      • 녹동균 세포외막 단백질 백신 CFC-1-101의 안정성 및 면역원성 검토 : 임상 제 Ⅰ/Ⅱa상 시험

        목적 : 제일제당에서는 녹농균의 세포외막 단밸질을 유효성분으로 하는 백신인 CFC-101을 개발하였으며, 동물시험에서 이 백신의 안전성과 유효성을 입증하였다. 본 연구에서는 이 녹농균 백신의 인체에 대한 안전성과 면역원성을 평가하는 동시에 인체 접종시의 최적 투여 용량을 결정하기 위하여 제 I/Ⅱa상 임상시험을 수행하였다. 방법 : 건강한 성인 남자를 피험자로 선별하여 각 용량군에 백신투여자 6명, 위약투여자 2명을 배정하였다. 백신 투여군은 0.25mg, 0.5mg 또는 1.0mg 용량의 녹농균 백신을 7일 간격으로 3회에 걸쳐 근육주사 하였으며, 위약 투여군에게는 세포외막 단백질을 제외한 동일한 성분을 투여하였다. 백신접종 후 국소적 또는 전신적인 반응의 발생여부를 관찰하고, 혈액시료를 체취하여 백신의 역가와 유효성을 검정하였다. 결과 : 녹농균 백신 CFC-101은 모든 접종자에서 양호한 내약성을 보였다. 또한 0.5mg 과 1.0mg 백신 투여군에서는 100%의 항체양전율을 나타내었다. 생성된 항체는 녹농균 세포외막단백질에 특이성을 보였고, 녹농균 감염에 대해 방어효능이 있었다. 결론 : 이와같은 결과로부터 이 녹농균 백신은 인체에 안전하게 투여할 수 있으며, 높은 항체 생성능으로 감염방어 효능을 보이고 0.5mg과 1.0mg이 최적용량인 것으로 판단되었다. Background : We developed a Pseudomonas aeruginosa outer membrane protein(OMP) vaccine CFC-101, and the prophylactic efficacy of which has been demonstrated in animal models. In order to evaluate the safety and immunogenicity of the P. aeruginosa vaccine, we carried out a phase I/Ⅱa clinical trial in healthy male volunteers. Methods : Groups of eight volunteers, including two placebo subjects, were vaccinated intramuscularly with three doses of 0.25, 0.5 or 1.0 mg of the vaccine at one week intervals. Sings of systemic and local reactions observed after vaccination were recorded for each vaccinee for 5 days. Physical examinations were performed on days 0, 1, 7, 8, 14, 15, 21, and 42, and clinical laboratory tests were done on days 0, 3, and 21. Blood samples for assay of serum antibody levels were obtained up to 42 days after the first vaccination. Results : The vaccine was generally well tolerated by all vaccinees, showing no significant side effects. In the three dosage groups, all vaccinees, except one receiving the 0.25 mg dose, showed significant elevation in serum IgG antibody titers against the vaccine proteins, indicating 100% seroconversion in 0.5 and 1.0 mg groups. The human antibodies induced by the vaccine were specific for P. aeruginosa OMPs, as confirmed by western blot analysis and immunoprecipitation assays. The capacity of the human antisera to enhance opsonophagocytic killing activity by polymorphonuclear leukocytes and to confer protection against P. aeruginosa infections indicates that the antibodies elicited by the vaccine have protective efficacy. Conclusion : We conclude that the P. aeruginosa OMP vaccine is safe and effective for human use and its optimal dose to be 0.5 or 1.0 mg.

        흰쥐에서 반복적 스트레스가 신경활성 스테로이드의 GABAA 수용체 조절작용에 미치는 영향

        연 구 : 신경활성 스테로이드는 대뇌피질 세포막의 GABAA 수용체 복합체에서 GABAA 수용체 효현제 양상으로 작용하여 [³H]flunitrazepam이 GABAA 수용체에 결합하는 항진 작용이 관찰되었으나, 반복적으로 신경활성 스테로이드를 처치한 세포에서는 이러한 신경활성 스테로이드의 [³H]flunitrazepam 결합 항진효과가 소실되었다는 보고가 있었다. 이에 본 연구에서는 반복적인 스트레스에 부하된 흰쥐를 사용하여 신경활성 스테로이드의 GABAA 수용체에 대한 배위자(ligand)의 결합반응 양상을 정상 실험동물과 비교, 관찰하여 보고자 하였다. 방 법 : 본 연구에서, 벤조다이아제핀 수용체 결합반응(receptor binding assay)을 위한 [3H]flunitrazepam 결합반응을 위해서는 GF/B 여과지(Whatman) 상에서 Brandel M-24R을 사용하여 측정했고, GABAA 수용체 결합반응 (receptor binding assay)을 위한 [3H]muscimol 결합반응에서는 각 시험관 당 100㎕의 대뇌피질 조직(약 0.32mg 단백에 해당)을 사용하였으며, 상기 벤조다이아제핀 수용체 결합반응과 동일한 실험방법을 실시하였다. 결 과 : 신경활성 스테로이드인 pregnanolone은 대뇌피질 세포막의GABAA수용체 복합체에 대한 [³H]flunitrazepam 결합반응을 항진시켜 GABAA 수용체 효현제 양상으로 작용하는 것으로 관찰되었다. 또 pregnanolone이 GABAA 수용체 복합체상의 GABA 결합부위에 대한 결합력을 알아보기 위하여 GABAA 수용체 효현제인 [³H]muscimol 결합반응을 실시하였을 때 pregnanolone은 50%정도의 억제율을 보여주는 GABA와는 그 결과가 다르게 [³H]muscimol 결합반응을 억제하지 않음으로써. muscimol과 경쟁적으로 같은 결합부위에 작용하지 않는다는 것을 보여주었다. 또 반복적인 부동화 스트레스에 부하된 흰쥐의 대뇌 피질조직을 사용하여 pregnanolone의 [³H]flunitrazepam 결합반응을 관찰하였을 때, 대조군에 비하여 pregnanolone의 [³H]flunitrazepam 결합 항진작용이 감약되는 것을 관찰하였다. 결 론 : 이상의 결과로부터 반복적인 스트레스에 부하된 흰쥐의 대뇌피질조직에서는 정상 흰쥐에 비하여, pregnanolone이 GABA성 신경전달에 미치는 항진작용이 감약되었으며, 이는 반복적인 스트레스와 연관된 불안장애의 정신생리 규명에 중요한 정보라고 사료된다. Objectives : Pregnanolone is a potent positive modulator of the γ-aminobutyric acid(GABA) response that enhances the binding of [³H]flunitrazepam to the GABAA receptor. Recently, it was reported that chronic treatment with pregnanolone uncouples allosteric interactions between steroid and benzodiazepine recognition sites. The present study was designed to assess the effect of repeated stress on the modulation of neuroactive steroids on the GABAA receptor. Methods : The effect of steroids on the ligands binding to GABAA receptor was investigated using cerebral cortices of unstressed and repeatedly immobilized rats. Male Sprague-Dawley rats, weighing 200-250g were forced to suffer an immobilization stress for 2 hours. Results : Pregnanolone enhanced the binding of [³H]flunitrazepam to GABAA receptor in both of unstressed and repeatedly stressed rats. However, repeatedly stressed rats showed significantly higher values in EC50 and lower values in Emax of enhancement binding of [³H]flunitrazepam than those of unstressed rats. Conclusions : From these findings, it can be concluded that repeated stress reduced the positive modulation of neuroactive steroid no the GABAA-receptor complex.

        In the 21th century. everyone feels that the multimedia system is close at hand in real life due to the rapid advance of the computer processing ability and high speed and high quality of communication services. Also the limited frequencies resource will be optimized due to rapid advances in digital video technology which is believed superior to analogue technology in information engineering. MEPG-2 has been introduced for broadcasting use such as digital TV Thus it features the high-definition and hyper-low bit rate. But. because of much throughput it has been implemented by high-priced private ASIC chip and is not in general use yet. But in this research. noticing the rapid enhancement of PC processor performance comparing with the price. MPEG-2 was developed by real time software MPEG-2 had been known impossible to implement with S/W. but the research proved the possibility of the S/W implementation and below are the pictures also in the research was improved Motion Vector and Compensation Algorithm which requires the most operations and DCT was made possible real time process. Multimedia Info Society has settled and accompanied by the rapid advance of image-processing technology and lots of standards.

        개인방사선피폭선량평가

        최근, 개인방사선피폭선량평가에 관한 기술기준이 과학기술처에 의해 고시됨에 따라 국내에서도 개인선량계를 이용한 체외피폭선량평가결과의 신뢰성 문제가 크게 대두되고 있다. 한국원자력연구소에서는 이러한 구내의 상황을 인식하고 자체적인 신뢰성 확보를 위해 미국의 Oak Ridge국립연구소의 주관하에 개인피폭선량의 평가에 관한 국제상호비교검증시험(Personnel Dosimetry Intercomparison Study ; PDIS) 을 수행하였다. 비교 검증시험에는 한국원자력연구소에서 사용하고 있는 Teledyne PB-3 열형광선량계가 사용되였으며 선량게산 알고리즘은 Teledyne PB-3 version1.5-1989를 사용하였다. 본 연구에서는 지금까지 실시된 PDIS의 결과를 요약하고 현재 PB-3 시스템의 개인선량평가성능에 대해 고찰하였으며, 선량평가절차에 대한 문제점 도출을 통하여 직업적 방사선피폭선량 평가능력의 향상을 위한 방안을 제시하였다. Recently, the Ministry of Science and Technology issued a Ministerial Ordinance (N0 1992-15) about the technical criteria on personnel radiation dosimetry. In today's climate, it is important to demonstrate and document that the processor's systems and services to others meet national standards of quality. The purpose of this study is to verify the performance of the Teledyne PB-3 personnel dosimetry system that is generally used in Korea Atomic Energy Research Institute (KAERI) by intercomparison with Oak Ridge National Laboratory. The KAERI has been participated in this personnel dosimetry intercomparison study(PDIS) program since 1991 and it could be possible to test and calibrate personnel monitoring system. This report presents a summary and analysis of by about 50 does equivalent measurements reported for PDIS-16 through 18 (1991-1993) with emphasis on neutron does equivalent sensitivity, accuracy and methods to improve personnel dosimetry performance are also discussed.

        The suction pressure of an abnormal condition spark ignition engine which has non adjusting intake valves becomes reduced and residual gas increases at a greater rate than under normal combustions. This reduction in suction pressure is caused by the distribution of in equalable air in a cylinder leading to abnormal combustion. If this condition continues, the engine ceases due to an increase in engine heat loss. In this study, we conducted tests to determine the combustion characteristics of a spark ignition engine under both norman and abnormal conditions. The results of this study will establish basic data analysis for developing an automotive diagnosis system by analyzing some of the factors including pressure in the cylinder, output signal of the MAP Sensor and Exhaust gases.

      • R-12 냉매용 냉동장치의 성능에 미치는 R-12와 R-134a 냉매의 효과

        High pressure, pressure ratio, refrigerating effect, heat transfer from the condenser and the power of the compressor etc. of a self-made refrigeration equipment for R-12 are investigated when R-12 and R-134a are used as the coolants. The comparison between the performance for R-12 and that for R-134a is made. As a result, R-134a is better than R-12 in the view of high pressure, refrigerating effect and the coefficient of performance and vice versa in the view of pressure ratio, exit gas temperature from the compressor and heat transfer from the condenser.

      • 세라믹튜브 및 패킹형 플라스틱 여재충전 혐기성 여상의 유가공 폐수 처리특성 및 반응조 내부거동

        This study was carried to develop of anaerobic reactor to be able to maximize the organic removal efficiency using laboratory-scale anaerobic filter. The ceramic tube and pall ring media were tested for the applicability as packing media of anaerobic filter for the treatment of a dairy waste. This study was conducted to determine the anaerobic treatability of a dairy waste, and investigate the effects of ceramic tube and pall ring media. Anaerobic filters were packed with packing volume of 65% of total working volume. When treating a dairy waste, the organic removal efficiency of anaerobic filter packed with pall rings was a little higher than that of anaerobic filter packed with ceramic tubes. This was because the reactor packed with ceramic tube had clogging problems and substrate transfer limitations. It seemed that the reactor packed with ceramic tubes had uneven packing due to structural characteristics. The COD removals of ceramic tube and pall ring anaerobic filters were greater than about 91% and 94%, respectively, at organic loading rate lower than 5kgCOD/㎥/d. The COD removals of both reactors decreased to 80% with organic loading rate of 10kgCOD/㎥/d. Biomass capture and solid-liquid-gas separation efficiency seemed to vary with the type of media.

        This paper researched the scale of subjective speech quality measurement over VoIP(Voice over IP) service which is a component of broadband networks. Though objective parameters of multimedia services like PSNR or Jitter can be easily measured and defined, those factors are not easily met the users' perceptual recognition. It is suggested that the scale of speech quality measurement through the subjective measurement is more adequate for end-to-end speech quality which is composed of sender-side quality, transmission quality, receiver-side quality, which provides the degree of correctness in speakers' representation, the degree of impairment caused by various factors, and the degree of recognition processed speech, respectively. It's also examined by the proposed method and verified by it's availability.
