        무 추출물이 식품관련 미생물의 증식에 미치는 영향

        To understand the effect of radish on growth of food born microorganisms, mashedflesh radishes were extracted by using acetone and distilled water. Their effect was assayed by measuring the optical density of cultural broth of food born microorganisms, In the experiment, seven strains of food born bacteria and one strain of yeast were used as the test organism. Acetone extract inhibited growth of the cells of L. plantarum, L. sake and Danmuji film yeast. Growth of the film yeast was drastically inhibited in the concomitant presence of 0.03% extract, while other microbes such as L. faecalis. P. pentosaceus, B. subtilis and E. coil grew by succeeding cultivation for 4 to 8 hours after addition of the extract. Water extract, on contrast to acetone extract, at the concentrations of 0.1~1.5% stimulated the growth of lactic acid bacteria, Culture of L. faecalis and L. sake showed an optical density higher than that of control by 40~50 times. The effect was not so apparent against E. coli, S. aureus and Danmuji film yeast.

        구성주의 교수-학습전략이 초등학교 자연과 교과태도 및 자아효능감에 미치는 영향

        The purpose of this study was to examine the effects of constructive instruction on children's academic attitude and self-efficacy of natural science subject. The constructive method emphasizes the learner's self-guiding, cooperative learning activities, the teacher's assistant and scaffolding role. To analyze how much the constructive methods do effect the variables, 88 subjects of 6 grade on elementary school sampled and divided two groups, experimental and comparative. The results are as followings: In the effect on academic attitude, the experimental group is significantly higher than the comparative group. Also, in the effect on self-efficacy, the experimental group is significantly higher than the comparative group.

        보육시설 과학 특별활동 프로그램의 개발과 적용

        The purpose of this study was to develop early childhood science activities, which could be utilized as an extracurricular program in the day-care center. The program is based on the Constructivist theory(Piaget and Vygotsky) and the nation-wide survey data on the extracurricular programs and science activities in the day-care center. After the manuals for the program was monitored and corrected by the professionals, the application of each activities was empirically tested by the teachers in the day-care setting. The effects of the sample program(6 activities) were conducted using Tegano, Sawyers & Moran(1989)'s Scientific problem-solving scale and Lee(2001)'s Scientific attitude scale. Subject of thirty nine children were administered by the pre-test and post-test and experimental group participated the program for six weeks. The results showed that the program was effective for enhancement of scientific problem-solving ability and scientific attitude of young children in day-care setting.

      • VTV: Real Time Obstacle Avoidance of Mobile Robots for Local Path Planning Using LMS

        In this paper, a new algorithm, called Virtual Tangential Vector (VTV), for real time mobile robot obstacle avoidance for local path planning in indoor environment is proposed and demonstrated. It is for a 2-dimensional space using a 2D laser measurement system (LMS) to detect obstacles. VTV consists of two processes. The first defines a virtual encircled obstacle from three points obtained by a LMS. The second decides movement direction combining the gravity vector to the given target position and the tangential vector obtained from the encircled obstacle. The magnitude of tangential vector is taken proportional to the square of the radius of the encircled obstacle to generate a shorter path. The VTV is tested by several cases to verify the performance of the algorithm and shown to be better when compared with a popular algorithm, the so-called virtual force field (VFF) algorithm.

        유아의 기질 , 어머니의 통제책략과 유아의 자기통제행동과의 관계

        The purpose of this study was to investigate whether children`s temperament and maternal control strategies were related to the self-control behaviors of 3-year-old children. The sample was comprised of 50 young children, ranging from 37 to 50 months of age, and their mothers. Mothers were asked to complete a questionnaire on children`s temperament. Situational observation was conducted for obtaining data on maternal control strategies and for children`s self-control behaviors in a resistance to temptation situation. All the subjects` activities were videotaped for 20 minutes. Differences were found in maternal control strategies and children`s self-control behaviors by gender of child and by age and education of mothers. Relationships were found between children`s temperament and maternal control strategies. However, children`s temperament had an indirect effect on their self-control behaviors through maternal control strategies. The findings indicated that the self-control development of children in this sample were in a transitional stage.

        신성요붕증 가계에서 바소프레신 V2 수용체(AVPR2) 유전자 분석 : AVPR2 유전자 R202C 돌연변이의 발견

        목적 : 신성 요붕증(Nephrogenic diabetes insipidus, NDI)은 바소프레신(arginine vasoporessin, AVP)에 대한 신세뇨관의 저항성으로 인하여 요농축의 장애를 특징으로 하는 드문 유전성 질환이다. 반성유전형 신성 요붕증은 바소프레신 V2수용체(AVPR2)의 장애에 기인하며, NDI 환자에서 지금까지 다양한 AVPR2의 돌연변이가 보고되었다. 저자들은 임상적으로 반성 유전형 신성 요붕증으로 진단된 가계에서 AVPR2 유전자의 돌연변이를 발견하기 위하여 분자유전학적 검사를 실시하였다. 방법 : 대상환자의 백혈구에서 추출한 DNA로 AVPR2유전자를 polymerase chain reaction-single strand conformational polymorphism(PCR-SSCP)분석하여 이상이 발견된 부분은 클론닝하여 염기서열을 분석하였다. 같은 PCR 산물을 Hae III로 처리하여 PCR-RFLP(restriction fragement length polymorphism) 분석을 하였다. 결과 : AVPR2 유전자를 PCR-SSCP 분석하였을 때 PCR 산물의 정상인과 이동거리의 차이가 발견되어 환아에서 돌연변이가 있고 환아의 어머니는 보인자임을 예측하였고, 염기서열을 분석하여 675번째 염기 A가 G로 치환됨으로 tryptophan이 cysteine으로 바뀌는 R202C 점돌연변이를 발견하였다. 같은 PCR 산물을 PCR-RFLP 분석을 하였을 때 돌연변이로 인한 Hae III의 인지부위의 상실을 확인하였고 환아의 어머니가 이종접합보유자 (heterozygote)임을 확인하였다. 결론 : 저자들은 임상적으로 신성 요붕증으로 확인된 환아와 어머니의 V2 수용체 유전자를 분석하여 R202C 돌연변이를 확인하였다. 신성 요붕증은 진단이 지연되면 성장장애, 정신박약과 사망을 초래할 수 있는 심각한 질환이나, 태생기 또는 신생아기에 진단하면 후유증을 예방할 수 있으므로 조기진단 및 보인자 발견에 분자유전학적 진단 방법을 적극 활용하여야 하겠다. Purpose : Nephrogenic diabetes insipidus (NDI) is a rare X-linked disorder associated with renal tubule resistance to arginine vasopressin (AVP). The hypothesis that the defect underlying NDI might be a dysfunctional renal AVPR2 has recently been proven by the identification of mutations in the AVPR2 gene in NDT patients. To investigate the association of mutations in th AVPR2 gene with NDI, we analyzed the AVPR2 gene located on the X chromosome. Methods : We have analyzed the AVPR2 gene in a kindred with X-linked NDI. The proband and proband's mother were analyzed by polymerase chain reaction-single strand conformational polymorphism(PCR-SSCP) and DNA sequencing of the AVPR2 gene. We also have used restriction enzyme analysis of genomic PCR product to evaluate the AVPR2 gene. Results : C to T transition at codon 202, predictive of an exchange of tryptophan 202 by cysteine(R202C) in the third extracellular domain was identified. This mutation causes a loss of Hae III site within the gene. Conclusion : We found a R202C missense mutation in the AVPR2 gene causing X-linked NDI, and now direct mutational analysis is available for carrier screening and early diagnosis.

        서울 시내 요식 업소의 위생 상태 및 급식되는 음식의 미생물적 품질 개선을 위한 연구

        Four types of restaurants in Seoul city area were assessed in terms of the sanitary conditions and practices, and the microbiological quality. Sanitary check-lists were developed to evaluate the sanitary condition of sampled restaurants. Subjective samples were randomly selected based on the distribution factors of areas, types, and sizes. Microbiological tests on foods, equipments, and utensils were done according to standard procedures and included total plate count and coliforms. Singnificant differences among types or sizes were determined by using one-way analysis of variance. Correlation coefficients were calculated to determine significant relationships between sanitary scores and microbiological counts. The results of the study are summarized as follows: 1) Sanitary condition of kitchen and dining areas as well as the sanitary practices of employees were evaluated as the unsatisfactory state with potentially hazardous practices observed. 2) The microbiological quality of food items with high cooking temperature was in good condition, but most food items showed high levels of microbiological counts mainly due to the improper food handling practices of employees. 3) The sanitary conditions of equipment and utensils which were used at preparation and cooking phases, and food containers which were used at the serving phase, were crucial. Serveal guidelines were suggested for the improvement of the working environment as well as the food quality.

      • 돼지췌장 Lipase 가 체다치즈 숙성중 조직의 물리적 , 관능적(官能的) 특성에 미치는 영향

        本 實驗은 lipase의 添加量을 달리하여 製造한 Cheddar 치즈를 7℃, 13℃, 21℃에서 各各 20週, 15週, 10週間 熟成시키면서 熟成過程中에 일어나는 texture의 變化를 Instron 및 官能檢査로 評價하여 lipase가 熟成期間中 Cheddar 치즈의 texture에 미치는 影響을 糾明하고자 實施하였다. 1. Fracturability, springiness 및 chewiness는 熟成溫度가 높고, 添加한 lipase의 양이 많아질수록 빠르게 減少하였다. 2. Hardness, cohesiveness 및 gumminess는 平價한 時期에 따라 增減의 幅이 컸으나 그 特性値는 control群, LPL群, HPL群의 順으로 減少하였다. 3. 官能檢査結果 openness와 corky property는 熟成 6週까지 急速한 減少를 보였으나 그 以後로는 거의 變化가 없었다. 4. Shortage는 熟成溫度 및 lipase의 添加에 의해 影響을 받지 않았으며, crumbly body는 control群에 비해 lipase 添加群에서 急速한 減少를 보였으며 熟成溫度가 높아질수록 減少速度 또한 빨라졌다. 5. Lipase를 添加하여 熟成시킨 Cheddar 치즈의 texture 變化를 物理的 官能的으로 評價한 實驗結果 lipase가 Cheddar 치즈의 熟成을 促進시키는 데 效果的인 것으로 觀察되었다. Effects of commercial food grade porcine pancreatic lipase were studied on textural changes in Cheddar cheese which was ripened at various temperatures(7℃, 13℃, 21℃). The physical properties of cheese were measured by an Instron Universal Testing Machine and the sensory characteristics of cheese were evaluated by a sensory panel. The results obtained are summarized as follows: 1. Fracturability, springiness and chewiness were decreased as ripening temperature and amount of lipase increased. 2. Hardness, cohesivness and gumminess were varied as different measuring time, but values of characteristics were increased in order of the amount of lipase. 3. As the results of sensory evaluation, openness and corky property were decreased rapidly during six weeks ripening periods but no variation was observed thereafter. 4. Shortage was not affected by ripening temperatures and addition of lipase, crumbly body was further decreased in the cheese added lipase than control and as ripening temperature was increased, this characteristics was also decreased. 5. Lipase was effective in the acceleration of cheese ripening by physical and sensory evaluation on the texture of Cheddar cheese.
