        New Method for Simultaneous Quantifi cation of 12 Ginsenosides in Red Ginseng Powder and Extract

        For quality control of components in Korean red ginseng powder and extract, a new method for simultaneous quantifi cation of 12 ginsenosides (Rg₁, Re, Rf, Rh₁, Rg₂[S], Rg₂[R], Rb₁, Rc, Rb₂, Rd, Rg₃[S], and Rg₃[R]) was studied. Compared to the offi cial method for quantifi cation of marker substances (ginsenosides Rg1 and Rb₁), the proposed methods were guaranteed by in-house method validation. Several criteria such as linearity, specifi city, precision and accuracy were evaluated. For red ginseng powder, recovery (averaging 95% to 105%) was calculated, and analysis of variance was carried out to estimate the relative standard deviation (0.20% to 2.12%). For red ginseng extract, the average recovery rate was 90% to 99% and the relative standard deviation was 0.39% to 2.40%. These results indicate that the proposed method could be used in the laboratory for determination of 12 ginsenosides in red ginseng powder and extract. In addition, this method was found to be suitable for quality control of ginseng products and potentially offer time and cost benefi ts.

        링레이저 자이로 기반 2축 회전형 관성항법장치 오차해석에 대한 연구

        관성항법장치 순수항법 성능을 개선하는 방법으로 관성센서 오차가 상호 상쇄되도록 관성센서뭉치를 회전시켜 항법 성능을 개선하는 방법이 있으며 이러한 원리로 동작하는 항법장치를 회전형 관성항법장치라 한다. 관성센서 오차에 의한 회전형 관성항법장치의 정확한 항법 성능 분석을 위해서는 이에 대한 이론적 오차해석이 요구되나 기존의 많은 연구에서는 지구회전 각속도 및 중력 가속도에 의한 영향을 무시하고 오차해석을 수행하여 회전형 관성항법장치의 정확한 항법 성능 분석이 수행되지 않았다. 본 논문에서는 링레이저 자이로 기반 회전형 관성항법장치의 정확한 항법 성능 분석을 위하여 지구회전 각속도 및 중력 가속도 항을 포함한 이론적인 오차해석을 수행하고 이를 기반으로 회전형 관성항법장치의 항법 성능 분석 결과를 제시하였다. There is a method to enhance the pure navigation performance of INS(Inertial Navigation System) through the rotation of inertial measurement unit to compensate error sources of inertial sensors each other and that INS using this principle of operation is called rotational INS. In this paper, the exact error analysis of rotational INS based on ring laser gyro considering the coupling effect with gravity and earth rate is performed to evaluate the navigation performance by inertial sensor error sources. And error analysis and performance evaluation result confirmed by modelling and simulation is also proposed in this paper.

        Apoptotic Effect of Co-treatment with Chios Gum Mastic and HS-1200 on G361 Human Melanoma Cell Line

        Chios gum mastic (CGM) is a resinous exudate obtained from the stem and the main leaves of Pistacia lenticulus tree native to Mediterranean areas. Recently it reported that CGM induce apoptosis in a few cancer cells in vitro. Bile acids and their synthetic derivatives induced apoptosis in various kinds of cancer cells and anticancer effects. It has been reported that the synthetic chenodeoxycholic acid (CDCA) derivatives showed apoptosis-inducing activity on various cancer cells in vitro. This study was undertaken to investigate the synergistic apoptotic effect of cotreatment with a natural product, CGM and a CDCA derivative, HS-1200 on G361 human melanoma cells. To investigate whether the co-treatment of CGM and HS-1200 compared with each single treatment efficiently reduced the viability of G361 cells, MTT assay was conducted. To investigate augmentation of apoptosis in G631 cells co-treated with CGM and HS-1200, DNA electrophoresis, Hoechst staining, proteasome activity assay, flow cytometry, Westen blot analyses, immunofluorescent staining and confocal microscopy were performed. In this study, G361 cells co-treated with CGM and HS-1200 showed several lines of apoptotic manifestation such as nuclear condensations, DNA fragmentation, the reduction of MMP and proteasome activity, the decrease of DNA content, the release of cytochrome c into cytosol, the translocation of AIF and DFF40 (CAD) onto nuclei, activation of caspase-9, caspase-3, PARP and DFF45 (ICAD), and up-regulation of Bax whereas each single treated G361 cells did not. Although the single treatment of 40 μg/mL CGM or 25 μM HS-1200 for 24 hrs did not induce apoptosis, the co-treatment of them induced prominently apoptosis. Therefore, combination therapy of CGM and HS-1200 could be considered, in the future, as an alternative therapeutic strategy for human melanoma.

        We report a fetal therapy of two cases of type I congenital cystic adenomatoid malformation(CCAM) of the lung. The two fetuses were treated by thoracoamniotic shunting, using a basket-shaped catheter at 25 weeks' and 31 weeks' gestation, respectively. Cystic lesions of the both fetal lung were successfully decompressed after the shunt operation. Healthy babies were delivered at 41 weeks' and 38 weeks' gestation with normal Apgar score. Surgical resections of the affected lobe of the lung in the both babies were performed without complication at 1st and 7th day of birth, respectively. Shunt operation in-utero is considered as a safe and effective therapy for type I CCAM of the fetal lung.

      • Berberry Extract Reduces Neuronal Damage and <i>N</i>-Methyl-<small>D</small>-aspartate Receptor 1 Immunoreactivity in the Gerbil Hippocampus after Transient Forebrain Ischemia

        <P>In the present study, we studied the neuroprotective effects of berberry extract (BE) against ischemic damage and the temporal and spatial alterations of <I>N</I>-methyl-<SMALL>D</SMALL>-aspartate receptor type 1 (NR1) and NR2A/2B immunoreactivities in the gerbil hippocampal CA1 region after transient ischemia to examine anti-ischemic effects and its role in transient forebrain ischemia. In the vehicle-treated group, the percentage of cresyl violet positive pyramidal cells in the CA1 region was about 11.4% compared to the sham-operated group 4 d after ischemic insult. BE showed neuroprotective effects against ischemic damage after ischemia-reperfusion. In the BE-treated groups, about 60—75% of CA1 pyramidal cells were stained with cresyl violet 4 d after ischemic insult. We observed the percentage of berberine (7.45+0.85 mg/g in BE) by HPLC, which is active ingredient of BE. NR1 immunoreactivity in the stratum pyramidale of the CA1 region in the vehicle-treated group was significantly increased at 30 min after transient forebrain ischemia, while at this time the NR1 immunoreactivity in the BE-treated groups was significantly low compared to the vehicle-treated group. The pattern of NR2A/B immunoreactivity in the stratum pyramidale of the BE-treated group and its protein levels were similar to that in the vehicle-treated group after ischemic insult. These results suggest that BE has potent neuroprotective effects against ischemic damage <I>via</I> the reduction of NR1 activity.</P>

        Digestive Physiological Characteristics of the Gobiidae

        In this study, we investigated the characteristics of CCK-producing cells and mucus-secreting goblet cells with respect to stomach fish and stomachless fish of the Gobiidae in order to provide a basis for understanding the digestive physiology. Hairychin goby (Sagamia geneionema), which is stomachless fish, the numbers of mucus-secreting goblet cells is highest in the posterior intestine portion (P<0.05), while CCK-producing cells are scattered throughout the intestine. Gluttonous goby (Chasmichthys gulosus), which is stomach fish, mucus-secreting goblet cells are most abundant in the mid intestine portion (P<0.05), whereas CCK-producing cells are observed only in the anterior and mid intestine portion. Trident goby (Tridentiger obscurus) which is stomach fish, mucus-secreting goblet cells were most abundant in the mid intestine portion (P<0.05). CCK-producing cells are found in the anterior and mid intestine portion. Giurine goby, Rhinogobius giurinus which is also stomach fish, the largest number of mucus-secreting goblet cells showed in anterior intestine portion except for esophagus (P<0.05). CCK-producing cells are present only in the anterior and mid intestine portion. In S. geneionema, digestive action occurs in the posterior intestine portion to protect and functions to activate digestion. In contrast, in C. gulosus, T. obscurus and R. giurinus, their digestive action occurs in the anterior and mid intestine portion to protect and functions to activate digestion. Further studies of the modes of food ingestion by these fish, the contents of their digestive tracts, and the staining characteristics of the goblet cells need to be carried out.

      • Disorders of Small and Large Intestine : Expression Pattern Of E-And N-Cadherin In Colorectal Adenocarcinoma With Liver Metastasis

        Background Aims: Colorectal carcinoma (CRC) is known to be metastasized to the liver frequently and is the major cause of cancer-related death. Recent studies have identified several biomolecular alterations that influence prognosis after resection of liver matastases, including treatment-related and clinicopathologic factors. However, the relationship between the expression pattern of E- & N-cadherin and liver metastasis of CRC is poorly understood. Methods: The present study included 45 patients who underwent hepatic resection for colorectal hepatic metastasis. Immunohistochemical analysis of the expression of E- & N-cadherin protein was carried out for evaluating the metastatic potential of CRC. The methylation status of the E- & N-cadherin promoter was investigated by utilizing the methylation-specific polymerase chain reaction (MSP) assay in 45 patients. The correlation between E- & N-cadherin expression and clinicopathologic parameters was also analysed. Results: Abnormal E-cadherin expressions observed 42.2% (19/45) in primary cancers and 46.7% (21/45) in metastatic tumors. Abnormal N-cadherin expressions were noted 71.1% (32/45) in primary cancers and 82.2% (37/45) in metastatic tumors. Methylated alleles for E-cadherin presented 73.3% (33/45) in primary cancers and 64.4% (29/45) in metastatic tumors. Methylated alleles for N-cadherin presented 11.1%(5/45) in primary cancers and 8.9% (4/45) in metastatic tumors. Abnormal E-cadherin expression was significantly associated with hypermethylation of E-cadherin in primary CRC (P=0.036). Recurrence rate was significantly associated with liver mass size (>3 cm) and abnormal E-cadherin expression in liver (p=0.02). Survival rate was significantly associated with liver metastasis number (multiple)(p=0.01). Conclusions: Abnormal expression of E-cadherin was significantly associated with E-cadherin promoter methylation in CRC. Liver metastatic mass size and metastasis number were significantly associated with survival rate but E-cadherin, N-cadherin and methylation state in liver were not significantly associated with survival rate.

      • Differences in conjuctival microbiome between patients with atopic keratoconjuctivitis and healthy individuals

        Background: Atopic keratoconjuctivitis (AKC) is an allergic ocular diasese which cause severe damage to cornea and vision loss. AKC is associated with atopic dermatitis (AD), so alteration of conjuctival microbiome may be associated with immunologic imbalance. Objectives: We analyzed floral communities of conjuctiva in patients with AKC, and compared those in healthy induviduals. Methods: 16S ribosomal ribonucleic acid (rRNA) gene sequencing and data processing were used to aquire library of compositional information of patients with AKC and healthy individuals. The microbial and mycobial communities of conjuctiva in AKC were charaterized by average taxonomic composition, relative abundance and diversity analysis. Results: In AKC patients, the conjuctival taxonomic composition is more diverse than those in healthy individuals. Beta diversity in patients with AKC was higher compared to healthy control group. Conclusion: Composition and diversity of microbial and mycobial communities in AKC patients were different from those of healthy individuals. Although the bacterial genus that cause AKC is not uncertain, alteration of conjuctival microbiome in AKC patients may have an effect on local immunologic dysfunction in AD. Our findings suggested dysbiosis in AKC patients can be associated with mucosal barrier dysfunction as same as in AD patients.
