Because of a sudden increase of vehicles, the limit of road capacity occurred. We are, therefore, constructing the road tunnel over l㎞ for the maximization of land use. To supply safe driving conditions in the road tunnel, we are controlling the exhaust emissions with a forced ventilation facility. The purpose of this study is to investigate the ventilation efficiency and the characteristics of tunnel on the change of natural ventilation force. To carry out this study, Sola program is transformed to fit for the road tunnel and it was simulated. The tunnel with ventilation system of transverse type is used as a objective tunnel. The results of this study are as follows; 1. The larger a natual ventilation force, the better a ventilation efficiency. 2. It is required the ventilation system dealing with quickly a change of the tunnel surroundings because of accumulation of 5m/sec. 3. The contaminant distributions in the near of ventilation system are highly stagnant in the left-below parts of tunnel, the below parts of air supply system and the below parts of ventilation system.

Recently, several new methods for the detection of genetic damages in vitro and in vivo based on molecular biological techniques were introduced according to the rapid progress in toxicology combined with cellular and molecular biology. Among these methods, mouse lymphoma thymidine kanase (tk) gene forward mutation assay, single cell gel electrophoresis (comet assay) and transgenic animal and cell line model as a target gene of lac I (Big Blue) and lac Z (Muta Mouse) gene mutation are newly introduced based on molecular toxicological approaches. The mouse lymphoma tk^(+/-) gene assay (MOLY) using L5178Y tk+i- mouse lymphoma cell line is one of the mammalian forward mutation assays, and has many advantages and more sensitive than hprt assay. The target gene of MOLY is a heterozygous tk^(+/-) gene located in 11 chromosome, so it is able to detect the wide range of genetic changes like point mutation, deletion, rearrangement, and mitotic recombination within tk gene or deletion of entire chromosome 11. The comet assay is a rapid, simple, visual and sensitive technique for measuring and analysing DNA breakages in mammalian cells. Also, transgenic animal and cell line models, which have exogenous DNA incorporated into their genome, carry recoverable shuttle vector containing reporter genes to assess endogenous effects or alteration in specific genes related to disease process, are powerful tools to study the mechanism of mutation in vivo and in vitro, respectively. Also in vivo acridine orange supravital staining micronucleus assay by using mouse peripheral reticulocytes was introduced as an alternative of bone marrow micronucleus assay. In this respect, there was an International workshop on genotoxicity procedure (IWGTP) supported by OECD and EMS (Environmental Mutagen Society) at Washington D, C, in March 25-26, 1999. The objective of IWGTP is to harmonize the testing procedures internationally, and to extend to finalization of OECD guideline, and to the agreement of new guidelines under the International Conference of Harmonization (ICH) for these methods mentioned above. Therefore, we introduce and review the principle, detailed procedure, and application of MOLY, comet assay, transgenic mutagenesis assay and supravital staining micronucleus assay.
소나무 잎과 가지의 정유와 그 성분들의 항산화 및 항노화 작용

To investigate effects of pine essential oil isolated by steam distillation from Pinus densiflora needles (PN) and twigs (PT) on anti-oxidant and anti-aging activities, the experiments were carried out to determine anti-oxidant and antiaging activities on DPPH radical scavenging activity, NBT/xanthine oxidase-superoxide scavenging activity, silica-induced intracellular H2O2 and hydroxyl radical generation in RAW264.7 cells, hyaluronidase and elastase activities in vitro. Essential oil of PN and PT were contained 0.225 and 0.176 (w/v) %, respectively. PN was contained with 30 kinds of essential oil and its major constituent is α-pinene (21.5%). Further PT was contained with 40 kinds of essential oil and its major constituent is β-pinene (22.4%) in GC/MS assay. Other essential oils of PN were β-pinene, camphene, myrcene, limonene, terpinolene, α-terpineol, bornyl acetate, α-copaene, caryophellene and humulene and PT were α-pinene, camphene, phellandrene, limonene, terpinolene, α-terpineol, bornyl acetate, α-copaene, caryophellene and humulene. The essential oil of PT have more active than that of PN in anti-oxidant activity which has significant DPPH radical and superoxide scavenging activity, and significant inhibitory activities on silica-induced intracellular H2O2 and hydroxyl radical generation, as well as, significantly inhibited elastase and hyaluronidase activities. Further, phellandrene, myrcene and α-pinene have DPPH radical and superoxide scavenging activities, β-pinene, terpinolene, myrcene and phellandrene inhibited silica-induced intracellular H2O2 and hydroxyl radical generations. And also phellandrene and β-pinene inhibited hyaluronidase and elastase activities. In conclusion, the essential oils isolated from PN and PT have anti-oxidant and anti-aging activities.

Background: EUS is an accurate diagnostic tool to detect common bile duct (CBD) stones in the patients with acute pancreatitis. This study was to evaluate the necessity for performing endoscopic retrograde cholangiopancreatography (ERCP) in the patients with suspicious gallstone pancreatitis associated with microlithiasis or none in the CBD by endoscopic ultrasound (EUS). Methods: Consecutive fi fty-three patients (M:F, 30:23; mean age, 57 18 years) with suspicious gallstone pancreatitis and no evidence of CBD stones by MDCT underwent EUS within 24 hours of admission between 2012 and 2014. Immediate ERCP (within 24 hours after EUS) was performed in the patients with CBD stones, microlithiasis, or CBD dilation by EUS fi ndings. The patients` medical records were retrospectively analyzed based on clinical symptoms, biochemical data, and hospital courses.Results: CBD stones, microlithiasis, and none were identifi ed in 21(39.6%), 7(13.2%), and 25(47.2%) patients by EUS. ERCP was performed in 21 patients with CBD stones, 7 patients with microlithiasis, and 8 patients with only CBD dilatation without stones. CBD stones were extracted by ERCP in 15 out of 21 (71.4%) patients, 1 out of 7 (14.3%) patient, and 1 out of 7 (12.5%) patient, respectively. All patients were improved and hospital days were not different with or without ERCP. However, 4 patients who performed ERCP in the condition of microlithiasis or CBD dilation only showed transient aggravation of acute pancreatitis after ERCP. Conclusions: Half of patients with gallstone pancreatitis showed no evidence of CBD stones and they improved with only conservative management. Immediate ERCP in patients with microlithiasis or CBD dilation only is not necessary in the EUS era.
