      • 탄수화물에서 syn-과 anti-2, 3-디히드록시 알데히드의 합성 : L-Factor와 Muricatacin의 합성

        천연물합성에 있어서 합성중간체로서 유용한 광학활성의 O-보호된 syn-과 anti-2, 3-디히드록시 알데히드의 네 가지 입체이성질체들을 쉽게 구할 수 있는 D-글루코스나 D-크실로스에서 유도된 푸라노스를 변형시켜 합성하였다. 예컨대 불포화 퓨라노스계의 이중결합에 수소화붕소 첨가반응-산화반응에 의해 입체선택적으로 히드록시퓨라노스를 얻고 이를 O-베질화후 탈아세톤화, 산화성절단반응에 의해 (2S, 3S)-3-포르옥시-2-벤질옥시알칸알을 얻었으며 그 외 (2S, 3R)-, (2R, 3S)-, (2R, 3R)-디히드록시알칸알을 얻었다. 이러한 중간체들을 이용하여 L-factor와 muricatacin을 합성하였다. Four stereosiomers of optically active O-protected syn- and anti-2, 3-dihydroxy aldehydes, chiral synthons for natutal product synthesis, were synthesized from readily avilable D-glucose and E-Xylose. (2S, 3S)-3-Formylosy-2-benzylxoy alkanals were synthesized by stereoselective hydroboration of the unsaturated furanoses followed by O-benzylation, deprotection, and oxidative cleavage. (2S, 3R)-, (2R, 3S)-, and (2R, 3R)-Dihydroxy alkanals were also synthesized. Using these chiral synthons, (4R, 5S)-(-)- and (4S, 5S)-(+)-L-factors, the proposed autoregulators from Strpiomyces griseus and muricatacin, a biologically active constituent from the seeds of Annona muricata L. were synthesized.

        탄수화물로부터 유도된 불포화 퓨라노스 고리계를 입체 선택적인 수소화 반응에 의해 chiral synthon인 광학활성의 (S)-3-포르밀옥시 알데히드를 합성하였고, 탄수화물에서 유도된 3-히드록시 퓨라노스를 Barton-McCombie의 deoxygenation반응에 의해 (R)-3-포르밀옥시 알데히드를 합성하였다. (S)-e-Formyloxyaldehydes, chiral synthons for natural products, were synthesized via highly stereoselective hydrogenation of the unsaturated furanose ring system derived from D-glucose or D-xylose. Alternatively, (R)-3-formyloxyaldehydes were prepared via deoxygenation of 3-hydroxyfuranoses.

      • Comparative AFLP Profiles among Strains of Korean Races of Xanthomonas oryzae pv. oryzae.

        We used an amplified fragment length polymorphism (AFLP) analysis, a novel PCR-based technique, to differentiate Xanthomonas oryzae pv. oryzae (Xoo) of Korean races. The 6 strains of Xoo K1, K2, K3 races were tested with 81 AFLP primer combinations to identify the best selective primers. The primer combinations were selected according to their reproducibility, number of polymorphic bands and polymorphism detected among Xoo strains. 18 strains of Xoo K1, K2 and K3 races were analyzed with the selected combinations of primer set. Some primer combinations (Eco R I +1 / Mse I+1) could differentiate Xoo of Korean races that were not distinguished by other fingerprinting analysis. Thus AFLP fingerprinting permitted very fine discrimination among different races.

      • 枸杞 Lycium chinense Miller의 果實이 實驗的 糖尿에 미치는 影響

        The influence of fruit of Lycium chinense Miller on the blood sugar levels and serum transaminase (SGP-T, SGO-T) activities from alloxan-diabetic rabbits were observed. The Lycii Fructus were extracted with methanol, and the extracted sample was orally administrated to normal and alloxan diabetic rabbits. As for the blood sugar levels and serum transaminase activities of normal rabbits no significant variation were detected by administration of the extracted sample. The blood sugar levels and serum transaminase activity of normal rabbits were significantly increased by administration of the alloxan. The increased blood sugar levels of 'the alloxan-diabetic rabbits were gradually decreased by administration of the extracted samples, and were returned to the control levels after 12 day. The increased transaminase activities in serum of the alloxan-diabetic rabbits were significantly decreased after 96 hours. According to the above results, it would be concluded that the extract of Lycii Fructus influences for the recovery of the increased amount of blood sugar levels and serum transaminase activities by alloxan.

        The primary production of phytoplankton in oyster farming bays were seasonally measured by ^14C method in Hansan-Koje Bay and Kamagyang Bay for one year period. In Hansan-Koje Bay, mean daily primary production was 0.52gC/㎡/day(0.22∼1.02gC/㎡/day), and it was relatively high in summer and low in winter. Annual primary production was estimated to be 189gC/㎡/yr. Mean chlorophyll α content was 1.70mg/㎥, and it was relatively high in summer. In Kamagyang Bay, mean daily primary production was 0.91gC/㎡/day(0.11∼3.61gC/㎡/day), and it was maximum in summer and minimum in fall. Annual primary production was estimated to be 334gC/㎡/yr. Mean chlorophyll α concentration was 2.34mg/㎥, and it was maximum in summer and relatively high in winter. Considering the water area of the two osyter farming bays, annual carbon production by phytoplankton were 9,450 ton in Hansan-Koje Bay and 37,000 ton in Kamagyang Bay. In general pattern of primary production in these bays, high production occurs in summer with increasing trend since spring.

        뇨(尿)중 3-Methylhistidine 함량의 측정과 이용

        기존의 방법을 일부 수정하여 뇨중 3-methylhistidine을 분석하였다. 뇨중 3-methylhistidine을 fluorescamine 유도체화하여 HPLC에 주입하고 C_18 column과 10 mM acetonitrile/sodium phosphate buffer(pH 7.5)로 분리·용출시켜 형광검출기로 측정하였다. 3-methylhistidine의 체류시간은 7분 이내이었으며, histidine과의 분리상태도 서로 간섭함이 없이 양호하였다. 뇨에 3-methylhistidine을 첨가하고 이를 분석하였을 때의 회수율은 93∼106%로 높은 수준이었다. 체육학과 남학생중 웨이트 트레이닝 단련자와 비단련자를 대상으로 조사한 단기간의 웨이트 트레이닝에 따른 뇨중 3-methylhistidine 함량의 변화는 두 집단 모두 웨이트 트레이닝후의 3-methylhistidine 분비량이 유의하게 증가하였다. A modified method is given for the precolumn derivatization and subsequent high-pressure liquid chromatographic seperation of 3-methylhistidine from urine. The elution contained isocratic solution with acetonirile and 10 mM sodium phosphate(pH 7.5) requires less than 7 min. The recoveries of 3-methylhistidine from urine control were 93% to 106%. 3-Methylhistidine determinations were performed on urine samples from volunteers who were both male trained and non-trained physical undergraduates. As the result, urinary 3-methylhistidine content of volunteers increased significantly after weight training.

        Direct Examination of the Dietary Preference of the Copepod Calanus helgolandicus Using the Colorimetric Approach

        The food selectivity of tethered females of the copepod Calanus helgolandicus was examined by using the colorimetric approach. First, feeding behavior of the copepod did not show any differences between the red-color stained with neutral red and non-stained diets using the diatom Coscinodiscus curvatulus. Then, the copepods were fed a mixtures of two diets, the diatom C. curvatulus, stained with neutral red, and the dinoflagellate Gymnodinium sanguineum for $14\sim60$ minutes of feeding duration. The foregut colors of females were examined using a stereo-microscope and a video monitor. The foreguts of animals fed with a high density of diatoms in mixed diets showed a dark red color, whereas those fed with a high density of dinoflagellate in mixed diets were a dark yellow. The results suggest that this species of copepod may not selectively feed either one of the diets used for this study. Their feeding activity may be more likely related to the density of available prey in their environment. Therefore, this quick and easy colorimetric approach could provide very useful information, like the pre-screening procedure before designing and conducting the time-consuming and complex feeding experiments to understand the feeding ecology of copepods.

        Effects of Enhanced pCO<sub>2</sub> and Temperature on Reproduction and Survival of the Copepod Calanus sinicus

        We tested the combined effects of increased partial pressure of $CO_2$ ($pCO_2$) and temperature on the reproduction and survival of the copepod Calanus sinicus from Asan Bay, the Yellow Sea under laboratory conditions to understand the impact of acidification on copepods. Egg production rate, survival rate, and fecal pellet production of C. sinicus were not affected by 1305 ppm $pCO_2$ or with combined treatments of temperature and $pCO_2$, including $8^{\circ}C$ and 289 ppm $pCO_2$ (ambient), $8^{\circ}C$ and 753 ppm $pCO_2$ (high $pCO_2$), $12^{\circ}C$ and 289 ppm $pCO_2$ (high temperature), and $12^{\circ}C$ and 753 ppm $pCO_2$ (greenhouse), for 5 or 10 d of exposure. However, egg hatching success of C. sinicus decreased significantly in the greenhouse treatment compared with the ambient or the high $pCO_2$ treatments. These results suggest that a combined treatment ($pCO_2$ and temperature) affected egg viability more than a single treatment($pCO_2$).
