Whole-genome resequencing analysis of 20 Micro-pigs

Background Miniature pigs have been increasingly used as mammalian model animals for biomedical research because of their similarity to human beings in terms of their metabolic features and proportional organ sizes. However, despite their importance, there is a severe lack of genome-wide studies on miniature pigs. Objective In this study, we performed whole-genome sequencing analysis of 20 Micro-pigs obtained from Medi Kinetics to elucidate their genomic characteristics. Results Approximately 595 gigabase pairs (Gb) of sequence reads were generated to be mapped to the swine reference genome assembly (Sus scrofa 10.2); on average, the sequence reads covered 99.15% of the reference genome at an average of 9.6-fold coverage. We detected a total of 19,518,548 SNPs, of which 8.7% were found to be novel. With further annotation of all of the SNPs, we retrieved 144,507 nonsynonymous SNPs (nsSNPs); of these, 5968 were found in all 20 individuals used in this study. SIFT prediction for these SNPs identified that 812 nsSNPs in 402 genes were deleterious. Among these 402 genes, we identified some genes that could potentially affect traits of interest in Micro-pigs, such as RHEB and FRAS1. Furthermore, we performed runs of homozygosity analysis to locate potential selection signatures in the genome, detecting several loci that might be involved in phenotypic characteristics in Micro-pigs, such as MSTN, GDF5, and GDF11. Conclusion In this study, we identified numerous nsSNPs that could be used as candidate genetic markers with involvement in traits of interest. Furthermore, we detected putative selection footprints that might be associated with recent selection applied to miniature pigs.

Lead leached out from the metal lead-sinker attached to fishing nets or contained in ropes are likely to cause the environmental contamination, consequently, resulting in ecotoxicity. However, it is very difficult to find out the data on the disposal reality of fishing nets, lead-containing form of/in fishing net and its content. In previous studies, we presented the containing form of lead and the lead concentration leached out by the containing forms. As a result, the leaching concentration of lead was the highest under the supposition condition of lead-sinker deposited on sandy tidal flat. In this study, as a subsequent study, we investigated long-term leaching characteristics from lead-sinker deposited on sandy tidal flat. We used a serial batch leaching experiment to investigate the long-term leaching characteristics, and the experimental conditions were as follows; solid to liquid ratio 1:20, shaking time 24 hours (repetition of 7 times), room temperature, and shaking rate 170 rpm. As a result, lead leaching concentration after shaking for 24 hours was 0.215 mg/L, showing the highest leaching concentration, and then the leaching concentrations between 2-7 times were greatly decreased. Analysis result by Visual Minteq 3.1 said most of the leached lead component could be extracted as crystal such as Anglesite (PbSO4), Cerrusite (PbCO3), Cotunnite (PbCl2), and Pb(OH)2 by sulfate and carbonate in seawater.

<P>Postmortem pH is a main factor influencing the meat quality in pigs. This study investigated the association of postmortem pH with single-nucleotide polymorphisms (SNPs) in the fourth member of the short-chain dehydrogenase/reductase family (DHRS4), the first member of serpin peptidase inhibitor, clade G (complement inhibitor) (SERPING1), and the apolipoprotein R precursor (APOR) genes in Berkshire pigs. The study included 437 pigs, and genotyping was conducted using the GoldenGate Assay (Illumina, San Diego, CA, USA). DHRS4, SERPING1, and APOR polymorphisms were significantly associated with pH(45) or pH(24) (p<0.05). SERPING1 was also statistically significantly associated with water holding capacity (p<0.05), which is closely associated with postmortem pH. These results suggest that SNPs in the DHRS4, SERPING1, and APOR genes have potential for use as genetic markers for the meat quality in pigs.</P>

J. Anim. Sci. 95: 31-36. The PGM1 gene from four porcine breeds (Berkshire, Duroc, Landrace, and Yorkshire) is highly expressed in liver tissue at the transcriptional level. Single nucleotide polymorphisms (SNPs) of PGM1 were examined to analyze association with increased expression of PGM1 gene in the Berkshire liver. A Leu525 synonymous SNP of Chr6:137174682A@@>@@G (c.1575A@@>@@G) was identified and showed significant (P@@<@@0.05) differences to backfat thickness, drip loss, protein content, fat content, Warner-Bratzler shear force, and post-mortem pH24h. Therefore, it is concluded that PGM1 synonymous SNP is an important factor regulating meat quality. </P>
괭생이모자반 에탄올 추출물이 3T3-L1 지방전구세포의 분화 및 adipogenesis에 미치는 영향

본 연구에서는 괭생이모자반 에탄올 추출물의 항비만 효과에 대해 알아보고자 3T3-L1 preadipocyte에서 Oil Red O staining과 triglyceride content, adipogenesis의 발현에 대해 조사하였다. MTT assay를 통해 괭생이모자반 에탄올 추출물의 세포 독성을 확인 해 본 결과 200~1,000 μg/ml 농도에서 90% 이상의 세포 생존율을 확인 할 수 있었다. 또한 insulin, dexamethasone과 IBMX 복합물을 처리하여 분화를 유도하였을 때는 분화유도제를 처리하지 않은 군에 비해 활발하게 지방구를 형성하였으나, 괭생이모자반 에탄올 추출물을 250, 500, 1,000 μg/ml 농도로 처리하였을 때는 농도 의존적으로 지방구 형성을 억제하였다. 이를 정량하여 triglyceride content (%)로 나타낸 결과 농도별로 13%, 16%, 23% 억제 효과를 나타내었다. 또한 Western blot assay를 통해 adipogenic transcription factor의 발현양상을 비교해 본 결과, 분화유도제 처리에 의해 C/EBPα, C/EBPβ와 PPARγ의 발현이 증가하였으나, 괭생이모자반 에탄올 추출물을 처리하였을 때는 이러한 전사인자들의 발현이 농도 의존적으로 감소됨을 확인 할 수 있었다. 이상의 결과를 통해 괭생이모자반 에탄올 추출물은 adipogenic transcription factor의 발현을 조절하여 지방세포의 분화를 억제하였으며, 지방구와 triglyceride 생성을 감소시켜 최종적으로 비만억제 효과를 나타낸다는 것을 알 수 있었다. Sargassum horneri (Turner) C. Agardh is a marine brown algae widely distributed in the North Pacific Ocean. It is known for its anti-inflammatory and anti-atopic effects. In this study, we determined the effects of ethanol extract of Sargassum horneri (Turner) C. Agardh (EESH) on anti-obesity activities in 3T3-L1 preadipocytes. Our results indicated that treatment with EESH decreased the differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet content observed by oil red O staining. The concentrations of cellular triglycerides were also reduced in 3T3-L1 cells after treatment with EESH. Triglyceride content was inhibited by 13%, 16%, and 23% after treatment with 250, 500, and 1,000 μg/ml of EESH in 3T3-L1 cells, respectively. Western blotting analysis showed that EESH suppressed adipogenic transcription factor expression in a dose dependent manner. Specifically, it suppressed cytidine-cytidine-adinosine-adenosine-thymidine (CCAAT) /enhancer binding proteins (C/EBP)α, C/EBPβ and peroxisome proliferator-activated receptor (PPAR)γ. This indicated that EESH could control the expression of adipogenic transcription factors and inhibit the differentiation of adipocytes. Taken together, these findings demonstrated that EESH showed anti-obesity effects and could have potential uses in the field of nutraceuticals.

<P>Schisandrin A is a bioactive lignan occurring in the fruits of plants of the <I>Schisandra</I> genus that have traditionally been used in Korea for treating various inflammatory diseases. Although the anti-inflammatory and antioxidant effects of lignan analogues similar to schisandrin A have been reported, the underlying molecular mechanisms have remained elusive. In the present study, schisandrin A significantly suppressed the lipopolysaccharide (LPS)-induced production of the key pro-inflammatory mediators nitric oxide (NO) and prostaglandin E<SUB>2</SUB> by suppressing the expression of inducible NO synthase and cyclooxygenase-2 at the mRNA and protein levels in RAW 264.7 macrophages. Furthermore, schisandrin A was demonstrated to reduce the LPS-induced secretion of pro-inflammatory cytokines, including tumor necrosis factor-α and interleukin-1β; this was accompanied by a simultaneous decrease in the respective mRNA and protein levels in the macrophages. In addition, the LPS- induced translocation of nuclear factor-κB (NF-κB), as well as activation of mitogen-activated protein kinases (MAPKs) and phosphatidylinositol-3 kinase (PI3K)/Akt pathways were inhibited by schisandrin A. Furthermore, schisandrin A significantly diminished the LPS-stimulated accumulation of intracellular reactive oxygen species, and effectively enhanced the expression of NF erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1). These results suggested that schisandrin A has a protective effect against LPS-induced inflammatory and oxidative responses in RAW 264.7 cells by inhibiting the NF-κB, MAPK and PI3K/Akt pathways; these effects are mediated, at least in part, by the activation of the Nrf2/HO-1 pathway. Based on these results, it is concluded that schisandrin A may have therapeutic potential for treating inflammatory and oxidative disorders caused by over-activation of macrophages.</P>
