      • Selection and Configuration Optimization for Customizable Cloud Services

        Cloud services is a typical trait of cloud computing. It allows tenants rent customizable services transparently, and guarantees substantial cost savings for the providers. With the significant number of cloud services, a key challenge when providing customizable cloud services is how to manage those services and find an optimized configuration service rapidly that maximizes tenants’ preferences, subject to resource constraints. To help address this challenge, this paper introduces a novel approach to support the modeling and configuration of customizable cloud services. In particular, we propose a customizable service selecting and configuring optimization approach based on a heuristic approach, which can quickly derive an optimized valid service selection by evaluation different configurations that optimize tenants’ preferences under resource limitations. The scalability and performance of the algorithms is investigated in detail. And the obtained experimental results demonstrate the feasibility of the proposed method.

        Cerebrosides from Sea Cucumber Protect Against Oxidative Stress in SAMP8 Mice and PC12 Cells

        Alzheimer's disease (AD) is a neurodegenerative disorder. Emerging evidence implicates β-amyloid (Aβ) plays a critical role in the progression of AD. In this study, we investigated the protective effect of cerebrosides obtained from sea cucumber against senescence-accelerated mouse prone 8 (SAMP8) mice in vivo. We also studied the effect of cerebrosides on Aβ-induced cytotoxicity on the rat pheochromocytoma cell (PC12) and the underlying molecular mechanisms. Cerebrosides ameliorated learning and memory deficits and the Aβ accumulation in demented mice, decreased the content of malondialdehyde (MDA), 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-OHdG), 8-hydroxy-2′-deoxyguanosine (8-oxo-G), and nitric oxide (NO), and enhanced the superoxide dismutase (SOD) activity significantly. The neuroprotective effect of sea cucumber cerebrosides (SCC) was also verified in vitro: the cerebrosides increased the survival rate of PC12 cells, recovered the cellular morphology, downregulated the protein levels of Caspase-9, cleaved Caspase-3, total Caspase-3, and Bax, and upregulated the protein level of Bcl-2, revealing that cerebrosides could inhibit Aβ-induced cell apoptosis. The results showed the protective effect of SCC was regulated by the mitochondria-dependent apoptotic pathway. Our results provide a new approach to developing the marine organisms as functional foods for neuroprotection.

        DYNAMIC ANALYSIS OF MISFIRE AND STATOR SHORT CIRCUIT COUPLING FAULT IN ELECTRIC VEHICLE RANGE EXTENDER

        Due to the complex structure and wide excitations of range extender, the misfire and stator short-circuit coupling fault is easy to occur. Therefore, it is necessary to study the coupling fault mechanism of range extender, analyze the cause of the fault and the evolution law of fault, and diagnose the coupling fault through coupling fault characteristics ultimately. In this paper, a range extender is taken as research object. By establishing a nonlinear dynamic model and using Runge-Kutta method with variable step size, the dynamic response characteristics of system under different working conditions are analyzed to study the single fault and coupling fault mechanism. Then through the finite element analysis and practical experiments, the feasibility of coupled fault diagnosis is verified. The results show that when misfire fault and stator short-circuit fault occur at the same time, the vibration intensity of coupling fault is greater than that of single fault, and the 0.5- and 3.0-order harmonic amplitudes of horizontal vibration speed increase with the coupling fault.

        Association of MMP-9 C-1562T polymorphism with risk of Henoch-Schönlein purpura nephritis in children of southeast China

        Henoch-Schönlein purpura nephritis (HSPN) is the most prevalent vasculitis in childhood worldwide. Although the etiology of HSPN is not yet fully elucidated, it is believed to be closely related with infections and genetic predispositions. In this study, a cohort of children from southeast China, including 108 healthy controls and 184 patients were involved in. We scanned the promoter region and the whole gene for genetic variations. A C/T transition located in the promoter region of MMP-9 gene, C-1562T was identified. Subjects were genotyped by using SNaPshot sequencing and the frequencies of CC, CT and TT genotypes were 43, 33 and 24% in patients and 53, 37 and 10% in healthy controls. Allele distribution was found to be in Hardy–Weinberg equilibrium (P = 0.17). Association analysis revealed that the polymorphism was significantly related with HSPN risk (P < 0.05) and T allele was shown to be a risk factor (P = 0.003, odds ratio (OR) 1.63, 95% confidence interval (CI) 1.13–2.34). Additionally, we measured the serum MMP-9 concentration and quantified the mRNA expression. Results showed that an elevated protein level was observed in patients compared with controls in both ELISA (P = 0.0029, 136.5 μg/L vs. 103.9 μg/L) and Western blot assays. Furthermore, we also detected 1.93-fold increase in the mRNA transcription in patients carrying homozygous TT genotype, indicating enhanced promoter activity caused by the polymorphism. Based on the incidence risk, a cut-off value of serum MMP-9 served as HSPN diagnosis was suggested at 155 μg/L. Conclusively, our findings suggested that MMP-9 polymorphism was significantly associated with HSPN susceptibility, and C-1562T T allele contributed to HSPN development.

        Unscented Kalman Filtering for Nonlinear State Estimation with Correlated Noises and Missing Measurements

        The unscented Kalman filtering problem is investigated for a class of nonlinear discrete stochastic systems subject to correlated noises and missing measurements. Here, a random variable obeying Bernoulli distribution with known conditional probability is introduced to depict the phenomenon of missing measurements occurring in a stochastic way. Due to taking the correlation of noises into account, a one-step predictor is designed by applying the innovative analysis and unscented transformation approach. And then, based on one-step predictor and the minimum mean square error principle, a new unscented Kalman filtering algorithm is proposed such that, for the correlated noises and missing measurements, the filtering error is minimized. By solving the recursive matrix equation, the filter gain matrices and the error covariance matrices can be obtained and the proposed results can be easily verified by using the standard numerical software. We finally provide a numerical example to show the performance of the proposed approach.

        Transcriptional profiling reveals the molecular bases of immune regulation in Apis mellifera in response to Ascosphaera apis infection

        Chalkbrood, which results from Ascosphaera apis infection, is one of the bee diseases that causes serious damage to the bee colony. Understanding the molecular bases underlying immune response to chalkbrood disease would facilitate the genetic breeding of bees by selecting races with superior chalkbrood resistance. In this study, transcriptome sequencing was performed to identify genes and pathways involved in the immune response to As. apis infection in A. mellifera larvae. In total, 2,890 differentially expressed genes (DEGs) (FDR < 0.001) were identified between the healthy and As. apis infected bee larvae, including 2,214 up‐regulated and 676 down‐regulated unigenes. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway classification of the DEGs revealed association with development, energy metabolism, signal transduction, disease, and immune response. Among the immune‐related genes, p38, toll‐like receptors (TLRs), integrin, and antimicrobial peptides were up‐regulated under As. apis infection. This study provides valuable gene transcriptional information towards the investigation of molecular mechanisms related to chalkbrood immune response and host pathogenesis in A. mellifera.

        Efficient biosorption of Pb(II) from aqueous solutions by a PAH-degrading strain Herbaspirillum chlorophenolicum FA1

        For the first time, biosorption of lead(II) using a PAHs-degrading bacterium, Herbaspirillum chlorophenolicum FA1, was investigated as a function of initial lead concentration, biomass dosage, pH, and temperature in batch conditions. Results showed that FA1 was highly resistant to lead and grew well even at lead concentration of 200 mg L−1. The kinetic and isotherm data were well described by commonly used models, such as the pseudo-second-order and Langmuir models. Adsorption thermodynamics was spontaneous and endothermic, and FA1 exhibited a high sorption capacity of 151.52 mg g−1. SEM-EDS and FTIR analysis was conducted to further explore the interaction mechanisms.

        Ferroptosis: A Novel Anti-tumor Action for Cisplatin

        Purpose Ferroptosis is a new mode of regulated cell death, which is completely distinct from other cell death modes based on morphological, biochemical, and genetic criteria. This study evaluated the therapeutic role of ferroptosis in classic chemotherapy drugs, including the underlying mechanism. Materials and Methods Cell viability was detected by using the methylthiazoltetrazlium dye uptake method. RNAi was used to knockout iron-responsive element binding protein 2, and polymerase chain reaction, western blot was used to evaluate the efficiency. Intracellular reduced glutathione level and glutathione peroxidases activity were determined by related assay kit. Intracellular reactive oxygen species levels were determined by flow cytometry. Electron microscopy was used to observe ultrastructure changes in cell. Results Among five chemotherapeutic drugs screened in this study, cisplatin was found to be an inducer for both ferroptosis and apoptosis in A549 and HCT116 cells. The depletion of reduced glutathione caused by cisplatin and the inactivation of glutathione peroxidase played the vital role in the underlying mechanism. Besides, combination therapy of cisplatin and erastin showed significant synergistic effect on their anti-tumor activity. Conclusion Ferroptosis had great potential to become a new approach in anti-tumor therapies and make up for some classic drugs, which open up a new way for their utility in clinic.

        Stenotrophomonas maltophilia HW2 enhanced cucumber resistance against cucumber green mottle mosaic virus

        Cucumber green mottle mosaic virus (CGMMV) is a major limiting factor in the production of cucumber plants worldwide. In the present study, we use plant growth-promoting rhizobacteria (PGPR) to control this virus effectively. Stenotrophomonas maltophilia HW2 was isolated from healthy cucumber root, exhibited a good biocontrol efficacy against CGMMV. Here, it is documented that 20 d after virus inoculation, the biocontrol efficacy of HW2 reached 52.61%. HW2 can effectively colonize in cucumber rhizosphere, and also promoted cucumber plants growth. We also examined the effect of HW2 on viral replication and its mechanism. Compared with the control, HW2 pre-treated plants could delay virus replication for more than 3 d and inhibit viral protein genes (CP, MP, Rep) expression in the cucumber leaf. The expression of antioxidant enzyme genes (SOD and CAT) and defense-related genes (PR1 and PR5) were quickly induced by HW2. These results suggest that HW2 induced plant defense responses to CGMMV by increasing the expression of defense response genes. We report for the first time that Stenotrophomonas maltophilia improved cucumber resistance against CGMMV, which highlights the applying of PGPR on controlling of virus diseases.
