        A Case Study on Physical Characteristics of High School Male Elite Athletes according to Sport Type: Focusing on the Target Elite Athletes of Seoul Sports Science Research Institute

        Since 2015, Korea has been operating regional sports science centers in 10 cities across the country, and is pursuing a plan to systematically link athletes’ physical fitness through physical fitness measurements according to the athlete’s life cycle. For 243 elite athletes whose physical strength was measured by the Seoul Sports Science Center, the development direction of each sport type of the elite athletes in Seoul is analyzed through classification and grade. As a result of the study, in terms of basic physical strength, the difference in physical strength by sport type was significant in all items of muscle strength, muscular endurance, muscle power, cardiopulmonary endurance, agility, balance, and coordination except flexibility. As a result of analyzing the difference in the level of the physical strength factor according to the application of the fitness grade criteria for each sport type, the A type showed significantly higher differences in grip strength and back strength than the B and D types. It was significantly higher in sit-up than type A. Type B showed significantly higher physical endurance, muscle power, cardiopulmonary endurance, agility, coordination, and equilibrium compared to the A and D types, and D type showed higher physical strength level than the A type in cardiopulmonary endurance, agility, and balance. Based on the results of this study, it is considered necessary to compare the performance and physical strength of male high school students in Seoul and the characteristics of athletes by sport type to review the training method and improvement of the performance.

        Methylobacterium terrae sp. nov., a radiation-resistant bacterium isolated from gamma ray-irradiated soil

        A Gram-stain-negative, asporogenous, aerobic rods, motile by means of a single polar flagellum, catalase- and oxidase-positive, methylotrophic bacterium, designated 17Sr1-28T, was isolated from gamma ray-irradiated soil. The 16S rRNA gene sequence analysis showed that strain 17Sr1-28T was phylogenetically related to Methylobacterium currus PR1016AT (96.8%), Methylobacterium platani PMB02T (96.2%), Methylobacterium aquaticum DSM 16371T (96.3%), Methylobacterium tarhaniae N4211T (96.4%), Methylobacterium frigidaeris IER25-16T (95.8%), and Methylobacterium organophilum JCM 2833T (92.7%). The G+C content calculated based on genome sequence was 71.6%. The average nucleotide identity and in silico DNA-DNA hybridization values between strain 17Sr1- 28T and M. currus, M. platani, M. aquaticum, M. tarhaniae, M. frigidaeris, and M. organophilum were 77.7–90.4% and 22–39.6%, respectively. The major fatty acids of strain 17Sr1- 28T were summed feature 8 (C18:1 ω7c and/or C18:1 ω6c), and summed feature 3 (C16:1 ω7c and/or C16:1 ω6c). The predominant quinone was ubiquinone 10 and the major polar lipids were diphosphatidylglycerol, phosphatidylcholine, phosphatidylethanolamine, and phosphatidylglycerol. On the basis of the data from phenotypic tests and genotypic differences between strain 17Sr1-28T and its close phylogenetic relatives, strain 17Sr1-28T represents a new species belonging to the genus Methylobacterium, for which the name Methylobacterium terrae sp. nov. (= KCTC 52904T = NBRC 112873T) is proposed.

        Chronic extensor hallucis longus (EHL) tendon rupture is relatively rare, but in such cases, surgical repair is necessary to prevent hallux dysfunction. To the best of our knowledge, reconstruction of chronic EHL rupture using a split tibialis anterior tendon autograft has not been previously reported. Here we present a case of spontaneous EHL tendon rupture with a 5 cm gap in a healthy 57-year-old woman. At the 1-year follow-up evaluation, hallux function was restored, and the patient was well satisfied with results.

        족부족관절 분야의 환자 자가 보고 결과 평가법

        The patient-reported outcome measures (PROMs) are important in assessing the patient’s overall health, lesion-specific outcomes, and condition-specific outcomes. PROMs are also known as “scoring systems” and are usually in questionnaires. There are almost a hundred different PROMs available in foot and ankle surgery. Each PROM has its merits, demerits, and validity. Selecting an appropriate PROM is important for adequately evaluating a patient’s health status. This article summarizes the most frequently used PROMs in the literature on foot and ankle surgery and presents the authors’ recommendations.

        Modified Weil Osteotomy for the Treatment of Freiberg’s Disease

        Background:Numerous metatarsal osteotomies have been developed for the treatment of Freiberg’s disease. The purpose of this study was to evaluate the clinical outcomes of modified Weil osteotomy in the treatment of Freiberg's disease. Methods: From November 2001 to July 2008, nineteen patients (twenty feet), treated surgically for Freiberg's disease, were included in this study. The average age of the patients was 33.6 years (range, 17 to 62 years), the mean follow-up period was 71.6 months (range, 41 to 121 months). Clinical outcomes were evaluated according to visual analogue scale (VAS), American Orthopaedic Foot and Ankle Society (AOFAS) lesser metatarsophalangeal-interphalangeal scale, the patients’ subjective satisfaction and range of motion (ROM) of metatarsophalangeal (MTP) joint. In the radiologic evaluation, initial metatarsal shortening by Freiberg’s disease compared to opposite site, metatarsal shortening after modified Weil osteotomy compared with preoperative radiography and term for radiologic union were observed. Results: VAS showed improvement from 6.2 ± 1.4 to 1.4 ± 1.5 at last follow-up (p < 0.0001). Points of AOFAS score increased from 63.3 ± 14.9 to 80.4 ± 5.6 (p < 0.0001). ROM of MTP joints also improved from 31.3 ± 10.1 to 48.3 ± 13.0 degrees at last follow-up (p < 0.0001). According to Smillie's classification system, there was no significant improvement of VAS, AOFAS score and ROM between early stages (stage I, II, and III) and late stages (stage IV and V). Out of twenty cases, nineteen (95%) were satisfied, reporting excellent or good results. Conclusions: Modified Weil osteotomy is believed to be a useful method for the treatment of Freiberg's disease, not only in the early stages but also in the late stages. It relieves pain and improves function via shortening of metatarsals and restoration of MTP joint congruency.

        Preparation and Polymerization Behavior of Polymeric Dental Restorative Materials Containing High Molecular Weight Diluent System

        Preparation of polymeric dental restorative materials (PDRM) showing a lower polymerization shrinkage was carried out using the hybrid-filler and 2,2'-bis-[4-(3-methacryloxy-2-hydroxypropoxy) phenyl]propane resin matrix that is diluted with a high molecular weight dimethacrylate monomer. A visible light system was utilized to activate the resin matrix for the polymerization. In order to enhance the miscibility of the inorganic hybrid-filler with organic resin matrix and to conduct the homogeneity of the filler in the PDRM, its surface was hydrophobically treated with y -methacryloxypropyltrimethoxysilane. The degree of conversion (DC), depth of cure, and polymerization shrinkage of the PDRM were investigated. The results revealed that PDRM prepared herein showed markedly lower polymerization shrinkage values. Besides, regardless of the filler loading, both DC and depth of cure values decreased with an increase in the amount of less flexible diluent in the resin matrix.

        Molecular Mechanism of Atopic Dermatitis Induction Following Sensitization and Challenge with 2,4-Dinitrochlorobenzene in Mouse Skin Tissue

        Laboratory animal models have been developed to investigate preventive or therapeutic effect of medicinal products, or occurrence or progression mechanism of atopic dermatitis (AD), a pruritic and persistent inflammatory skin disease. The murine model with immunologic phenomena resembling human AD was introduced, which demonstrated skewedness toward predominance of type-2 helper T cell reactivity and pathophysiological changes similar as human AD following 2,4-dinitrochlorobenzene (DNCB) sensitization and challenge. Molecular mechanism on the DNCB-mediated AD was further evaluated. Skin tissues were collected from mice treated with DNCB, and each tissue was equally divided into two sections; one for protein and the other for mRNA analysis. Expression of filaggrin, an important protein for keratinocyte integrity, was evaluated through SDS-PAGE. Level of mRNA expression for cytokines was determined through semi-quantitative reverse transcriptase polymerase chain reaction. Expression of filaggrin protein was significantly enhanced in the mice treated with DNCB compared with the vehicle (acetone : olive oil = 4 : 1 mixture) treatment group or the normal group without any treatment. Level of tumor necrosis factor-alpha and interleukin-18 mRNA expression, cytokines involved in activity of type-1 helper T (TH1) cell, was significantly downregulated in the AD group compared with other control groups. These results suggest that suppression of TH1 cell-mediated immune response could be reflected into the skin tissue of mice treated with DNCB for AD induction, and disturbance of keratinocyte integrity might evoke a compensatory mechanism.

      • Synthesis and biological evaluation of 3-(2-aminoethyl) uracil derivatives as gonadotropin-releasing hormone (GnRH) receptor antagonists

        <P><B>Abstract</B></P> <P>We investigated a series of uracil analogues by introducing various substituents on the phenyl ring of the <I>N</I>-3 aminoethyl side chain and evaluated their antagonistic activity against human gonadotropin-releasing hormone (GnRH) receptors. Analogues with substituents at the ortho or meta position demonstrated potent in vitro antagonistic activity. Specifically, the introduction of a 2-OMe group enhanced nuclear factor of activated T-cells (NFAT) inhibition up to 6-fold compared to the unsubstituted analogue. We identified compound <B>12c</B> as a highly potent GnRH antagonist with moderate CYP inhibition. Compound <B>12c</B> showed potent and prolonged LH suppression after a single dose was orally administered in castrated monkeys compared to a known antagonist, Elagolix.         Isolation of Angiotensin Converting Enzyme Inhibitors from Compositae Plants

        Thirty plants of the Compositae family were screened for inhibitory activity of angiotensin converting enzyme (ACE). Among them, Chrysanthemum boreale Makino and Ixeris dentate Nakai were selected for further investigation since they had the highest inhibitory activity. Crude water extracts of the flowers of Chrysanthemum boreale Makino and the roots of Ixeris dentate Nakai were prepared by heating at 95℃ and 60℃ for 2 hr, respectively, followed by centrifugation at 8000×g for 30 min. Crude extracts were then filtered using YM-3 and YM-1 membranes. The ACE inhibitors were isolated using consecutive chromatographic methods including: Sephadex G-15, ion exchange, FPLC, and reverse phase HPLC. The inhibitors were identified to have molecular masses of 204 and 283 daltons, respectively, by mass spectrometry. These results demonstrate that crude extracts of Compositae plants may be useful as functional food ingredients with anti-hypertensive properties.

      • Cellular phosphatase activity of C1-Ten/Tensin2 is controlled by Phosphatidylinositol-3,4,5-triphosphate binding through the C1-Ten/Tensin2 SH2 domain

        <P><B>Abstract</B></P> <P>Regulation of tyrosine phosphorylation on insulin receptor substrate-1 (IRS-1) is essential for insulin signaling. The protein tyrosine phosphatase (PTP) C1-Ten/Tensin2 has been implicated in the regulation of IRS-1, but the molecular basis of this dephosphorylation is not fully understood. Here, we demonstrate that the cellular phosphatase activity of C1-Ten/Tensin2 on IRS-1 is mediated by the binding of the C1-Ten/Tensin2 Src-homology 2 (SH2) domain to phosphatidylinositol-3,4,5-trisphosphate (PtdIns(3,4,5)P<SUB>3</SUB>). We show that the role of C1-Ten/Tensin2 is dependent on insulin-induced phosphoinositide 3-kinase activity. The C1-Ten/Tensin2 SH2 domain showed strong preference and high affinity for PtdIns(3,4,5)P<SUB>3</SUB>. Using site-directed mutagenesis, we identified three basic residues in the C1-Ten/Tensin2 SH2 domain that were critical for PtdIns(3,4,5)P<SUB>3</SUB> binding but were not involved in phosphotyrosine binding and PTP activity. Using a PtdIns(3,4,5)P<SUB>3</SUB> binding-deficient mutant, we showed that the specific binding of the C1-Ten/Tensin2 SH2 domain to PtdIns(3,4,5)P<SUB>3</SUB> allowed C1-Ten/Tensin2 to function as a PTP in cells. Collectively, our findings suggest that the interaction between the C1-Ten/Tensin2 SH2 domain and PtdIns(3,4,5)P<SUB>3</SUB> produces a negative feedback loop of insulin signaling through IRS-1.</P> <P><B>Highlights</B></P> <P> <UL> <LI> C1-Ten's function as a PTP for IRS-1 in insulin signaling is controlled by PI3K. </LI> <LI> The C1-Ten SH2 domain binds to PtdIns(3,4,5)P<SUB>3</SUB> with high affinity. </LI> <LI> The PtdIns(3,4,5)P<SUB>3</SUB> binding of C1-Ten is necessary for its function as PTP. </LI> </UL> </P>
