백합의 품종별 소자구 형성에 미치는 절편 및 생장조절물질의 영향

백합 오리엔탈 및 아시아틱 계통의 구근으로부터 소자구의 기내대량번식을 시도할 때 영향을 미치는 절편 종류 및 부위, 생장조절물질로서 cytokinin류의 농도별, auin으로서 NAA를 농도별로 처리하여 자구의 증식과 비대에 미치는 영향을 검토하였다. 품종별 자구발생율은 'Lereve'와 'Casa Blanca'에서, 자구의 비대는 'Pesaro'에서 양호하게 이루어졌다. 절편부위로는 화경과 인편조직에서 자구의 발생이 많았으나 자구의 비대는 생장점 부위, 화경, 인편조직의 순으로 나타나 품종과 절편부위에 따라 자구의 생육상이 다르게 나타났다. 처리된 생장조절물질 가운데 BAP 첨가가 자구발생과 구의 비대에 적절하였으며 kinetin은 전 품종에 대해 억제작용을 나타내었다. NAA를 0.3 ㎎/L로 첨가하였을 때 'Dame Blanche'와 'Casa Blanca'의 자구발생을 촉진하였으나 'Cherry Blossom'과 'Acapulco'에 대해서는 자구의 비대에 촉진적으로 작용하여 품종별로 기관분화에 각각 다르게 영향하는 것으로 나타났다. Effect of explant sources, growth regulators such as cytokinins and auxin on in vitro induction and growth of bulblets from several cultivars of Lilium were investigated. 'Lereve' and 'Casa Blanca' showed the excellent abilities of bulblet regeneration and 'Pesaro' exhibited maximal increment of bulblet weight among tested. Though there were some difference according to cultivars, flower stems and/or bulb scale were more desirable to use as explant sources for the mass proliferation of bulblets, BAP was found to be the most effective cytokinin for bulblet formation. But bulblet growth was severely inhibited by kintin treatment. Different response were observed among cultivars when bulblets were grown in the medium containing NAA. Low concentration of NAA not only showed stimulation effect on bulblet formation of 'Dame Blanche', 'Casa Blanca' but also enhanced increment of bulblet weight of 'Cherry Blossom' and 'Acapulco'. These results suggested that NAA may play a role in oranogenesis in Lilium.

Somatic hybridization between axenic shoot culture of Dendranthema grandiflorum and albino cell suspension of Salpiglossis sinuata was investigated. Out of six fusion protocols and three culture media, electrofusion and KP8 medium were found to be most effective in generating somatic hybrids. Hybrid selection strategy, based on complementation for chlorophyll proficiency by the fusion of a non-dividing mesophyll protoplast system and a cytoplasmic albino mutant protoplast system was effective. Following plant regeneration, the hybridity of the regenerated plants was confirmed by morphological, cytological and biochemical analyses. Morphologically, these hybrids resembled D. grandiflorum. The somatic chromosome numbers of these plants were 2n=60. Leaf peroxidase and esterase isoenzyme analyses showed that the regenerated plants contained the banding patterns of D. grandiflorum, plus two bands associated with S. sinuata in the peroxidase profiles.
CO₂-Enrichment and Photosynthetic Photon Flux Affect the Growth of In Vitro-Cultured Apple Plantlets

Micro-cutting (shoots with two small leaves) of cultivar M9 apple were cultured in-vitro for 40 d under CO2-enriched and non-enriched (i.e., ambient air) conditions, and at a PPF of 40 or 100 μmol m-2 s-1. Afterward, shoot length, number of leaves, leaf area, chlorophyll content, shoot and root fresh weights, and % survival were recorded. Those plantlets grown under CO2 -and PPF-enriched treatments were healthy and vigorous, and showed higher values for their growth parameters. In contrast, those grown without supplemental CO2 or PPF often showed hyperhydricity. We also demonstrated that CO2 enrichment and a relatively high PPF during in-vitro culture promoted normal photosynthesis and growth after ex-vitro transplantation.
