In this study, the saponin-rich fractions of five individual (two Red and three Black) sea cucumbers (Apostichopus japonicus) in South Korea were investigated for their antiproliferative effect against HL-60, B16F10, MCF-7, and Hep3B tumor cell lines. The red sea cucumber saponin-rich fraction (SSC) from Jeju Island (JRe) decreased the growth of HL-60 with an IC50 value of 23.55 ± 3.40 lg/mL, which represented the strongest anticancer activity among the extracts. Further, SSC downregulated B-cell lymphoma extra-large (Bcl-xL), while upregulating, to different degrees, Bcl-2-associated X protein (Bax), caspase-9, caspase-3, PARP cleavage, and apoptotic bodies in cancer cells. Evidence for SSC inducing apoptosis via the mitochondria-mediated pathway was found. The contents of SSCs were determined using ultra high-performance liquid chromatography coupled with a quadrupole orbitrap mass spectrometry to comparatively evaluate the regional influence. In West Sea, the total SSC content of A. japonicus was 15.5 mg/g, representing the highest content, while A. japonicus in the South Sea yielded the lowest content at 8 mg/g. The major saponin constituent in SSC was identified as Holotoxin A1, which may the anti-tumor compound in A. japonicus.

<P>A(3) adenosine receptor (AR) ligands including A(3) AR agonist, N-6-(3-iodobenzyl)adenosine-5'-N-methyluronamide (1a, IB-MEGA) were examined for adiponectin production in human bone marrow mesenchymal stem cells (hBM-MSCs). In this model, 1a significantly increased adiponectin production, which is associated with improved insulin sensitivity. However, A(3) AR antagonists also promoted adiponectin production in hBM-MSCs, indicating that the A(3) AR pathway may not be directly involved in the adiponectin promoting activity. In a target deconvolution study, their adiponectin-promoting activity was significantly correlated to their binding activity to both peroxisome proliferator activated receptor (PPAR) gamma and PPAR delta. They functioned as both PPAR gamma partial agonists and PPAR delta antagonists. In the diabetic mouse model, la and its structural analogues A(3) AR antagonists significantly decreased the serum levels of glucose and triglyceride, supporting their antidiabetic potential. These findings indicate that the polypharmacophore of these compounds may provide therapeutic insight into their multipotent efficacy against various human diseases.</P>

Purpose: Uptakes of Tc-99m MIBI(MIBI) and Tc-99m tetrofosmin (tetrofosmin) in human non-small cell lung cancer A549, multidrug-resistance associated protein (MRP) expressing cell, were investigated in vitro and in vivo. Materials and Methods: Western blot analysis and immunohistochemistry were used for detection of MRP in A549 cells with anti-MRPr1 antibody. Cellular uptakes of two tracers were evaluated at 100 μM of verapamil (Vrp), 50 μM of cyclosporin A (CsA) and 25 μM of butoxysulfoximide(BSO) after incubation with MlBl and tetrofosmin for 30 and 60 min at 37℃, using single cell suspensions at 1×10^(6) cell/ml. Radioactivities in supernatants and pellets were measured with gamma well counter. A549 cells were inoculated in each flanks of 24 nude mice. Group 1(Gr1) and Gr3 mice were treated with only MlBl or tetrofosmin, and Gr2 and Gr4 mice were treated with 70mg/kg of CsA i.p. for 1 hour before injection of 370KBq of MlBl or tetrofosmin. Mice were sacrificed at 10, 60 and 240 min. Radioactivities of organs and tumors were expressed as percentage injected dose per gram of tissue(%ID/gm). Results: Western blot analysis of the A549 cells detected expression of MPRr1 (190 kDa) and immunohistochemical staining of tumor tissue for MPRr1revealed brownish staining in cell membrane but not P-gp. Upon incubating A549 cells for 60 min with MlBl and tetrofosmin, cellular uptake of MlBl was higher than that of tetrofosmin. Coincubation with modulators resulted in an increase in cellular uptakes of MlBl and tetrofosmin. Percentage increase of MlBl was higher than that of tetrofosmin with Vrp by 623% and 427%. CsA by 753% and 629% and BSO by 219%and 140%, respectively. There was no significant difference in tumoral uptakes of MlBl and tetrofosmin between Grl and Gr3. Percentage increases in MlBl (114% at 10 min, 257% at 60 min. 396% at 240 min) and tetrofosmin uptake (110% at 10 min, 205% at 60 min, 410% at 240 min) were progressively higher by the time up to 240 min with CsA. Conclusion: These results indicate that MlBl and tetrofosmin are suitable tracers for imaging MRP-mediated drug resistance in A549 tumors. MlBl may be a better tracer than tetrofosmin for evaluating MRP reversal effect of modulators.

Ginsenoside Rb,, at a concentration of 10 $\mu\textrm{g}$/ml and over, initiated the cycle of oscillation of ion flux in erythrocytes after the cells had been treated with a protonophore, carbonyl cyanide p-trifluoro-methoxyphenyl hydrazone (FCCP) and then with a $Ca^{2+}$ ionophore, A23,3,. Its action was similar to the additional portion of $Ca^{2+}$-ionophore or $Ca^{2+}$ ion to the erythrocytes. Effects of $Rg_1$ and Rf were different from that of Rb,. They did not induce the oscillation. They, however, increased the extracellular $K^{+}$ concentration and pH without returning to the initial state in the erythrocytes processed with FCCP and $A_{23187}$. We established that ginsenosides from 20-(5)-panaxatriol family induced the membrane hyperpolarization in erythrocytes, which was attenuated by the pretreatment of $Rb_1$, a major component of 20-(5)-panaxadiol.

Imatinib mesylate (IM) is used to treat a wide range of diseases, including Philadelphia chromosome-positive chronic myeloid leukemia (CML), on account of its high tolerability and low incidence of minor adverse events. Hemorrhage is thought to be a rare complication of IM. Recently, IM has been associated with reduced a2-plasmin inhibitor and platelet dysfunction. We report here the case of a 33-year-old female patient with CML who experienced subdural hematoma after an incremental increase in IM dosage due to a loss of complete molecular response. This case indicates that physicians should be alert to this atypical cause of headache in patients taking high-dose IM.

Objective To evaluate the clinical, hormonal and metabolic characteristics according to luteinizing hormone (LH)/follicle stimulating hormone (FSH) ratio in women with polycystic ovary syndrome (PCOS). Methods A total of 225 women with PCOS were included in this study. They were divided into two groups according to LH/FSH ratio; group A (LH/FSH < 2, n=160) and group B (LH/FSH > 2, n=65). We compared clinical, hormonal and metabolic characteristics including age at menarche, body mass index (BMI), blood pressures, ovarian volume, and serum androgen levels between the two groups. Serum glucose and insulin levels, fasting glucose/insulin ratio, homeostatic model assessment of insulin resistance (HOMA-IR), and lipid profi les were also compared between the two groups. Pearson correlation coeffi cients were used to evaluate correlation between the LH/FSH ratio and various parameters. Results Age of menarche in the group B was signifi cantly later than that in the group A. BMI, waist-hip ratio and blood pressures were signifi cantly lower in the group B compared to those of the group A. Ovarian volume and serum levels of LH, estradiol, total testosterone, sex-hormone binding globulin, 17-hydroxyprogesterone (OHP), and high density lipoprotein were signifi cantly higher in the group B. Postprandial 2 hours glucose, insulin and HOMA-IR were signifi cantly higher in the group A. After adjustment of BMI, the LH/FSH ratio was signifi cantly positively correlated with age at menarche, ovarian volume, total testosterone levels, and 17-OHP levels. Conclusion The inappropriate gonadotropin secretion may be negatively correlated with BMI, and positively with age of menarche and increased ovarian androgen production irrespective of BMI in women with PCOS. However, it may not be associated with metabolic characteristics.

Purpose of this study : In 2001, this study that investigate actual conditions about adaptation degree of Korean social life of the North Korean defectors was carried out to 151 people that succeed in chases among 200 the North Korea defectors answer in study. Purpose of this study is as following. First, we wished to search change aspect of actual conditions about South Korea society adaptation of the North Korean defectors who take part in the first investigation for past 3 years. Second, we wished to search difference of adaptation aspect by populational and social element. Third, We wished to suggest things that can be beaconed to efficient support policy of government or private organization and successful South Korea social adaptation method which support the North Korean defectors on the basis of study finding about change of adaptation aspect of the North Korea defectors. Methods of Study : We designated 200 Panels to study and survey actual adaptation conditions of the North Korean defectors on South Korea society synthetically and for long-term. So, chasing about 151 people was possible in 2004. We are fertilized and supplement the questionnaire forming in 2001 and composed the new questionnaires to part of a life, mental state, Body and health. This study among them investigated a standard of living of the North Korean defectors. In this process, interviewer directly asked and recorded questions to reduce individual variation of people who receive interview. Study Results : Government's residence support policy about North Korean secession inhabitants was generally attained as effective and suitable for North Korean inhabitants' early South Korea settlement. Only, government need to discuss about North Korea secession inhabitants' residence support more variously for long-term. Second, education should be supported more actively and variously. Suitable point of education for their needs should make a effect of vocational training or education maximize. This is a very important part. Third, they need guidance and education that can receive more easily law support. Fourth, medical supports and health cares should be achieved continuously and systematically to them since they arrived in South Korea. To supports and studies North Korean secession inhabitants has the large significance to gain a foothold about reunification of North and south. Their South Korea society adaptation is becoming topic of conversation among South Korean. An atmosphere of unity is elated. And interest degrees about North Korean are actually rising as exchanges between South and North Korean is activated. Therefore, the adaptation study about North Korean secession inhabitants will have to be expanded in studies for South Korean, not to be limited in studies for North Korean.
학령전기 아동의 글씨 쓰기에 영향을 미치는 요인에 관한 연구

목적 : 학령전기 아동의 글씨 쓰기 명료도 및 속도와 지각-운동 기술의 상관관계를 확인하고, 글씨 쓰기에 가장 영 향을 주는 요인을 알아보고자 하였다. 연구방법 : 덴버 발달 선별 검사를 통하여 정상 발달로 선별된 23명의 학령 전기 아동(평균 연령=68.61개월, SD=2.04)을 대상으로 글씨 쓰기 명료도 및 속도 측정을 위하여 한글 자모 쓰기 검사지를 사용하여 자음과 모음을 쓰도록 하였다. 시지각과 시각-운동 통합 수행 능력 측정을 위하여 시지각 발달 검사를 사용하였고, 소운동 협응 수행 능력 측정을 위하여 Grooved pegboard test를 사용하였다. 결과 : 글씨 쓰기 명료도와 전반적인 시지각, 비운동성 시지각, 시각-운동 통합, 우세손 소동작 협응의 상관관계는 없었으며, 글씨 쓰기의 속도는 전반적 시지각과 우세손 소동작 협응 항목에서 유의한 상관관계를 보였다. 회귀 분 석 결과 글씨 쓰기 속도에 가장 큰 영향을 주는 요인은 전반적 시지각 항목이었다. 결론 : 만 5~6세 학령 전기 아동의 전반적 시지각 능력의 발달을 향상시키기 위한 중재를 제공하는 것이 글씨 쓰기 의 속도 능력을 향상 시킬 수 있다는 것을 알 수 있다. Objective : This study investigated the relationships of handwriting legibility and perceptual-motor skills, and handwriting speed and perceptual-motor skills. And identified the predictors that most affect the handwriting of preschool children. Methods : Twenty-three typically developing preschool aged children (mean age: 68.61 months, SD=2.04) were selected through the Korean-Denver Developmental Screening Test-2(K-DDST-2). The children were tested with regard to handwriting legibility, visual perception, visual-motor integration and fine-motor coordination. Results : First, a significant relationship was not found among handwriting legibility, visual perception, visualmotor integration and fine-motor coordination. Second, a significant relationship was found among handwriting speed, visual perception and fine-motor coordination. Third, stepwise multiple regression analyses showed that general visual perception were significant predictors for handwriting speed. Conclusion : Occupational therapists should evaluate children's visual perception levels utilizing a standardized test, and focus on general visual perception in order to improve handwriting skill(speed). Also, occupational therapists are expected to play an important role in the management and treatment of children's handwriting skills.
Systematic characterization of A-to-I RNA editing hotspots in microRNAs across human cancers

<P>RNA editing, a widespread post-transcriptional mechanism, has emerged as a new player in cancer biology. Recent studies have reported key roles for individual miRNA editing events, but a comprehensive picture of miRNA editing in human cancers remains largely unexplored. Here, we systematically characterized the miRNA editing profiles of 8595 samples across 20 cancer types from miRNA sequencing data of The Cancer Genome Atlas and identified 19 adenosine-to-inosine (A-to-I) RNA editing hotspots. We independently validated 15 of them by perturbation experiments in several cancer cell lines. These miRNA editing events show extensive correlations with key clinical variables (e.g., tumor subtype, disease stage, and patient survival time) and other molecular drivers. Focusing on the RNA editing hotspot in miR-200b, a key tumor metastasis suppressor, we found that the miR-200b editing level correlates with patient prognosis opposite to the pattern observed for the wild-type miR-200b expression. We further experimentally showed that, in contrast to wild-type miRNA, the edited miR-200b can promote cell invasion and migration through its impaired ability to inhibit <I>ZEB1/ZEB2</I> and acquired concomitant ability to repress new targets, including <I>LIFR</I>, a well-characterized metastasis suppressor. Our study highlights the importance of miRNA editing in gene regulation and suggests its potential as a biomarker for cancer prognosis and therapy.</P>
