<B>ABSTRACT</B><P>Apurinic/apyrimidinic endonuclease 1 (Ape1/Ref-1) dysregulation has been identified in several human tumors and in patients with a variety of neurodegenerative diseases. However, the function of Ape1/Ref-1 is unclear. We show here that Ape1/Ref-1 increases the expression of glial cell-derived neurotropic factor (GDNF) receptor α1 (GFRα1), a key receptor for GDNF. Expression of Ape1/Ref-1 led to an increase in the GDNF responsiveness in human fibroblast. Ape1/Ref-1 induced <I>GFRα1</I> transcription through enhanced binding of NF-κB complexes to the <I>GFRα1</I> promoter. GFRα1 levels correlate proportionally with Ape1/Ref-1 in cancer cells. The knockdown of endogenous Ape1/Ref-1 in pancreatic cancer cells markedly suppressed GFRα1 expression and invasion in response to GNDF, while overexpression of GFRα1 restored invasion. In neuronal cells, the Ape1/Ref-1-mediated increase in GDNF responsiveness not only stimulated neurite outgrowth but also protected the cells from β-amyloid peptide and oxidative stress. Our results show that Ape1/Ref-1 is a novel physiological regulator of GDNF responsiveness, and they also suggest that Ape1/Ref-1-induced GFRα1 expression may play important roles in pancreatic cancer progression and neuronal cell survival.</P>

The pattern of choice among respondents appeared to be InMyunMunWonWaDang. They preferred HanBok design to be the combination of traditional patterns and typical HanBok design. They considered practical utility, design, and comfortableness as factors that they pay attention to when purchasing HanBok. The survey results of household items found that respondents purpose of purchase was for souvenir and gift. They suggested the increased number of item models and representation of locality for market success of household items. They wished to apply geometric patterns for household item design. The color preference for household items included colorless transparent-tone, followed by pastel-tone and blue-tone. They preferred such household items as dolls for decoration, bags, wallets, and sofa/chair pillows. Based on the survey results, we designed and created products of HanBok and household items. We transformed patterns of PoRyuSuGeumMun and BoSangWhaMun for daily outfits (HanBok) and KyiMyunMun and InMyonMunWonWaDang for household items.

Background/Aims: Heat shock proteins (HSPs) protect rats from cerulein-induced acute pancreatitis (AP) by preventing the subcellular redistribution of cathepsin B and the activation of trypsinogen. Autophagy plays a critical role in the secretion of digestive enzymes and triggering of ceruleininduced AP via the colocalization of trypsinogen and lysosomes. Therefore, using a rat cerulein-induced AP model, we investigated whether HSPs prevent AP by regulating autophagy. Methods: Twelve hours after fed standard laboratory chow and water, the experimental groups (cerulein, water-immersion [WI]-cerulein and heat-shock [HS]-cerulein) and the control groups (control, WI, and HS) received one intraperitoneal injection of cerulein (50 μg/kg) or saline, respectively. All of the rats were sacrifi ced at 6 hours after injection. The severity of the AP was assessed based on the serum amylase level and the histological and electron microscopy findings. Western blotting was also performed for HSP60/70 and LC3B-II. Results: WI and HS induced HSP60 and HSP70, respectively. The induced HSP60/70 effectively prevented the development of cerulein-induced AP. Autophagy developed in the rats with cerulein-induced AP and was documented by the expression of LC3-II and electron microscopy fi ndings. The WI-stressed rats and HS-treated rats did not develop cerulein-induced autophagy. Conclusions: HSPs exert protective effects against cerulein-induced AP in rats by inhibiting autophagy. 면역세포화학적 방법을 이용한 고양이 중간뇌에서 Calbindin D-28k 와 Parvalbumin을 함유한 GABA의 분포상에 관한 연구

간추림 고양이 중간뇌에서 calbindin D-28k (CB) 와 parvalbumin (PV), 그리고 gamma-aminobutyric acid (GABA) 의 분포상과 세포의 형태 및 크기를 관찰하고, 동일세포 E 안에서 이들의 공존을 확인하기 위하여 본 실험을 시행 하였다. CB, PA 및 GABA 에 대한 특이성을 지난 단세포군항체를 이용한 면역세포화학적 염색을 하여 관찰한 결과 다음과 같은 결론을 얻었다. 1 적색핵, 흑색질, 눈돌림신경핵, 청색핵에서 CB-IR, PV-JRGABA-IR 세포를 가장 많이 관찰할 수 있었다. 2 적색핵에서는 PV-IR 세포가, 흑색질에서는 CB-IR 세포가 더 많이 출현하였다. 3. PV-IR 과 GABA-IR 이 통일세포에 공존하는 것을 관찰할 수 있었다. 4. CB-IR, PV-IR, GABA- IR 세포의 세포체의 형태는 방추형, 타원형, 원형이었고 그 크기는 15-20 μm 이었으며 세포의 모양을 흩극, 이극, 뭇극신경원이었다.

Background: Panax ginseng has a wide range of biological activities including anti-inflammatory, antioxidant, and immunomodulatory functions. Wild ginseng cambial meristematic cells (CMCs) were obtained from P. ginseng cambium. This study examined the protective mechanism of wild ginseng CMCs against $\small{D}$-galactosamine (GalN)-induced liver injury. GalN, a well-known hepatotoxicant, causes severe hepatocellular inflammatory damage and clinical features similar to those of human viral hepatitis in experimental animals. Methods: Hepatotoxicity was induced in rats using GalN (700 mg/kg, i.p.). Wild ginseng CMCs was administered orally once a day for 2 wks, and then 2 h prior to and 6 h after GalN injection. Results: Wild ginseng CMCs attenuated the increase in serum aminotransferase activity that occurs 24 h after GalN injection. Wild ginseng CMCs also attenuated the GalN-induced increase in serum tumor necrosis factor-${\alpha}$, interleukin-6 level, and hepatic cyclooxygenase-2 protein and mRNA expression. Wild ginseng CMCs augmented the increase in serum interleukin -10 and hepatic heme oxygenase-1 protein and mRNA expression that was induced by GalN, inhibited the increase in the nuclear level of nuclear factor-kappa B, and enhanced the increase in NF-E2-related factor 2. Conclusion: Our findings suggest that wild ginseng CMCs protects liver against GalN-induced inflammation by suppressing proinflammatory mediators and enhancing production of anti-inflammatory mediators.
공침법에 의한 BiDy₂Fe_(5)O_(12) 결정형 나노입자 분말의 제조 및 응용 연구

금속산화물들은 각종 산업재료로서 유용하며 더욱이 그것들의 결정형 나노입자를 경제적으로 제조하는 기술은 21세기 첨단산업에 매우 중요하다. 저자들은 前報에서 화학합성법에 의한 각종 금속산화물의 나노입자를 제조하는 표준공정법을 연구하여 보고하였다. 본 연구에서는 그 표준공정법을 이용하여, 가넷의 일종인 BiDy_(2)Fe_(5)O_(12) 결정 나노입자 분말을 제조하고 그것의 자성체로의 응용을 위해 제품특성을 조사하었다. 가네트 제조과정에 필요한 침전물은 제품설계에 의거 몰비로 계산된 Bi, Dy, Fe의 질산염들의 혼합수용액에 침전제로서 pH NH_(3)-NH_(4)CI 완충용액 일정량을 가하여 얻어졌다. 이렇게 제조된 침전은 열처리과정을 거처 BiDy_(2)Fe_(5)O_(12) 결정형 나노입자 분말로 만들어졌다. 얻어진 제품의 입자크기, 조성, 결정구조 및 광자기성 등 특성시험이 SEM, ICP, XRD, 그리고 VSM 등을 사용하여 실시되었다. 각종 분석법을 사용하여 제품의 특성을 조사한 결과, 최초에 설계한 대로의 자성체가 나노 크기의 분말로 얻어졌으며 이것을 통하여 전보에서 확립한 표준공정법의 유효성도 입증되었다. Metallic oxides are useful as various industrial materials. Futhermore, the cost-effective techniques of fabricating the crystalline nanopartides of metallic oxides are very important for the high-tech industries of the 21 century, In the previous study, the authors reported the standard process for the preparation of crystalline nanoparticles of metallic oxides by chemical synthesis, In this study, BiDyzFesOiz nanoparticles were prepared by using the reported standard process and the characteristics of the product were investigated for applicability as magnetic substance. The precipitation needed for manufacturing process of garnet was made by adding the NH_(3)-NH_(4)CI buffer solution of pH 10 to the mixed solution of bismuth (III) nitrate, dysprosium(III) nitrate and iron(III) nitrate. The amount of each nitrate was calculated by the designed mole ratios. The nano-size crystalline powders of BiDy_(2)Fe_(5)O_(12) were earned by heating the precipitation at 800℃ for 4 hours in air. The characterizations, such as particle size, composition, crystal structure and magneto-optical property of garnet were tested by SEM, ICP, XRD, and VSM analyses. The results of the analyses showed the following two facts. First, a good magnetic substance of BiDy_(2)Fe_(5)O_(12) was earned as crystalline nano-size powders according to the original design. Next, the established standard process for the fabrication of METALLIC oxides nanonartieles was approved as a very effective method.

<P><B>Abstract</B></P> <P>The epithelial to mesenchymal transition (EMT), a hallmark of chronic kidney disease, is a key event in the conversion from tubular epithelial cells to myofibroblasts in renal fibrosis. Epstein-Barr virus (EBV) is a γ-herpes oncovirus associated with chronic kidney disease. However, the relationship between EBV and the EMT process in renal tubular epithelial cells is not well understood. Among EBV-latent genes, EBV-encoded latent membrane protein 1 (LMP1) induces EMT by regulating a variety of molecules in EBV-induced oncogenic transformation. In this study, we investigated EBV-encoded LMP1 and EMT process markers in human proximal tubule epithelial cell line HK-2. LMP1 overexpression induces cell morphological changes via the epithelial to mesenchymal process in HK-2 cells, and these changes accelerate cell proliferation, cell motility, and invasion. Furthermore, VSIG4 upregulation by EBV-LMP1 induced LMP1-mediated EMT, cell motility, and invasion. VSIG4 upregulation by LMP1 was regulated at the transcriptional level via the NF-kB signaling axis. These results suggest that EBV-encoded LMP1 regulates EMT through the NF-kB-VSIG4 axis in HK-2 cells, and VSIG4 is a potential target in EBV-induced chronic kidney diseases.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Epstein Barr virus-encoded LMP1 overexpression induces epithelial to mesenchymal transition in renal epithelial HK-2 cells. </LI> <LI> Upregulation of VISG4 expression by EBV-LMP1 plays an important role in LMP1-induced EMT process in HK-2 cells. </LI> <LI> VSIG4 upregulation by EBV-LMP1 is regulated at transcriptional level via LMP1-induced NF-kB activation. </LI> <LI> The interruption of VSIG4 expression could be a potential strategy for treatment of EBV-infected renal diseases. </LI> </UL> </P>
