Distribution of (-)-Yatein in Cupressaceae Family Analysed by High Performance liquid Chromatography

The method for the chiral analysis of (-)-yatein was developed and the distribution of this component in the plants of three genera like Juniperus, Thuja and Chamaecyparis belonging to Cupressaceae family was examined. The chiral analysis of (-)-yatein from the plants was carried out by high performance liquid chromatography on (R,R)-Whelk-O1 column using 81 v/v% methanol as mobile phase. The yatein content in the leaves of Juniperus was the highest in compare with that of the other two genera, providing the possibility of the chemical discrimination of the plants in Juniperus from the other plants in the Cupressaceae family. In general, the yatein content in the leaves was much higher than that in the twigs. This method could be applied for the quality control of (-)-yatein in the plants belonging to Cupressaceae family.

Brazilin [7,11b-dihydrobenz[b]indeno[1,2-d]pyran-3,6a,9,10(6H)-tetrol] inhibited thrombin-, collagen- and ADP-induced aggregation of washed rat platelets. T hrombin- and collagen-induced ATP release were also inhibited by brazilin in a concentration-dependent manner. Brazilin inhibited the formation of platelet thromboxane $A_2$ caused by thrombin, whereas it had no effect on the prostaglandin $D_2$ formation. Brazilin inhibited $^3H$-arachidonic acid liberation from membrane phospholipids of thrombin-stimulated platelets. Brazilin inhibited the rise of intracellular free calcium caused by thrombin. These results indicate that the inhibition of phospholipase ($PLA_2$) activity and [$[Ca^{2+}]_1$ elevation might be at least a part of antiplatelet mechanism of brazilin.

To elucidate the preventive effect of oriental medicine Gungchi-hwadamsun (GH) on stress, we investigated the physiological change of rats which were applied immobilization stress. For immobilization stress, rats were placed in restrainer for 12 hours a day for 3 days. During application of stress, body weight of rats was measured. After sacrifice, 8 organs were taken for measurement of organ weight. Brain was sectioned into 4 parts that are Frontal Cortex, Corpus Striatum, Hypothalamus and Hippocampus. Each part was homogenated and its catecholamine and serotonin contents were measured with HPLC. In our study, stress mainly induced increase of concentration of neurotransmitters in brain, but had mild effect on other physical function of rats. GH inhibited stress induced changes of neurotransmitter content in brain.

The oxidative modification of low density lipoprotein(LDL) has been implicated in the development of atherosclerosis. Oxidized LDL are found in macrophage foam cell, and it can induce an macrophage proliferation in atherosclerotic plaque. In this study, we investigated the hypothesis that gamisoyosan(GS) may reduce atherosclerosis by lowering the oxidiazability of LDL, To achive this goal, we examined the effect of GS on LDL oxidation, nitric oxide production in mouse macrophage cell line, RAW264.7, and the effect of GS on cupuric sulfate-induced cytotoxicity, LDH release, and macrophage activity. GS inhibited the generation of oxidized LDL from native LDL in RAW264.7 cell culture, and decreased the release of LDH from cupric sulfate-stimulated RAW264.7 cell. In other experiments, GS activated RAW264.7 cell, and prolonged the survival time, and increased nitric oxide production in Raw 264.7 cells.
