Antitumor Constituents of Polyporus giganteus

The polysaccharides obtained from the shake-cultured mycelia of eight species of edible basidiomycetes of Korea were subjected to antitumor test against sarcoma 180 in vivo. The polysaccharides from pluteus cervinus, Pleurotus pulmonarius, Hydnum repandum, and Laccaria laccata showed antitumor activities. Of these four species of basidiomycetes, ata P. cervinus was selected fnrther study. Four fractions were obtained from mycelia and three fractions from the culture filtrate of the mushroom. Among them, the water-soluble polysaccharide fraction of the hot water extract of the mycelia showed ehe highest activity. The inhibition ratios of this fraction against sarcoma 180 were 73.9, 86.3 and 87.8% at the three dose levels of 10 ㎎, 20 ㎎ and 50 ㎎/㎏/day. This water-soluble polysaccharide was subjected to Sephdex G-200 gel filtration and separated into two fractions. The higher molecular weight polysaccharide which showed a sharp anthrone positive peak on Sephadex G-200 was found to be the active fraction and tentatively named as cerviran A. When this was chemically analysed, it was found to consist of a polysaccharide and a protein. The polysaccharide moiety of cerviran A was mostly composed of glucose (92.4%) and small quantities of galactose (3.6%) and mannose (4.0%) and traces of fucose and xylose.
