      • GO-32 : Therapeutic Targeting of C-MET in Ovarian Clear-Cell Carcinoma

        목적: Recent reports revealed that c-Met activation is specifically associated with epithelial ovarian carcinoma and is association with poor prognosis. In this study, we investigated c-Met expression and the effects of its therapeutic targeting in ovarian clear-cell carcinoma (CCC). 방법: Expression levels of c-Met in the CCCs, serous carcinoma and normal ovarian tissues were evaluated using real-time PCR. To test c-Met inhibitors in cell lines including ES2, RMG1, RMG2, OVISE and OVSAHO, we performed in vitro experiment including MTT and apoptosis assay. We performed Western blots to evaluate the c-Met expression and down-stream pathway. Moreover, we performed the in vivo therapy experiment in patient-derived xenograft models (PDX, Avatar) of ovarian CCC to confirm these effects. 결과: The c-Met expression was significantly increased in CCCs compared with serous carcinoma and normal ovarian tissues (p<0.05). When we treated c-Met inhibitor (SU11274, Crizotinib) to ovarian CCC cells, cell viability was significantly decreased and apoptosis was significantly increased. Western blot assay demonstrated that the protein expressions for c-Met signaling pathway were decreased by c-Met inhibitors. Moreover, we found that c-Met inhibitor (SU11274) had significant decreased tumor weight compared with control in a case of PDX (Avatar) models for ovarian CCC. 결론: These results revealed that c-Met inhibitor have significant anti-tumor effects in ovarian CCC, and suggested that c-Met may have a potentials of therapeutic target for ovarian CCCs.

        Quercetin의 급여가 육계 다리육의 산화 안정성에 미치는 영향

        본 연구는 quercetin의 급여가 닭고기 다리육의 지방산패 억제 효과에 미치는 영향을 구명하고자 실시하였다. 1일령 Ross 320마리 육계 병아리를 8처리군으로 나누어 대조군(T1), 항생제 처리군(T2), 비타민 E 20 ppm 처리군(T3), 비타민 E 200 ppm 처리군(T4), quercetin 20 ppm(T5), quercetin 200 ppm(T6), methoxylated quercetin 20ppm(T7), methoxylated quercetin 200 ppm(T8) 처리구로 나누어 35일간 급여하였다. 급여 후 36일째 도계하여 다리육만 분리하였고, 0, 3, 7일 냉장 저장 기간 동안의 단백질 변패도(VBN), 지방산패도(TBARS), 지방산 조성 변화, 미생물학적 특성을 조사하였다. 저장 3일에는 대조군보다 항생제처리군(T2), 비타민 E처리군(T3, T4)와 quercetin 20 ppm 처리군(T5)에서 유의적인 감소를 나타내어 quercetin 20 ppm의 높은 단백질 산패 억제 효과를 보였다(P<0.05). 도계 직후 다리살의 지방산패도는 대조군보다 비타민 E 200 IU 급여한 T4에서 가장 낮은 값을 나타내었으며, 이 값은 항생제 처리구인 T2, quercetin 200 ppm 첨가구인 T5와 유의적인 차이를 나타내지 않아, 비타민 E 200 IU와 유사한 항산화력을 나타냄을 보여주었다(p<0.05). 그러나 저장 7일에 quercetin 20 ppm 처리군(T5)과 200 ppm 처리군(T6)은 항생제 처리군보다도 낮은 총 균수를 나타내어 높은 항균력을 나타내었다(P<0.05). 대장균군은 도계 직후부터 냉장 저장 7일까지 quercetin 처리에 의한 유의적인 감소 효과는 없었다. This study was carried out to determine the effect of dietary supplementation of quercetin and methoxylated quercetin extracted from onions on oxidation of chicken thigh during cold storage. For 35 days, 1-day-old 320 broiler chicks (Ross) were divided into 8 groups and supplemented the diet; basal diet only (T1), T1 with antibiotics (T2), vitamin E 20 IU (T3), vitamin E 200 IU (T4), quercetin 20 ppm (T5), quercetin 200 ppm (T6), methoxylated quercetin 20 ppm (T7), methoxylated quercetin 200 ppm (T8). After slaughtering the broilers, thigh meats were collected and analyzed the oxidation stability during storage at 4℃ for 7 days. Volatile basic nitrogen (VBN) was ranged from 10.98~15.37 mg% until day 3 and quercetin 20 ppm (T5) significantly reduced (p<0.05) the VBN value of thigh as much as antibiotics treatment group (T2). The supplementation of quercetin at a 200 ppm level significantly reduced 2-thiobarbituric acid reactive substances value when compared with the control. Supplementation of 20 ppm quercetin also affected fatty acid composition of chicken thigh and resulted in increase of C18:1w9 and decrease of C18:2w6 on day 3. In microbes of chicken thigh during storage, supplementation of 20 ppm quercetin significantly reduced total microbes compare to control on day 7 (p<0.05). There was no effect on the number of coliforms in thigh with quercetin diet. From these results, the dietary supplementation of 20 ppm of quercetin may have effects on enhancing oxidation stability.

      • The effect of dietary α-lipoic acid, betaine,l-carnitine, and swimming on the obesity of mice induced by a high-fat diet

        <P>We evaluate the effect of supplementation, at 300 mg kg<SUP>−1</SUP>body weight (BW), with the antioxidants α-lipoic acid (AL), betaine (BT),l-carnitine (LC), and the combination of these and exercise on obesity induced by a 9 week high-fat diet (HFD) in mice. Healthy 5 week-old male C57BL/6J mice were divided into 9 groups: (1) CON, control group fed with a commercial mice chow containing 10% crude fat; (2) HFD, high fat diet group fed with a commercial mice chow containing 60% crude fat; (3) HFD-AL, HFD group fed with AL; (4) HFD-BT, HFD group fed with BT; (5) HFD-LC, HFD group fed withLC; (6) HFD-SW, HFD with swimming as an exercise; (7) HFD-SWAL, HFD-AL with swimming; (8) HFD-SWBT, HFD-BT with swimming, and (9) HFD-SWLC, HFD-LC with swimming. The BW of mice withLCand swimming reduced the increase of BW after 9 weeks. Relative adipose tissue weights were reduced by the combinations of antioxidant supplementation and swimming. Levels of serum glucose and leptin were reduced in the HFD-SWLC group when compared with the HFD group. Serum triglyceride and total cholesterol and the size of adipose were also decreased in the HFD-LC and HFD-SWLC groups. These results show thatLCat a dose of 300 mg kg<SUP>−1</SUP>BW was the most effective for reducing fat accumulation in mice with HFD for 9 weeks. In addition, exercise should be given in combination to enhance the BW reduction and serum lipid level.</P>

        Protection effect of donkey hide gelatin hydrolysates on UVB-induced photoaging of human skin fibroblasts

        <P><B>Abstract</B></P> <P>UVB irradiation is a potent photoaging factor and leads to the formation of skin wrinkles. The objective of this study was to investigate the mechanisms of UVB photoaging inhibition of low molecular weight peptides derived from donkey hide gelatin hydrolysates (LDGH) on human skin fibroblasts. The donkey hide was hydrolyzed using proteases, and the LDGH exhibited higher antioxidant activity than the original hydrolysate and inhibited collagenase and elastase activities. We also found that LDGH increases viability of Hs68 cells after UVB irradiation at 100 mJ/cm<SUP>2</SUP>. Moreover, LDGH prevented the decrease in procollagen type I levels caused by exposure to UVB irradiation in Hs68 cells and reduced the up-regulated phosphorylation of p38, ERK, and JNK in the mitogen-activated protein kinase (MAPK) signaling pathway.         The Hi-pass, which is the representative of Green house gas reduction from the road, succeeded to reduce the serious CO₂ emission and fuel with vehicle increase for last years. The increase of user who experiences the convenient and fuel saving effect by Hi-pass occurred. This study was investigated CO₂ emission comparing on the Hi-pass and the normal roads. To measure CO₂ emission, 4 types of test vehicles which is compact, small, medium and large size were used. CO₂ emission was calculated by fuel consumption from test vehicles. The normal roads had 315,835 ton CO₂ emission. However, The Hi-pass roads had 293,057 ton. These test results show that Hi-pass has reduction effect of CO₂ emission of 22,778ton.

        Method of Tier2 is calculated by the CO₂ emission factor and calorific values for intrinsic carbon content of domestic fuel. However, until the present, we have been applying the CO₂ emission factor of IPCC(Intergovernmental Panel On Climate Change) guidelines and calorific values for intrinsic carbon content of domestic fuel to calculate CO₂ emission of Tier2 level. So we think that estimated CO₂ emission is not accurate. In this study, the CO₂ emission factor and calorific values of gasoline, diesel and LPG according to intrinsic carbon content of domestic fuel are developed and be calculated CO₂ emission to be accurate estimation of CO₂ emission. As a results, the CO₂ emission factors of gasoline, diesel and LPG are 72,221㎏/TJ, 73,187㎏/TJ, 66,053㎏/TJ in each. The CO₂ emissions of gasoline, diesel and LPG are also 22,546,595ton, 41,200,441ton, 13,581,060ton in each per year.

        Gallic Acid와 Linoleic Acid로부터 합성한 Octadeca-9,12-dienyl-3,4,5-trihydroxybenzoate의 유전독성학적 안전성 및 화장품 제형을 통한 생리 기능성 평가

        Gallic acid와 linoleic acid의 합성물인 octadeca-9,12-dienyl-3,4,5-trihydroxybenzoate(GA-LA)의 화장품산업에서의 이용 가능성을 알아보기 위하여 유전독성학적 안전성을 확인하고 크림 베이스 유화물을 사용하여 전자공여능, FRAP 및 tyrosinase 활성 억제능력과 같은 GA-LA의 생리 기능성 실험을 실시하였다. 그 결과 유전독성학적 안전성이 확인되었고, 산화에 의한 피부의 노화를 억제하기 위한 항산화 능력과 미백과 관련된 tyrosinase 활성 억제 능력이 있음이 확인되었다. 따라서 GA-LA는 식품산업뿐만 아니라 화장품산업에서 유용한 기능성 물질로 쓰일 수 있을 것이라고 생각되어진다. The objective of this study was to investigate the genotoxicological safety and biological functions of octadeca-9,12-dienyl-3,4,5-trihydroxybenzoate (GA-LA) in cream-based emulsion for future application as a functional cosmetic material as well as food. GA-LA was synthesized chemically from gallic acid and linoleic acid. The Ames test showed that GA-LA did not have mutagenical toxicity. The control cream-based emulsion containing GA-LA was prepared by commercial method and tested for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Ferric reducing antioxidant power (FRAP) and inhibition effect against tyrosinase of the emulsions were tested for the evaluation of antioxidative and skin-whitening activities. The results showed that DPPH radical scavenging activity in the cream-based emulsion containing GA-LA was higher (52.65%) than that of the control (4.30%). The FRAP value of the sample was 12.85%, however, no activity was found in control. The inhibition effect of tyrosinase showed also a higher value (26.29%) when compared to the control. The results indicate that GA-LA, which showed superior antioxidative and skin-whitening activities in cream-based emulsion, is a useful functional material applicable in cosmetic products as well as food.

        약용식물 추출물 급여가 닭 다리육의 항산화 기능에 미치는 영향

        항균 및 항산화 활성을 갖는 약용식물 추출물을 육계에 급여하는 과정에서 약용식물이 갖는 항산화 활성이 계육에 전이될 수 있는가를 확인하기 위하여 다리육을 냉장 저장하면서 항산화 활성과 관능적 특성 변화를 조사하였다. 약용식물 추출물을 0.3%와 1%로 첨가한 사료를 급여시킨 육계의 다리육에서 수분, 조단백질, 조지방 및 조회분의 함량은 차이를 보이지 않았으나, 다리육의 총 페놀 함량을 측정한 결과 처리구가 대조구에 비해 유의적으로 높았으며, 0.3과 1.0% 처리구간 차이는 발견되지 않았다. 닭 다리육의 DPPH 라디칼 소거능은 0.3% 및 1.0% 약용식물 추출물 첨가군에서 대조군보다 약간 높은 라디칼 소거능을 보이는 것으로 나타났다. 또한 TBARS 값은 0.3% 및 1.0% 첨가군의 경우에는 대조군에 비하여 약간 높은 TBARS 값으로 시작하였으나 저장기간에 따른 변화를 보면 대조군에 비해서는 다소 낮은 TBARS 값을 유지하는 것으로 나타났다. 관능검사 결과를 보면 저장 7일 이후에 색, 맛, 냄새 그리고 종합적 기호도에서 약용식물 첨가군이 대조군에 비하여 다소 높은 관능점수를 얻어 계육의 저장 중 산화적 변질이 급여한 약용식물 추출물에 의해 감소되는 것을 확인할 수 있었다. This study was performed to investigate the effect of dietary medicinal herb extract mix (MHEM, mulberry leaf : Japanese honeysuckle : goldthread=48.5:48.5:3.0) on antioxidative activity of chicken thigh meat. The dietary treatments consisted of a corn-soybean meal basal diet (control), basal diet with 0.3% and 1% MHEM. At the end of the feeding trial, thigh meat samples were collected and stored in a refrigerator at 4oC to be analyzed on day 0, 3, 7 and 14. The MHEM did not affect proximate composition of the thigh meat. Total phenol contents of the thigh meats in treatment groups were higher than that of the control (p<0.05). 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity were higher in dietary treatment group. 2-Thiobarbituric acid reactive substances values of the treatment group were lower than that of control at day 7 and 14 and the added level also affected the inhibition of lipid oxidation of thigh meat. In sensory test, panelists preferred treatment group at day 7 and 14. Therefore, results indicate that dietary MHEM added to chicken may retard the oxidative deterioration of chicken thigh meat during cold storage.

        Method of Tier2 is calculated by the CO₂ emission factor and property of domestic fuel. However, until the present, we have been applying the CO₂ emission factor of IPCC(Intergovernmental Panel On Climate Change) guidelines and calorific values for intrinsic carbon content of domestic fuel to calculate CO₂ emission of Tier2 level. So we think that estimated CO₂ emission is not accurate. In this study, the CO₂ emission factor and calorific values of gasoline, diesel and LPG according to property (density, calorific values, intrinsic carbon content) of domestic fuel is developed and be calculated CO₂ emission to be accurate estimation of CO₂ emission. As a results, the CO₂ emission factors of Gasoline, Diesel, LPG and CNG are 72,940kg/TJ, 73,170kg/TJ, 66,070kg/TJ, 55,380kg/TJ in each.
