소맥분과 물엿을 첨가한 침엽수 톱밥배지에서의 꽃송이버섯 생산

꽃송이버섯은 베타글루칸 함량이 높아 버섯재배자들 사이에서 재배를 희망하는 농가들이 부쩍 늘어나고 있다. 하지만 이에 대한 실용적인 재배법이 확립되지 않아 까다로운 버섯으로 인식하여 재배를 기피하고 있어 본 실험에서는 침엽수 톱밥과 소맥분, 물엿을 이용한 간편하고 경제성 있는 배지조성법을 연구하였다. 톱밥 접종원과 액체 접종원을 사용한 경우의 균사 생장에는 뚜렷한 차이가 없었으며, 4℃에서 1일 동안 저온충격을 주는 방식이 원기 형성을 위하여 가장 좋은 방법으로 생각되었다. 또한 자실체 발생용 최적배지는 '미송+소맥분+옥수수+면실박+10% 물엿 수용액'으로 41%의 회수율을 보였으나, '낙엽송+소맥분+옥수수+면실박+10% 물엿 수용액'에서도 회수율이 37%로 자실체 발생량이 많았다. 또한 회수율과 버섯배지 중량 감소율과의 관계를 보면 회수율이 높을수록 중량감소율이 높아 서로 비례한 것으로 조사되었다. Cauliflower mushroom(Sparassis crispa) contains much amount of β-glucan, which make lots of farmer want to cultivate the mushroom. Since a practical cultivation method is not provided yet, the mushroom is considered as a difficult crop to deal with. In this study, we tried to develope a simple method to cultivate the mushroom by use of coniferous sawdust-based medium with wheat flour and molasses. There was no significant differences between the sawdust spawn and the liquid spawn for the mycelial growth of the mushroom. The cold shock in 4℃ for a day was thought to be the best way to seduce primordium formation. The sawdust medium of Pseudotsuga menziesii mixed with wheat flour, corn chip, cottonseed meal and 10% molasses was showed the best yield with 41% followed by that of Larix leptolepis mixed with the same additives with 37% of yield. There was good relations between the fruit-body production and the weight loss of the sawdust substrate.
A study on anti-diabetic effects of Mycoleptodonoides aitchisonii extract

Mushrooms including Mycoleptodonoides aitchisonii are used as foods and employed as folk remedies for diabetes and inflammatroy diseases. This study analyzed anti-diabetic effects of Mycoleptodonoides aitchisonii, which grows in some areas such as Gangwon-do and Jeju-do in Korea, as insulin-derived phosphorylation. When 100 ng/ml of IL-6, inflammatory cytokine was given to SK-hep1, HepG2, Akt phosphorylation by insulin was found to be remarkably reduced. In addition, metformin, Antidiabetics serving as positive control in liver was used. Mycoleptodonoides aitchisonii used for analyzing anti-diabetic effects in this test didn’t give a great impact on the decrease in Akt phosphorylation by IL-6 at high concentration. However, fruit body in Mycoleptodonoides aitchisonii inhibited the decrease in Akt phosphorylation by IL-6 according to the concentration. At the highest concentration 100㎍/ml, it had an effect of increasing Akt phosphorylation to 77%, which was decreased by 50% compared to the insulin treated group by IL-6. Therefore, Mycoleptodonoides aitchisonii fruit body used in this test activated Akt phosphorylation inhibited by IL-6 and showed a possibility of significant anti-diabetic effects compared to positive control(metformin).
Purification and Characterization of Protease from the Hepatopancreas of Shrimp, Penaeus orientalis

A protease without tryptic and chymotryptic activities was purified from the hepatopancreas of shrimp, Penaeus orientalis, using Q-Sepharose ionic exchange, benzamidine Sepharose-6B affinity, Mono-Q, and gel chromatography. Molecular weight (M.W.) of the protease was estimated to be 27kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDSPAGE). The amino acid composition of the protease was different from that of protease from P. japonicus or trypsin from P. orientalis. The protease was completely inhibited by benzamidine, $N\alpha-p-tosyl-L-lysine$ chloromethyl ketone (TLCK), and phenylmethylsulfonyl fluoride (PMSF) and was not affected by leupeptin, pepstatin, N-tosyl-L-phenylalanine chloromethyl ketone (TPCK), iodoacetate, and ethylenediamine tetra acetate (EDTA). The enzyme did not have any activity against Na-benzoyl-DL-arginine p-nitroanilide (BAPNA) or N-benzoyl-L-tyrosine ethyl ester (BTEE) which are specific substrates of trypsin and chymotrypsin, respectively. However, the protease showed hydrolytic activity for a carboxyl terminal of Tyr, Trp, Phe, Glu, and Cys.
Study on mycelial growth and β-glucan content by adjusting the culture medium compositions

Edible mushrooms have been used and researched as medicinal ingredients. They improve immunity and contain excellent anticancer components with large amounts of minerals such as iron, calcium, and potassium. Due to this reason, it has been hailed as a raw material for functional foods. Especially, β-glucan, known to be contained in Ganoderma incidum Karst, Coriolus versicolor, and Phellinus linteus, was reported to inhibit proliferation of cancer cells by activating immune function (macrophages, natural killer cells and T-cells). In addition, mushroom polysaccharides dissolve in water but are undigested ingredients, resulting in stimulating the immune system as well as staying of parasympathetic nervous system in the stomach for a long time. As a result, they induce increase of T-cells and NK cell that attacks cancer cell and has effect on the discharge of body's waste products, blood purification, and constipation improvement. In this study, we report the culture characteristics of Sparassis crispa as to the medium growth compositions, yeast, and elicitor treatments to investigate the optimal condition for the highest β-glucan production in mushrooms.

S. crispa is a good source of food and nutraceuticals, or dietary supplements, due to its rich flavor compounds and beta-glucan content. This review is a comprehensive summary of its distribution, growth, management, general constituents, functional ingredients, as well as its current and potential medicinal and other applications.</P>
