        Dislocation Density-Based Constitutive Model and Processing Map for T2 Copper During Isothermal and Time-Variant Deformation

        Isothermal compression tests were carried out to study the hot deformation behavior of T2 copper under various conditions. The deformation parameters, such as temperature and strain rate, have strong influence on flow stress and microstructureevolution of the alloy. A unified dislocation density-based model considering dynamic recovery and recrystallization wasestablished. And material parameters of the developed model were optimized by genetic algorithm. Comparisons betweenthe experimental and model data demonstrates that the developed model can precisely describe the flow behavior at quitewide range of deformation conditions. Meanwhile, the designed iterative procedure allows the model to be applied in timevariantdeformation conditions. Processing map and microstructure examination were constructed to optimize the processingwindow of the studied alloy. According to the processing maps, flow instability mainly appeared at low temperatures of500–650 °C and strain rates higher than 0.1 s−1. The optimum deformation parameters of T2 copper was concluded as thetemperature range of 700–800 °C and the strain rate of 0.1–1 s−1.

        Characterization, Cloning, and Heterologous Expression of a Subtilisin-Like Serine Protease Gene VlPr1 from Verticillium lecanii

        The entomopathogenic fungus Verticillium lecanii is a wellknown biocontrol agent. V. lecanii produces subtilisin-like serine protease (Pr1), which is important in the biological control activity of some insect pests by degrading insect cuticles. In this study, a subtilisin-like serine protease gene VlPr1 was cloned from the fungus and the VlPr1 protein was expressed in Escherichia coli. The VlPr1 gene contains an open reading frame (ORF) interrupted by three short introns, and encodes a protein of 379 amino acids. Protein sequence analysis revealed high homology with subtilisin serine proteases. The molecular mass of the protease was 38 kDa, and the serine protease exhibited its maximal activity at 40°C and pH 9.0. Protease activity was also affected by Mg2+ and Ca2+ concentration. The protease showed inhibitory activity against several plant pathogens, especially towards Fusarium moniliforme.

        Suppression of MED19 expression by shRNA induces inhibition of cell proliferation and tumorigenesis in human prostate cancer cells

        MED19 is a member of the Mediator that plays a key role in the activation and repression of signal transduction or the regulation of transcription in carcinomas. To tested the functional role of MED19 in human prostate cancer, we downregulated MED19 expression in prostate cancer cells (PC-3 and DU145) by lentivirus- mediated short hairpin (shRNA), and analyzed the effect of inhibition of MED19 on prostate cancer cell proliferation and tumorigenesis. The in vitro prostate cancer cell proliferation, colony formation, and in vivo tumor growth in nude mice xenografts was significantly reduced after the downregulation of MED19. Knock- down of MED19 caused S-phase arrest and induced apoptosis via modulation of Bid and Caspase 7. It was suggested that MED19 serves as a novel proliferation regulator that promotes growth of prostate cancer cells. [BMB reports 2011; 44(8): 547-552]

        토끼에서 경부 교감신경절의 무수 에틸 알코올에 의한 화학적 차단

        N/A Blockade of cervicothoracic sympathetic ganglion (stellate ganglion controls pain on face, head, neck, shoulder, upper limbs, and upper chest, including their viscera and sympathetically maintained pain. This procedure also increases blood flow to the above areas and relieves hyperreactivity of sympathetic nervous system. Clinically, repeated stellate ganglion blocks with local anesthetic agent may become difficult with complications such as accidental intravascular or subdural injection, recurrent laryngeal nerve or bracheal plexus paralysis, pneumothorax and edema on injection site. Therefore, at times long-term cervicothoracic ganglion block with neurolytics is necessitated but its applica- tions are prohibited by the critical structures surrounding ganglion. There are also few reports of neurolytic stellate ganglion block. This study was performed to observe the complications, gross changes of surrounding struc- tures, and microscopic findings of ganglion cells after neurolytic block and to certify the possi- bility of clinical use of neruolytic stellate ganglion block. The unilateral superior cervical sym- pathetic ganglion of rabbit was blocked with absolute ethyl alcohol 0.4 ml at the level of cricoid cartilage. Normal ganglion was used as a control and 5 animals were sacrificed at each inter- vals of 7, 15 and 50 days after block. The results were as follows; 1) All experimental animals showed no specific changes of behavior, motor function. No ne- crotic tissues were present in the block area during the observation period. There were some gross scar tissues along the fascia of muscles surrounding the needle injection site, but gross atrophy of muscles or injured major vessels were not found. 2) Microscopically, structures of normal ganglion of rabbit were very similar to those of hu- mans. Seven days after absolute ethyl achohol injection there were marked edema of ganglion cells and nuclei with irregular nuclear membrane. Some of the ganglion cells lost their nuclei and showed degenerative changes. Fifteen days after block, cell edema were decreased and loss of the Nissl's body was prominant. The ganglion cell structures looked close to normal but the cytoplasm and nucleus were generally contracted 50 days after block. These results suggest absolute ethyl alcohol injection on cervical sympathetic ganglion with above method mainly blocks pre- and post-synaptic fibers and the long-term neurolytic block- ade of this ganglion may be possible in rabbits.

        Environmental Microbiology/Microbial Diversity : Isolation and Characterization of Bacteriophages Against Pseudomonas syringae pv. actinidiae Causing Bacterial Canker Disease in Kiwifruit

        Pseudomonas syringae pv. actinidiae causes bacterial canker disease in kiwifruit. Owing to the prohibition of agricultural antibiotic use in major kiwifruit-cultivating countries, alternative methods need to be developed to manage this disease. Bacteriophages are viruses that specifically infect target bacteria and have recently been reconsidered as potential biological control agents for bacterial pathogens owing to their specificity in terms of host range. In this study, we isolated bacteriophages against P. syringae pv. actinidiae from soils collected from kiwifruit orchards in Korea and selected seven bacteriophages for further characterization based on restriction enzyme digestion patterns of genomic DNA. Among the studied bacteriophages, two belong to the Myoviridae family and three belong to the Podoviridae family, based on morphology observed by transmission electron microscopy. The host range of the selected bacteriophages was confirmed using 18 strains of P. syringae pv. actinidiae, including the Psa2 and Psa3 groups, and some were also effective against other P. syringae pathovars. Lytic activity of the selected bacteriophages was sustained in vitro until 80 h, and their activity remained stable up to 50oC, at pH 11, and under UV-B light. These results indicate that the isolated bacteriophages are specific to P. syringae species and are resistant to various environmental factors, implying their potential use in control of bacterial canker disease in kiwifruits.

        위암화과정에서 Helicobacter pylori 감염이 위점막 세포 역동학과 아포토시스 관련 단백질 발현에 미치는 영향

        Background/Aims: We investigated apoptosis, cell proliferation and the expression of apoptosis-related proteins in the gastric epithelial cells of Helicobacter pylori (H. pylori)-negative and positive patients. Methods: TUNEL staining, immunohistochemical staining for Ki-67, p53, Bcl-2, Bcl-XL and Bax were performed on paraffin-embedded specimens obtained from 11 H. pylori-negative controls, 20 H. pylori-positive chronic gastritis, 10 chronic gastritis with intestinal metaplasia (IM), 11 adenoma, and 7 adenocarcinoma patients. Apoptosis and proliferation were expressed as apoptosis index (AI) and proliferation index (PI). Results: In H. pylori-positive groups, apoptosis and proliferation were increased compared with controls. However, an AI/PI ratio was highest in chronic gastritis cases and then gradually reduced according to the progress from IM to cancer. p53 was expressed in 28.6% of adenocarcinoma cases. Expression of Bax was significantly increased in chronic gastritis and IM patients compared with controls. Conclusions: H. pylori infection induces marked increase in apoptosis and proliferation of gastric mucosa. In the premalignant lesions, the balance between apoptosis and proliferation is altered, and this imbalance may play a key role in the gastric carcinogenesis. (Korean J Gastroenterol 2003;42:12-19)

        반월상 사구체신염 (Crescentic Glomerulonephritis)의 병리적 분석

        배 경 : 반월체는 보우만 강을 전체적으로 혹은 부분적으로 채우는 두 층 이상의 세포로 정의 되며, 이러한 반월체가 보이면서 심한 염증성 사구체 손상이 주로 나타나며 급속히 진행되는 신장 질환을 반월상 사구체신염이라고 한다. 이 질환은 병리학적 분류에 따라 치료 및 임상적 진행이 다르다. 한 임상 기관에서의 반월상 사구체신염의 병리 경험을 모아서 분석하였다. 방 법 : 저자들은 1987년 1월부터 2002년 2월까지 한양대학교병원 병리과에서 신생검으로 진단된 반월상 사구체신염 25예를 항사구체기저막 질환 (Ⅰ군), 면역복합체 사구체신염 (Ⅱ군), ANCA와 연관된 사구체신염 (Ⅲ군) 그리고 특발성 반월상 사구체신염 (Ⅳ군) 4개의 군으로 나누고, 질환에 따라 다시 세부 분류하여 진단시 임상 및 생검 소견의 조직학적 차이점을 조사하였다. 결 과 : 총 25예 중 항사구체기저막 질환으로 분류되는 예는 없었고 면역복합체 사구체신염군으로 분류된 8예 증 감염 후 사구체신염이 3예, IgA nephropathy가 3예, Henoch-Scho¨nlein purpura가 2예가 있었으며, 그리고 ANCA와 연관된 사구체신염으로 분류된 12예 중 현미경적 다발성 혈관염이 7예, Wegener 육아종증이 4예, ANCA 관련 사구체신염 1예가 있었으며, 특발성 반월상 사구체신염이 5예가 있었다. 총 25예 중 남녀의 비율은 비슷하였고 평균 연령은 Ⅱ군이 32.0세, Ⅲ군이 59.3세, Ⅳ군은 39.0세였으며 생검 당시의 혈청 크레아티닌의 평균값은 각각 9.1, 5.2, 8.8 ㎎/dL이었다. 광학 현미경 소견상 각 질환군에서 사구체 반월체의 평균 빈도 (%)는 Ⅱ군 64.4%, Ⅲ군 43.7%, Ⅳ군 51.2%였으며 신세뇨관의 염증 세포 평균 침윤 정도 (score)는 Ⅱ군 0.8점, Ⅲ군 0.4점, Ⅳ는 0.6점이었고 신간질의 평균 섬유화 정도 (score)는 Ⅱ군 1.0점, Ⅲ군 0.8점, Ⅳ군 1.2점이었으며 동맥 및 세동맥의 평균 경화증 정도 (score)는 Ⅱ군 1.4점, Ⅲ군 0.9점, Ⅳ군 1.6점이었다. 면역형광 현미경 소견상 주로 Ⅱ군에서 A 및 C3에 대한 침착을 관찰할 수 있었다. 이상의 결과에서 광학 현미경 소견상 Ⅱ군에서는 반월체 빈도가 높게 관찰되었고 Ⅲ군에서는 섬유화 정도가 적게 관찰되었으며 동맥 경화증의 경우 Ⅲ군 에서 적은 빈도로 관찰되었다. 결 론 : 반월상 사구체신염의 임상 및 병리학적 소견을 재검토함으로써 유사한 증례의 조기 진단에 도움이 되리라 사료되며 앞으로 좀 더 많은 증례를 대상으로 분석하면 각 질환군의 병리적 특징과 예후 인자를 밝히는데 도움이 될 것으로 생각한다. Background: Crescentic glomerulonephritis is expressed pathologically by cresecent formation in Bowman`s capsule and clinically by rapidly progressive loss of renal function. The pathologic experience of crescentic glomerulonephritis in one institution was analyzed here. Methods: We classified 25 cases of crescentic glomerulonephritis patients into 4 categories and reviewed the cases pathologically and clinically. Results: We found no case with group 1 (anti-GBM disease), 8 cases in group Ⅱ (immune complex glomerulonephritis) including 3 patients with IgA nephropathy, 2 patients with Henoch-Scho¨nlein purpose and 3 patients with APSGN, 12 cases in group Ⅲ (ANCA-associated glomerulonephritis) including 7 patients with microscopic polyangitis, 4 patients with Wegener`s granulmatosis and 1 patient with ANCAGN, and 5 cases in group Ⅳ (idiopathic crescentic glomerulonephritis). The mean ages of patients with proup Ⅱ, Ⅲ, and Ⅳ were 32.0 59.3 years old respectively, and mean serum creatinine levels at the time of biopsy were measured as 9.1, 5.2, 8.8mg/dL in each group. On light microscopic findings, the frequency of crescents in glomeruli was 64.4% in group Ⅱ, 43.7% in group Ⅲ, and 51.2% in group Ⅳ. The score of infiltration into tubules of inflammatory cells was 0.8 in group Ⅱ, 0.4 in group Ⅲ, and 0.6 in group Ⅳ and the score of fibrosis in interstitium was 1.0 in group Ⅱ, 0.8 in group Ⅲ and 1.2 in group Ⅳ. The score of aherosclerosis in arteries was 1.4, 0.9 and 1.6 in each group. Conclusion: We conclude that the precise diagnosis and classification of crescentic glornerulone phritis by an early renal biopsy and clinical assessments are important in the management of rapidly progressive(crescentic) glomerulonephritis. Since the number of the cases was not so enough, we could not analyze the statistical significance between morphologic differences of each group of crescentic glomerulonephritis, but if more case were collected, acknowledgements of differences and prognostic factors in pathologic findings could be possible.
