로 질적지표 충족 수준이 상대적으로 높게 분석되었다. 결론: 본 연구의 결과를 바탕으로 지적장애 대상 비디오 모델링 중재연구의 질적 수준을 확보하고 효과적인 증거기반 중재 제공을 위한 함의를 논하였다. Purpose: This study aims to synthesize video modeling(VM) intervention studies using quality indicators to examine effects of VM for individuals with intellectual disabilities. Method: To this end, a total of 21 Korean articles published between 2000 and 2020 were selected for analysis. Results: (a) about 70% of VM intervention studies for people with intellectual disabilities were conducted between 2006 and 2015; (b) majority of studies were practiced in single subject design; (c) VM interventions were often provided in school settings to improve independent living skills at the primary school level; (d) findings from the analysis of quality indicators also concluded that only 1 out of 3 quasi-group-design studies showed sufficient level, whereas 17 out of 18 single-subject-design studies showed a relatively high level of satisfaction with an average of 2.0 points or higher. Conclusion: Based on the results, implications for securing the quality level of VM intervention studies for people with intellectual disabilities and providing effective evidence-based interventions were discussed.

        Vegetable consumption is associated with health benefits. Recently, the interest in these food groups has grown among due to the identification of new compound in their chemical composition. Most of the vegetables are cooked by blanching, boiling, steaming, microwaving before consumed. These cooking processes may change not only food physical characteristics but also chemical composition. The present study was undertaken to investigate the effect of different cooking methods on phytochemical content and antioxidant capacity in Ipomoea batatas (L.) Lam stem. The result of study, Total polyphenol of phytochemical in Ipomoea batatas (L.) Lam stem was 6.88-12.65 mg GAE/100 g and flavonoid was 3.16-10.46 mg CE/100 g. Compared with the raw of Ipomoea batatas (L.) Lam stem, all cooking methods were found to significant correlations between the antioxidant activity assessed by ABTS, DPPH and polyphenol(p<0.05). These results can be provide the basic data how to prevent lose of nutrition in Ipomoea batatas (L.) Lam stem when cooking.

        Transient Plant Expression of Recombinant Rabies Virus Glycoprotein: A strategy for Vaccine Production

        The rabies virus causes neurological infections, resulting in the deaths of over 60,000 people worldwide each year. Currently available rabies vaccines are attenuated virus vaccines, which carry the risk of transmission among both humans and animals. An exciting alternative to these traditional vaccines is the use of recombinant plant-based vaccines. In this study, we applied the rabies virus glycoprotein (RVGP) as a recombinant vaccine to be transiently expressed in plants. To achieve this, we fused human immunoglobulin G Fc with the KDEL sequence, a motif for endoplasmic reticulum (ER) retention (FcK), to the RVGP, to generate RVGP-FcK. We then cloned the RVGPFcK gene expression cassette into the pEAQ vector and agro-infiltrated Agrobacterium tumefaciens (LBA4404) carrying the pEAQ RVGP-FcK vector into the leaves of Nicotiana benthamiana for transient expression. A RT-PCR analysis confirmed the transcription of the RVGP-FcK gene, which was evident as early as four days post-infiltration (dpi). To optimize the spatial and temporal aspects of RVGP-FcK production, we conducted analyses of its expression levels at different leaf positions (top, middle, and base) and dpi. A western blot analysis demonstrated that the RVGP-FcK protein reached its highest expression level at 7 dpi in the top leaf position and at 5 dpi in the middle leaf position. However, no detectable expression was observed in the bottom leaves at any time point. Subsequently, we validated the functionality of RVGP-FcK through an ELISA analysis. The results revealed that RVGP-FcK was expressed and assembled into its functional form most effectively at 5 and 7 dpi in the top leaf position and at 5 and 7 dpi in the middle leaf position. Our findings demonstrate that the transient expression of a functional RVGP-FcK protein can be optimized spatially and temporally in plants.

        Transient exposure to hydrogen peroxide inhibits the ubiquitination of phosphorylated IκBα in TNFα-stimulated HEK293 cells

        <P>During ischemia-reperfusion injury, brief pre-exposure to oxidative stress renders organs resistant to subsequent severe damage. NF-κB is a transcription factor that is involved in reperfusion-induced inflammatory and immune responses. The activity of NF-κB has been shown to be modulated by oxidative stress in various cell types through different pathways. We studied the effect of pre-exposure to oxidative stress on subsequent NF-κB activation in TNFα-stimulated HEK293 cells. The cells were transiently exposed to 0.5 mM H<SUB>2</SUB>O<SUB>2</SUB> for 20 min, prior to stimulation with TNFα, and the subsequent expression of NF-κB-dependent genes and the levels of NF-κB signaling molecules were measured. Pre-exposure to H<SUB>2</SUB>O<SUB>2</SUB> significantly delayed the TNFα-induced expression of an NF-κB reporter gene and inflammatory proteins (intercellular adhesion molecule-1 and IL-1β). The degradation of inhibitor of NF-κB α (IκBα) and the nuclear translocation of NF-κB were also delayed by H<SUB>2</SUB>O<SUB>2</SUB> treatment, whereas IκBα phosphorylation and IκB kinase activity were not changed. When we examined the ubiquitin/proteosome pathway in H<SUB>2</SUB>O<SUB>2</SUB>-treated cells, we could not detect significant changes in proteosomal peptidase activities, but we were able to detect a delay of IκBα poly-ubiquitination. Our results suggest that transient exposure to oxidative stress temporally inhibits NF-κB-dependent gene expression by suppressing the poly-ubiquitination of phosphorylated IκBα in HEK293 cells.</P>

        Overexpression of PMA1 enhances tolerance to various types of stress and constitutively activates the SAPK pathways in Saccharomyces cerevisiae

        <P>PMA1 encodes a transmembrane polypeptide that functions to pump protons out of the cell. Ectopic PMA1 overexpression in Saccharomyces cerevisiae enhances tolerance to weak acids, reactive oxygen species (ROS) and ethanol, and changes the following physiological properties: better proton efflux, lower membrane permeability, and lessened internal hydrogen peroxide production. The enhanced stress tolerance was dependent on the mitogen-activated protein kinase (MAPK) Hog1 of the high osmolarity glycerol (HOG) pathway, but not the MAPK Slt2 of the cell wall integrity (CWI) pathway; however, a PMA1 overexpression constitutively activated both Hog1 and Slt2. The constitutive Hog1 activation required the MAPK kinase kinase (MAP3K) Ssk2 of the HOG pathway, but not Ste11 and Ssk22, two other MAP3Ks of the same pathway. The constitutive Slt2 activation did not require Rom2 and the membrane sensors of the CWI pathway, whereas Bck1 was indispensable. The PMA1 overexpression activated the stress response element but not the cyclic AMP response element and the Rlm1 transcription factor. PMA1 overexpression may facilitate the construction of industrial strains with simultaneous tolerance to weak acids, ROS, and ethanol.</P>

        Patterns of Users’ First Turns with a Service Chatbot: A Conversation Analytic Perspective

        The present study examines patterns of users’ first turns in their interaction with a service chatbot developed by a team in the Department of Computer Science and Engineering at Sogang University, South Korea. The user’s first turn is where the user produces their initial request to the chatbot based on a task prompt that is given to them. This is a crucial site that can project the trajectory of the conversational dialogue in the next turns. The data for this study is a corpus of 456 conversational dialogues between human users and the service chatbot for the task of scheduling (e.g., creating/deleting/changing schedules). Analyses reveal three main patterns emerging from users’ first turns: (1) naming main request; (2) prefacing another request; and (3) aggregating task prompt. These patterns are described as users’ sense-making practices which demonstrate their understanding of the task prompt presented to them as well as how they interpret the underlying mechanism of the chatbot. The first and second patterns, in particular, are illustrative of discrepancies in assumptions between human users and chatbot developers. The study provides practical implications for chatbot developers and discusses the utility of Conversation Analysis (CA) as a methodology to investigate human-chatbot interaction.

        Hydrogen production from the photocatalytic hydrolysis of sodium borohydride in the presence of In–, Sn–, and Sb–TiO2s

        To enhance the evolution of hydrogen from the photocatalytic hydrolysis of sodium borohydride solution, three photocatalysts; 0.5 mol% In, Sn, and Sb, incorporated into TiO2, were prepared using a hydrothermal method. The particle sizes of well-developed anatase were about 5.0 nm, except those for Sb–TiO2, which were above 5.0 nm, with closed to cubic form shapes, except for pure TiO2 that was spherical. In the X-ray photoelectron spectra (XPS) for Ti2p3/2, a band at 460.34 eV, attributed to partial MTiO3, was seen in Sn–TiO2. Additionally, two bands of Sn3d5/2 at 484.5 and 486.8 eV, assigned to the Sn and SnO or SnO2, respectively, appeared over Sn–TiO2, with the ratios of metal–OH/metal–O in the O1s bands, imply a hydrophilic property, also increased in this compared with the other catalysts. The hydrogen evolution from sodium borohydride (NaBH4) hydrolysis was enhanced in the Sn–TiO2 photocatalytic system compared to those for the other catalysts; hydrogen was evolved at 140 ml/g h over Sn–TiO2, which was different from the 100 ml/g h for the hydrolysis of pure NaBH4, and increased to 1350 ml/g after 5 h. In particular, the evolved hydrogen increased with increasing amounts of Sn–TiO2, and the production continued for 150 h.

      • Microbiota-Derived Lactate Accelerates Intestinal Stem-Cell-Mediated Epithelial Development

        <P><B>Summary</B></P> <P>Symbionts play an indispensable role in gut homeostasis, but underlying mechanisms remain elusive. To clarify the role of lactic-acid-producing bacteria (LAB) on intestinal stem-cell (ISC)-mediated epithelial development, we fed mice with LAB-type symbionts such as <I>Bifidobacterium</I> and <I>Lactobacillus</I> spp. Here we show that administration of LAB-type symbionts significantly increased expansion of ISCs, Paneth cells, and goblet cells. Lactate stimulated ISC proliferation through Wnt/β-catenin signals of Paneth cells and intestinal stromal cells. Moreover, <I>Lactobacillus plantarum</I> strains lacking lactate dehydrogenase activity, which are deficient in lactate production, elicited less ISC proliferation. Pre-treatment with LAB-type symbionts or lactate protected mice in response to gut injury provoked by combined treatments with radiation and a chemotherapy drug. Impaired ISC-mediated epithelial development was found in mice deficient of the lactate G-protein-coupled receptor, Gpr81.         Toluene Decompositions over Al-W-incorporated Mesoporous Titanosilicates Photocatalysts

        This study investigated the decomposition activities of toluene on 10 mol% Al-W-incorporated mesoporous titano (15 mol %) silicates. The mesopore sizes observed in the transmission electron microscopy images ranged from 2.0 to 5.0 nm, and the pores were irregular on the addition of 10 mol% Al or W ions, but changed to regular hexagonal forms with the simultaneous additions of Al and W. The X-ray photon spectroscopy results showed a shift of the special peak for Ti2p in Al-incorporated mesoporous titanosilicates to a stronger binding energy compared to those of mesoporous titanosilicates and Al-incorporated mesoporous titanosilicates. Three O1s peaks in the spectra of the Al and W coexisted samples were observed at 530.5 and 531.7, 533, and 533.7eV, which were assigned to Ti-Os in TiO2 and Ti2O3, Si-O in SiO2 and Al-O in Al2O3, respectively. The toluene molecules desorbed at lower temperatures over W-incorporated mesoporous titanosilicates, and the amounts of toluene desorbed were also small; however, Al-incorporated mesoporous titanosilicates adsorbed much more toluene, particular over Al7.5-W2.5-Ti15-Si75. The photocatalytic decomposition of toluene was more enhanced over Al7.5-W2.5-Ti15-Si75 than over Al- or W-incorporated mesoporous titanosilicates only.

        Immunogenicity of lipid nanoparticles and its impact on the efficacy of mRNA vaccines and therapeutics

        Several studies have utilized a lipid nanoparticle delivery system to enhance the effectiveness of mRNA therapeutics and vaccines. However, these nanoparticles are recognized as foreign materials by the body and stimulate innate immunity, which in turn impacts adaptive immunity. Therefore, it is crucial to understand the specific type of innate immune response triggered by lipid nanoparticles. This article provides an overview of the immunological response in the body, explores how lipid nanoparticles activate the innate immune system, and examines the adverse effects and immunogenicity-related development pathways associated with these nanoparticles. Finally, we highlight and explore strategies for regulating the immunogenicity of lipid nanoparticles.
