      • Approximating bone ECM: Crosslinking directs individual and coupled osteoblast/osteoclast behavior

        <P>Osteoblast and osteoclast communication (i.e. osteocoupling) is an intricate process, in which the biophysical profile of bone ECM is an aggregate product of their activities. While the effect of microenvironmental cues on osteoblast and osteoclast maturation has been resolved into individual variables (e.g. stiffness or topography), a single cue can be limited with regards to reflecting the full biophysical scope of natural bone ECM. Additionally, the natural modulation of bone ECM, which involves collagenous fibril and elastin crosslinking via lysyl oxidase, has yet to be reflected in current synthetic platforms. Here, we move beyond traditional substrates and use cell-derived ECM to examine individual and coupled osteoblast and osteoclast behavior on a physiological platform. Specifically, preosteoblast-derived ECM is crosslinked with genipin, a biocompatible crosslinker, to emulate physiological lysyl oxidase-mediated ECM crosslinking. We demonstrate that different concentrations of genipin yield changes to ECM density, stiffness, and roughness while retaining biocompatibility. By approximating various bone ECM profiles, we examine how individual and coupled osteoblast and osteoclast behavior are affected. Ultimately, we demonstrate an increase in osteoblast and osteoclast differentiation on compact and loose ECM, respectively, and identify ECM crosslinking density as an underlying force in osteocoupling behavior.         HVPE 방법에 의해 r-plane 사파이어 기판 위의 선택 성장된 GaN/AlGaN 이종 접합구조의 특성

        본 논문에서는 혼합소스(mixed-source) HVPE(hydride vapor phase epitaxy)방법으로 선택성장(SAC: selective area growth) GaN/AlGaN 이종접합구조의 발광다이오드를 r-plane 사파이어 기판 위에 제작하였다. SAG-GaN/AlGaN DH(double heterostructure)는 고온 GaN 버퍼층, Te 도핑된 AlGaN n-클래딩층. Gan 활성층. Mg 도핑된 AlGaN p-클래딩층. Mg 도핑된 GaN p-캡층으로 구성되어있다. GaN/AlGaN 이종접합구조의 발광다이오드의 특성을 알아보기 위해 SEM을 통한 구조적 분석과 전류-전압 측정(I-V: current-voltage measurement), 전류-광출력(EL: electroluminescence) 측정을 통하여 전기적, 광학적 특성을 평가하였다. In this paper, a selective area growth (SAG) of a GaN/AlGaN double heterostructure (DH) has been performed on r-plane sapphire substrate by using the mixed-source hydride vapor phase epitaxy (HVPE) with multi-sliding boat system. The SAG-GaN/AlGaN DH consists of GaN buffer layer, Te-doped AlGaN n-cladding layer, GaN active layer, Mg-doped AlGaN p-cladding layer, and Mg-doped GaN p-capping layer. The electroluminescence (EL) characteristics show an emission peak of wavelength, 439 nm with a full width at half maximum (FWHM) of approximately 0.64 eV at 20 mA. The I-V measurements show that the turn-on voltage of the SAG-GaN/AlGaN DH is 3.4 V at room temperature. We found that the mixed-source HVPE method with a multi-sliding boat system was one of promising growth methods for III-Nitride LEDs.

        Purpose: The purpose of this study was to identify the relationship between satisfaction toward appearance and the level of mental health among female middle school students. Methods: The total study sample was 432 consisting of 3rd year female students who attended co-educational or girls’ middle schools in Korea. Data were collected from August 8 to August 16, 2012. Statistical analyses were conducted using t-test and Pearson’s Correlation Coefficient with SPSS18.0Windows software. Results: There was correlation between appearance satisfaction and mental health, especially with personal sensitivity (r= -.447, p= .000) and depression (r= -.350, p= .000). The level of mental health significantly differed according to type of school (t= -2.668, p= .008). However, appearance satisfaction did not differ according to type of school. Conclusion: The results of this study implied the importance of appropriate education and nursing intervention which would improve satisfaction toward the appearance of female middle school students.

      • 여대생의 우울, 스트레스와 문제도박과의 관계

        Purpose: The purpose of this study was to learn about problem-gambling level, depression and stress among female college students. Method: In this cross-sectional design study, a convenient sample of 325 female college students were recruited between September and October, 2014. Measurement used for this study were the CPGI(Canadian Problem Gambling Index), the CES-D(Center for Epidemiologic Studies Depression Scale), and the Life stress scale for college students. Data were statistically analyzed using descriptive statistics, t-test, ANOVA and chi-square test. Result: About 58.5 percent of the participants had experience of gambling; lottery was the most frequently used gambling among participants. 36 participants(11.1%) were problem gamblers. Relationship of smoking status to problem gambler group was statistically significant. There were statistically significant relationships among problem-gambling level(Non-problem, Low-risk, Moderate-risk, High-risk), depression and stress. The problem gambler group had significantly higher trait stress scores compared to the normal group. Conclusion: Problem gambling was a risk factor for both depression and stress among female college students. The proposals of this study are as follows; first, in depth-research are required with more expanded sampling as sample of this study was conveniently gathered, it is hard to generalize the result. Second, a problem gambling prevention program is recommended for low-risk problem gamblers as they have risk to become problem gambler later. Third, nursing interventions for problem gamblers should be investigated with regard to depression and stress level.

      • 전류측정형 배열화 센서를 위한 다중전극 정전위 전해시스템의 개발

        다성분 동시분석용 배열화 전류감지센서(Amperometric Sensor Array)를 연구하고 적용하기 위해 다중전극용 정전위전해시스템을 설계 제작하였다. 이 시스템은 마이크로 컴퓨터와 인터페이스 되어 Turbo C 2.0 언어로 작성된 프로그램에 의해 16개의 측정 전극의 전위를 동시에 조절할 수 있으며 각 전극의 전류를 측정하고 데이터를 처리할 수 있다. 각 측정전극의 전위는 -2V~+2V 범위에서 2mV 간격으로 임의로 정할 수 있으며, 전류감도는 1mA/V~10nA/V의 여섯 범위 중에서 선택할 수 있고, 측정값의 풀 스케일은 ±10V, 분해능은 0.3mV 이다. 이 전해시스템과 16개의 백금전극이 RDE에 대해 동심원 상으로 배열된 다중전극을 사용하여 다중전극 PAD 방법으로 0.2N NaOH 용액에 녹아있는 과산화수소의 산화환원 특성을 조사하였다. To study and apply the amperometric sensor array for the simultaneous determination of multielements, we designed and fabricated multiple electrode potentiostat system. This system, interfaced with microcomputer, can control potentials of 16 working electrodes simultaneously, measure currents and process the data by the program-programed with Turbo C 2.0 language. Potential of each electrode can be controlled over the range from +2V to - 2V at 2mV step. Current sensivities of 1mA/V to 10nA/V in 6 ranges are selectable, and the full-scale of indication is ±10V with 0.3mV resolution. Using this sytem with 16 Pt electrode array (circle concentric to RDE), we investigated reduction- oxidation characteristics of hydrogen peroxide, dissoved in 0.2N NaOH solution, by multiple electrode PAD method.

        Purpose: Precedent studies about the relationship between trauma and other variables was focused on children and middle aged women. This study however, aims to study the female college student population who are exposed to depression and low self-esteem from traumatic events of childhood and adolescence Methods: Using a descriptive correlation study design, 205 female college students attending female colleges were recruited. The instruments Post-traumatic Diagnostic Scale by Jacoz&Perry(1997), Self-esteem Scale by Rosenberg(1979), Family Support &Conflict Scale by Kim, and Depression Scale by Radloff(1977) were used. The questionnaires were distributed with Google Survey online. The data w re analyzed by average, standard deviation, percentage, and t-test using the SPSS/WIN 21.0 program. Results: The degree of trauma and self-esteem, family support and conflict showed negative correlation, and the degree of trauma and depression showed positive correlation. In other words, female college students with higher trauma tend to have lower self-esteem, more cognizance of family conflict, less feeling of family support, and more depression. Conclusion: This study is different in that it analyzes correlation between the degree of trauma and other variables, not the absence or frequency of trauma. This study is also meaningful as it consider the psychological changes from traumatic experience and provides the basic data for the prevention and therapeutic intervention in clinical field.

        <P>There are concerns about the combined estrogenic effects of chemicals since mixtures of these chemicals exist in our environment. This study investigated potential additional interactions between bisphenol A (BPA) and isobutylparaben (IBP), which are major xenoestrogens used in the manufacture of plastics, cosmetics, drugs, and other products. The combined effects of these two chemicals were analyzed by measuring the expression of calbindin-D(9k) (CaBP-9k) in rat pituitary cancer GH3 cells. GH3 cells were treated with single and combination doses of both chemicals (BPA single doses: 10(-7), 10(-6) and 10(-5) M; IBP single doses: 10(-7), 10(-6) and 10(-5) M, and each of the BPA and IBP doses combined). Prior to treatment, cells were temporarily transfected with a plasmid containing an ERE-luciferase reporter gene. Luciferase activity was measured as an indicator of ER activation by 17β-estradiol (E2), BPA, and IBP. BPA (10(-5) M) combined with IBP (10(-7) M and 10(-6) M) induced a significant increase in the luciferase activity. Twenty-four hours after treatment, dose-dependent effects were observed in both single and combined dose groups, and several combination doses induced significant increases in the expression of CaBP-9k and progesterone receptor (PR) at both transcriptional and translational levels. Pre-treatment with ICI 182,780, a pure estrogen antagonist, significantly reversed BPA- and IBP-induced CaBP-9k and PR upregulation in GH3 cells. Taken together, these results indicate that BPA and IBP may have additionally increased estrogenic potency via an estrogen receptor-mediated pathway.</P>

      • 다중전극 정전위 전해시스템을 이용한 H₂O₂, L-ascorbic acid, uric acid, D-(+)-glucose 분석에 관한 연구

        Using the multiple electrodes potentiostat system with 16 Pt electrodes array, the electrochemical behaviour of and feasibility for simultaneous determination of H_(2)O_(2), L-ascorbic acid, uric acid and D-(+)-glucose were investigated. From 3 dimensional data of potential-current-time abtained by pulsed amperometric detection (PAD) method using this multipotentiostat system, the concentration of glucose can be directly measured without the interference of L-ascorbic acid. But it was abserved that uric acid affects at glucose oxidation not by oxidation-reduction reaction but by surface adsorption on Pt electrode.

        von Hippel–Lindau protein promotes Skp2 destabilization on DNA damage

        Germline mutations in the von Hippel–Lindau (VHL) tumor suppressor gene cause VHL disease, a rare and autosomal-dominant genetic syndrome. Because VHL protein (pVHL) is the master regulator of hypoxia-inducible factor alpha (HIFα), the most prominent feature of VHL disease is the deregulation of HIFα proteins. However, the precise mechanism by which the loss of pVHL function contributes to tumorigenesis is not fully understood. Here, we show that pVHL destabilizes the F-box protein Skp2, a chief component of Skp, Cullin, F-box-containing complex that promotes DNA synthesis in the S phase. The β-domain of pVHL interacts with Skp2, stimulating proteasome-dependent Skp2 degradation, but the destabilization of Skp2 does not depend on the E3 ubiquitin ligase activity of pVHL. Notably, the generation of DNA damage induces Skp2 degradation, which is attenuated by the suppression of endogenous pVHL expression. One possible mechanism of pVHL-dependent Skp2 degradation entails the antagonizing of Akt-mediated Skp2 phosphorylation, which maintains Skp2 stability. Reintroduction of VHL into VHL-null renal cell carcinoma (RCC) cells decreased Skp2 levels and restored DNA damage-dependent Skp2 degradation. These results identify the tumor suppressor function of pVHL in delaying the S-phase progression to inhibit cell proliferation on DNA damage. Clinically, this report explains as to why Skp2 accumulates abnormally in RCC tissues.
