Expression of antimicrobial peptide PAJE in Escherichia coli

The antibiotic peptide PAJE (RWKIFKKPFKISIHL-NH2), designed incorporating the N-terminal α-helical segments of papiliocin and jelleine, is a 15-residue hybrid peptide that has a broad spectrum of activity against Gram-negative, positive bacteria and fungi. In this study, we successfully expressed bioactive PAJE in Escherichia coli cells that are highly sensitive to this peptide. For the efficient production of peptide, we synthesized gene encoding PAJE, and fused the sequence in-frame to ketosteroid isomerase (KSI) gene to construct an expression vector pET29b-PAJE-KSI, which was then used to transform E. coli BL21 (DE3). The fusion protein PAJE-KSI was expressed as inclusion body at high level (more than 30% of the total proteins). Recombinant PAJE was easily released by cleavage of the fusion protein with cyanogen bromide (CNBr). Subsequently, we purified the recombinant PAJE by FPLC chromatography. The purified PAJE displayed considerably antibacterial activity identical to that previously reported for chemically synthesized PAJE. The results indicated that successful expression of PAJE in E. coli cells and efficient procedure for purification may lead to a cost-effective platform for the mass production of PAJE.

The purpose of this study was to collect the basic data about oral health of residents in Chollabuk-do Province of Korea. 11 sampling areas were chosen by Board of National Statistics in this province. 5 areas were city areas including capital city of Chollabuk-do and 6 areas were rural areas. The number of total sampling person was 676 adults aged 18 years old and above, The survey was conducted by individual home-visiting method. The contents of survey were, caries prevalence, community periodontal index, number of present tooth. The data were collected and analyzed by SPSS/PC. The obtained results were as follows ; 1. The DMF rate of residents was 89.5% in all age groups, The DMF rate was lowest in 30-34 year group as 74.5% and highest in 75 over year group as 100.0%. 2, The average DMFT index per person was 8.72 teeth in all age groups. But that was 7.39 teeth in male and 9.73 teeth in female, there was a statistically different in the average DMF index. 3. The average percent of level 2 by community periodontal index (needing calcalus removal and oral prophylaxis) was 69.9%. 4. The average number of present tooth in oral cavity was 22,72 teeth in all age group. But that of was 14.55 teeth in 65-74 aged group, and 10.80 teeth in 75 over aged group, the number of present teeth was decreased rapidly after 65 year old.

The potentiometric measurement result for a high resistance up to 100 PΩ using a low resistance, a low voltage source and a commercial digital multimeter(DMM) is presented. With the method, a resistance can be easily, fast and economically measured. Using the method, resistance ranges over the 10 GΩ range which is difficult to measure using a commercial DMM and resistance ranges between 100 TΩ and 100 PΩ which cannot measure using an insulation tester were measured within accuracy of a few percent. It is expected that it can be useful to determine the temperature and voltage effect of a high resistance and an insulation material because it uses a reference resistance with a low resistance, very low temperature and voltage effect. Besides, it is expected that it can be useful to calibrate a dc high voltage divider with a large resistance ratio and a very low resistance because arbitrary resistance ratio measurements are possible with it.

<P>The well-defined hierarchical superstructures constructed by the self assembly of programmed supramolecules can be organized for the fabrication of remote-controllable actuating and rewritable films. To realize this concept, we newly designed and synthesized a benzene-1,3,5-tricarboxamide (BTA) derivative (abbreviated as BTA-3AZO) containing photoresponsive azobenzene (AZO) mesogens on the periphery of the BTA core. BTA-3AZO was first self-assembled to nanocolumns mainly driven by the intermolecular hydrogen-bonds between BTA cores, and these self-assembled nanocolumns were further self-organized laterally to form the low-ordered hexagonal columnar liquid crystal (LC) phase below the isotropization temperature. Upon cooling, a lamello-columnar crystal phase emerged at room temperature via a highly ordered lamello-columnar LC phase. The three-dimensional (3D) organogel networks consisted of fibrous and lamellar superstructures were fabricated in the BTA-3AZO cyclohexane-methanol solutions. By tuning the wavelength of light, the shape and color of the 3D networked thin films were remote-controlled by the conformational changes of azobenzene moieties in the BTA-3AZO. The demonstrations of remote-controllable 3D actuating and rewritable films with the self-assembled hierarchical BTA-3AZO thin films can be stepping stones for the advanced flexible optoelectronic devices.</P>

N/A Lung cancer has become one of the most common cancers in Korea. Chest X-rays show solitary or diffuse infiltrative lesions but these arc not confirmed by sputum chtology and bronchoscopic exam. It is extremely necessary to establish a tissue diagnosis. Transbronchial lung biopsy and transthoracic needle lung biopsy were compared with respect to diagnostic accuracy and complications. The age distribution of the patients was from nineteen to seventy-five years and the male to female ratio was about 6:1. The diagnostic yields of the transbronchial lung biopsy and the transthoracic needle lung biopsy were 68% and 86% respectively. Common diseases were primary malignancy, pulmonary tuberculosis and interstitial fibrosis; next in order were pneumonia and lung abscess. Among the nonspecific diagnoses, 7 cases were confirmed as malignant diseases by subsequent diagnostic procedures. Complications of the transthoracic needle lung biopsy were pneumothorax in 5 cases (8%) hemothorax in one case (2%) and hemoptysis in 2 cases (4%) Complications of the transbronchil lung biopsy were hemothorax in one case (2%), and hemoptysis in two cases (4%). We think that the transthoracic needle lung biopsy represents minor, safe, accurate and simple procedure for peripheral lung lesions and the transbronchial lung biopsy is the procedure of choice for central lung lesions.

On previous studies, we constructed a transgenic silkworm which produces the chimeric silk fused green fluorescent protein (EGFP), but the transgenic silkworm has decreased commercial feasible traits such as convenience of breeding and productivity of silk. In this study, we performed cross fertilization between green fluorescent silk transgenic silkworm and colored cocoon silkworm descents to make the transgenic the transgenic silkworm producing improved fluorescence cocoon. In the result, we found out a bit valuable cross fertilization manners (female × male) in respect of silk productivity such as T59B × Jam126, Jam329 × T59W, T59W × Jam329, and T59W × Jam178. The color-difference of offspring cocoons were measured according to different cross manners using by CIE Lab-based formulae with a Xrite VS450. In the result, the depth of green color of cocoons was a little high at cross manners as Jam329 × T59W, T59W × Jam178. Meanwhile, the depth of yellow clolor of cocoons was remarkable at cross manners as Jam178 × T59W, T59W × Jam178, respectively.

Prostate cancer has been a critical health problem due to an increase of prostate cancer-related deaths worldwide. Also, a frequent treatment option for prostate cancer is androgen ablation, but this treatment has a limited scope, especially for hormone-refractory cancer. There is an urgent need for the identification of alternative therapeutic strategies for prostate cancer. Previously, over one hundred species of dried-plant methanol extracts were tested for inhibitory effects on proliferation. One of them, Piper longum Linn. was selected based on its potent anti-proliferation effect. The dried root of P. longum Linn. was extracted with 100% methanol for 2-3 days and its extract was fractionated using chloroform. The chloroform layer was then subjected to column chromatography on silica gel, reverse phase-18 (RP-18) and Sephadex LH-20, in turn. Finally, the pure compound was obtained and identified as pipernonaline by NMR spectroscopic and physico-chemical analysis. In this study, anti-proliferation and cell cycle arrest effects of pipernonaline on human prostate cancer PC-3 cells were investigated using the MTT and PI staining, respectively. Our findings suggest that pipernonaline represents a dose-dependent growth inhibition pattern on PC-3 cells and, moreover, its growth inhibition is associated with sub-G1 and G0/G1 cell cycle accumulation in PC-3 cells. Also, these results provide an anticancer candidate for human prostate cancer.

절연재료 및 1 Tohm(1E12 ohm)이상의 높은 저항기의 절연저항을 정확하게 측정하기 위한 누설저항 효과를 분석하였다. 여러 절연저항 측정 방법을 비교하고 직접 누설저항을 측정한 결과, 정전압법을 사용하면 누설효과 없이 절연저항을 짧은 시간에 정확하게 측정할 수 있음을 확인하였다.
