        Evaluation for Impacts of Nitrogen Source to Groundwater Quality in Livestock Farming Area

        We investigated 52 livestock farming complexes in Gyeong-Gi and Incheon provinces based on low, medium, and high livestock density and groundwater quality. The objective of this study was to evaluate a relationship between nitrate N concentration in groundwater and animal factors, such as livestock density and animal species. 2,200 groundwater samples for 3 years from 2012 to 2014 at Gyeong-Gi and Incheon provinces were collected and analyzed for pH, EC, DO, ORP, temperature, major anions and cations, such as NO₃-N, HCO₃<SUP>-</SUP>, PO₄<SUP>-</SUP>, SO₄<SUP>2-</SUP>, Cl<SUP>-</SUP>, NH₄-N, K<SUP>+</SUP>, Na<SUP>+</SUP>, Ca<SUP>2+</SUP>, Mg<SUP>2+</SUP>, T-N, and TOC. Average concentration of total N for generated load density was 23,973 g day<SUP>-1</SUP> km<SUP>-2</SUP> for cattle, 51,551 g day<SUP>-1</SUP> km<SUP>-2</SUP> for pig, and 52,100 g day<SUP>-1</SUP> km<SUP>-2</SUP> for poultry. For animal feeding species, average ratio for generated load over discharge load was 16.1% for cattle, 7.8% for pig, and 7.1% for poultry. Therefore, cattle feeding region is highly vulnerable for water pollution compared to pig and poultry feeding areas. The concentrations of chloride, nitrate, and total N in the groundwater samples were higher at high animal farming regions than other regions. The average concentration of nitrate, and chloride in groundwater samples was 5.0 mg L<SUP>-1</SUP>, 16.6 mg L<SUP>-1</SUP> for low livestock density, 6.9 mg L<SUP>-1</SUP>, 17.7 mg L<SUP>-1</SUP> for medium livestock density and 7.6 mg L<SUP>-1</SUP>, 22.7 mg L<SUP>-1</SUP> for high livestock density and total nitrogen (T-N) was 7.7 mg L<SUP>-1</SUP> for low livestock density, 9.4 mg L<SUP>-1</SUP> for medium livestock density, 10.7 mg L<SUP>-1</SUP> for high livestock density. In conclusion, based on this research, for managing groundwater quality near livestock farming regions, Ca-(Cl+NO₃) group from the Piper diagram is more efficient than using 19 factors for water quality standard.

In conclusion, based on this research, for managing groundwater quality near livestock farming regions, $Ca-(Cl+NO_3)$ group from the Piper diagram is more efficient than using 19 factors for water quality standard.

        Excision with Temporary Interphalangeal Joint Pin Fixation for Toe Ganglion Cysts

        Background: Toe ganglion cysts are often symptomatic and recurrent. Communicating lesions between ganglion cysts and the interphalangeal joint (IPJ) or tendon sheath make it difficult to prevent a recurrence. Temporary restriction of the joint and tendon motion can facilitate surgical site healing. This study analyzed the clinical results of temporary pin fixation of the IPJ after toe ganglion cyst excision. Methods: Sixteen patients with symptomatic toe ganglion cysts underwent surgical treatment. Excision alone was initially performed on 10 patients. Six patients underwent temporary pin fixation of the IPJ after ganglion cyst excision. Repeat excision with pin fixation was performed for recurrence in two patients after excision only. Clinical evaluations and postoperative complications were analyzed. Results: Fourteen of 16 toe ganglion cysts were located near the IPJ. Two cysts not adjacent to the joint completely healed after excision alone. Seven of 14 cysts near the joint recurred after initial excision alone and required repeated reoperation. Eight cysts did not recur after excision with pin fixation, including 2 that recurred after excision alone. Conclusions: Temporary IPJ pin fixation after excision for ganglion cysts can be effective for preventing the recurrence of ganglion cysts adjacent to toe IPJ.

        Ethyl Acetate Fraction from <i>Cudrania Tricuspidata</i> Inhibits IL-1β-Stimulated Osteoclast Differentiation through Downregulation of MAPKs, c-Fos and NFATc1

        <P><B>Background/Aims</B></P><P>The present study was performed to determine the effects of the ethyl acetate extract of <I>Cudrania tricuspidata</I> (EACT) on interleukin (IL)-1β-stimulated receptor activator of NF-κB ligand (RANKL)-mediated osteoclast differentiation.</P><P><B>Methods</B></P><P>Bone marrow cells were harvested from 6-week-old male imprinting control region mice, and the differentiation of osteoclasts from these cells was evaluated by tartrate-resistant acid phosphatase and resorption pit formation assay. Phosphorylated extracellular signal regulated kinase (p-ERK), phosphorylated p38, phosphorylated c-Jun amino-terminal kinase, NF-κB (p65), IκBα, c-Fos, and nuclear factor of activated T-cells c1 (NFATc1) expression was examined by immunoblotting and quantitative reverse transcription-polymerase chain reaction.</P><P><B>Results</B></P><P>EACT inhibits IL-1β-stimulated RANKL-mediated osteoclast differentiation. EACT also inhibits IL-1β-stimulated RANKL-mediated phosphorylation of ERK 1/2, p38 mitogen activated protein kinase, and expression of c-Fos and NFATc1.</P><P><B>Conclusions</B></P><P>These results suggest that EACT may be involved in the inhibition of bone loss by preventing osteoclast formation and may be used to manage bone destruction in inflammatory diseases, such as rheumatoid arthritis.</P>

        <P>Objectives: The objective of this study is to determine the effects of Ethyl acetate fraction from Cudrania tricuspidata (EACT) on the interleukin-1b (IL-1b)-induced proliferation of rheumatoid synovial fbroblasts (RASFs) and production of matrix metalloproteinases (MMPs), cyclooxygenase (COX) and prostaglandin E2 (PGE2) by RASFs. Materials and Methods: The proliferation of RASFs was evaluated with CCK-8 reagent in the presence of IL-1b with/without EACT. The expression of MMPs, TIMP-1, COXs, PGE2 and intracellular MAPK signalings, including p-ERK, p-p38, p-JNK and NF-kB were examined by immunoblotting or semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and ELISA in conditions as described above. Results: EACT inhibits IL-1β-induced proliferation of RASFs and MMP-1, 3, COX-2 mRNA and protein expression, PGE2 production induced with IL-1b. EACT also inhibits the phosphorylation of ERK-1/2, p38, JNK and activation of NF-kB by IL-1b. Conclusions: These results suggest that EACT might be involved in synovial fbroblast proliferation and MMPs, COX-2, and PGE2 production, which are involved in joint destruction in rheumatoid arthritis (RA), indicating that this might be a new therapeutic modality for management of rheumatoid arthritis.</P>

        Directing vanillin production from ferulic acid by increased acetyl-CoA consumption in recombinant Escherichia coli

        <P>The amplification of gltA gene encoding citrate synthase of TCA cycle was required for the efficient conversion of acetyl-CoA, generated during vanillin production from ferulic acid, to CoA, which is essential for vanillin production. Vanillin of 1.98 g/L was produced from the E. coli DH5α (pTAHEF-gltA) with gltA amplification in 48 h of culture at 3.0 g/L of ferulic acid, which was about twofold higher than the vanillin production of 0.91 g/L obtained by the E. coli DH5α (pTAHEF) without gltA amplification. The icdA gene encoding isocitrate dehydrogenase of TCA cycle was deleted to make the vanillin producing E. coli utilize glyoxylate bypass which enables more efficient conversion of acetyl-CoA to CoA in comparison with TCA cycle. The production of vanillin by the icdA null mutant of E. coli BW25113 harboring pTAHEF was enhanced by 2.6 times. The gltA amplification of the glyoxylate bypass in the icdA null mutant remarkably increased the production rate of vanillin with a little increase in the amount of vanillin production. The real synergistic effect of gltA amplification and icdA deletion was observed with use of XAD-2 resin reducing the toxicity of vanillin produced during culture. Vanillin of 5.14 g/L was produced in 24 h of the culture with molar conversion yield of 86.6%, which is the highest so far in vanillin production from ferulic acid using recombinant E. coli.         Effects of various concentrations of garlic powder and garlic extract in the diets on growth, serum chemistry and immune response of juvenile olive flounder Paralichthys olivaceus

        Effects of various concentrations of garlic powder and garlic extract in the diets on growth, serum chemistry and immune response of olive flounder were determined. Thirty-five juvenile fish averaging 5.1 g were randomly distributed into 21 of 180 L flow-through tanks. Seven experimental diets with various concentrations of garlic powder (GP) and garlic extract (GE) were prepared in triplicate: GP-0 without garlic supplementation, GP-0.5, GP-1, GP-2, GP-3 and GP-5 diets containing garlic powder at the concentrations of 0.5, 1, 2, 3 and 5%, respectively at the expense of wheat flour and finally, GE-0.4 diet containing 0.4% garlic extract were prepared. At the end of the 8-week feeding trial, serum chemistry of fish was measured. In addition, twenty fish from each tank were artificially infected with E. tarda for the following 96 h to monitor cumulative mortality. Weight gain of fish fed GP-0 diet was higher than that of fish fed GP-1, GP-2, GP-3 and GP-5 diets. No difference in serum criteria (total protein, glucose, glutamate oxaloacetate transaminase, cholesterol and triglyceride levels) of olive flounder was found among the experimental diets except for glutamate pyruvate transaminase. Lysozyme activity of fish fed GP-0, GP-1, GP-3 and GE-0.4 diets was higher than that of fish fed GP-5 diet. The highest cumulative mortality was 93.3% in fish fed GP-0 diet at 96 h after E. tarda infection, followed by GP-3, GP-1, GP-5, GP-2, GP-0.5 and GE-0.4 diets. In considering these results, dietary inclusion of garlic powder and garlic extract has no distinctive positive effect on improvement in growth, serum chemistry and immune response of olive flounder in this experimental conditions, therefore, its application should be carefully considered.

        Functional characterization of NtCDPK1 in tobacco.

        <P>We previously showed that NtCDPK1, a tobacco cal-cium-dependent protein kinase, interacts with and phosphorylates the Rpn3 regulatory subunit of the 26S proteasome, and that both NtCDPK1 and Rpn3 are mainly expressed in rapidly proliferating tissues, in-cluding shoot and root meristem. In this study, we ex-amined NtCDPK1 expression in roots using GUS ex-pression in transgenic Arabidopsis plants, and investi-gated its function in root development by generating transgenic tobacco plants carrying a sense NtCDPK1 transgene. GUS activity was first detected in roots two days after sowing. In later stages, strong GUS expres-sion was detected in the root meristem and elongation zone, as well as the initiation sites and branch points of lateral roots. Transgenic tobacco plants in which NtCDPK1 expression was suppressed were smaller, and their root development was abnormal, with reduced lateral root formation and less elongation. These re-sults suggest that NtCDPK1 plays a role in a signaling pathway regulating root development in tobacco.</P>

        Temporal expression of AMP-activated protein kinase activation during the kainic acid-induced hippocampal cell death.

        <P>Kainic acid (KA)-induced seizure induces the hippocampal cell death. There are reports that AMP-activated protein kinase (AMPK), which is an important regulator of energy homeostasis of cells, has been proposed as apoptotic molecule. In this study, we investigated the altered expression of AMPK cascade in the hippocampus of mice during KA-induced hippocampal cell death. Mice were killed at 2, 6, 24 or 48 h after KA (30 mg/kg) injection. Histological evaluation of KA-treated hippocampus revealed hippocampal cell death first at 6 h and appearing prominently by 48 h after KA injection. Immunoreactivity of Ca(2+)/calmodulin-dependent protein kinase kinasebeta (CaMKKbeta) was increased after KA treatment. In Western blot analysis, AMPK activation was increased 2 h after KA treatment. The proteins of downstream AMPK, including those of glucose transporter1 (GLUT1) and phosphorylation of Acetyl CoA Carboxylase (ACC) were increased in the hippocampus after KA treatment. These results indicate that sustained AMPK activation might be a mechanism by which KA-induced seizure causes hippocampal cell death of mice.</P>

        Characterization and Genomic Analysis of Novel Bacteriophage ΦCS01 Targeting Cronobacter sakazakii

        Cronobacter sakazakii is an opportunistic pathogen causing serious infections in neonates. In this study, a bacteriophage ΦCS01, which infects C. sakazakii, was isolated from swine feces and its morphology, growth parameters, and genomic analysis were investigated. Transmission electron microscopy revealed that ΦCS01 has a spherical head and is 65.74 nm in diameter with a 98.75 nm contracted tail, suggesting that it belongs to the family Myoviridae. The major viral proteins are approximately 71 kDa and 64 kDa in size. The latent period of ΦCS01 was shown to be 60 min, and the burst size was 90.7 pfu (plaque-forming units)/ infected cell. Bacteriophage ΦCS01 was stable at 4-60°C for 1 h and lost infectivity after 1 h of heating at 70°C. Infectivity remained unaffected at pH 4-9 for 2 h, while the bacteriophage was inactivated at pH < 3 or >10. The double-stranded ΦCS01 DNA genome consists of 48,195 base pairs, with 75 predicted open reading frames. Phylogenetic analysis is closely related to that of the previously reported C. sakazakii phage ESP2949-1. The newly isolated ΦCS01 shows infectivity in the host bacterium C. sakazakii, indicating that it may be a promising alternative to antibacterial agents for the removal of C. sakazakii from powdered infant formulas.
