        A new compound, 1H,8H-pyrano[3,4-c]pyran-1,8-dione, suppresses airway epithelial cell inflammatory responses in a murine model of asthma.

        <P>Clinical and experimental studies have established eosinophilia as a sign of allergic disorders. Activation of eosinophils in the airways is believed to cause epithelial tissue injury, contraction of airway smooth muscle and increased bronchial responsiveness. As part of the search for new antiasthmatic agents produced by medicinal plants, the effects of 270 standardized medicinal plant extracts on cytokine-activated A549 human lung epithelial cells were evaluated. After several rounds of activity-guided screening, the new natural compound, 1H,8H-Pyrano[3,4-c]pyran-1,8-dione (PPY), was isolated from Vitex rotundifolia L. To elucidate the mechanism by which the anti-asthmatic responses of PPY occurred in vitro, lung epithelial cells (A549 cell) were stimulated with TNF-alpha, IL-4 and IL-1beta to induce the expression of chemokines and adhesion molecules involved in eosinophil chemotaxis. PPY treatments reduced the expression of eotaxin, IL-8, IL-16 and VCAM-1 mRNA significantly. Additionally, PPY reduced eotaxin secretion in a dose-dependent manner and significantly inhibited eosinophil migration toward A549 medium. In addition, PPY treatment suppressed the phosphorylation of p65 and ERK1/2, suggesting that it can inhibit the MAPK/NF-KB pathway. To clarify the anti-inflammatory and antiasthmatic effects of PPY in vivo, we examined the influence of PPY on the development of pulmonary eosinophilic inflammation in a murine model of asthma. To accomplish this, mice were sensitized and challenged with ovalbumin (OVA) and then examined for the following typical asthmatic reactions: an increase in the number of eosinophils in BALF; the presence of Th2 cytokines such as IL-4 and IL-5 in the BALF; the presence of allergen-specific IgE in the serum; and a marked influx of inflammatory cells into the lung. Taken together, our results revealed that PPY exerts profound inhibitory effects on the accumulation of eosinophils into the airways while reducing the levels of IL-4, IL-5, and IL-13 in the BALF. Therefore, these results suggest that PPY may be useful as a new therapeutic drug for the treatment of allergic asthma.</P>

      • Purification and functional characterization of the first stilbene glucoside-specific β-glucosidase isolated from Lactobacillus kimchi

        This study aimed to develop viable enzymes for bioconversion of resveratrol-glucoside into resveratrol. Out of 13 bacterial strains tested, Lactobacillus kimchi JB301 could completely convert polydatin into resveratrol. The purified enzyme had an optimum temperature of 30-40<SUP>o</SUP>C and optimum pH of pH 5.0 against polydatin. This enzyme showed high substrate specificities towards different substrates in the following order: isorhaponticin>>polydatin>>mulberroside A>oxyresveratrol-3-O-glucoside. Additionally, it rarely hydrolyzed astringin and desoxyrhaponticin. Based on these catalytic specificities, we suggest this enzyme be named stilbene glucoside-specific β-glucosidase. Furthermore, polydatin extracts from Polygonum cuspidatum were successfully converted to resveratrol with a high yield (of over 99%). Stilbene glucoside-specific β-glucosidase is the first enzyme isolated from lactic acid bacteria capable of bio-converting various stilbene glucosides into stilbene.

        Prevalence of Anaplasma and Bartonella spp. in Ticks Collected from Korean Water Deer(Hydropotes inermis argyropus)

        Deer serve as reservoirs of tick-borne pathogens that impact on medical and veterinary health worldwide. In the Republic of Korea, the population of Korean water deer (KWD, Hydropotes inermis argyropus) has greatly increased from 1982 to 2011, in part, as a result of reforestation programs established following the Korean War when much of the land was barren of trees. Eighty seven Haemaphysalis flava, 228 Haemaphysalis longicornis, 8 Ixodes nipponensis, and 40 Ixodes persulcatus (21 larvae, 114 nymphs, and 228 adults) were collected from 27 out of 70 KWD. A total of 89/363 ticks (266 pools, 24.5% minimum infection rate) and 5 (1.4%) fed ticks were positive for Anaplasma phagocytophilum using nested PCR targeting the 16S rRNA and groEL genes, respectively. The 16S rRNA gene fragment sequences of 88/89 (98.9%) of positive samples for A. phagocytophilum corresponded to previously described gene sequences from KWD spleen tissues. The 16S rRNA gene fragment sequences of 20/363 (5.5%) of the ticks were positive for A. bovis and were identical to previously reported sequences. Using the ITS specific nested PCR, 11/363 (3.0%) of the ticks were positive for Bartonella spp. This is the first report of Anaplasma and Bartonella spp. detected in ticks collected from KWD, suggesting that ticks are vectors of Anaplasma and Bartonella spp. between reservoir hosts in natural surroundings.

      • Matrix Metalloproteinase-8 Plays a Pivotal Role in Neuroinflammation by Modulating TNF-α Activation

        <P>Matrix metalloproteinases (MMPs) play important roles in normal brain development and synaptic plasticity, although aberrant expression of MMPs leads to brain damage, including blood–brain barrier disruption, inflammation, demyelination, and neuronal cell death. In this article, we report that MMP-8 is upregulated in LPS-stimulated BV2 microglial cells and primary cultured microglia, and treatment of MMP-8 inhibitor (M8I) or MMP-8 short hairpin RNA suppresses proinflammatory molecules, particularly TNF-α secretion. Subsequent experiments showed that MMP-8 exhibits TNF-α–converting enzyme (TACE) activity by cleaving the prodomain of TNF-α (A<SUP>74</SUP>/Q<SUP>75</SUP>, A<SUP>76</SUP>/V<SUP>77</SUP> residues) and, furthermore, that M8I inhibits TACE activity more efficiently than TAPI-0, a general TACE inhibitor. Biochemical analysis of the underlying anti-inflammatory mechanisms of M8I revealed that it inhibits MAPK phosphorylation, NF-κB/AP-1 activity, and reactive oxygen species production. Further support for the proinflammatory role of microglial MMP-8 was obtained from an in vivo animal model of neuroinflammatory disorder. MMP-8 is upregulated in septic conditions, particularly in microglia. Administration of M8I or MMP-8 short hairpin RNA significantly inhibits microglial activation and expression/secretion of TNF-α in brain tissue, serum, and cerebrospinal fluid of LPS-induced septic mice. These results demonstrate that MMP-8 critically mediates microglial activation by modulating TNF-α activity, which may explain neuroinflammation in septic mouse brain.</P>

        정신보건심판위원회 퇴원명령 대상자 추적조사

        Objectives The purpose of this study was to examine the current situation and the limitations of the Mental Health Review Board and to identify the effectiveness of the community linkage for discharged patients. Methods The data was collected from the 116 patients who were discharged from August 2005 to July 2007 by the order of the Mental Health Review Board. The survey was conducted by telephone with using a structured questionnaire. Results 51 patients (44.0%) were able to be referred to community mental health services when they discharged. The readmission rate was 50.9% and the mean duration to the first readmission was 51.73 days (S.D.=103.5). The mean duration to the first readmission was significantly longer for the group that was referred to community services and that had a negative correlation with the stability of health security. The outpatient maintenance rate and the current admission status were also significantly different between the group that was referred to community services and the group that was not referred. Conclusion The activities of the mental health review board have definite limitations in Korea such as the high rate of immediate readmissions after the discharge orders. A hospital-community integrated service system could be one possible solution to improve the maintenance of mental health patients in the community and to prevent their readmission.

        Accurate acquisition of surface geometries such as machined surfaces, biological surfaces, and deformed parts have been very important technique in scientific study an engineering, expecially for system design, manufacturing and inspection. Two camera method keeps accuracy more than double than mechanical method. In this paper, we describe the processes surface data of teeth for automatic processing, transformation that changes to coping data of possible numeric data form, and processed inlay teeth. As the result of the inlay manufacture, which can process with high resolution, therefore we construct the automatic processing system that depends on manual.

      • 국내산 과일 및 채소 추출물을 이용한 체중 조절 관련 기능성 식품소재 탐색

        Obesity can be defined as a disorder of lipid metabolism and the control of this process is regulated by enzymes involved in lipid metabolic pathway. Obesity is also characterized increases in the number and size of adipocyte differentiated from fibroblastic pre-adipocytes in adipose tissue. The aim of this study was to screen ethanol extracts of 21 fruits and vegetables cultivated in Republic of Korea for identifying natural sources of lipase inhibitor and to investigate the effects of several extracts resulting in suppression of triglyceride (TG) accumulation in 3T3-L1 cells. Among the tested extracts, Diospyros kaki L. showed the highest contents of total sugar and the content of total phenolic compounds was detected 17 species while there was not detected in Ixeris dentate root and Allium fistulosum C. etc. We were screened 26 extracts of fruits and vegetables for pancreatic lipase inhibitory activity compared with orlistat. The strongest lipase inhibitory activities were shown by extract of Allium sativum L. stem, Allium sativum L. bulb, Apteryx australis, Diospyros kaki L. Malus Pumila, Perilla frutescens and Prunus mume regardless of concentrations treated. Eight extracts including Brassica juncea var. integrilolia had no effect on lipase inhibition. During the differentiation of 3T3-L1 preadipocyte to mature adipocyte, the cells were treated with carrier or 7 extracts(1, 5 and 10 ㎍/ml), which was shown to highest lipase inhibitory effects. We were quantified lipid contents secreted mature adipocytes and stained with Oil Red O. All cells treated with the Apteryx australis and Prunus mume significantly decreased TG contents than the control. These findings suggested that the screened fruit and vegetable could be a promising source as the effective and safe lipase inhibitors for isolating bioactive ingredients and developing functional foods.

        해마리아 난 종자의 기내배양에 의한 대량증식 체계를 확립하기 위해 적합한 수분 시기를 구명하고, 식물생장조절물질 및 종자 저온전처리가 기내발아 및 근경에 미치는 효과를 조사하였다. 종자를 획득하기 위해 다양한 시기 즉개화 당일에서 1일 후, 2일에서 5일 후, 6일에서 10일 후 및 11일에서 15일 후에 자가수분 시킨 후 35일이 경과되었을 때 자연 상태에서 착과율을 조사한 결과 11일에서 15일(A4)후에 수분 시켰을 때 71.4%로 가장 높았다. 종자의 성숙정도별 기내발아에 미치는 효과를 조사하기 위해 개화 당일에서 15일까지 시기별로 자가수분을 실시하여 35일이 경과한 후 꼬투리 내 형성된 미성숙 및 성숙 종자를 1.0 mg·L<sup>-1</sup> NAA, 1.0 mg·L<sup>-1</sup> TDZ가 혼용 첨가된 1/2 MS 배지에 배양한 결과 기내 배양에서의 발아율은 개화 10일 후에 수분시킨 종자에서 100일 및 200일이 경과되었을 때 25.2% 및 36.1%의 발아율로 가장 효과적이었다. 기내 발아율 및 근경 형성에 미치는 식물생장조절물질은 1/2 MS 기본배지에 1.0 mg·L<sup>-1</sup> NAA와 1.0 mg·L<sup>-1</sup> TDZ 혼용첨가한 배지에서 발아율이 48.9%로 효과적이었으며, BA, 2ip 및 kinetin을 각각 1.0 mg·L<sup>-1</sup> 씩 혼용 처리한 결과 NAA와 BA 혼용처리에서는 근경이 66.3개로 형성되었다. 종자의 저온전처리가 기내발아 및 근경 발생에 미치는 효과를 조사한 결과 착과된 종자꼬투리를 5℃의 냉장고에서 15일간 처리한 결과 배양 60일 후에 발아수가 271.9개, rhizome은 85.7개가 발생되어 가장 효과적이었다. To establish a mass production system of Haemaria discolor Lindl., we investigated the several factors including optimum time of pollination to induce the seed formation and ratio of fruit set and effect of seed maturity, low temperature treatment, and plant growth regulators on in vitro germination. To obtain seeds, self-pollination was done in various times after flowering, i.e. 0-1 days, 2-5 days, 6-10 days, and 11-15 days after flowering (DAF). The half strength of MS medium was used as the basic, and PGRs included NAA in combination with TDZ, BA, 2ip, and kinetin. The results showed that the highest fruit set (71.4%) occurred when pollinated 11-15 DAF. The in vitro seed germination was highest (25.2%) when the immature seeds, collected 11 to 15 days after pollination done 6-10 DAF, were cultured on the MS medium containing 1.0 mg·L<sup>-1</sup> NAA and 1.0 mg·L<sup>-1</sup> TDZ. The rhizome production from the seed culture was highest in the mixed treatment of 1.0 mg·L<sup>-1</sup> NAA and 1.0 mg·L<sup>-1</sup> BA. The effect of low temperature treatment of seed on germination and rhizome production was most effective when seed capsules were stored at 5˚C for 15 days.
